Changes of Quality in the Osmotic Dehydration of Cherry-Tomatoes and optimization for the Process (방울토마토의 삼투건조시 품질의 변화와 공정의 최적화)

  • This study was carried out to determine the effect of osmotic dehydration as pretreatment on the qualities of dried cherry-tomatoes. The weight reduction and solid gain in osmosed cherry-tomato were increased by increasing sugar concentration, immersion temperature and time; among three parameters, the immersion temperature affected more than sugar concentration and immersion time did. The moisture content was decreased as increasing sugar concentration, immersion temperature and time, and it was the lowest at the osmotic conditions of 7$0^{\circ}C$, 60$^{\circ}$Brix and 11hr. To determine the optimum processing condition by RSm, the polynomial optimum models were established. The regression models was significant (p<0.05). It was used contour plots to optimize osmotic dehydration. The optimum condition for osmotic dehydration as pretreatments for drying of cherry-tomatoes were immersion temperature of 47~53$^{\circ}C$, sugar concentration of 39~43$^{\circ}$Brix, and immersion time of 7hr, in which process conditions were 78~86% moisture content, 8.5~10$^{\circ}$Brix sugar content and 80~86% weight reduction.

Optimal Culture Conditions on the Tyrosinase Inhibitor Production by Actinomycetes F-97 (방선균 F-97에 의한 Tyrosinase 저해제 생성 최적 배양 조건)

  • A Actinomycetes F-97 producing tyrosinase inhibitor was isolated from soil samples. The optimum culture condition for 쇼rosinase inhibitor production was investigated and the results were as follows. The best carbon source for tyrosinase inhibitor production was shown as soluble starch, the optimum concentration was 3.0%. The best nitrogen source for tyrosinase inhibitor production was shown as peptone, the optimum concentration was 0.36%. As effect of metal ions on the production of tyrosinase inhibitor, K$_2$HPO$_4$ was shown the best and the optimum concentration was 0.1 mM. The optimum pH and temperature was shown 7.0 and 30${\circ}$C, respectively. And the highest tyrosinase inhibitor production was observed at 70hr cultivation under optimum conditions in jar fermentor scale.

  • We report that the optimum substrate temperature to fabricate a-Si:H $n^+-p-p^+$ cell decreases with increasing the boron concentration in the Player. The results can be explained as the dependence of substrate temperature for the relaxation of silicon atoms and the bonded hydrogen concentration in the player.

  • A calculation method of optimum parameters on dual adsorption isotherm system was examined. The optimum parameters were obtained by non-linear regression analysis based upon a limited solute concentration of dual adsorption isotherm. The results were analyzed with adducing experimental data of formerly reported treaties. The percentage mean deviation of dual adsorption equation calculated with optimum parameters was less than about 5% of experimental data, which was far less than results obtained with parameters of the adduced treatises.

  • A three variables by three level factorial design and response surface methodology were used to determine optimum conditions for osmotic dehydration of banana. The moisture loss, solid gain, weight loss and reduction of moisture content after osmotic dehydration were increased as temperature, sugar concentration and immersion time increased. The effect of concentration was more significant than those of temperature and time on mass transfer. Color difference and titratable acidity were decreased by higher concentration. Sweetness was increased by increasing sugar concentration, temperature, immersion time during osmotic dehydration. The regression models showed a significant lack of fit (p>0.5) and were highly significant with satisfying values of R2. To optimize osmotic dehydration, based on surface response and contour plots, superimposing the individual contour plots for the response variables. the optimum conditions for this process wire 26$^{\circ}C$, 44 $^{\circ}$brix and 2 hrs for moisture content, sweetness and color difference are less than 72%, 24 obrix and 10 degree.

  • In order to utilize unsorted ONP, which contains leaflets (printed coated papers), as a raw material to produce pulp mold, optimum conditions for repulping were investigated with the pilot Helico pulper at Centre Technique du Paper (CTP), France. Two major process factors were focused: repulping concentration and rotor speed. Repulping at a higher concentration showed more rapid defibering kinetics. Increasing the rotational speed of rotor at the optimum repulping concentration accelerated the defibering kinetics while it also led to higher fines generation and faster decrease in drainage property of the produced pulp. Hence, an alternative way was suggested: starting repulping at a conventional rotor speed and then accelerating the rotor speed for the last minute(s) of repulping.

  • This research aims the production of anti-tumor substances through in vitro culture of hairy roots transformed by Agrobacterium rhizogenes in Artemisia sylvatica MAX and the effect of culture conditions on optimum growth of hairy roots. We investigated the optimum medium, pH, carbon source, sucrose, light, Fe and polyamine conditions of various lines of hairy roots (NK3, NK4, YX. NK3-10) induced from Artemisia sylvatica to increase the optimum growth of hairy roots. MS medium was the best for optimum growth of hairy root clone, NK3-S10. The optimum culture period was 4 weeks for NK3-S10. The optimum sucrose concentration was 3.5%. The optimum concentration of FeSO$_4$, spermine and spermidine was 0.1 mM, 10 mM and 100 mM, respectively.

  • Characteristics of cell growth and poly(3-hydroxybutyrate-co-4-hydroxybutyrate) [P(3HB-co-4HB)] synthesis was investigated through flask and batch cultures of Alcaligenes latus and Comamonas acidovorans. The specific growth rate of C. acidovorans increased with yeast extract concentration and decreased with 1,4-butanediol concentration. Optimum glucose concentration for growth of C. acidovorans was 20 g/L. In one-step flask cultures of C. acidovorans, final dry cell weight and PHA content decreased with the ratio of 1,4-butanediol to glucose, while the 4HB fraction in copolymers gradually increased to 100 $mol\%$ with an initial 1,4-butanediol concentration of 20 g/L as single carbon source. The specific growth rate of A. latus decreased with v-butyrolactone concentration and optimum sucrose concentration for growth was 10 g/L. In batch cultures of A. latus, 4HB fraction increased with initial v-butyrolactone concentration. P(3HB-co-4HB) with 19 $mol\%$ 4HB was obtained when the initial ratio of v-butyloractone (g/L) to sucrose (g/L) was 10 : 10.

  • The performance of ozone contactor in ozone-BAC advanced water treatment process was evaluated by the degree of decomposition of organic matters. The degree was measured by the analyses of $UV_{254}$ absorbance and the concentrations of DOC and BDOC for the sand filtered water and the ozone treated water, respectively. In addition, the ozone concentration in the contactor, required for the maximum BDOC concentration, was selected as the optimum concentration, and the appropriate residential time of ozone treated water in a reservoir was recommended based on the residual ozone concentration in the treated water. The following results were obtained from the pilot scale experiments. By ozonation $UV_{254}$ absorbance was decreased, and BDOC concentration was increased. The change of DOC concentration by ozonation was negligible, but the excess input of ozone resulted in the removal of the small amount of BDOC by complete oxidation. The optimum ozone concentration was 0.58mg $O_3/mg$ DOC. In order to remove residual ozone, 20minutes of the residential time were enough after ozonation.

Optimization of Processing Conditions of Chinese Smoke-cured Bacon (Larou) with a New Natural Coating Solution during Storage Period

  • The study aims to determine the optimum sterilization rate and water activity of Chinese traditional smoke-cured bacon product (Larou) in the preservation with natural coating solution. With the response surface methodology (RSM), we analyzed 3 factors of processing conditions (the concentration of lysozyme, concentration of sodium alginate, and concentration of chitosan) and 2 response factors (sterilization rate and water activity). Sterilization rate and water activity of Larou were largely affected by the concentration of lysozyme, concentration of sodium alginate, and concentration of chitosan. The final optimum concentrations of lysozyme, sodium alginate, and chitosan were 0.09, 1.40, and 1.60% and realized the high sterilization rate. Water activity of sliced Larou was significantly correlated with the sterilization rate. Low-field nuclear magnetic resonance analysis verified the optimum processing conditions. The coating resulted in 99.69% rate of reducing bacteria after 30-day storage. The data of the total number of colony, peroxidation value, moisture content, pH, and sensory evaluation provided the theoretical basis for extending the shelf life of Chinese traditional smoke-cured bacon product (Larou) with natural coating solution.
