Quantitative Real-time PCR using Lactobacilli as Livestock Probiotics (Real-time PCR을 이용한 가축생균제용 유산균 정량분석)

  • This study was conducted using quantitative real-time PCR using Lactobacilli as probiotics. Quantitative real-time PCR (RT PCR) was conducted via a method involving SYBR Green 1 and a probe. Plasmid DNA was cloned using the 16S-23S rRNA intergenic species region. Gene clones were diluted from $10^2$ to $10^{10}$. Standard curves were constructed via Ct values obtained from the results of Real-time PCR via the aforementioned SYBR Green 1 and probe method. Plasmid DNA was also cloned using the 16S-23S rRNA intergenic species region and the gene clones were diluted from $10^2$ to $10^{10}$ copy numbers via the probe method. Using RT PCR, a standard curve of plasmid DNA copy numbers was also determined. The slope value for the Y-axis intercept and $R^2$ value were measured as -3.346, 33.18, and 0.993, respectively, via the first method. For the second method, the slope value for the Y-axis intercept and $R^2$ were -3.321, 31.10 and 0.995, respectively. The PCR inhibitor could not express the detection curve at a copy number over $10^{10}$ via either method, owing to high DNA density. The DNA extract from probiotics was diluted without pre-culturing, and 16 products were amplified via both methods. The Ct value was 11.06~18.12 in the first method and 16.74~22.11 in the second method. Measured probiotics and log copy values were largely similar among the methods used. It was concluded that both methods are effective for analysis, but further research will be required to verify the optimal method.

A Study on Left-turn Queues Analysis using Queueing Theory under Permissive Left-turn Signal System (비보호좌회전 신호체계운영에 따른 좌회전 대기행렬분석에 관한 연구)

  • In this study, the optimal length of left-turn lane in permissive left-turn signal system at the signalized intersection which has a left-turn bay is estimated. It is a simulation analysis using the queueing theory that estimate the length of left-turn lane. Traffic density conform to the standards of operating a permissive left-turn system of the Practical Manual Traffic Safety Facilities. And each of a left-turn arrival rate, a left-turn service rate, left-turn average queueing time, for green time average queueing vehicle, for red time average queueing vehicle and average queueing vehicle cycle is calculated. As a result of this study, we would learn how much the space should be secured at the signalized intersection which has a left-turn bay. The methodology using the queueing theory to work out the optimal length of waiting lane in the permissive left-turn signal system was presented.

Design of Midongsan Ecological park (미동산 산림환경 생태원 설계)

  • This report describes the first prize design of Midongsan Eco-park, which is located in Chungcheongbuk-do (North Chungcheong) province. Criteria of the winning design are as follows: 1) The design should include a neighborhood park as well as a portion of a green zone, in order to harmonize the area ecosystem, gradually assimilate the surroundings, and serve as an education tool for the public. 2) According to an analysis of area maps and a site-inspection, a preserved area, a buffer area, a restored area, and a developed area should be developed. In the buffer and restored areas, major facilities and activity spaces should be set up. 3) Observation trails should be accessible to visitors, and a variety of adventure programs should be available. 4) The eco-park should be designed to provide a habitat for the Gorani (Chinese water deer) which used to live around Midongsan. 5) For sensuous and vivid visitor experiences, diverse programs are to be designed to accommodate different seasons, visitor ages, and required time. Environment and eco-education are to be emphasized to maximize the effect of the eco-programs. 6) Optimal activities for visitors of different ages, optimal time intervals and eco-programs by age are also suggested. 7) Facilities are zoned into three sections according to level of development. Each section has a main area and supplementary areas for better understanding. 8) Nature-friendly arrangement of facilities and construction should minimize earthwork and adverse effects on the local ecology. Throughout the planning, the following things were learned: longer-term analysis including all season's observation is essential; planning, designing, bui Iding and managing should be based on this careful, extended observation. The contest should have taken plate over a longer period of time in order enable more detailed planing of the eco-park For improved management and activities, a volunteer system in linked with schools eco-institutions and NGOs should be developed with eco-interpreters.

A Study on Bicycle Route Selection Considering Topographical Characteristics (지형적 특성을 고려한 자전거 경로 선정에 관한 연구)

  • As interest in green growth picks up, the importance of bicycles which are an environment friendly means of communication has been magnified. However, bicycle routes which are the base of bicycles are designed without considering topographic elements and thus many users are experiencing inconvenience in using bicycles. The present study presents a routing technique to select optimal routes when selecting routes in commuting to school utilizing bicycles. To this end, a formula for optimum route calculation considering slope and intersections was drawn and a method to select optimum routes by applying modified Dijkstra Algorithms was studied. According to the results, the bicycle routes for commuting to school should be selected by the shortest time rather than the shortest distances to the destination, because it required reach the destination faster. Therefore when selecting the routes, it must be based on the shortest time considering waiting time due to crosswalks or crossroads and speed variations due to slopes.

Potential Impact of Timber Supply and Fuel-Wood on the Atmospheric Carbon Mitigation : A Carbon Cycle Modeling Approach (목재공급과 연료용 목재가 대기에 축적된 탄소저감에 미치는 잠재적 영향 : 탄소순환모형 접근법)

  • There is general agreement that global warming is occurring and that the main contributor to this probably is the buildup of green house gasses, GHG, in the atmosphere. Two main contributors are the utilization of fossil fuels and the deforestation of many regions of the world. The burning of fossil fuels increases atmospheric carbon while the burning of fuel-wood reducing fossil fuel consumption along with its forest source maintain an atmospheric carbon level. The standing timber in the forests is a carbon sink, as are wood buildings and structures, and fossil fuel in the ground. This paper is designed to examine a number of current issues related to mitigating the global warming problem through forestry. For this purpose, we develop a modeling approach by integrating timber market, fossil fuel market and carbon cycling model. We use discrete time optimal control theory to identify optimal time paths, the laws of motion, and stationary stats solutions of endogenous variables in the model. On the basis of these results, we identify the optimal amounts of subsidies to be provided or taxes to be imposed by the regulatory agency to mitigate atmospheric carbon accumulation. We also present a numerical example to help illustrate the characteristics of variables in the model when the social cost for atmospheric carbon incrementally shifts upward. A surprising result is that the social cost function for atmospheric carbon has a very smaller impact on the optimal rotation period than previous literature suggested.

Development of Miniaturized Culture Systems for Large Screening of Mycelial Fungal Cells of Aspergillus terreus Producing Itaconic Acid

  • The task of improving a fungal strain is highly time-consuming due to the requirement of a large number of flasks in order to obtain a library with enough diversity. In addition, fermentations (particularly those for fungal cells) are typically performed in high-volume (100-250 ml) shake-flasks. In this study, for large and rapid screening of itaconic acid (IA) high-yielding mutants of Aspergillus terreus, a miniaturized culture method was developed using 12-well and 24-well microtiter plates (MTPs, working volume = 1-2 ml). These miniaturized MTP fermentations were successful, only when highly filamentous forms were induced in the growth cultures. Under these conditions, loose-pelleted morphologies of optimum sizes (less than 0.5 mm in diameter) were casually induced in the MTP production cultures, which turned out to be the prerequisite for the active IA biosynthesis by the mutated strains in the miniaturized fermentations. Another crucial factor for successful MTP fermentation was to supply an optimal amount of dissolved oxygen into the fermentation broth through increasing the agitation speed (240 rpm) and reducing the working volume (1 ml) of each 24-well microtiter plate. Notably, almost identical fermentation physiologies resulted in the 250 ml shake-flasks, as well as in the 12-well and 24-well MTP cultures conducted under the respective optimum conditions, as expressed in terms of the distribution of IA productivity of each mutant. These results reveal that MTP cultures could be considered as viable alternatives for the labor-intensive shake-flask fermentations even for filamentous fungal cells, leading to the rapid development of IA high-yield mutant strains.

Application of Three-phase Hollow Fiber LPME using an Ionic Liquid as Supported Phase for Preconcentration of Malachite Green from Water Samples with HPLC Detection

  • A novel three-phase hollow fiber liquid phase microextraction was developed for the determination of malachite green (MG) in environmental waters, which selected [BMIM][$PF_6$] mixed with 1% trioctylphosphine oxide (TOPO) as supported phase. Several parameters (accepter phase pH, sample pH, supported phase membrane, volume of accepter phase, salinity, extraction time) that could affect extraction performance were investigated. Under the optimal extraction conditions, the established approach showed excellent characters as: high enrichment factor (212), wide linear range ($0.20-100{\mu}gL^{-1}$), low detection limit ($0.01{\mu}gL^{-1}$), good reproducibility (RSD, 8.9%, n=5) and satisfactory recovery (84.0-106.2%). The method was applied to detect MG at Yangtze River and pond waters in Zhenjiang, Jiangsu province, and 4 sites among 15 sampling sites were found MG with the concentration of $1.73-11.06{\mu}gL^{-1}$, which confirmed that the proposed environmentally friendly method was simple and effective for monitoring MG in aquatic system.

Effects of Green Tea Extract on Sperm Quality, Reactive Oxygen Species and Lipid Peroxidation in Long-term Liquid Preservation of Boar Spermatozoa

  • During storage, boar spermatozoa undergo several changes including diminished motility and viability and accumulated reactive oxygen species (ROS). In this study, we investigated the effects of green tea extract (GTE) supplementation in the Sui Dil extender on the sperm motility, viability, ROS and lipid peroxidation (LPO) of long-term preserved boar semen at $17^{\circ}C$. A total number of eight boars were used for this experiment. Pooled ejaculates were diluted to $20{\times}10^6sperm/ml$ in the Sui Dil extender containing 0 (control), 1, 10, 100 or 500 mg/l GTE and were preserved at $17^{\circ}C$ for 24, 72, 120 and 168 h, respectively. At each storage time, sperm motility and viability were estimated by microscopic examination and the fluorescent double stain $Fertilight^{(R)}$, respectively. Sperm ROS level and LPO were assessed using the 2', 7'-dichlorodihydrofluorescein diacetate ($H_2DCFDA$)/propidium iodide (PI) and C11-BODIPY581/591/PI with flow cytometry, respectively. Compared to that of the 500 mg group, there were higher sperm motility and viability in the 1, 10 and 100 mg GTE groups during the preservation from 24 to 168 h (p < 0.05). The ROS levels of the 10 and 100 mg groups during the 168 h preservation were lower than those of the 0, 1 and 500 mg groups (p < 0.05). There were no significant differences in LPO regardless of the preservation period or the GTE concentration. In conclusion, the optimal concentrations (10 and 100 mg/l) of GTE that led to lower ROS levels may be useful for liquid boar sperm preservation at $17^{\circ}C$ for a period of 168 h.

  • A GA is one of the best method to find optimal value in searching area. A GA is driven by probabilistic selection that based on the survival of the fittest. So this algorithm need a huge solving time even if it can be used lots of optimizing problem such as structural design, machine learning, system's identification and so on. This GA's characteristic constrain the program to drive offline. Some studies try to use this algorithm on online or reduce the GA's running time with parallel GA or micro GA. Unfortunately these studies still didn't reduce amount of fitness solving. If the chromosome was imported to the system, it affected system's stability. And when the control system uses online GA, it also doesn't have enough learning time. In this study, try to find stability criterion to reduce the chromosome's affection and find the characteristic of the number of population and generation when GA was driven into the wide searching area.

An analysis of the Wi-Ni Carbide Alloy Diffusion Bonding technique in its application for DME Engine Fuel Pump

  • Dimethyl Ether(DME) engine use a highly efficient alternative fuel having a great quantity of oxygen and has a advantage no polluting PM gas. The existing DME fuel cam material is a highly expensive carbide alloy, and it is difficult to take a price advantage. Therefore the study of replacing body area with inexpensive steel material excluding piston shoe and contact area which demands high characteristics is needed. The development of WC-Ni base carbide alloy optimal bonding composition technique was accomplished in this study. To check out the influence of bonding temperature and time, bonding characteristics of sintering temperature was experimented. The hardness of specimen and bonding rate were measured using ultrasound equipment. The bonding state of each condition was excellent, and the thickness of mid-layer, temperature and maintaining time were measured. The mid-layer thickness according to bonding temperature and maintaining time were observed with optical microscope. We analyzed the micro-structural analysis, formation of bonding specimen, wafer fabrication and fuel cam abrasion test. Throughout this study, we confirmed that the fuel cam for DME engine which demands high durability against velocity and pressure is excellent.
