Characterization of Dextranase Produced by Flavobacterium multivorum HL-1 (Flavobacterium multivorum HL-1 이 생산하는 Dextran 분해효소의 특성)

  • Dextranase produced by Flavobacterium multivorum HL-1 isolated from soil were characterized. Optimum growth condition for the production of the enzyme by the strain appeared to be at the 1.0% concentration of dextran. When NH$_{4}$N0$_{3}$ and beef extract was added to the culture media, the enzyme activity increased upto 570 and 680 units per ml respectively, and other nitrogen sources did not increase the activity. Urea, casamino acid, (NH$_{4}$)$_{2}$SO$_{4}$ and KNO$_{3}$ inhibited the activity. In the presence of Mg^{2+} in culture media, the enzyme activity increased 124%, but its activity was inhibited in the presences of Ca$^{2+}$, Co2$^{2+}$, Hg^{2+} and Zn$^{2+}$ . The optimum temperature for the enzyme activity was 45-55$\CIRC$C . In the ranges of pH 4 to 10, the activity of the enzyme appeared approximately similar.

Producton of Yeast Spores from Rice Wine Cake. (주박으로부터 효모포자의 생산)

  • Rice wine cake (RWC) is the solid waste obtained after rice wine fermentation. For the mass production of the spores of yeast Saccharomyces from RWC, the optimum pretreatment condition of RWC, the optimum composition of culture medium, and the optimum culture condition were examined. For sporulation, yeast cells were grown in the pre sporulation medium (PSM), transferred into sporulation medium (SM) containing 1 % potassium acetate, and incubated in a rotary shaking incubator at $25^{\circ}C$ for 4 days. The supernatant of the mixture of RWC and water was used as the presporulation medium (PSM). The optimum temperature and time for the pre-incubation of the mixture of RWC and water (1:2) to obtain maximum sporulation yield were $V^{\circ}C$ and 24 hr, respectively, and optimum culture time in PSM was 48 hr. Using these optimum conditions, the asci number obtained was 0.72$ 1.06${\times}$10^{8}$$m\ell$. The addition of wheat coat koji into SM increased the final number of asci to beTEX>$10^{8}$ $m\ell$. Spores were formed in the SM with the initial pH of 7-11, but no spores were formed in the SM with the initial pH of 5. To save the time and effort to pretreat the RWC, 2% and 0.5% RWC without any pretreatment were directly added into PSM containing 1 % brown sugar and SM, respectively, and the maximum asci number of $1.27${\times}$10^{8}$ /$m\ell$ was obtained.

Optimal Condition of Mycelial Growth of Grifola frondosa (잎새버섯 균주의 균사체 생육 최적조건)

  • This study was carried out to obtain basic data on mycelial growth characteristics for an artificial cultivation of Grifola frondosa. Ten strains of G. frondosa were collected from Korea, China and Japan and investigated its optimal culture condition. Among four kinds of mushroom culture media, PDA medium was selected as the suitable culture medium. The optimal conditions for the mycelial growth of G. frondosa in PDA medium were $25^{\circ}C$ and $4{\sim}5$ of pH, respectively. The carbon and nitrogen sources for the optimum mycelial growth were fructose and peptone, respectively, and the highest mycelial growth was observed when the C/N ratio was $10{\sim}20$.

Yeast Production from Soybean Curd Waste Water (두부 폐수를 이용(利用)한 효모(酵母) 배양(培養))

  • As a primary study for SCP production from soybean curd waste water, selection of yeast and optimum cultivation condition of selected yeast on soybean curd waste water were investigated. Eighteen strains of the genus Candida and Saccharamyces were tested to compare their abilities to grow on soybean curd waste water. Candida utilis YUFE 1508 and Candida guilliermondii KFCC 35120 grew most successfully. Optimum pH and optimum temperature of the basal medium for growth of the two strains were $6.0{\sim}6.5$ and $25^{\circ}C$, respectively. The optimum culture medium of the two yeasts was soybean curd waste water supplemented with molasses 2.5% (as total sugar), ammonium acetate 0.1-0.3% (as nitrogen), $KH_2PO_4$ 0.1-0.2% (as phosphorus), and $K_2HPO_4$ 0.05% (as phosphorus). But yeast growth was not affected by metal salts. Under the optimum cultivation condition, the maximum cell weights of Candida utilis YUFE 1508 and Candida guillfermondii KFCC 35120 were 1.313g and 1.322g/100ml of culture broth respectively after 48 hr of cultivation. The cell yields of Candida utilis YUFE 1508 and Candida guilliermondii KFCC 35120 were 68.4% and 74.2%, respectively, based on utilized sugar. On the other hand, crude protein of dry yeast produced by Candida utilis YUFE 1508 and Candida guilliermondii KFCC 35120 under optimum condition was 54.0% and 56.8%, respectively.

Factorial design에 의한 Acetobacter xylinum KJ1의 Bacterial cellulose 생산조건의 최적화

  • Acetobacter xylinum KJ1 efficiently producing bacterial cellulose(BC) in shaking culture was isolated from a rotten grape. The strain was used to investigate optimum operating conditions for increasing BC production and factorial design model was employed for the optimization. The results of experiments were statistically analyzed by SAS program. Reciprocal effects of each factors(carbon source concentration, shaking speeds(rpm), oxygen pressure, and CSL concentration) and culture condition of BC production were examined by getting regression equation of the dependent variable. Comparisons between experimental results and predicted results about BC concentration were done in total 24 experiments by combination of each factors using SAS program, and the correlation coefficients of BC concentration and BC yield were 0.91 and 0.81, respectively. The agitated cultures were performed in various operation conditions of factors which affected considerably to BC production in jar fermentor. The results showed that BC concentration was 11.67g/ L in 80 hours cultivation under the condition of carbon source concentration shaking speeds(rpm) : oxygen pressure: CSL concentration = 4% : 460rpm : 0.28 : 6%. On the other hand BC yield was 0.42g/g in 80 hours cultivation under the condition of carbon source concentration shaking speeds(rpm) : oxygen pressure: CSL concentration = 4% : 564rpm : 0.21 : 2%. The BC production could be enhanced up to more than 65.3% by factorial design. The result of a verifying experiment under the optimal conditions determined by the factorial design to the BC production showed that the model was appropriate by obtaining BC concentration of 11.02g/L in the optimum condition

Establishment of Optimum Condition for the Coagulation and Antimicrobial Activity of Konjac Jelly (곤약의 최적 응고 및 항미생물 활성을 위한 조건 확립)

  • This study was conducted to establish optimum conditions for coagulation of konjac jelly as well as antimicrobial activity by concentration of $Ca(OH)_2$. Hardness, gumminess, and chewiness of konjac jelly increased according to concentration of konjac powder, the key material of konjac jelly. The highest sensory evaluation score was acquired with konjac jelly made with 3% konjac powder. A $Ca(OH)_2$ concentration of 0.4-0.6% as a coagulation agent was optimum for coagulation of konjac jelly. Further, sensory score was highest at a $Ca(OH)_2$ concentration of 0.6%. All populations of bacteria, yeast, and mold in konjac jelly were restrained by $Ca(OH)_2$ in a concentration- dependent manner. Furthermore, all tested microorganisms were strictly restrained at $1.0{\times}10^{-2}$ N of $Ca(OH)_2$.

Production of Lignin-Degrading Enzymes by White Rot Fungi Immobilized in a Rotating Bioreactor (회전생물반응기에 고정화된 백색부후균에 의한 리그닌 분해효소의 생산)

  • The objective of this study is to investigate optimum condition for lignin peroxidase production by white rot fungi Phanerochaete chysosporium IFO 31249 immobilized in a rotating bioreactor. The maximum lignin peroxidase activity of batch culture in rotating bioreactor was 300 U/L. The optimum rotating speed and packing ratio of support for lignin peroxidase production in a rotating bioreactor were 1 rpm and 20%, respectively. The optimum concentration of $MnSO_4$$\cdot$$H_2O$ for manganese-dependent peroxidase production in a rotating bioreactor was 50 ppm. The sufficient supply of oxygen was the most important factor to achieve maximum lignin peroxidase production. It was possible to produce lignin peroxidase (LiP) and manganese-dependent peroxidase (MnP) for at least 3 times successive repeated-batch cultures, respectively.

  • The purpose of this study was to determine the organoleptic charateristics of cauliflower pickles made in various compounding ratio according to central conposite design for optimum organoleptic characteristics of the cauliflower pickles. The optimum mixing condition of cauliflower pickles were optimized, using central composite design with 3 variables and 3 levels, by response surface methodology. The various kinds of cauliflower pickle were made in various compounding ratio of vinegar, salt and sucrose - critical ingredients of pickle recipe - and were presented to reliable panels, who graded the subjects in 7 degrees for 4 items : color, flavor, hardness and overall quality. The optimum mixing conditions of cauliflower pickle were 603.50g of vinegar, 80.13g of salt and 251.07g of sucrose in the maximum point of overall quality.

Fusarin C Production by Fusarium moniliforme in Liquid Media (액체배지에서의 Fusarium moniliforme에 의한 Fusarin C생성에 관한 연구)

  • This study was carried out to find the optimum condition for production of fusarin C, Known as a mutagenic and toxic agent. Three liquid media, Czapek-kox, MYRO, GYEP and microorganism, Fusarium moniliforme F84 isolated by Bjeldanes lab. in U.C. Berkeley, were used in this experiment. Fusarin C amounts were determined upon PH and fluctuating time/temperature. The results were obtained as follows; 1. The largest amounts of fusarin C were shown in Czapek-Dox medium and the amounts were about 1/10 of fusarin C amounts in corn culture. 2. In Czapek-Dox medium, the best condition for fusarin C production was at $28^{\circ}C$ for 2 weeks culture, and in corn culture, at $28^{\circ}C$ for l week culture. 3. The best initial PH for fusarin C production was 6.5 in Czapek-Dox medium and also at the initial pH 6.3, 5.9 the fusarin C amounts produced were much higher than other initial PH.

Effect of Seeding Rate on Growth and Yield of Ginseng Plant in Direct-Sowing Culture (인삼 직파재배에서 파종밀도가 생육 및 수량에 미치는 영향)

  • To know possibility of raw ginseng production for white- and red-ginseng by direct-sowing culture, seeds were directly sowed or seedlings were transplanted at soil condition of sandy loam in ginseng field. After cultivation, the characters of 5-year-old ginseng were investigated. Number of survived plants was increased with increase the seeding quantity in direct-sowing culture, the survived plants in direct-sowing culture was higher than that of transplanting one. Rate of the numbers of survived plants to numbers of seeds sowed in plots of 134 or 90 seeds sowed per tan, 180 x 90cm area, inspire of high number of survived plant was high compared to that of transplanting culture. Occurrence rate of rusty root of ginseng in direct-sowing culture was low significantly compared to that of transplanting culture. Root yield showed in the order of 134, 268, 90 seeds sowed, the values of yield in direct-sowing culture were high obviously compared to that of transplanting one. Individual root weight was increased with decrease the seeding quantity, however, the root weight in plot of 90 seeds sowed showed almost equal the weight in transplanting culture. The number of usable raw ginseng for white- and red-ginseng was increased with decrease of the seeding quantity; the numbers were higher than that of transplanting culture remarkably. We concluded that optimum seedling rate in direct-sowing culture of ginseng was 90 to 134 seeds per tan considering the yield per area and production rate of large root.
