Production of poly-$\beta$-hydroxybutyric acid(PHB) from Liquefied Natural Gas using an Obligatory Methanotroph Methylosinus trichosporium OB3b (메탄자화균 Methylosinus trichosporium OB3b를 이용한 액화 천연가스로부터 poly-$\beta$-hydroxybutyric acid(PHB)의 생산)

  • An obligatory methanotroph Methylosinus trichosporium OB3b was cultivated for the production of poly-${\beta}$-hydroxybutyric acid(PHB) in shake-flask using liquefied natural gas(LNG) as the sole source of carbon and energy. The maximal specific growth rate decreased by 40% using LNG compared with that obtained with pure methane. This is attributed to the inhibition by ethane and propane presents in the LNG as impurities. For the production of PHB, two-stage culture separating the production stage from the growth stage was carried out. PHB accumulation was observed after switching nutrient-sufficient to nutrient-limited condition of non-carboneous component (NO3-, PO43-, K+, Na+, Fe2+, or Mg2+). The limitation of K+ or Mg2+ resulted in relatively high PHB content, but the highest content was obtained by nitrate limitation. The optimal pH and temperature for PHB accumulation was 7.0 and $30^{\circ}C$. Under the optimal condition the maximal PHB content was about 45% after 4-day cultivation.

  • This study was carried out to examine the optimal cultural condition in enzyme activities of CMCase, FPase and xylanase in selected strains which secret extracellular enzymes for using deinking agent to old newsprint. The results of this study were as follow: The production of enzyme by Bacillus pumilus I was maximal as grown on the medium, containing of rice bran+xylan $2.0\%$, peptone $0.8\%,\;K_2HPO_4\;0.1\%\;and\;CaCl_2\;0.06\%$ at pH 8.0 and $28^{\circ}C$ for 72 hours. Optimal cultural condition of B. subtilis I was avicel+xylan $3.5\%,\;urea\;0.4\%,\;K_3PO_4\;0.1\%\;and\;CaCl_2\;0.015\%$ at pH 9.0 and $28^{\circ}C$ for 36 hours. The maximal enzyme production was observed in the medium, containing of avicel+xylan $3.5\%,\;urea\;1.6\%\;and\; K_2HPO_4\;0.125\%$ with pH 9.0 when B. pumilus II was cultured at $28^{\circ}C$ for 60 hours. The production of enzyme by B. subtilis IT was maximal as grown on the medium, containing of xylan $2.0\%,\;yeast\; extract\;0.6\%,\;K_2HPO_4\;0.1\%\;and\;ZnSO_4\;0.04\%$ at pH 8.0 and $34^{\circ}C$ for 36 hours. The activities of FPase and xylanase in tested 4 strains were not much different with Thermomonospora fusca.

The Growth of L.acidophilus and B. bifidum in Fermented Milk Product (Lactobacillus acidophilus와 Bifidobacterium bifidum의 발효 유제품내의 성장)

  • The optimal condition for manufacturing the yoghurt by using B. bifidum and L. acidophilus was studied. Optimal growth condition of B. bifidum and fermentation effects of B. bifidum, L. acidophilus and Str. thermophilus in mixed culture were investigated separately. Fermentation conditions of B. bifidum were optimized in 12% reconstitued skim milk. The addition of BIOS 2000 to 12% skimmilk was effective on the growth and acid production of B. bifidum. There was asymbiosis among B. bifidum, L. acidophilus and Str. thermophilus in mixed culture which resulted in higher viable cell numbers and acid production. The optimum production of yoghurt was achieved by incubation at $37^{\circ}C$ for 8 hours with 5% mixed starter culture (B. bifidum+L.acidophilus+Str. thermophilus). In this case the effective ratio of B. bifidum: L. acidophilus: Str. thermophilus was 2:1:1 on both acid production and viable cell counts.

High Yield Production of Cyclofructan by Deletion Mutant Enzyme of Cycloinulooligosaccharide Fructanotransferase (Cycloinulooligosaccharide fructanotransferase의 결손변이효소에 의한 cyclofructan의 고효율 생산)

  • This study investigated the optimal conditions of high yield production of cyclofructan (CF) using recombinant deletion mutant enzyme CFT108 which is constructed by N-terminal deletion from cycloinulooligosaccharide fructanotransferase (CFTase) gene of Penibacillus polymyxa. The production yield was dependent on reaction time, substrate concentration and enzyme concentration. The optimum reaction time for industrial purpose was achieved at 3 hr reaction. The optimal concentrations of substrate and enzyme were found to be $2\%$ inulin and 40 unit/ g inulin, respectively. At optimum condition, 9.5 g/l of maximum yield and $47.5\%$ of conversion efficacy were achieved. For purification of CF produced, the reaction mixture was treated with 1 unit/ml exoinulinase and then added $3\%$ CaO three times with blowing $CO_2$ gas, resulted in $95\%$ purity.

  • Trehalose is a non-reducing disaccharide in increasing demand for applications in food, nutraceutical, and pharmaceutical industries. Single-step trehalose production by trehalose synthase (TreS) using maltose as a starting material is a promising alternative process for industrial application due to its simplicity and cost advantage. Pseudomonas monteilii TBRC 1196 was identified using the developed screening method as a potent strain for TreS production. The TreS gene from P. monteilii TBRC 1196 was first cloned and expressed in Escherichia coli. Purified recombinant trehalose synthase (PmTreS) had a molecular weight of 76 kDa and showed optimal pH and temperature at 9.0 and 40℃, respectively. The enzyme exhibited >90% residual activity under mesophilic condition under a broad pH range of 7-10 for 6 h. Maximum trehalose yield by PmTreS was 68.1% with low yield of glucose (4%) as a byproduct under optimal conditions, equivalent to productivity of 4.5 g/l/h using enzyme loading of 2 mg/g substrate and high concentration maltose solution (100 g/l) in a lab-scale bioreactor. The enzyme represents a potent biocatalyst for energy-saving trehalose production with potential for inhibiting microbial contamination by alkaline condition.

Joint Production and Disposal Decisions for Sustainable Operations of the Hybrid Production System (혼성 생산 시스템의 지속 가능 운영을 위한 신제품 생산과 회수제품 수용 통제의 통합 구현)

  • We consider a reverse supply chain with a production facility and a recovery facility, and address the joint control of production and disposal decisions for sustainable operations. Demands are satisfied from on-hand inventory of serviceable products, replenished via manufacturing or remanufacturing. Sold products may be returned after usage and each returned product is disposed of or accepted for recovery. Accepted returned products are converted into serviceable products after remanufacturing process. Formulating the model as a Markov decision process, we characterized the structure of the optimal production and disposal policy as two monotone switching curves under a special condition. Three types of heuristic policies are presented and their performance is numerically compared.

  • Streptomyces themocyaneovio-laceus producing the thermostable xylanase was used for the production of xylooligosaccharides from xylan. The optimal conditions for the xylanase production were investigated in jar fermentor, which operated at 2 vvm aera-tion and 400 rpm agitation speed at $50^{\circ}C$ for 24 h. The optimal reaction condtion for the production of xylooli-gosaccharides with xylanases which were prepared by the percipitation with ammonium sulfate were obtained by the reaction at $60^{\circ}C$ for 12 h in the mixture composed of 10% birchwood xylan in 50 mM sodium phosphate buffer (pH 6.0)and 10 unit/ml of xylanase. In this optimal condition for the xylooligosaccharides production the mixture of xylooligosaccharides (58.8 g/I) which were composed of 20.1 g/I of xyobiose, 8.9 g/I of xylotriose 4.5 g/I of xylotetraose 16.2g/I of xylopentaose and 9.1 g/I xylohexaose and 5.0 g/I of xylose was produced from 100 g/I of birchwood xylan by the xylanases of S thermocyaneoviolaceus .

Media Optimization of Corynebacterium glutamicum for Succinate Production Under Oxygen-Deprived Condition

  • Corynebacterium glutamicum is one of the well-studied industrial strain that is used for the production of nucleotides and amino acids. Recently, it has also been studied as a possible producer of organic acids such as succinic acid, based on its ability to produce organic acids under an oxygen deprivation condition. In this study, we conducted the optimization of medium components for improved succinate production from C. glutamicum under an oxygen deprivation condition by Plackett-Burman design and applied a response surface methodology. A Plackett-Burman design for ten factors such as glucose, ammonium sulfate, magnesium sulfate, potassium phosphate ($K_2HPO_4$ and $KH_2PO_4$), iron sulfate, manganese sulfate, biotin, thiamine, and sodium bicarbonate was applied to evaluate the effects on succinate production. Glucose, ammonium sulfate, magnesium sulfate, and dipotassium phosphate were found to have significant influence on succinate production, and the optimal concentrations of these four factors were sequentially investigated by the response surface methodology using a Box-Behnken design. The optimal medium components obtained for achieving maximum concentration of succinic acid were as follows: glucose 10 g/l, magnesium sulfate 0.5 g/l, dipotassium phosphate ($K_2HPO_4$) 0.75 g/l, potassium dihydrogen phosphate ($KH_2PO_4$) 0.5 g/l, iron sulfate 6 mg/l, manganese sulfate 4.2 mg/l, biotin 0.2 mg/l, thiamine 0.2 mg/l, and sodium bicarbonate 100 mM. The parameters that differed from a normal BT medium were glucose changed from 40 g/l to 10 g/l, dipotassium phosphate ($K_2HPO_4$) 0.5 g/l changed to 0.75 g/l, and ammonium sulfate ($(NH_4)_2SO_4$) 7 g/l changed to 0 g/l. Under these conditions, the final succinic acid concentration was 16.3 mM, which is about 1.46 fold higher than the original medium (11.1 mM) at 24 h. This work showed the improvement of succinate production by a simple change of media components deduced from sequential optimization.

  • Recently Neural Network techniques have widely used in adaptive and learning control schemes for production systems. However, generally it costs a lot of time for learning in the case applied in control system. Furthermore, the physical meaning of neural networks constructed as a result is not obvious. And in practice since it is difficult to the PID gains suitably lots of researches have been reported with respect to turning schemes of PID gains. A Neural Network-based PID control scheme is proposed, which extracts skills of human experts as PID gains. This controller is designed by using three-layered neural networks. The effectiveness of the proposed Neural Network-based PID control scheme is investigated through an application for a production control system. This control method can enable a plant to operate smoothy and obviously as the plant condition varies with any unexpected accident.
