Antimicrobial Activity of Oleanolic Acid, Ursolic Acid, and Sophoraflavanone G against Periodontopathogens

  • In general, oleanolic acid (OA) and ursolic acid (UA) have antimicrobial effect against Gram-positive bacteria but not Gram-negative bacteria whereas sophoraflavanone G has antimicrobial activity against both bacterial types. However, the antimicrobial effects of OA, UA, and sophoraflavanone G against periodontopathogens have not been studied to any great extent. The aim of this study was to investigate antimicrobial effect of OA, UA, and sophoraflavanone G against 15 strains (5 species) of oral Gram-negative bacteria, which are the major causative bacteria of periodontal disease. The antimicrobial activity was evaluated by minimal inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) determinations. OA and UA showed antimicrobial effects against all of the Porphyromonas gingivalis strains tested and also Prevotella intermedia ATCC $25611^T$. Interestingly, P. intermedia ATCC 49046 showed greater resistance to OA and UA than P. intermedia ATCC $25611^T$. In contrast, sophoraflavanone G had antimicrobial activity against all strains, with MIC and MBC values below $32{\mu}g/ml$, except Aggregatibacter actinomycetemcomitans. These results indicate that sophoraflavanone G may have potential for use in future oral hygiene products such as dentifrices and gargling solution to prevent periodontitis.

개구제한시 하악운동연습의 효과에 관한 근전도학적 연구 (An Electromyographic Study of the Efficacy of Mandibular Movement Esercise on Opening Limitation)

  • 52 Dental students without masticatory problems were selected for this study. They were trained on several mandibular position and mandibular movement exercise, that is, rest position, light bite, tapping, hinge opening, habitual opening, opening limitation, stretch exercise, resistance exercise and clenching. The objectives of this study was to investigate the effects of mandibular movement exercise, especially stretch and resistance, on the experimentally guided limited mouth opening. Muscle activity of the anterior temporalis and the masseter on above mentioned position or exercise were recorded with bioelectric processor EM2(Myotronics, U.S.A.) and the data were processed with SPSS. The obtained results were as follows : 1. Activity of the muscles at rest position were decreased with mandibular movement exercise. 2. Forceful mouth opening on opening limitation increased muscle activity greatly, especially of the masseter. 3. On opening limitation, stretch or resistance exercise was very efficient for decrease of muscle activities. 4. There were no difference of muscle activity between on hinge opening and on habitual opening. Therefore, for muscle relaxation, the two movement exercise can be used interchangeably.

Peroxidase가 Lysozyme 활성에 미치는 영향 (Influences of Peroxidase on Lysozyme Activity)

  • 타액에 존재하는 여러 항균물질은 상호작용을 통하여 부가적이거나 상승작용을 나타내고 때로는 저해작용을 나타내는 것으로 알려져 있다. 본 연구는 in vitro 상에서 lysozyme과 peroxidase 사이의 상호작용을 효소활성 측면에서 조사하고자 시행되었다. Lysozyme과 peroxidase의 상호작용은 hen egg-white lysozyme(HEWL)과 bovine lactoperoxidase(bLP)를 혼합하는 방법으로 조사되었고, peroxidase system이 lysozyme에 미치는 영향은 potassium thiocyanate와 hydrogen peroxide를 추가적으로 첨가하는 방법으로 조사되었다. Lysozyme 활성은 Micrococcus lysodeikticus 기질용액의 혼탁도 변화를 측정하는 방법으로 측정되었고, peroxidase 활성은 NbsSCN 법으로 측정되었다. Wilcoxon signed rank 법을 이용하여 lysozyme과 peroxidase 효소활성 변화를 대조군과 비교하였다. 생리적 농도범위에서 bLP는 HEWL의 효소활성을 증가시켰으며(P < 0.05), 그 효과는 bLP의 농도증가에 따라 영향을 받았다. 하지만 HEWL는 bLP의 효소활성에 영향을 주지 못하였다. Thiocyanate는 HEWL의 효소활성에 영향을 주지 못하였고, potassium thiocyanate와 hydrogen peroxide를 추가한 peroxidase system도 HEWL 효소활성의 추가적인 상승을 유도하지는 못하였으며, hydrogen peroxide의 농도변화도 영향을 미치지 못하였다. 본 연구의 결과는 타액 항균 물질을 포함하고 있는 구강건강용품이나 구강에서 lysozyme과 peroxidase의 상호작용을 이해하는데 필요한 중요한 정보를 제공해준다.

Antimicrobial Activity of Mulberry Leaf against Mutans Streptococci and Periodontopathogens

  • This study investigated the antimicrobial activity of methanol extract of mulberry leaf against 16 strains of mutans streptococci and four species of periodontopathogens: Porphyromonas gingivalis, Prevotella intermedia, Fusobacterium nucleatum, and Aggregatibacter actinomycetemcomitans. The antimicrobial activities of the crude extracts or silica gel chromatography fractions of methanol-extracted mulberry leaf were evaluated by determining minimal inhibitory concentrations using an established microdilution method. The cytotoxicity of the extracts of mulberry leaf on KB cells was tested by the methyl thiazolyl tetrazolium assay. Chromatography fraction 12 displayed the most potent antimicrobial activity against all 16 strains of mutans streptococci, P. gingivalis, and P. intermedia. No KB cell cytotoxicity was evident up to $128{\mu}g/ml$ of fraction 12. The methanol extract had no antimicrobial activity against F. nucleatum and A. actinomycetemcomitans. These results suggest chromatography fraction 12 methanol extract of mulberry leaf could be useful in the development of oral hygiene products, such as dentifrice and oral hygiene solution, for the prevention of dental caries.

죽력(竹瀝)(죽초액(竹酢液)) 경구투여(經口投與)와 간유(肝兪) 담유(膽兪) 약침(藥鍼)이 Alcohol 대사(代謝) 및 간기능(肝機能)에 미치는 영향(影響) (Effects of BCL oral administation and herbal acupuncture at BL18, BL19 on Liver function changes induced by Alcohol in the mice)

  • This dissertation was designed to evaluate the effect of BCL(refinded Bambusae Caulis in Liqua-men) oral administration and herbal acupuncture on alcohol metabolism and liver function. For this study. mice were damaged by a large quantity of alcohol and received treatment of either BCL 1 mg/kg in oral or BCL 250㎍/kg in herbal acupuncture-BL18 . BL19 bilateral. and then such parameters as GOT. GPT. catalase and superoxide dismustase(CuZn-SOD, Mn-SOD) were measured. The results of the experiments were summarized as follows. 1. Compared with control group, the proper degree of alcohol in serum was not significantly differ from oral administration group and herbal acupuncture group. 2. Compared with control group. the activity of GOT in serum was significantly reduced both oral administration and herbal acupuncture group. 3. Compared with control group. the activity of GPT in serum was significantly reduced both oral administration and herbal acupuncture group. 4. The activity of catalase in liver cell tissue, compared with control group. was not sigificantly affected either by oral administration and herbal acupuncture group. 5. The activity of CuZn-SOD in liver cell tissue was not significantly change in herbal acupuncture and oral administration group. The activity of Mn-SOD was significantly increased in oral administration group. while it was not the case in acupuncture group. In conclusion. we consider that BCL oral administration and herbal acupuncture is highly effetive in recovering alcohol metabolism and liver disfunction induced by alcohol.

구강활동을 병행한 감각통합치료가 발달장애 아동의 호흡 및 구강협응능력에 미치는 효과 (The Effects of Oral Activity With Sensory Integration Intervention on Breathing and Oral Diadochokinetic Movement of a Child With Developmental Disability)

  • 목적 : 본 연구는 구강활동을 병행한 감각통합치료가 발달장애 아동의 호흡과 구강협응능력에 미치는 효과를 알아보고자 하였다. 연구 방법 : 연구대상은 G시에 거주하는 만 6세 7개월의 남아로, 발달장애 진단을 받았다. 연구는 2012년 11월부터 2013년 2월까지 실시하였다. 실험은 개별실험연구 중 AB디자인을 사용하였다. 실험과정은 기초선(A) 3회기, 중재(B) 16회기이었다. 기초선 기간 동안에는 감각통합치료를 실시하였고, 중재기간동안에는 감각통합치료에 구강활동을 병행하여 실시하였다. 호흡기능은 폐활량 측정계로 측정하였으며, 구강협응능력은 "파타카(pataka)"를 10회 반복할 때 소요된 시간을 초단위로 측정하였다. 결과 : 아동의 호흡과 구강협응능력은 기초선과 비교하여 중재기간 동안 유의하게 증가하였다. 결론 : 구강활동을 병행한 감각통합치료가 아동의 호흡과 구강협응능력 향상에 긍정적이었다.

영남지역 일부 주민들의 일반적인 특성 및 건강생활에 따른 주관적인 구강보건과 삶의 질 평가 (General aspect of residents in Youngnam area and subjective evaluation about oral health and quality of life along with health activity)

  • This survey is to increase the quality of life by improved oral health. Total 572 Youngnam area residence joined the survey and answered for the subjects of general aspect & health activity, self-determined oral health status, oral health knowledge, oral health activity, food behavior, OHIP-14,THI, QOL. Survey was analyzed by SPSS (Ver. 12.0). Result of the study is summarized as follows; 1. Among total 572 residents, women were 58% and 20~29 aged were 35.5%, highest. In academic background, High school graduated were 59.6%. In average monthly income view, Lower than 1.5 million KRW was 43.5%, 49.3% replied drinks 1~3 times a month. No smoking group was 73.1%. No exercise group was 35.5% and every day exercise group was 11.4%. In self evaluating about oral health status question 40.4%(231) replied as bad, which was highest. 2. Women have higher score about oral health knowledge. Woman and Every day exercise group have better score about oral health practice. Man, 40~49 aged, more than 2.51 million KRW avg' income, every day exercise groups has good score about diet related question. 3. OHIP-14 has lower score as age is older, as more income. THI shows higher score from woman, 20~29, more income group and QOL has higher score from more than college educated, every day exercise groups. 4. Similar correlation indexes on QOL are OHIP-14, diet, oral health knowledge, oral health practice listed in order of effect size. Self-determined oral health index is important to improve quality of life along with health activity, especially oral health recognition index is different depends on age and monthly income that addresses it is necessary to deliver training with carefully designed oral health training program development.

한국 경제활동자의 구강 건강과 자살 생각의 관련성 융합연구 (Convergence Relationship of Oral Health and Suicidal ideation in Korea Economic activity)

  • 본 연구는 경제활동자의 구강 건강과 자살 생각과의 관련성을 알아보기 위해 우리나라 국민건강영양조사의 자료를 바탕으로 연구하여 경제활동자의 자살 예방과 구강건강 증진을 위한 기초자료로 활용하고자 한다. 자료분석은 복합표본 빈도분석, 복합표본교차분석, 복합표본 로지스틱 회귀분석을 통계프로그램 IBM SPSS Statistics 20.0으로 사용하여 분석하였다. 연구결과, 구강 건강과 자살 생각과의 관련성은 저작 불편 있는 경우 2.49배로 유의하게 높게 나타났다. 따라서 경제활동자들의 자살생각을 감소시킬수 있는 요인으로 구강건강관리가 필요함을 알 수 있다.

Protective Effect of EGCG Against Reactive Oxygen Species-induced Stress

  • EGCG[(-)-epigallocatechin gallate], is a major component of green tea has been considered as a major antioxidant constituent. It has been considered as potential chemopreventive and chemotherapeutic agents. However, very little is known about the cellular actions by which EGCG mediates its therapeutic effects. Various aspects of antioxidant activity of EGCG were evaluated in this study. EGCG itself did not show significant cytotoxicity. Significant 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was observed in all ranges of concentration ($0.8-100{\mu}g/ml$) used in this study. Protective effect of EGCG against hydrogen peroxide induced cell death was observed. Relatively high lipid peroxidation inhibitory activity were detected ($IC_{50}$ was about $20{\mu}g/ml$). EGCG also dose-dependently enhanced the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) in V79-4 cells. In concentrations of $100{\mu}g/ml$ of EGCG, activities of SOD, CAT and GPX were measured as 36.9 U/mg of protein, 22.9 U/mg of protein and 17.8 U/mg of protein, respectively. When these values were compared with those of the control groups (24.9 U/mg of protein, 14.9 U/mg of protein and 11.7 U/mg of protein), the relative increases were calculated as 48, 54 and 52%, respectively. Taken together, our findings suggest that EGCG can act as an antioxidant by scavenging radicals and enhancing antioxidant enzyme activities.

구강암에 대해 항암효과를 나타내는 methanol 자화 방선균의 분리 및 동정 (Isolation and Identification of Methylotrophic Actinomycetes capable of Producing Anti-oral Cancer Activity)

  • An appropriate amount of samples, collected from three each paddy forest, field and riverside soil near Taegu city, was suspended in sterile water and then diluted in order to isolation of antagonistic to oral cancer. The diluted samples were inoculated on separating medium in the routing spreading method. So, seven hundred and eighteen strains were isolated on HV agar and 220 strains were on methanol medium from soil samples. So, during the screening of anti-oral cancer activity from soil, we isolated microorganisms showing powerful antagonistic activity. Among them, No. 78 strain exhibited the most strongly anti-oral cancer activity. Microbiological properties were investigated by the methods described in the Bergey's Manual of Systematic Bacteriology and experimental methods of identification of actinomycetes by Hamada et al. As a result, a methylotrophic actinomycetes strain No. 79 was estimated as Amycolatopsis sp. based on taxonomic studies.
