Trends in MEA-based Neuropharmacological Drug Screening (MEA 기반 신경제약 스크리닝 기술 개발 동향)

  • The announcement of the US Environmental Protection Agency that it will stop conducting or funding experimental studies on mammals by 2035 should prioritize ongoing efforts to develop and use alternative toxicity screening methods to animal testing. Toxicity screening is likely to be further developed considering the combination of human-induced pluripotent-stem-cell-derived organ-on-a-chip and multielectrode array (MEA) technologies. We briefly review the current status of MEA technology and MEA-based neuropharmacological drug screening using various cellular model systems. Highlighting the coronavirus disease pandemic, we shortly comment on the importance of early prediction of toxicity by applying artificial intelligence to the development of rapid screening methods.

  • Inhalation toxicity, mutagenicity, and immunotoxicity tests were performed using a smoke generation system to investigate the safety of Herbrette, a tobacco substitute made with the leaves of Perilla frutescens. ICR mice were exposed to nicotine-free Herbrette smoke with concentrations of 0 (control), 4.08 $\pm$ 1.32 mg/$m^3$ (low dose), 7.72 $\pm$ 2.14 mg/$m^3$ (medium dose) and 12.83 $\pm$ 1.69 mg/$m^3$ (high dose) total particulate matters (TPM) for 4 weeks. When compared to the control group, the body weights, organ weights in the exposed groups did not show any significant differences. However, certain change of several serum chemical data and biochemical parameters were observed, however, the changes were within normal physiological ranges. Moreover, no changes in organ weight, and no gross/microscopic changes were observed between the exposed and control groups. Salmonella typhimurium reverse mutation, in vivo chromosomal aberration and micronucleus assays revealed that Herbrette did not induce mutagenicity. Upon evaluation of peripheral cellular immunity of mice through in vitro lymphocyte proliferation assay, no significant difference was observed in mean stimulation index between the exposed and control groups. Taken together, our results strongly suggest that Herbrette may not cause toxicity on mice under current condition.

  • Under pathological stress stimuli, dynamics of a biological system can be changed by alteration of several components such as functional proteins, ultimately leading to disease state. These dynamics in disease state can be modeled using differential equations in which kinetic or system parameters can be obtained from experimental data. One of the most effective ways to restore a particular disease state of biology system (i.e., cell, organ and organism) into the normal state makes optimization of the altered components usually represented by system parameters in the differential equations. There has been no such approach as far as we know. Here we show this approach with a cardiac hypertrophy model in which we obtain the existence of the optimal parameters and construct an optimal system which can be used to find the optimal parameters.

  • Cyclosporin A (CyA) is widely used in the inhibition of graft rejection in organ transplantation. However, the bioavailability of CyA after oral administration is very low due to its poor solubility and dispersability hi water. To improve the solubility of CyA, microemulsion systems were developed and its physicochemical characteristics were evaluated by phase studies, solubility and dispersability tests. Phase studies on the systems composed of ethyl oleate (EO), PPG-20 methyl glucose ether (GP-20), poloxamer 123 (PL) and water U) were carried out to make stable w/o emulsions. Besides, based on CyA solubility test in various compositions of surfactant systems, a reasonable surfactant composition (GP-20/PL=4/1) was selected to enhance its solubility.

Nociplastic pain

  • Nociplastic pain refers to pain arising from altered nociception without evidence of tissue or somatosensory damage. It encompasses various clinical conditions with shared neurophysiological mechanisms involving different organ systems. Nociplastic pain can occur independently or alongside chronic pain conditions with a nociceptive or neuropathic origin. This review introduces the concept of nociplastic pain, its clinical manifestations and the underlying pathophysiology. Taking a biopsychosocial approach can lead to a better understanding of nociplastic pain and improved treatment outcomes for affected individuals.

  • The avian pineal as well as the retina has been known to contain several types of photoreceptors with different visual pigments such as rhodopsin, iodopsin and the pineal specific opsin, pinopsin. These organs are also known to have circadian clock to regulate melatonin production. Exposure of animals to light causes a decline of the melatonin level and the phase shifts of melatonin rhythms in the pineal and retina. Therefore, the circadian clock system of these organs seem to consist of three elements, i.e., light input, oscillator and melatonin output systems. In birds, it was suggested that rhodopsin might be involved in the entrainment of pineal melatonin rhythms from the action spectrum experiment for controlling NAT activity rhythms. However, there are much more pinopsin-immunoreactive (Pino-IR) cells than rhodopsin (Rho-IR) and iodopsin (Iodo-IR) cells in the avian pineal. We found that Pino-IR cells appeared earlier embryonic stages than Rho-IR and Iodo-IR cells. So, we tried to identify the visual pigments involved in the circadian melatonin rhythms in the pineal and retina. Organ cultured pineals were exposed to monochromatic light to find out which opsin participates in regulation of melatonin rhythms. The action spectra showed a peak at 475nm, suggesting that pinopsin is the major photopigment to regulate melatonin production in birds.

  • This study has the purpose to investigate other countries' arbitrator systems to compare with the current KCAB's system and to find their merits and demerits so that we can make up for the demerits of KCAB's arbitrator system and to make the most use of its merits. The most important factor of arbitral procedure is the arbitrator. If we compare clauses related with the arbitrator to KCAB's arbitration rules, expand the merits of it and apply the merits of other arbitrating organs, KCAB's arbitration can avoid criticism which it has got so far while it was handling the international cases. Also, we may need to grow up the role of the executive office in the range of respecting the self-government of the concerned party for the rapid proceeding of arbitral procedure. According to the foreign countries' international arbitration rules, they go with the process that they firstly give the concurrence period to the concerned parties, especially related with the arbitral procedure such as selection of arbitrator or filling the vacancy of the arbitrator, and as for the concerned party who doesn't fulfill within that period, the arbitrating organ or the other one corresponding to the pertinent arbitrating organ in case of the ad-hoc arbitration rules fulfill instead.

The Impact of Implementation of ISO9000: 2000 on Technology Improvement: A Case study

  • ISO9000 set of standards has been widely applied in Iranian Automotive manufacturing industry. Regarding the effects of TQM and ISO9000 set of standards various studies have been conducted and different quantitative conclusions have been made. Due to lack of quantitative information, in many of the cases, using qualitative information becomes the best option. The qualitative nature of data in such cases, requires qualitative analysis methods that might lead to some challenging computational issues. This paper examines the impact of ISO9000: 2000 certification and its perceived benefits for an automotive manufacturing company. Using an empirical approach, the paper seeks to ascertain if certification has indeed improved the performance of the company. Our null hypothesis rotates around the effect of ISO9000: 2000 on the 4W's of an enterprise. To carry out the research, first we developed a couple of questionnaires including all criteria of ISO9000 and 4W's. Second, the questionnaires were discussed with two researchers knowledgeable in the field, and then submitted to the quality practitioners and executives of Iran Khodro Enterprise-a leading company in Iranian automotive manufacturing industry. Finally the null hypothesis was tested and the technology improvement dimensions were ranked through nonparametric tests. The results illustrated a reasonable cause and effect relationship, suggesting that ISO9000: 2000 has positive effect on the 4W's of company result. In this work we investigate the effect as technology improvement viz., the improvement of techno-ware, human-ware, info-ware, and organ-ware.

Pressure Monitoring System in Gastro-Intestinal Tract

  • Diseases in the gastro-intestinal tract are on an increasing trend. In order to diagnose a patient, various signals of the digestive organ, such as temperature, pH, and pressure, can offer the helpful information. Among the above mentioned signals, we choose the pressure variation as a monitoring signal. The variation of a pressure signal of the gastro-intestinal tract can offer the information of a digestive trouble or some clues of the diseases. In this paper, a pressure monitoring system for the digestive organs of a living pig is presented. This is why a pig's gastro-intestinal tract is very similar as human's. This system concept is to transmit the measured biomedical signals from a transmitter in a living pig to a wireless receiver that is positioned out of body. The integrated solution includes the swallow type pressure capsule and the receiving set consisting of a receiver, decoder circuit. The merit of the proposed system is that the monitoring system can supply the precise and a durable characteristic to measure and to transmit a signal in the gastro-intestinal tract. We achieved the pressure tracings in digestive organs and verified the validity of system after several in-vivo tests using the pressure monitoring system. Through various experiments, we found each organ has its own characterized pressure fluctuation.

Chemopreventive Effects of Ginseng on Rat Carcinogenesis

  • The chemopreventive effects of ginseng on rat carcinogenesis models were investigated, In the present study, the inhibitory effects of white and red ginseng on tumor development were examined using medium-term liver, initiation and medium-term multi-organ carcinogenicity bioassay systems. No modifying potential of the ginsengs was evident in terms of the numbers or areas of glutathione S-transferase placental form (GST -P)-positive foci, which is a marker of preneoplastic lesion in rat livers. However, white ginseng, but not red ginseng was found to decrease the incidences of adenocarcinoma of the small intestine and colon in the medium-term multi-organ carcinogenesis model. These results indicate that white ginseng may have inhibitory effects on progression stage of rat intestinal carcinogenesis, but the influence is not strong. Ginseng is unlikely to have promoting or inhibitory effects in other organs under the present type of experimental conditions. Possible application on ginseng for chemoprevention of colon cancer in humans, can be concluded given the lack of obvious adverse effects.
