  • A novel ceramic synthesis technique, combustion synthesis, is explored to produce a orange-emitting $SnO_{2}:Eu^{3+}$ phosphors. This technique involves a reaction of metal salts with a citric acid as an organic fuel. The variation of the ratio of citric acid to metal introduces change in reaction temperature and atmosphere of the combustion reaction, which in turn influences crystallinity and luminescence properties of the final products seriously. And, the heat treatment of the as-synthesized product increases its luminescence intensity. Especially, the samples treated at above $1100^{\circ}C$ show a orange-emission peak, which is attributed to the formation of single phase and well-crystallized $SnO_{2}:Eu$ oxides in the rutile structure

450-500nm의 최대흡수를 가지는 순수 폴리올레핀 소재용 초소수성 오렌지 염료의 합성 (Synthesis of Super Hydrophobic Orange Dyes Having Maximum Absorption at 450-500nm for Pure Polyolefin Fibers)

  • Novel super hydrophobic orange dyes having maximum absorption band at 450-500nm were synthesized to dye polyolefin fibers such as polypropylene and ultra high molecular weight polyethylene fibers, using 4-alkylanilines and ${\beta}$-naphthol. Their absorption spectra at visible range showed almost the same, which meant that the alkyl substituents introduced to chromophore did not affect on color appearance of the dyes. Considering both color strength and wash fastness, the decyl-substituted dye was determined as the optimum one practically. From the dyeing results at various conditions, the optimum dyeing was $130^{\circ}C$ for 1 hour with 5% owf of dyes. The good fastness ratings to washing, rubbing were obtained showing 4-5 for both fibers. Light fastness was also acceptable giving rating 3-4 for polypropylene fibers and rating 3 for ultra high molecular weight polyethylene fibers.

  • This work wes performed to investigate the photocatalytic decolorization of waste water from textile industries. Methyl orange was used as a target dye with suspended Hombikat TiO$_2$ photocatalyst with a recirculating annular photoreactor. 1 wt % Pt-doped Hombikat thin film tubular reactor with parabolic reflector also wes usedin this experiment. The pH effect and flow rate effect on photobleaching of 0.012 g/l methyl orange solution, AtpH=3 Colour of methyl orange was completely bleached in 30 min with a 20 W UV lamp.

Selective production of red azaphilone pigments in a Monascus purpureus mppDEG deletion mutant

  • The Monascus azaphilone (MAz) pigment is a well-known food colorant that has yellow, orange and red components. The structures of the yellow and orange MAz differ by two hydride reductions, with yellow MAz being the reduced form. Orange MAz can be non-enzymatically converted to red MAz in the presence of amine derivatives. It was previously demonstrated that mppE and mppG are involved in the biosynthesis of yellow and orange MAz, respectively. However, ${\Delta}mppE$ and ${\Delta}mppG$ knockout mutants maintained residual production of yellow and orange MAz, respectively. In this study, we deleted the region encompassing mppD, mppE and mppG in M. purpureus and compared the phenotype of the resulting mutant (${\Delta}mppDEG$) with that of an mppD knockout mutant (${\Delta}mppD$). It was previously reported that the ${\Delta}mppD$ strain retained the ability to produce MAz but at approximately 10% of the level observed in the wildtype strain. A chemical analysis demonstrated that the ${\Delta}mppDEG$ strain was still capable of producing both yellow and orange MAz, suggesting the presence of minor MAz route(s) not involving mppE or mppG. Unexpectedly, the ${\Delta}mppDEG$ strain was observed to accumulate fast-eluting pigments in a reverse phase high-performance liquid chromatography analysis. A LC-MS analysis identified these pigments as ethanolamine derivatives of red MAz, which had been previously identified in an mppE knockout mutant that produces high amounts of orange MAz. Although the underlying mechanism is largely unknown, this study has yielded an M. purpureus strain that selectively accumulates red MAz.

항균소재 함유 박스로 포장한 방울토마토와 밀감의 저장중 품질 특성 (Quality Characteristics of Cherry Tomato and Unshiu Orange Packaged with Box Incorporated with Antimicrobial Agents)

  • 항균소재를 함유한 포장지로 포장하여 실험실 조건하에서 저장하였을 때 방울토마토와 밀감의 저장기간 연장효과가 있는 것으로 나타난 항균포장지가 대규모 저장 및 유통시에도 적용이 가능한지를 알아보기 위하여 항균포장지를 함유한 박스에 방울토마토와 밀감을 담아서 냉장 창고에서 저장하면서 품질 변화를 고찰하였다. 즉 분말 형태의 식물성 천연항균제인 자몽추출물제재와 제올라이트를 각각 3% 및 6%를 넣어서 만든 항균포장지로 방울토마토와 밀감을 포장하여 $5^{\circ}C$에서 저장하면서 품질특성의 변화를 측정하였다. 저장 기간중 품질 변화의 지표로 중량감소율, pH, 총산, 가용성고형물, 총균수, 효모 및 곰팡이수 등을 측정하였다. 방울토마토의 pH는 저장 20일까지는 다소 증가하였으나 저장 40일에는 급격하게 감소하였다. 가용성 고형물은 포장 직후 5.5 $^{\circ}Brix$였으며, 저장 10일에 5.8 $^{\circ}Brix$까지 증가하였다. 포장 직후 방울토마토의 총균수는 $10^{2.87}CFU/g$, 효모 및 곰팡이는 $10^{2.76}CFU/g$ 이었으며, 저장 기간이 경과함에 따라 지속적으로 증가하였다. 저장 30일에 대조구는 11.5%, 항균포장지를 함유한 박스로 포장한 시험구는 1.5%의 부패가 발생하여 항균포장지를 함유한 박스포장이 방울토마토의 부패를 억제하는 효과가 있는 것으로 확인하였다. 밀감은 저장 직후 pH 3.78, 총산 0.68%였으며 저장 30일까지는 대체적으로 감소하였으나 가용성 고형물은 저장 직후 10.7 $^{\circ}Brix$ 였으며, 저장 30일까지는 다소 증가하였다. 포장 직후 밀감의 총균수는 $10^{4.01}CFU/g$이었으며, 효모 및 곰팡이는 $10^{3.99}CFU/g$이었고,저장기간이 경과함에 따라 지속적으로 증가하였다. 저장 20일에 대조구는 1.3%의 부패율을 나타내었으나 항균포장지를 함유한 박스에 포장한 밀감은 저장 30일에 부패가 발생하여, 대조구에 비하여 10일 정도 저장기간의 연장 효과가 있었다.

잿빛곰팡이병 추출물을 이용한 순무배양세포의 Indole-3-ylmethyl glucosinolate의 생합성유도와 병원성연구 (Elicitation of Indole-3-ylmethyl Glucosinolate Biosynthesis in Turnip Culture Cells and Their Relationship with Plant Resistance to Botrytis cinerea)

  • 8종의 잿빛곰팡이병 균주를 순무잎에 접종하여 병반의 크기를 확인한 결과 가장 강한 감염력을 보인 '포도-01' 균주와 병반의 확산이 가장 적은 '오랜지'를 선발하였다. 순무잎이 저항성을 보인 '오랜지'균주를 처리한 잎이 감수성을 보인 '포도-01'균주를 처리한 잎보다 indole-3-ylmethyl glucosinolate (I3M-GLS) 함량이 무처리 보다 2.5배 이상 높았으나 '포도-01' 균주를 처리한 잎에서는 무처리 보다 낮은 함량을 보였다. 균주의 메탄올 추출액과 물추출물을 식물배양세포에 처리한 결과 '오랜지'균주의 추출물이 '포도-01' 균주의 추출물보다 배양세포의 생장을 더 강하게 억제 한 것으로 나타났는데 '오랜지' 균주의 메타놀 및 물 추출물 처리에서 배양세포의 활력은 각각 22.7% 및 16.5% 감소시키는 것으로 나타났다. 한편 '오랜지'균주 추출물을 처리한 배양세포에서 I3M-GLS의 생합성이 '포도-01' 균주 추출물보다 현저히 높은 것으로 나타났다. 본 결과로 보아 식물체내에 생합성되는 I3M-GLS 함량은 잿빛곰팡이균에 대한 식물세포의 저항성과 밀접한 관계가 있는 것으로 판단된다.

  • Aspergillus saitoi가 생산(生産)하는 섬유소분해효소(纖維素分解酵素)의 생산조건(生産條件)과 밀감내과피(內果皮)의 탈피효과(脫皮效果)를 검토한 결과(結果)는 다음과 같다. 1. Asp. saitoi의 배양최적(培養最適) 온도(溫度)는 $30^{\circ}C$, 시간(時間)은 72시간(時間)에서 섬유소(纖維素) 생성능(生成能)이 가장 좋았다. 2. 본효소(本酵素)를 10배(倍) 농도로 희석하여 (희석효소액 : 밀감 2 : 1) $40^{\circ}C$의 항온조(恒溫槽)에서 $3.5{\sim}4$시간(時間) 작용(作用)시킨 것이 탈피정도(脫皮程度), 풍미(風味), 광택(光澤)이 우수하여 밀감 syrup 통조림 제조조건으로 최적(最適)이었다.

베트남 참전 제대 군인의 범주화된 에이전트 오렌지 개인 폭로량과 혈청 다이옥신 측정치와의 상관성에 관한 연구 (A Study on the Correlation between Categorization of the Individual Exposure Levels to Agent Orange and Serum Dioxin Levels Among the Korean Vietnam Veterans)

  • Objectives : In an epidemiologic study on the health impact of Agent Orange exposure, the valid estimation of exposure level is the most important step. Based on recent studies, we examined the correlation between exposure levels categorized by personal exposure estimates and serum 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7,8-TCDD, Dioxin), exploring the possibility of utilizing the exposure level as a surrogate for the estimate of exposure to agent orange. Methods : During the study period (Jan 1996-Feb 1996), blood specimens of 745 subjects taken randomly among 1,329 persons and kept frozen, were analyzed for 2,3,7,8-TCDO and six other dioxin congeners. The serum dioxin and congeners were measured in 1998 by CDC, adjusted for serum lipids. We categorized the total exposure scores into five groups based on Agent Orange exposure data collected by interview and military records. Pearson and Spearman's correlation coefficients & multiple regression analysis were used to identify the relationship of the exposure level categorized with serum concentration of 2,3,7,8-TCDD, and six other dioxin congeners. Results : Dioxin and the other congeners, except 1,2,4,6,7,8-HpCDD, showed significant correlations to exposure categories (p<0.005): 2,3,7,8-TCDD and OCDD showed positive correlations, whereas the other congeners did negative. The values of 2,3,7,8-TCDD differed according to exposure category and proportionally increased from the low exposure group to the high, a dose-response relationship, even after other possible confounding variables were adjusted for. In multiple regression analysis, age$(\beta=0.033)$, dioxin$(\beta=0.433)$, 1,2,3,7,8-PeCDD$(\beta=-0.998)$, 1,2,3,4,7,8-HxCDD$(\beta=-0.773)$, 1,2,3,6,7,8-HxCDD$(\beta=0.255)$, 1,2,3,7,8,9-HxCDD$(\beta=-3.468)$, 1,2,3,4,6,7,8-HpCDD$(\beta=0.109)$ we re found to be significantly related to the total exposure score(p<0.005). Conclusion : This study demonstrated that the use of such categorizations as a surrogate measure of agent orange exposure in identifying exposure degrees in a health impact study is valid.

Synthesis of Japanese Encephalitis Virus in Porcine Kidney Stable Cells Observed by Fluorescent Antibody Technique and Autoradiography

  • The site of the synthesis of Japanese encephalitis virus(JEV) in the actinomycin-treated and infecter PS Y15 cells(a porcine kidney stable cell line) was observed by the immunofluorescent antibody technique, acridine orange staining, and the autoradiographic analysis. In the parallel studies by immunofluorescent technique and acridine orange staining it the infected cells, Viral protein(as an antigen) and viral RNA were detected at the same site of cytoplasm. In the autoradiographic analysis, the cytoplasmic labeling of $^3H$-uridine was due to the synthesis of JEV-RNA, while the nucleolus and nucleus were not involved. In the autoradiographic studies on the secton of infected cells, the $^3H$-uridine was frequently incorporated around the cytoplasmic vacuoles. This localization of labeling agreed with the site of acridine orange positive granules. The results suggest that the syntheses of the viral RNA and viral protein occurred in the similar site of cytoplasm of the infected cells, and also the virus particles seem to be assembled in the sites of the viral RNA and protein syntheses.

반응표면 분석법을 이용한 스피루리나 첨가 마들렌 제조의 최적화 (Optimization of Spirulina Madeleine Using Response Surface Methodology)

  • The purpose of this study was to determine the optimum amount of four ingredients (spirulina, sugar, lemon and orange) for the preparation of spirulina madeleine using response surface methodology. Spirulina was added at a level of 3.5-6.0 g, sugar powder, at 40-60 g, while lemon and orange peel were included at a level of 0-8 g. The optimum mixing rates of spirulina powder, sugar powder, lemon and orange were 3.5, 40, 0, and 8 g for overall quality and 3.52, 54.42, 0, and 8 g for maximum score of overall organoleptic quality, respectively.
