Development of an Algorithm for Searching Optimal Temperature Setpoint for Lettuce in Greenhouse Using Crop Growth Model (작물생장모델을 이용한 상추의 온실 최적설정온도 탐색 알고리즘의 개발)

  • This study was conducted to develop a searching algorithm for optimal daily temperature setpoint greenhouse. An algorithm using crop growth and energy models was developed to determine optimum crop growth environment. The results of this study were as follows: 1. Mathematical models for crop growth and energy consumption were derived to define optimal daily temperature setpoint. 2. Optimum temperature setpoint, which could maximize performance criterion, was determined by using Pontryagin maximum principle. 3. Dynamic control of daily temperature using the developed algorithm showed higher performance criterion than static control with fixed temperature setpoint. Performance criteria for dynamic control models were with simulated periodic weather data and with real weather data, increased by 48% and 60%, respectively.

Photosynthesis and Growth Responses of Soybean (Glycine max Merr.) under Elevated CO2 Conditions (대기 중 CO2 상승 조건에서 재배되는 콩의 광합성과 생장 반응의 분석)

  • The effects of elevated atmospheric $CO_2$ on growth and photosynthesis of soybean (Glycine max Merr.) were investigated to predict its productivity under elevated $CO_2$ levels in the future. Soybean grown for 6 weeks showed significant increase in vegetative growth, based on plant height, leaf characteristics (area, length, and width), and the SPAD-502 chlorophyll meter value (SPAD value) under elevated $CO_2$ conditions ($800{\mu}mol/mol$) compared to ambient $CO_2$ conditions ($400{\mu}mol/mol$). Under elevated $CO_2$ conditions, the photosynthetic rate (A) increased although photosystem II (PS II) photochemical activity ($F_v/F_m$) decreased. The maximum photosynthetic rate ($A_{max}$) was higher under elevated $CO_2$ conditions than under ambient $CO_2$ conditions, whereas the maximum electron transport rate ($J_{max}$) was lower under elevated $CO_2$ conditions compared to ambient $CO_2$ conditions. The optimal temperature for photosynthesis shifted significantly by approximately $3^{\circ}C$ under the elevated $CO_2$ conditions. With the increase in temperature, the photosynthetic rate increased below the optimal temperature (approximately $30^{\circ}C$) and decreased above the optimal temperature, whereas the dark respiration rate ($R_d$) increased continuously regardless of the optimal temperature. The difference in photosynthetic rate between ambient and elevated $CO_2$ conditions was greatest near the optimal temperature. These results indicate that future increases in $CO_2$ will increase productivity by increasing the photosynthetic rate, although it may cause damage to the PS II reaction center as suggested by decreases in $F_v/F_m$, in soybean.

  • Plastic film houses are directly associated with increases in plant growth and yield of vegetable crops through a year round cultivation, however, at the same time temperature stresses are one of fates which are difficult to avoid during crop growth. The objective of this study was to examine the translocation and distribution of minerals (N, P, K) and carbohydrates as well as seasonal fluctuation of mineral uptake and carbohydrate production in cucumber plant grown under moderately high temperature. The temperature treatments consisted of 2-layers film houses (optimal temp.) and 3-layers (high temp.). Shoot growth of cucumber plants were linearly increased until 14 weeks after transplanting (WAT) without any significant difference between both temperatures, and the slowdown was observed from 16 WAT. The level of soluble sugar and starch was slightly greater in optimal temperature compared to the high. Cumulative accumulation of soluble sugar was significantly different before and after 12 WAT in both treatments, whereas starch level represented a constant increase. Monthly production of soluble sugar reached the peak between 12 to 16 WAT, and starch peaked between 4 to 8 WAT and 12 to 16 WAT. Total uptake of N, P and K in optimal and high temperature conditions was $18.4g\;plant^{-1}$ and 17.6 for N, 4.7 and 5.1 for P, and 37.7 and 36.2 for K, respectively, and the pattern of monthly N uptake between optimal and high temperatures was greater in early growth stage, whereas was greater in mid growth stage in both P and K. Thus, this study suggests that moderately high temperature influences much greater to photosynthesis and carbohydrate production than plant biomass and mineral uptake. On the basis of the present result, it is required to indentify analysis of respiration rates from plant and soil by constantly increasing temperature conditions and field studies where elevated temperatures are monitored and manipulated.

  • For polyhydroxyalkanoic acid (PHA) production, several microorganisms were isolated from sewage sludge. One of them, GB-77 strain, was chosen from its PHB/HV copolymer production on only fructose without cosubstrate. The isolated strain GB-77 was identified as the genus Alcaligenes. Optimal temperature and pH for cell growth were 36C and 6.8. Optimal medium composition was 10 g/l of fructose and 5 g/l of polypeptone, 1 $\times$ 10$^{-2}$M Na$^{2}$HP0$^{4}$, 1.3 $\times$ 10$^{-2}$M KH$^{2}$PO$^{4}$. To investigate the optimal condition for polyhydroxyalkanoic acid production two-stage culture technique was used; first stage for cell growth and second stage for PHA production on unbalanced growth conditions. Optimal conditions for high PHA production were C/N ratio 50, temperature 36$\circ$C and pH 6.8. To overcome fructose inhibition on cell growth, intermittent feeding fed-batch culture technique was used. Total cell concentration was 17.4 g/l with 9.1 g/l of PHA. The purified PHA was identified PHB/HV copolymer by NMR analysis.

A Study on the Cultural Characteristics of Pholiota nameko Mycelium (맛버섯 균사체의 배양 특성에 관한 연구)

  • This Study was carried out to investigate the optimal mycelial growth of Pholiota nameko. The optimal medium for the mycelial growth was ME medium. The optimal temperature and pH were $25^{\circ}C$$\pm$1 and 5.5, respectively. The modified optimal medium compositions were glucose 3% (w/v), malt extract 0.25% (w/v), yeast extract 0.25% (w/v), $KH_2PO_4$ 0.046% (w/v), $K_2HPO_4$ 0.1% (w/v), $MgSO_4$$7H_2O$ 0.05% (w/v). From the result of experiments on the optimal temperature, pH and nutritional requirements, the mycelial growth of modified optimal medium was higher than that of ME medium.

Xylanase Production by Bacillus sp. A-6 Isolated from Rice Bran

  • A Bacillus sp. A-6 strain that produced xylanase was isolated from rice bran. The optimal temperature and pH for xylanase activity of the culture supernatant of Bacillus sp. A-6 were 40$^{\circ}C$ and pH 7, respectively. The optimal temperature and pH for xylanase production in the xylan medium were 30$^{\circ}C$ and pH 9, respectively. The optimal concentrations of oat spelt xylan and peptone for xylanase production were 0.5% and 1.5%, respectively. The best nitrogen sources for xylanase production was beef extract, but xylanase production was also supported comparably by tryptone and peptone. The bacterial growth in the optimal xylan medium reached stationary growth phase after 12 h of incubation. The xylanase production in the culture supernatant increased dramatically during the initial 12 h exponential growth phase and then remained constant at 23.8-24.5 unit/ml during the stationary growth phase. The pH of the culture medium decreased from 8.8 to 6.7 during the exponential growth phase and subsequently increased to 8.1 during the stationary growth phase. Rice bran, sorghum bran, and wheat bran as well as oat spelt xylan induced xylanase production. The xylanase production was repressed when glucose was added to the xylan-containing medium.

  • To elucidate the optimal conditions of mycelial growth of Pholiota adiposa, the effect of a kinds of media, incubation temperature and shaking culture were investigated. Mushroom complete medium was selected as superior media for mycelial growth in this fungus. The optimal temperature for mycelial growth of this fungus shows at 25℃. Shaking culture could be obtain the mycelia in two or three times than the stationary culture.

Growth Characteristics and Plasmid Profiles of Yersinia enterocolitica lsolated from Springs Water (약수터수로부터 분리한 Yersinia enterocolitica의 성장특성 및 Plasmid 유형)

  • The studies were conducted to explore the dffects of growth or survival against various factors and plasmid profiles of 49 Y. enterocolitica isolated from springs water. In the presence of calcium hypochlorite, y. enterocolitica was entirely extinguished by exposure for 33 hours at 0.8 ppm concentration, and was grown up to 7% NaCl, but not at 95 NaCI. Y. enterocolitica was presented optimal growth at pH 7.0 anad 9.0, and not allowth the growth at pH3.0, 5.0 and 11.0. The optimal temperature for growth of Y. enterocolitica was 25$\circ$C and 35$\circ$C, and allowed the growth at refrigerant temperature, 5$\circ$C. Y. enterocolitica was remarkably decreased by exposure for 30 seconds under UV light, and entirely extinguished by exposure for 90 seconds. Therefore, UV light was effective for sterilization of Y. enterocolitica. Fourty-nine strains of Y. enterocolitica were harbor plasmid DNA of approximately 46 Kb molecular weight.

Effects of Temperature and Culture Media Composition on Sporulation, Mycelial Growth, and Antifungal Activity of Isaria javanica pf185

  • The fungal isolate Isaria javanica pf185 has potential as a mycopesticide because it demonstrates insecticidal activity against the green peach aphid and antifungal activity against Colletotrichum gloeosporioides. For commercialization of this isolate, determination of the optimal and least expensive culture conditions is required; however, these data are not currently available. This study describes the conditions for optimal development of conidia and production of metabolites for the biocontrol of the fungal pathogen. The optimal culture conditions were examined using cultures on solid agar and liquid media. High growth temperature enhanced spore formation but reduced antifungal activity in both solid and liquid media. The highest spore yield was obtained in a medium containing glucose as a carbon source and yeast extract as a nitrogen source. Soybean powder and wheat bran were effective nitrogen sources that promoted spore production and antifungal activity of the isolate. These results revealed the basic, cost-effective growth media for commercial production of a biopesticide with insecticidal and antifungal properties for use in integrated pest management.

Effect of Climate Change for Diatom Bloom at Winter and Spring Season in Mulgeum Station of the Nakdong River, South Korea (낙동강 물금 지점의 겨울 및 봄철 식물플랑크톤 생물량에 대한 기후변화 영향)

  • To confirm the relationship between climate change and Stephanodiscus in Mulgeum station of Nakdong River, Korea, this study was conducted. The temperature in crease by climate change was observed in the study site, where the temperature was gradually increased in most seasons, except for summer season. The mass proliferation of Stephanodiscus constantly appeared in every year, especially between November and March, and when Stephanodiscus abundance was above 90% in phytoplankton biomass. Among this period, phytoplankton biomass was high related with water temperature ($r^2$=0.249, P<0.01) than nutrient factors such as nitrogen and phosphorus in the study site. Finally, temperature by climate change can be regarded as the affecting factor for chl. a variation, because temperature was strongly related with water temperature ($r^2$=0.748, P<0.01). From 1997 to 2010, the annual maximum phytoplankton biomass was recorded in the range of temperature from $4.8^{\circ}C$ to $8.4^{\circ}C$, and the range was regarded as the temperature condition for the optimal growth of Stephanodiscus in the study site. On the optimal growth temperature, the trend of monthly average temperature corresponded to the trend of chl. a variation from November to March. In future, the increase of temperature by climate change can prolong Stephanodiscus blooming period in winter and spring seasons.
