A SCANNING ELECTRON MICROSCOPIC STUDY ON THE ADAPTATION OF ROOT CANAL FILLING MATERIAL TO ROOT CANAL WALL WITH AND WITHOUT SMEAR LAYER (Gutta percha 충전시 도말층 유무에 따른 근관벽과의 접합도에 관한 주사전자현미경적 연구)

  • The purpose of this study was to evaluate the adaptation of filling material to the dentinal walls of root canals with and without smear layer. Fifty extracted upper and lower anterior teeth were selected, and the root canals were instrumented with K - files 1mm short of the apical foramen by step - back method. The teeth were randomly divided into two groups of 25 each : in the group I, smear layers were not removed, and in the group II, smear layers were removed by 15% EDTA solution. Again the two groups 25 teeth were randomly divided into unfilled contol group and filling groups(lateral, ultrasonic, ULTRAFIL, McSpadden compaction group). Upon completion of root canal filling, the teeth were grooved on the both the labial and lingual surfaces and then split with mallet and chisel. Each specimens were examined with JSM - 840 Scanning Electron Microscope (JEOL., Japan). The results were as follows : 1. In the contol group, dentinal tubules of group I couldn't be distinguished in the canal wall, but those of group II appeared to be open and patent. 2. In the filling groups of group I, the tubular penetration of the sealer or gutta percha couldn't be seen, but in the filling groups of group II, it could be seen except McSpadden compaction group. 3. In the filling groups, ULTRAFIL group showed the best adaptation of filling material to root canal wall among the group I, and lateral and ultrasonic condensation group showed the best adaptation of filling material among the group II. McSpadden compaction group showed the worst adaptation in group I, II. 4. Generally, the group II showed better adapation of filling material to root canal wall than the group I.

A novel prediction model for post-fire elastic modulus of circular recycled aggregate concrete-filled steel tubular stub columns

  • The post-fire elastic stiffness and performance of concrete-filled steel tube (CFST) columns containing recycled aggregate concrete (RAC) has rarely been addressed, particularly in terms of material properties. This study was conducted with the aim of assessing the modulus of elasticity of recycled aggregate concrete-filled steel tube (RACFST) stub columns following thermal loading. The test data were employed to model and assess the elastic modulus of circular RACFST stub columns subjected to axial loading after exposure to elevated temperatures. The length/diameter ratio of the specimens was less than three to prevent the sensitivity of overall buckling for the stub columns. The gene expression programming (GEP) method was employed for the model development. The GEP model was derived based on a comprehensive experimental database of heated and non-heated RACFST stub columns that have been properly gathered from the open literature. In this study, by using specifications of 149 specimens, the variables were the steel section ratio, applied temperature, yielding strength of steel, compressive strength of plain concrete, and elastic modulus of steel tube and concrete core (RAC). Moreover, parametric and sensitivity analyses were also performed to determine the contribution of different effective parameters to the post-fire elastic modulus. Additionally, comparisons and verification of the effectiveness of the proposed model were made between the values obtained from the GEP model and the formulas proposed by different researchers. Through the analyses and comparisons of the developed model against formulas available in the literature, the acceptable accuracy of the model for predicting the post-fire modulus of elasticity of circular RACFST stub columns was seen.

The Evaluation of UV-induced Mutation of the Microalgae, Chlorella vulgaris in Mass Production Systems (자외선에 의해 유도된 Chlorella vulgaris 돌연변이 균주의 대량 생산 시스템에서의 평가)

  • The microalgae Chlorella vulgaris has been considered an important alternative resource for biodiesel production. However, its industrial-scale production has been constrained by the low productivity of the biomass and lipid. To overcome this problem, we isolated and characterized a potentially economical oleaginous strain of C. vulgaris via the random mutagenesis technique using UV irradiation. Two types of mass production systems were compared for their yield of biomass and lipid content. Among the several putatively oleaginous strains that were isolated, the particular mutant strain designated as UBM1-10 in the laboratory showed an approximately 1.5-fold higher cell yield and lipid content than those from the wild type. Based on these results, UBM1-10 was selected and cultivated under outdoor conditions using two different types of reactors, a tubular-type photobioreactor (TBPR) and an open pond-type reactor (OPR). The results indicated that the mutant strain cultivated in the TBPR showed more than 5 times higher cell concentrations ($2.6g\;l^{-1}$) as compared to that from the strain cultured in the OPR ($0.5g\;l^{-1}$). After the mass cultivation, the cells of UBM1-10 and the parental strain were further investigated for crude lipid content and composition. The results indicate a 3-fold higher crude lipid content from UBM1-10 (0.3%, w/w) as compared to that from the parent strain (0.1% w/w). Therefore, this study demonstrated that the economic potential of C. vulgaris as a biodiesel production resource can be increased with the use of a photoreactor type as well as the strategic mutant isolation technique.

  • In the system of GC-OTC/FID (Gas chromatography-Open Tubular Column/Flame Ionization Detector), DMSO (Dimethyl sulfide) solvent was used to separate the polar solvents (Alcohols). In this system DMSO was eluted later than the separated polar solvents. At this system to calculate chromatographic factors [adjusted retention time ($t_R^{\prime}=t_R-t_O$), capacity factor{$k^{\prime}=(t_R-t_O)/t_O$} and separation factor {${\alpha}=(t_{R2}-t_O)/(t_{R1}-t_O)$}], dead time($t_O$) is necessary, but the method to calculate it has not been reported yet. Therefore, we have tried to develop $t_O$. To calculate $t_O$, we conversed DMSO retention time (DMSO $t_R$) to logarithm ($f(x)={\log}\;t_{R(DMSO)}{\rightarrow}t_O$, $t_O={\log}$ 9.551=0.980). To confirm the optimization of the developed method, we compared with $CH_4\;t_R$ and ${\ln}\;t_{R(DMSO)}$. Both of the values calculated by $CH_4\;t_R$ and ${\ln}\;t_{R(DMSO)}$ were not suitable in the calculation k' and ${\alpha}$. The developed method in this study{${\log}\;t_{R(DMSO)}$} has satisfied both of the values k' criteria (1${\alpha}(1<{\alpha}<2)$. The developed calculation method in this study was easy and convenient, therefore it can be expected to be applied to these similar systems.

Morpho-molecular characterization of diatom Skeletonema pseudocostatum(Thalassiosirales, Bacillariophyta) from the Korean coast (규조류 Skeletonema pseudocostatum Medlin (Thalassiosirales, Bacillariohyta)의 형태적 특징과 분자계통학적 위치)

  • Morphology of a strain of unspecified Skeletonema species established from Korean coast was examined by light, fluorescence and scanning electron microscopy, and SSU(small subunit) and LSU(large subunit) rDNA of the strain were also sequenced. The specimen was characterized by solitary or short chains, and each cell contained 1-2 chloroplasts. The valve face was slightly convex, and the terminal fultoportula processes (TFPPs) were open and showed narrow distal ends that could be truncated or spiny with claw-like protrusions. The basal part of the TFPPs was tubular and oblique to the cell axis. The intercalary fultoportula processes (IFPPs) were also narrow, completely open, and joined in a 1 : 1 junction. Occasionally, one IFPP was connected with two opposing IFPPs. The morphological features of the specimen were identical to those of Skeletonema pseudocostatum. Molecular phylogeny based on SSU rDNA revealed that the Korean strain is nested within a clade comprising S. pseudocostatum and S. tropicum. However, based on D1-D2 LSU rDNA sequences, a clade including S. pseudocostatum and a Korean strain was separated from the S. tropicum clade. This indicates that the Korean strain can be identified as S. pseudocostatum. This species represents the first record from Korean coastal waters.

Development of a Solar Collector Performance of Cylindrical Parabolic Concentrating Solar Collector (태양열(太陽熱) 집열기개발(集熱器開發)에 관(關)한 연구(硏究) - 포물반사곡면(抛物反射曲面)으로된 2차원(二次元) 집광식(集光式) 태양열(太陽熱) 집열기(集熱器)의 성능분석(性能分析) -)

  • It is desirable to collect the solar thermal energy at relatively high temperature in order to minimize the size of thermal storage system and to enlarge the scope of solar thermal energy utilization. So far the concentrating solar collector has been developed to collect solar thermal energy at relatively high temperature, but it has some difficulties in maintaining the volumetric body of solar collector for long term utilization. On the other hand, the flat-plate solar collector has been developed to collect the solar thermal energy at low temperature, and it has advantages in maintaining the system for long term utilization, since it's thickness is thin and not volumetric. In this study, to develop a solar collector that has both advantages of collecting solar thermal energy at high temperature and fixing conveniently the collector system for long term period, a cylindrical parabolic concentrating solar collector was designed, which has two rows of parabolic reflectors and thin thickness such as the flat-plate solar collector, maintaining the optical form of concentrating solar collector. The characteristics of the concentrating parabolic solar collector newly designed was analysed and the results are summarized as follows; 1. The temperature of the air enclosed in solar collector was all the same as $50^{\circ}C$ in both cases of the open and closed loop, and when the heat transfer fluid was not circulated in tubular absorber, the maximum surface temperature of the absorber was $118-120^{\circ}C$, this results suggested that the heat transfer fluid could be heated up to $118^{\circ}C$. 2. In case of longitudinal installation of the solar collector, the temperature difference of heat transfer fluid between inlet and outlet was $4^{\circ}-6^{\circ}C$ at the flow rate of $110-130{\ell}/hr$, and the collected solar energy per unit area of collector was $300-465W/m^2$. 3. The collected solar energy per unit area for 7 hours was 1960 Kcal/$m^2$ for the open loop and 220 Kcal/$m^2$ for the closed loop. Therefore it is necessary to combine the open and closed loop of solar collectors to improve the thermal efficiency of solar collector. 4. The thermal efficiency of the solar collector (C.P.C.S.C.) was proportional to the density of solar radiation, indicating the maximum thermal efficiency ${\eta}_{max}=58%$ with longitudinal installation and ${\eta}_{max}=45%$ with lateral installation. 5. The thermal efficiency of the solar collector (C.P.C.S.C.) was increased in accordance with the increase of flow rate of heat transfer fluid, presenting the flow rate of $110{\ell}/hr$ was the value of turning point of the increasing rate of the collector efficiency, therefore the flow rate of $110{\ell}/hr$ was considered as optimum value for the test of the solar collector (C.P.C.S.C.) performance when the heat transfer fluid is a liquid. 6. In both cases of longitudinal and lateral installation of the solar collector (C.P.C.S.C.), the thermal efficiency was decreased linearly with an increase in the value of the term ($T_m-T_a$)/Ic and the increasing rate of the thermal efficiency was not effected by the installation method of solar collector.

Occlusion Effect of Dentinal Tubules of the Desensitizing Dentifrices Marketed in Korea (국내 시판중인 지각과민 완화 치약의 상아세관 폐쇄효과)

  • The aim of this study was to compare the effects of the short-term use of the desensitizing dentifrices marketed in Korea in vitro. Fifty human dentine specimens were wet ground with silicone carbide paper and etched with 6% citric acid for 90 seconds to allow complete opening of the dentinal tubule. Ten specimens from each group were brushed for 50 and 150 strokes with a V8 Cross Brushing Machine(Sabri Co., U. S. A). All the specimens were evaluated by SEM(${\times}3000$). The degree of occlusion of the dentinal tubules was quantified using an image analyzer. The results were analyzed by one-way ANOVA and Tukey's multiple comparisons using Window SPSS. The dentifrices containing nano-carbonate apatite, potassium nitrate and hydroxyapatite showed significantly higher occlusion effects than the other dentifrices after toothbrushing for 50 strokes(p<0.05). The Sensodyne freshmint$^{(R)}$dentifrice showed 34% fewer open tubular areas compared with the Sensodyne original$^{(R)}$dentifrice for 50 strokes. According to the short-term use of desensitizing dentifrices, the dentifrices containing nano-carbonate apatite, potassium nitrate and hydroxyapatite were most effective in occluding the dentinal tubules.

Analysis of Molecular Species of Vegetable Oil Triglycerides by Capillary Column GC-MS (Capillary Column GC-MS에 의한 식물유 트리글리세리드 분자종의 분석)

  • Triglyceride molecular species In some vegetable oils were analyzed by capillary column gas chromatography and electron impact ionization mass spectrometry utilizing selected ion monitoring. Triglycerides were separated according to their molecular weights and their degrees of unsaturation on $25m{\times}0.25mm$ fused silica open tubular capillary column coated with a phenylmethylsilicone gum stationary phase and in an analysis time less than 13 min. Triglyceride molecular species were identified by analyzing the fragment ions having the same time on the selected ion monitoring profile . The major triglyceride molecular species in each oils were $C_{18:1}.\;C_{18:2}.\;C_{18:2}(OLL:18.3%),\;C_{18:2}.\;C_{18:2}.\;C_{18:2}(LLL;\;14.3%),\;C_{18:0}.\;C_{18:2}.\;C_{18:2}(SLL;\;14.1%),\;C_{16:0}.\;C_{18:2}.\;C_{18:2}(PLL;\;13.2%),\;C_{16:0}.\;C_{18:2}.\;C_{18:1}(PLO;\;11.6%)$ in corn oil, $C_{18:2}.\;C_{18:2}.\;C_{18:2}(LLL;\;18.0%),\;C_{18:1}.\;C_{18:2}.\;C_{18:2}(OLL;\;18.0%),\;C_{16:0}.\;C_{18:2}.\;C_{18:2}(PLL;\;17.1%)$ in safflower oil, $C_{16:0}.\;C_{18:2}.\;C_{18:2}(PLL;\;23.5%),\;C_{16:0}.\;C_{18:2}.\;C_{18:1}(PLO;\;13.8%),\;C_{18:0}.\;C_{18:1}.\;C_{18:1}(SOO;\;13.5%),\;C_{18:1}.\;C_{18:2}.\;C_{18:2}(OLL;\;10.6%)$ in cottonseed oil.
