HAZ TOUGHNESS AND MICROSTRUCTURE IN HIGH NITROGEN AUSTENITIC STAINLESS STEEL

  • HAZ(Heat Affected Zone of weldm ents) properties were investigated for a high nitrogen austenitic stainless steel with a chemical composition of Fe-0.02C-0.15Si-6.00Mn-10.0Ni-23.0Cr-2.00Mo-0.48N-0.14V. Thermal cycle of HAZ was simulated by the thermal cycle simulator (Gleeble 1500). The heat treatment was applied to the Charpy test size sample without notch under various peak temperatures and/or the holding times condition. V-notch Charpy test was performed at the temperature range of 273~77 K. Metallographic examination also was carried out by using optical microscopy, scanning electron microscopy and transmission electron microscopy. The simulated specimens revealed a slight embrittlement compared with the base materials. The impact toughness of the specimens deteriorated with the decreasing test temperature. The results from Charpy V-notch test, however, showed that significant degradation of absorbed energy caused by brittle fracture was not observed for the specimen tested in the test temperature range.

Property of the HPHT Diamonds Using Stack Cell and Zn Coating with Pressure (적층형 셀과 아연도금층을 이용한 고온고압 합성다이아몬드의 압력변화에 따른 물성 연구)

  • Fine diamond powders are synthesized with a 420 ${\phi}$ cubic press and stack-cell composed of Kovar ($Fe_{54}Ni_{29}Co_{17}$) (or Kovar+7 ${\mu}m$-thick Zn electroplated) alloy and graphite disks. The high pressure high temperature (HPHT) process condition was executed at $1500^{\circ}C$ for 280 seconds by varying the nominal pressure of 5.7~10.6 GPa. The density of formation, size, shape, and phase of diamonds are determined by optical microscopy, field emission scanning electron microscopy, thermal gravimetric analysis-differential thermal ammnlysis (TGA-DTA), X-ray diffraction (XRD), and micro-Raman spectroscopy. Through the microscopy analyses, we found that 1.5 ${\mu}m$ super-fine tetrahedral diamonds were synthesized for Zn coated Kovar cell with whole range of pressure while ~3 ${\mu}m$ super-fine diamond for conventional Kovar cell with < 10.6 GPa. Based on $750^{\circ}C$ exothermic reaction of diamonds in TGA-DTA, and characteristic peaks of the diamonds in XRD and micro-Raman analysis, we could confirm that the diamonds were successfully formed with the whole pressure range in this research. Finally, we propose a new process for super-fine diamonds by lowering the pressure condition and employing Zn electroplated Kovar disks.

Recent advances in intravital microscopy for investigation of dynamic cellular behavior in vivo

  • Currently, most biological research relies on conventional experimental techniques that allow only static analyses at certain time points in vitro or ex vivo. However, if one could visualize cellular dynamics in living organisms, that would provide a unique opportunity to study key biological phenomena in vivo. Intravital microscopy (IVM) encompasses diverse optical systems for direct viewing of objects, including biological structures and individual cells in live animals. With the current development of devices and techniques, IVM addresses important questions in various fields of biological and biomedical sciences. In this mini-review, we provide a general introduction to IVM and examples of recent applications in the field of immunology, oncology, and vascular biology. We also introduce an advanced type of IVM, dubbed real-time IVM, equipped with video-rate resonant scanning. Since the realt-ime IVM can render cellular dynamics with high temporal resolution in vivo, it allows visualization and analysis of rapid biological processes.

Development of line-scanning two-photon microscopy based on spatial and temporal focusing for tryptophan based auto fluorescence imaging (고속 트립토판 자가형광 이미징을 위한 시공간적 집중 기반의 라인 스캐닝 이광자 현미경 개발)

  • Two-photon microscopy (TPM) is minimally-invasive 3D fluorescence microscopy based on nonlinear excitation, and TPM can visualize cellular structures based on auto-fluorescence. Line-scanning TPM is one of high-speed TPM methods without sacrificing the image resolution by using spatial and temporal focusing. In this paper, we developed line-scanning TPM based on spatial and temporal focusing for auto-fluorescence imaging by exciting the tryptophan. Laser source for this system was an optical parametric oscillator (OPO) and it made near 570 nm femtosecond pulse laser. It had 200fs pulse width and 1.72 nm bandwidth, so that the achievable depth resolution was 2.41um and field of view (FOV) is 10.8um. From the characterization, our system has 3.0 um depth resolution and 12.3 um FOV. We visualized fixed leukocyte cell sample and compared with point scanning system.

Formation of Ultrafine Grains in Cu-Fe-P Alloy by Accumulative Roll-Bonding Process (ARB법에 의한 Cu-Fe-P합금의 초미세결정립 형성)

  • A Cu-Fe-P copper alloy was processed by accumulative roll-bonding (ARB) for ultra grain refinement and high strengthening. Two 1mm thick copper sheets, 30 mm wide and 300 mm long, were first degreased and wire-brushed for sound bonding. The sheets were then stacked on top of each other and roll-bonded by about 50% reduction rolling without lubrication at ambient temperature. The bonded sheet was then cut into two pieces of the same dimensions and the same procedure was repeated for the sheets up to eight cycles. Microstructural evolution of the copper alloy with the number of the ARB cycles was investigated by optical microscopy (OM), transmission electron microscopy(TEM), and electron back scatter diffraction(EBSD). The grain size decreased gradually with the number of ARB cycles, and was reduced to 290 nm after eight cycles. The boundaries above 60% of ultrafine grains formed exhibited high angle boundaries above 15 degrees. In addition, the average misorientation angle of ultrafine grains was 30 degrees.

  • Precipitation characteristics of the Fe-36Ni based high strength Invar alloy for power transmission wire was investigated in this study. High strength can be obtained in this alloy through solution hardening, precipitation hardening and strain hardening by cold working. In the present study, ingots of Fe-36Ni based Invar alloys with the contents of C, Mo and V varied. Microstructure observations by OM, SEM, and TEM were carried out to validate the simulation results. BCC phase and $FeNi_3$ phase are also expected at lower temperatures below $500^{\circ}C$. Aging treatments were carried out at temperatures ranging from 400 to $900^{\circ}C$ for time intervals from 3 min to 100hrs. Peak aging condition was obtained as $400^{\circ}C$ and 1 hr. With temperature increased, peak strength was decreased abruptly. Microstructure observation was conducted by optical microscopy, scanning electron microscopy, and transmission electron microscopy.

Micro-defects in $LiNbO_3$ single crystals with congruent melting composition (조화용융성조성을 가진 $LiNbO_3$ 단결성의 미소결함)

  • Micro-defects in the undoped and MgO-doped $LiNbO_3$ single crystals, which were grown from a congruent melting composition (48.6 mol% $Li_2$O) by the CZ (Czochralski) method, were analyzed using microscopic techniques such as optical microscopy (OM), scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Effects of the dopant and g value (the fraction solidfied) on the domain structure and micro-defect were investigated to build a corelationship with a growth condition of crystal. It was observed that the micro-defect concentrated near the domain wall is caused by high stress. Especially, the micro-defect was observed to be biased toward a certain side of the domain wall.

  • 3-D visualization using confocal laser scanning microscopy (CLSM) in a chaotic micromixer was performed as a reproduction experiment and the feasibility of 3-0 imaging technique in the microscale was confirmed. For diagonal micromixer (DM) and two types of staggered herringbone micromixers (SHM) designed by Whitesides et al., to verify the evolution of mixing, cross sectional images are reconstructed at the end of every cycle. In a DM, clockwise rotational flow motion generated by diagonal ridges placed on the floor of micromixer is observed and this motion makes the fluid commingle. On the contrary, there are two rotational flow structures in the SHM and the centers of rotation exchange their position each other every half cycle because of the V shape of ridges varying their orientation every half cycle. Local rotational flow and local extensional flow generated by the complicate ridge pattern make the flow be chaotic and accelerate the mixing of fluid. The dominant parameter that influences on the mixing characteristic of SHM is not the length of micromixer but the number of ridges under the same flow configurations.

Synthesis and Characterization of Rutile TiO2 Powder by the Sulfuric Acid Method (황산법을 이용한 루틸형 TiO2 분말의 제조 및 특성 평가)

  • We investigated the experimental method that uses the homogeneous precipitation method to prepare mica flakes-coated rutile-type titania pearlescent pigment with urea as a precipitant. $TiO_2$ particles exhibit a high reflection of lights and optical properties with chemical stabilities, so they are appropriate for coating on luminescent pigments (mica). The coating principle of mi ca coated titania with various thicknesses was analyzed by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM) and tested by spectrophotometer. Mica with a particle size in the range of $40-60{\mu}m$ was suspended in water, and metal sulphates and urea were added to the mixture, which was heated to boiling. The change in pH was continuously followed. The metal oxide and crystal structure were affected by the conditions of $TiOSO_4$ concentration and reaction time with a sintering temperature the range of $800-1100^{\circ}C$.

Simultaneous Estimation of Spatial Frequency and Phase Based on an Improved Component Cross-Correlation Algorithm for Structured Illumination Microscopy

  • Accurate estimation of spatial frequencies and phases for illumination patterns are essential to reconstructing super-resolution images in structured illumination microscopy (SIM). In this manuscript, we propose the improved component cross-correlation (ICC) algorithm, which is based on optimization of the cross-correlation values of the overlapping information between various spectral components. Compared to other algorithms for spatial-frequency and phase determination, the results calculated by the ICC algorithm are more accurate when the modulation depths of the illumination patterns are low. Moreover, the ICC algorithm is able to calculate the spatial frequencies and phases simultaneously. Simulation results indicate that even if the modulation depth is lower than 0.1, the ICC algorithm still estimates the parameters precisely; the images reconstructed by the ICC algorithm are much clearer than those reconstructed by other algorithms. In experiments, our home-built SIM system was used to image bovine pulmonary artery endothelial (BPAE) cells. Drawing support from the ICC algorithm, super-resolution images were reconstructed without artifacts.
