Nickel recovered from the spent oil-hydrogenation catalysts was used in hydrogenation catalyst preparation. The spent catalyst contains approximately 21.8% Ni, 0.7% Mg, and small quantities of Al, Fe, and Zn. Nickel recovery was obtained by inorganic acid digestion in the order of HCI>$NHO_3$>$H_2SO_4$. For $HNO_3$, 3hour extraction with 3N solution was satisfactory. In the PH range of 6.5~9.0, Ni recovery was higher, but metallic impurities were found to be coprecipitated. The PH in the range of 7.0~9.0 seems to be the optimum condition for separation to obtain acceptable Ni precipitates without the decrease of purity. The catalysts prepared with reclaimed nickel by wet reduction methods showed catalytic activities close to those prepared using reagent nickel in the oil hydrogenation reaction. The surface areas of the support do not seem to affect the catalytic activity.
Rhodopseudomonas sp. was immobilized in three supports(agar, k-carrageenan, and PVA) in order to remove nitrate in wastewater coming from fish farm. Among them 3% agar was the most suitable support when denitrification rate and bead durability were tested. Optimum bead size was 4mm-diameter when the substrate transfer into the bead and shear stress for bead were considered, and optimum cell loading was 25mg dry $cells/cm^2$gel gel. Ethanol was the best as a carbon source, and optimum C:N ratio, temperature and pH were 1.5:1, $31^{\circ}C$,, and 6, respectively. Under these conditions the maximum denitrification rate in synthetic wastewater was $$345{\MU}{\ell};N_2/Cm^3 gel{\cdot}hr;and that in modified MYC medium was 450{\MU}{\ell}};N_2/Cm^3 gel{\cdot}hr $$.
The main objectives of this study were to fruit body of Phellinus linteus using liquid spawn. Highest mycelial growth and in vitro fruiting was observed when $180m{\ell}$ of water was added to 200g of brown rice in $1000m{\ell}$ PP bottle. Optimum inoculum amount was 100ml per bottle. Better mycelial growth and fruiting were observed when the amount of brown rice was lowered. For timber log culture of P. linteus, low contamination was observed when logs were sterilized for as low as 60 mins. But, best mycelial growth was observed when the logs were sterilized for 120mins. Among different log types, highest mycelial growth and fruiting were observed in oak and mulberry. Only few fruiting could be observed in birch, chestnut, alder, while no fruiting was produced in Aspen. Best fruiting was produced from isolate PH-211, although other isolates also produced few fruiting.

The objective of this study is to investigate the optimum conditions of extracting collagen without chrome ion from the leather waste. The effect of temperature, pH, and the concentration of alkaline solution on the collagen extraction has been studied. The result indicated that the incipient denatured temperature of collagen measured by viscosity was $25^{\circ}C$ and the complete denatured temperature was $31.5^{\circ}C$. The optimum solubilization condition for temperature was between $15^{\circ}C$ and $20^{\circ}C$, pH was 1.5, the concentration of alkaline solution was 3% of sodium hydroxide. The almost complete chrome ion separation was possible around the pH of 1.5. The separation efficiency of chrome ion from tannery waste was more than 99.5%. Extraction efficiency of crude protein from leather waste was about 89.5%. The hydroxyproline and collagen content in the extracted crude protein were 8.53% and 63.62%, respectively.
Endo-polygalacturonase was purified from tomato, Lycopersicon esculentum L. The purification procedures included gel filtration on Sephadex G-150 and DEAE-cellulose ion exchange chromatography. Yield of the enzyme purification was 12.74 %. Purified enzyme was confirmed as a active single band by the SDS-polyacrylamide gel electrophoresis. When the purified enzyme was applied to SDS-PAGE, the molecular weight was estimated about 50,000. The optimum pH for the enzyme activity was 5.0 and the range of its stability to the pH was 4.0 to 5.0. The optimum temperature was $50^{\circ}C$, while the enzyme was abruptly inactivated above $50^{\circ}C$. From the result of the study on the effects of metals ion, it was found that $Ag^+$, $Zn^{++}$ and $Mg^{++}$ increased on the enzyme activity. In contrast, $Ba^{++}$, $Hg^{++}$, $Pb^{++}$, $Ca^{++}$, $Mn{++}$, $Cu^{++}$, $Fe^{+++}$, $Na^+$ and $K^+$ decreased it. the reaction catalyzed by this enzyme followed typical Michaelis-Menten kinetics with the Km value of $1.43{\times}10^{-1}\;mol/l$.
For efficient micro scale syntheses of Rose $Bengal-^{131}I$, $Hippuran-^{131}I$, and $H.S.A.-^{131}I$, the dependence of labelling yields on pH, on salt contents, and on the volume of buffer solution in the reaction mixtures as well as the reaction apparatus were studied. pH of 5.6 was optimum for preparation of both Rose $Bengal-_{131}I$ and Hippuran $-^{13}I$ but pH of 8.5 was optimum for preparation of $H.S.A.-^{131}I$. Salt in the reaction mixtures hindered drastically the formation of $Hippuran-^{131}I$ but it slightly increased the labelling yield of H.S.A.. The compactly closed reaction vessels were effective for preparations of both Rose $Bengal-^{131}I$and $Hippuran-^{131}I$ in small volume. Thereupon, the labelling procedures were modified to bring about higher labelling yields and better reproducibilities. By these newly established procedures, the labelling yields of Rose $Bengal-^{131}I$ and $Hippuran-^{131}I$ could be increased even with the home-produced sodium $iodide-^{131}I$ solution containing reducing agent.
The selective absorption of metal ions by chelating agent-loaded resins was studied in aqueous media. The resins were prepared by loading the conventional anion exchange resin, Dowex 1-X8 (50 to 100 mesh) with chelating agents containing sulfonic group, such as 8-hydroxy-quinoline-5-sulfonic acid (HQS) and 7-nitroso-8-hydroxyquinoline-5-sulfonic acid (NHQS). The stability of the resin was markedly influenced by the following factors; (1) the affinity and concentration of anions in the external solution, (2) the pH of the media. The optimum conditions for the absorption of metal ions were determined with respect to the pH, shaking time, and the effect of anion concentration in the medium. Under the optimum condition the order of the absorption of metal ions such as Fe(Ⅲ), Cu(Ⅱ), Pb(Ⅱ), and Zn(Ⅱ) was in accord with that of the stability constants of the chelates. The total capacities of the resins were found in the range of 0.6∼1.6 mmole metal per gram.
Effects of various kinds of sawdusts, supplements and culture conditions on activities of several enzymes such as protease, phenoloxidase and cellulase produced from mycelium of P. ostreatus grown on sawdust medium were studied and the results are as follows; Higher specific activity of these enzymes was observed when oak tree sawdust and poplar tree sawdust were supplemented with rice bran or wheat bran at rate of 30%, 20% and 10% in total volume respectively. Higher total activities of protease, phenoloxidase and cellulase were observed at 70% of the moisture contents of culture media, while lower activity of these enzymes was observed with 40% moisture contents of sawdust culture medium. The pH 4 and 9 of the sawdust media appeared to be optimum pH for the. production of protease while pH 5 and 7 were optimal for the production of phenoloxidase. The pH 6 of the sawdust medium was optimal for the production of cellulase. The optimum incubating temperature for the production of protease, phenoloxidase and cellulase was $25^{\circ}C$. Higher total activities of protease and phenoloxidase were observed when culture medium was added with wood vinegar at the control, and 0.5% for cellulase.
This study was carried out to leach valuable metals from galena using indigenous bacteria with no optimum pH conditions at room temperature. Even in these conditions, the rod-shaped indigenous bacteria, ranging from $0.4{\times}0.2{\mu}m$ to $0.5{\times}1.7{\mu}m$, were attached to the surface of the galena. For the 19 days of the bioleaching experiment, the content of Ph, Fe, Zn ions was found to be 347, 222 and 1.7 times higher than that of the control leaching agent, respectively. Numerous hexagonal column crystals were observed on the surface of galena. Those crystals may be formed from the biooxidation of galena by the indigenous bacteria. XRD analysis showed the peaks of anglesite observed in the bioleached galena. It is expected that more valuable elements can be leached out of the galena, if the bacteria is used under optimum pH and temperature conditions in future bioleaching experiments.
This study was carried out to get more operational characteristics of Anoxic(anaerobic)-Oxic-Anoxic-Oxic $(AO)_2$ sequencing batch biofilm reactors (SBBRs) at the low TOC concentration, The operating time in anoxic (anaerobic) time to oxic time was I : I. Experiments were conducted to find the effects of the aeration time distribution on the organic matters and nutrients removal. Three lab-scale reactors were fed with synthetic wastewater based on glucose as carbon source. During studies, the operation mode was fixed. The first aeration time to the second aeration time in SBBR-I was 2 : 3, and those in SBBR-2 and SBBR-3 were I : 4 and 3 : 2, respectively. The organic removal efficiency didn't show large difference among three reactors of different aeration time distribution. However, from these study results, the optimum aeration time distribution in the first and the second aeration time for biological nutrient removal was shown as 3 : 2. The release of phosphorus was inhibited at the second non-aeration period because of the low TOC concentration and the nitrate produced by the nitrification at the first aeration period.
