Isolation and Diversity of Yeasts from Fruits and Flowers of Orchard in Sinam-myeon of Yesan-gun, Chungcheongnam-do, Korea (충남 예산군 신암면 일대 과수원의 과일과 꽃들로부터 효모의 분리 및 분포 특성)

  • Several kinds of yeasts from fruits and flowers of orchard in Yesan-gun of Chungcheongnam-do, Korea were isolated and identified by comparison of nucleotide sequences for PCR-amplified D1/D2 region of 26S rDNA using BLAST. Fourty eight yeast strains of twenty five species and one hundred eight yeast strains of fourty eight species were isolated from fruits and flowers of orchard in Sinam-myeon of Yesan-gun, Chungcheongnam-do, respectively. Among total one hundred fifty-six yeast strains, only sixteen species were overlapped from fruits and flowers.

The Impact of Air Temperature During the Growing Season on NEE of the Apple Orchard (사과 생육기의 기온이 사과원의 NEE에 미치는 영향)

  • Terrestrial ecosystem are a strong sink of carbon. Forest ecosystem, one of them, has been expected to play an important role in climate changing process by absorbing atmospheric carbon dioxide. On the other hand, agricultural ecosystem that consists mainly of annual crops is regarded as poor contributor to carbon accumulation, because its production (carbon hydrate) is decomposed into carbon at a short period, which is emitted to the atmosphere. However, it is thought that fruit tree plays a great role in decreasing atmospheric carbon dioxide concentration, same as forest. Net ecosystem exchange of $CO_2$ (NEE) was measured to estimate carbon fixation capacity using an eddy covariance (EC) system method in 2 years from 2005 to 2006 at an apple orchard in Uiseong, Gyeongbuk. Average air temperature values were higher in 2006 than in 2005 during the dormant season, and lower by about $5^{\circ}C$ over the growing season causing visible cold injuries. Accordingly, we investigated long-term exchange of carbon to determine how much difference of carbon fixation capacity was shown between 2006 and 2005 in terms of environmental and plant variables such as NEE, leaf area index (LAI), and Albedo. NEE was $4.8Mg\;C\;ha^{-1}yr^{-1}$ in 2005 and $4.7Mg\;C\;ha^{-1}yr^{-1}$ in 2006, respectively. Low temperature after July in 2006 decreased LAI values faster than those in 2005. Meanwhile, Albedo values were higher after July in 2006 than in 2005. These results show that the low temperature after July in 2006 apparently affected apple growth.

Correlation among Conformation Parameters in 'Fuji' Apple Fruit ('후지' 사과 과실의 형태 결정 요인 간 상관)

  • Factors affecting fruit conformation of 'Fuji' apples (Malus domestica Borkh.) were investigated through correlation study of fruit weight, seed numbers, and seed weight in relation to fruit conformation indices. Apples were harvested from two orchards in 'Andong' and 'Youngyang' areas at commercial maturity and grouped by fruit weight. Average number of seeds and seed weight were 8.9 and 0.64 g in 'Andong' orchard, while 6.3 and 0.47 g in 'Youngyang' orchard, respectively. When pooled data from the both orchards were analyzed, correlation of fruit weight with seed weight was significantly positive, whereas that with seed number was not significant. Fruit shape index determined by length/diameter ratio was positively correlated with fruit weight, while relationship of the shape index with seed number or with seed weight was not significant. Asymmetry index tended to be low in apples from the 'Andong' orchard in which higher seed numbers and seed weight were observed. The asymmetry index showed highly negative correlations with seed numbers and with seed weight.

UTILIZATION OF RICE STRAW BY RUMINANTS AS INFLUENCED BY GRASS HAY SUPPLEMENTATION

  • Twenty Korean native bulls averaging 181 kg body weight were fed a fixed amount (1.5% of live weight) of concentrate and free choice roughage cubes which had four ratios of rice straw and orchard grass hay; 100:0, 85:15, 70:30 and 55:45 on a weight basis. Five bulls were assigned to each treatment and fed for 98 days in a confinement house. Nutrient digestibility and available energy content of mixed rations (40% concentrate and 60% roughage cubes) was determined in a digestion trial with twelve wethers. In vitro dry matter digestibility (IVDMD) and in situ dry matter disappearance of the roughage cubes were also determined. The ratio of grass hay to rice straw did not influence dry matter intake. Significant improvements in body weight gains and feed/gain ratios were obtained as grass hay levels increased. Average daily gain and feed/gain for each treatment was 0.83, 0.88, 0.98 and 0.99 kg; 7.63m 7.59, 6.83 and 6.41, respectively. Digestibility of the nutrients was improved with increasing levels of grass hay in the cubes. The IVDMD of roughage samples having a ritio of 100:0, 85:15, 70:30 and 55:45 between rice straw and orchard grass hay were 31.0, 37.1, 41.8 and 43.4%, respectively. Grass hay improved the IVDMD of rice straw diets in a linear manner up to 30%. In situ dry matter disappearance rate was also increased as the level of orchard grass hay increased.

Characteristics of Schizandra chinensis Baillon Orchard Soils Located in Jangsu-gun, Jeollabuk-do (전라북도 장수군 오미자 재배과원 토양 특성)

  • This study was conducted to investigate the physico-chemical properties of Schizandra chinensis Baillon orchard soils located in Jangsu-gun, Jeollabuk-do. Surface (0 to 10 cm) soils were collected from 200 experimental sites located at Jangsu-eup (53 site), Gyenam-myeon (31), Chunchun-myeon (73), Janggye-myeon (12), Bunyam-myeon (31). The soil texture was mostly loamy sand, and the mean values of degree of soil aggregate and soil porosity were 33.1 and 59.9%, respectively. The pH, EC, total-N, available-P, soil organic matter, and cation exchange capacity of the soils were $5.51{\pm}0.54$, $290{\pm}139{\mu}S\;cm^{-1}$, $946.3{\pm}65.5mg\;kg^{-1}$, $319.6{\pm}29.2mg\;kg^{-1}$, $29.0{\pm}13.9g\;kg^{-1}$ and $4.11{\pm}0.34cmol_c\;kg^{-1}$, respectively. The concentrations of Pb, Cd, Cu and Zn were $3.48{\pm}0.55$, $0.09{\pm}0.04$, $6.90{\pm}0.91$ and $97.7{\pm}42.2mg\;kg^{-1}$, respectively. The presented data can be utilized in better managing Schizandra chinensis Baillon orchard soils in the studied areas.

Stink Bugs (Hemiptera) and Their Size, Collected near Jinju City, Gyeongnam Province (경남 진주 지역에서 채집된 노린재의 종류와 크기)

  • True bugs were surveyed at the forest, bank of rice fields, and sweet persimmon orchard near Jinju city, Korea in 2006 and 2007. Number of species (individuals) collected at the forest, bank of rice fields and persimmon orchard are 24 (817), 21 (425) and 15 (193) species, respectively. A dominant species was Nysius plebeius at the forest, and Cletus punctiger at the bank of rice fields and persimmon orchard. In all the three sites N. plebeius, C. punctiger, and Pachygrontha antennata were more abundant than other species, consisting 77.8% of the total collections. N. plebeius was mainly collected on July at the bank of paddy field, and on August and after October at the forest. Most number of C. punctiger was collected from early July to early September at three sites. P. antennata was mainly collected after October at the forest. Body sizes of the stink bugs were measured.

Exposure and Risk Assessment of Operators to Insecticide Acetamiprid during Treatment on Apple Orchard

  • Occupational exposure and risk assessment were conducted to evaluate the safety of operators when insecticide acetamiprid was applied to apple orchard using a speed sprayer. Dermal patches, cotton gloves, socks, and masks were used to monitor the dermal exposure, and personal air pump with solid sorbent was used to measure the potential inhalation exposure. In validation to analytical methods, the limit of detection and limit of quantitation were 0.25 ng and 1 ng, respectively. Good reproducibility (coefficient variation < 4%), linearity (coefficient determination > 0.999), and recovery (85.3-118.2%) were obtained. Trapping efficiency of solid sorbent was 96.4% while breakthrough did not occur. Only hand exposure was measured on the gloves during mixing/loading to give $33-1,132{\mu}g$. Exposure amount of operator 3 among 4 workers was noticeably high. The total volumes of spray liquid for operators were $535-1,235mL{\cdot}h^{-1}$, corresponding to 0.03-0.08% of the applied spray solution. Highest contaminated parts of body were thighs, chest, and lower legs. The inhalation exposure ratio to the total application amount was significantly low. However, wind seemed to affect the inhalation exposure of operator. For risk assessment, margin of safety was calculated by the application of cloth and dermal penetration rate to obtain values of much larger than 1 in all cases. Therefore, health risk of operators during treatment of acetamiprid in apple orchard could be of least possibility.

Effect of Agro-chemical Alternatives on the Yield and Fruit Quality of Apple (대체농업자재가 사과의 수량 및 품질에 미치는 영향)

  • This study was tried to examine the efficacy of apples that had grown with agro-chemical alternatives in an apple orchard. The chlorophyll content of apple leaves was a little higher in the plot of Charcoal powder, Chitosan, and Peat moss treatment among the other agro-chemical alternatives. On the contrary, the sugar content of apple fruits was higher in the plot of Amino acids, and Green ion calcium treatment, but there was quite a difference among them. Vitamin C content of apple fruits was high at the cell division period. It became lower at the hypertrophic period and then got higher again at the harvest. Vitamin C content was the highest in the chemical fertilizer plot as well. In an apple orchard where agro-chemical alternatives were used only, the commercial grade of ripened apples had declined by 25%, compared to those in customary cultivation. The commercial grade of ripened apples with agricultural chemicals using fertilizers and pesticides was over 90% with over 250g of its weight in the plot of Charcoal powder, Amino acids, and Vitamin C treatment. Therefore. we guessed the environment-friendly method of cultivation in an apple orchard shall be established by reducing the usage of agricultural chemicals gradually and increasing the agro-chemical alternatives at the same time.

Application of Sprinkler System for Control of Cone Insects on Korean Pine, Pinus koraiensis Seed Orchard (스프링클러시스템을 이용한 잣나무 채종원의 구과해충 방제에 관한 연구)

  • In order to prevent cones from being damaged by cone insects, Gravitarmata margarotana and Dioryctria abietella, sprinkler system was installed on the Pinus koraiensis Seed Orchard. Such a test was conducted at Chung-ju, Chungcheong buk-do, Korea in 2002. Insectides of Diflubenzuron 25% WP ${\times}2,500$ and Cyfluthrin 2%EC ${\times}\;1,000$ were sprayed at fifteen-day and twenty-day intervals during the period of insects adult occurrence by sprinkler system. The Gravitarmata margarotana generally emerged for about 53 days from middle April to early June, and peak emergence was 10 days from April 20 to April 30. Dioryctria abietella generally emerged for about 108 days from late April to early August, and peak emergence was 10 days from May 20 to May 30. Damage rate of cones were $8.7{\sim}9.7%$ and $10.9{\sim}12.7%$ on average with crown insecticide spraying with Cyfluthrin and Diflubenzuron, while 67.5% when not treated. Control effectiveness of this insecticide spraying was $85.6{\sim}87.1%$ and 83.9%와 81.2%, respectively. When it considers the forest environment, it is recommended diflubenzuron which is low toxic pesticide. In conclusion, effectual times and number of diflubenzuron 25% WP application with sprinkler system against cone insects were from early May through late August and six times at twenty-day interval.

Assessment of Pathogenic Variation against Pitch Canker Pathogen, Fusarium circinatum in Pinus thunbergii and Responses of Natural Selection Pinus × rigitaeda to Branch Inoculation in a Seed Orchard

  • A half-sib family of two 4-year-old seedlings of Pinus $\times$ rigitaeda was inoculated with each of 20 Pinus thunbergii isolates of Fusarium circinatum (syn. Fusarium subglutinans f. sp. pini) from two pitch canker damaged sites in Jeju Island, South Korea. Initial symptoms of needle damages were visible on most of the seedlings at 18 days after inoculation. The 20 tested isolates were not significantly different in virulence, based on lesion lengths at the site of inoculation (P = 0.217). The most virulent isolate FT-7 showed the longest lesion length. Some seedlings began to die 46 days after inoculation. All seedlings were dead by 68 days after inoculation except two seedlings inoculated with each of isolates FS-2 and FS-13, respectively. Using the FT-7, 38-year-old 11 P. $\times$ rigitaeda trees, which were survived from a seed orchard severely damaged by pitch canker, were inoculated on branches in the seed orchard in Jeju Island to assess differences in susceptibility to pitch canker. The 11 trees differed significantly (P < 0.001) in susceptibility to F. circinatum based on average lesion lengths measured 56 days after inoculation. It is possible that induced resistance contributed to their capacity to limit lesion development. The susceptibility of natural selection P. $\times$ rigitaeda trees are more likely affected by interaction with F. circinatum rather than environmental conditions.
