Effect of temperature on resting spore germination of Plasmodioprora brassicae was indirectly estimated based on examining temporal change of number of inactive resting spores. Resting spore germination was the highest at $28^{\circ}C$ reaching 55.6% and 82.5%, 24hr and 132hr after treatment, respectively. Optimum pH for resting spore germination was pH6, following pH7 and pH8, and the germination was inhibited at pH 4, and pH9. termination of resting spores was stimulated by root extracts of radish, Chinese cabbage and kidney bean, but inhibited by that of lettuce. Number of inactive resting spores was increased as temperature increases and time prolongs after temperature treatment. However, degree of inactivation of resting spores after 1hr at $40{\sim}65^{\circ}C$ was similar with $40{\sim}60%$, but rapidly increased to 91.5% at $70^{\circ}C$. When root galls were submerged in water, density of inactive resting spores was increased rapidly and reached 60.3% 9 days after treatment. Flooding of infested soil resulted in 30% reduction of survived resting spores 5 months later. Among the two registered fungicides, fluazinam was better for inactivation of resting spores than flusulfamide, but both fungicides were inferior to phosphoric acid.
Betula platyphylla var. japonica and Betula ermanii segregate vertically at an elevation of approximately 1,850 m on Mt. Neko in Nagano Prefecture, Japan. B. platyphylla var. japonica and B. ermanii were the dominant species below and above this altitude, at which the mean-annual and growing-season air temperatures were $4^{\circ}C$ and $14.1^{\circ}C$, respectively. Based on a modification of Kira's warmth index which employs cumulative temperature represented as $^{\circ}C$ day, leaf unfolding in both species was observed to be initiated at $58^{\circ}C$ day and $169^{\circ}C$ day, respectively. In 1996, leaf unfolding was initiated on 18 May in B. platyphylla var. japonica (+/-6 days) and on 5 June in B. ermanii (+/-8 days), shortly after the last frost which occurred on 5 May 1995 above 1,850 m; below this elevation there was no risk of frost at the time. At elevations above 1,850 m, the unfolded leaves of B. platyphylla were damaged by late frost, while B. ermanii escaped injury because the leaves were still protected by winter buds. The optimum temperature for seed germination in both B. platyphylla and B. ermanii was $30^{\circ}C$. Temperature alternation from 10 to $30^{\circ}C$ and moist storage of seeds at $4^{\circ}C$ (stratification) prior to incubation increased germination rates in both species. The seedlings of B. ermanii had a greater survival rates than those of B. platyphylla var. japonica when planted above 1,850 m. Comparisons of the timing of leaf unfolding and the latest frost at a site appeared to be the main factors affecting the vertical distribution of these species.
The fungus which belongs to Alternaria has been isolated from "damping-off" infected seedlings of pitch pine (Pinus rigida Mill.) in the nursery (seed bed) of Chung Puk College, Cheong-Ju. The author has carried out this study regarding to (1) the effects of H-ion concentration on the growth of fungus, (2) the effects of temperature and humidity on the conidia germination, (3) the effects of temperature and humidity on the conidia formation. The summary of obtained in this experiments is as follows: 1. The suitable H-ion concentration for growth of isolated fungus is in the range of PH 4.0-9.5, the most suitable of it is PH 6.5-8.0. 2. The germination of the conidia is suppressed at below $0^{\circ}C$ or at $40^{\circ}C$ and above, the range of optimum temperature for the germination is $5^{\circ}C-35^{\circ}C$, the percentage of germination is highest at $20^{\circ}C$. And under relative humidities of 10% or below, the germination is suppressed, favorable relative humidities $85%{\cdots}100%$. 3. The formation of the conidia is suppressed at below $10^{\circ}C$ or at $34^{\circ}C$ and above, the favorable condition of the condia formation is as follows; Temperature; $22^{\circ}C-28^{\circ}C$ Relative humidity:85%-100%.

Chitinase-producing Paenibacillus elgii strain HOA73 has been used to control plant diseases. However, the antimicrobial activity of its extracellular chitinase has not been fully elucidated. The major extracellular chitinase gene (PeChi68) from strain HOA73 was cloned and expressed in Escherichia coli in this study. This gene had an open reading frame of 2,028 bp, encoding a protein of 675 amino acid residues containing a secretion signal peptide, a chitin-binding domain, two fibronectin type III domains, and a catalytic hydrolase domain. The chitinase (PeChi68) purified from recombinant E. coli exhibited a molecular mass of approximately 68 kDa on SDS-PAGE. Biochemical analysis indicated that optimum temperature for the actitvity of purified chitinase was $50^{\circ}C$. However, it was inactivated with time when it was incubated at $40^{\circ}C$ and $50^{\circ}C$. Its optimum activity was found at pH 7, although its activity was stable when incubated between pH 3 and pH 11. Heavy metals inhibited this chitinase. This purified chitinase completely inhibited spore germination of two Cladosporium isolates and partially inhibited germination of Botrytis cinerea spores. However, it had no effect on the spores of a Colletotricum isolate. These results indicate that the extracellular chitinase produced by P. elgii HOA73 might have function in limiting spore germination of certain fungal pathogens.
Various hydration treatments have been devised to improve the rate and univormity of seed germination as well as seed viability. Hydropriming and priming, the imbibitaion of seeds in distilled water and osmotic solutions, respectively, are useful techniques for early establishment of uniform seedlings. In this study, the optimum conditions were determined for hysropriming of cucumber seeds. Effects of hydropriming were also compared to those of osmotic priming. Seeds, hydroprimed with distilled water and primed with 50 mM $NaNO_3$at 20 for 24 hour, showd shorter number of days required to reach 50% of the final germination percentage ($T_{50}$), and mean number of days to germinate (MDG) that those untreated seeds. Hydroprimed with 10 mL distilled water showed better results in improving the rate of germination thatn other treatments tested. Hydropriming of the aged seeds improved germination percentage at 15$^{\circ}C$ and 25$^{\circ}C$. The $T_{50}$ and MDG values were reduced significantly by hydropriming of the aged seeds regardless of germination temperature. In addition to being simple and inexpensive, the hydropriming resulted in early germination at low temperature. he hydropriming was also effective in increase of germination percentage and normal seedlings.

Exochorda serratifolia, broad-leaved shrub has beautiful flowers and is resistant to cold, shade and decease. New this native plant is considered to be worth being exploited as the outstanding plant for landscaping and horticulture. So this study was executed to utilize Exochorda serratifolia as the planting material for landscaping and horticulture through the survey of its habitat environment and the experimennt of its seed physiology and germination, vegetative propagation, culture and utilization, etc.. The results are as follows ; 1. The color of the flower is white, blooming in the early and middle of May and the seeds ripen late in Sep.. 2. The elevation, elevation, gradient and direction of the native habitat were 250m, 20-25$^{\circ}$and northern side respectively. 3. The soil pH of the natural habitat was 5.3 and soil fertility was poor. 4. Exochorda serratifolia appeared as indicator within P. densiflora community and its neighboring species were Q. mongolica, Q. dentata, R. mucronulatum, L. obtusiloba, E. oxyphillus, C. heterolphylla, var thunbergii, etc.. 5. The optimum temperature for seed germination was found at 20$^{\circ}C$ and the longer the stratification period at 5$^{\circ}C$ was, the lower the germination rate was. 6. The treat merits of GA and Kinetin increased the seed germination rate, especially under the dark condition but their high concentrations decreased the seed germination rate. 7. The rooted rate of the greenwood cutting was the highest at IBA 100ppm plot of vermiculite bed but its high concentration decreased the rooted rate conspicuously. 8. In the tissue culture, the each 1.0 ppd plot of NAA, Kinetin and NAA 1.0ppm + Kinetin showed the best growth. And the mixture of NAA 1.0ppm and Kinetin showed better growth than the single treatment of NAA or Kinetin did. 9. Transplanted Exochorda serratifolia showed healthy growth with shaded environmental condition(42.1% light intensity), therefore they can be cultivated as a shade tolerant landscape plant. 10. It was considered that Exochorda serratifolia was applicable to group planting at shade places or under trees in parks, homes, etc..
Melanose caused by Diaporthe citri is a serious disease on yuzu fruits. Moisture is the most important to infect after the pycnidiospores are released. In order to understand how dryness and moisture interruption affect on germination, the conidial suspensions on slide glass were dried for 0-48 h. For the moisture interruption experiment, moisture was supplied for 10 h then interrupted for 0-6 h depending on the treatments. Germinations on the treatments with longer than 15 h of dryness were less than 10%. And those with longer than 3 h of moisture interruption were less than 30%. Compared with other fungal phytopathogens, D. citri was xero-sensitive. Germination on yuzu feel medium was measured under 15, 20, 25, 30, $35^{\circ}C$ with 3, 6, 9, 12, 15, 18, and 24 h of wetness. The minimum wetness periods achieving 10% and 50% germinations were 4.5 and 13 h, and the optimum temperatures were $29.1^{\circ}C$ and $29.2^{\circ}C$, respectively. From the yuzu medium study, the regression equations to be required wetness period achieving 10% and 50% germinations on various temperatures were $Wh_{50%}=0.1093{\times}T^2-6.3762{\times}+106.08$ and $Wh_{10%}=0.082{\times}T^2-4.8025{\times}T+74.861$.
Heteropappus arenarius Kitam., an autumn-flowering biennial belonging to wild chrysanthemums, is found to be native in southeastern coastal area and Jeju island of Korea. It could play a good role for ground cover plants on a large-scale landscape area, especially, barren soil or sloping hillside. This study was initiated to screen optimum germination temperature influenced by local strain and harvesting stage of H. arenarius. The following was the response of seed germination between local strain and temperature. The average of final germination percentage (FG) was the highest in 'Guryongpo' (89.7%) among four local strains, followed by 'Gujwa' (87.3%), 'Gampo' (87.3%), and 'HKNU-I' (71.5%). The average of $T_{50}$ was shorter in 'Gujwa' (3.6 d) and 'Guryongpo' (4.0 d) than the others. The average of FG and $T_{50}$ was the highest as 76.2% and shortest as 3.6 d in $20^{\circ}C$, respectively, followed by $30^{\circ}C$, $25^{\circ}C$, and $15^{\circ}C$. In case of 'Gujwa', however, FG and T50 was higher in $20^{\circ}C$ and shorter in $15^{\circ}C$ than others. In the relationship between harvesting stage and temperature, the average of FG was greatly higher in Stage III (90.7%) and Stage IV (88.6%) than the others including Stage II (35.7%) and Stage I (26.0%). The average of $T_{50}$ was shorter in Stage IV (3.7 d) and Stage III (4.3 d) than the others, which showed less than 50% of FG. Nevertheless, the available range of seed harvesting stage was from Stage I to Stage IV because H. arenarius seeds could germinate at all stages. In conclusion, it was recommended that the optimum temperature and harvesting stage was $20^{\circ}C$ and Stage $III{\sim}IV$, respectively, for seed germination of H. arenarius.
This study was conducted to investigate the variation of cone, seed and germination characteristics among populations and among individuals within populations of Picea jezoensis(Siebold & Zuccarini) Carriere distributed in Korea. Cone collected from 25 trees in two natural populations(Mt. Jiri and Mt. Dokyu) and their thirteen cone and seed characteristics as well as three germination behaviors were analyzed. Statistical analysis showed significant differences among populations and among individuals within populations in 9 traits except for seed breadth, seed weight, index of seed wing and mean germination time. Generally, morphological variation of cone and seed wings from Mt. Jiri population has smaller and longer than those of Mt. Dokyu population. Percentage of sound seeds and Tetrazolium test from Mt. Jiri population showed 1.79 and 1.87 higher values than Mt. Dokyu population, respectively. The maximum germination percentage was obtained at the optimum temperature of $20^{\circ}C$ and in this case, mean germination time and germination rate showed 7.5 days and 2.9 ea./day, respectively. In seed germination behaviors, percentage(40.7%) and rate(0.90 ea./day) of germination in Mt. Jiri population were more higher and faster than those of Mt. Dokyu population(17.7%, 0.37 ea./day). According to correlation analysis, P. jezoensis populations with small restricted distribution may have been reduced because seed qualities were correlated with increased levels of inbreeding and disproportion flowering.
This experiment was conducted to investigate the optimum temperature and the effects of sulfuric acid, flowing water and $GA_3$ on the germination of Bupleurum falcatum. At the same time, the effect of germination-related compounds in Bupleurum falcatum seed was tested. Among the tested temperatures $(15,\;20,\;25\;and\;30^{\circ}C)$ the highest germination rate was obtained at $15^{\circ}C$. The optimum soaking time of sulfuric acid was 5 minutes at the concentration of both 0.1 and 1.0%. At the same time, the germination rate was highest when Bupleurum falcatum seed was washed for 2 days in the flowing water before germination test. The effect of $GA_3$ on the germination was highest at 100 ppm out of 10, 50 and 100 ppm. Regardless of treatments, the germination rate of Jeongseon species was higher than that of Mishima species. SEM photographs revealed that most of germination-inhibiting substances in the seed coat were washed away by rinsing in the flowing water for 2 days thus the seed coat became more smooth. The 4, 000 ppm alcohol extract of Bupleurum falcatum seeds inhibited the germination of lettuce seeds as much as 41.7% in Jeongseon species and 58.3% in Mishima species. The higher extract concentration from seed of Bupleurum falcatum, the lower the germination of lettuce seeds was obtained. Out of phenolic acids, the contents of orchinol, pyrogallol, and p-hydroxybenzoic acid were highest in both varieties. Compared with Mishima species, Jeongseon species showed much higher content of salicylic and vanillic acid.
