  • This study was conducted to characterize red worm(Lumbricus rubellus) and its cast reared six months using with pulp sludge, mixture of pulp sludge and methane sludge, and cow manure and methane sludge. The results obtained were as follows: 1. The optimum growth temperature ranged from $16^{\circ}C$ to $25^{\circ}C$ and monthly weight increase was about one kilogram per square meter. 2. The weight of red worm increased 9 to 11 percent reared with mixture of cow manure and methane sludge compared to pulp sludge. 3. Red worm contained large amount of amino acid, including. Lycine, which, might be a good source for a feed additive. 4. Yields of red worm cast ranged from 90 to 95 ton/10a/year on dry weight base. 5. CEC of red worm cast varied from 19.4 to 49.9 meg/100g and O.M content ranged from 26.4 to 35.1 percent. It contained lots of nutrients resulting in a good fertilizer source.

Characterization of Growth and Auxin Formation in a Bacteria Isolated from Waste bed of Agaricus bisporus (양송이 수확 후 배지로 부터 분리한 옥신생산 세균의 생육특성)

  • An auxin-producing bacteria (2SJ8-02) was isolated from waste mushroom bed of Agaricus bisporus in Buyeo-Gun, Chungnam. The strain 2SJ8-02 was classified as a novel strain of Pantoea rwandensis based on a chemotaxanomic and phylogeneticanalyses. The isolate was confirmed to produce indole-3-acetic acid (IAA), one of auxin hormones, by TLC and HPLC analyses. The maximum concentration of IAA, $122mg\;L^{-1}$ was detected from the culture in R2A broth containing 0.1% tryptophan for 24 h at $35^{\circ}C$. The molecular weight of the main peak obtained by LC-mass analysis was 175 corresponding to that of IAA. To investigate the growth-promoting effects to the crops, the culture broth of Pantoea rwandensis 2SJ8-02 was infected to water cultures and seed pots of mung bean. In consequence, the adventitious root induction and root growth of mung bean were two times higher than those of the control.

Evaluation of Physical Properties according to Mixing Ratio and the Survey of the Current Situation for Epoxy Resin used in Conservation (문화재 보존처리에 사용되는 에폭시수지의 사용현황과 배합비율에 따른 물성 변화 연구)

  • Two-component epoxy resin is widely used in the cultural heritage restoration field. However according to mixing ratio of resin and hardener, curing property, mechanical strength and chemical structure differ which have possibility to effect the stability of cultural heritage. Result of questionnaire survey shows hands-on workers in the conservation field tend to mix the epoxy resin with his or her eye measurement when the using amount is small or mix additional hardener to shorten the pot life of epoxy resin. This research aims to analyze the curing property, mechanical strength and chemical structure of rapid curing type epoxy resin and medium curing type one depending on relative ratio of 0.25~4 of hardener to resin. When the amount of hardener was 0.5~2 times more than the resin, exothermic heat and curing speed were both increased. In case of included hardener to resin was lower than official ratio, mechanical strength (tensile shear strength, tensile strength and compressive strength) became higher along with active cross-linking bonding of the epoxy resin. Medium curing type epoxy relatively had lower exothermic heat and slower reaction during curing process. It was observed to be put to definite point of mechanical strength under lower content of hardener than official ratio. While, hardener ratio more than twice the resin slowed down the curing greatly and lowered the adhesion strength also. In conclusion, under the lower mixing rate of hardener than official ratio would show relatively fast reaction with similar mechanical strength. Over the official ratio on the other hand, material property drops rapidly. Accordingly, mixing ratio of epoxy resin is expected to be influential to the stability of cultural heritage.

Soil Applications of Slaked Lime and Organic Fertilizer for Reducing 99Tc Transfer from Soil to Rice Seeds (99Tc의 토양-쌀알 전이 감소를 위한 소석회와 유기질 비료의 토양첨가)

  • To see if slaked lime and organic fertilizer applications to soil are useful as countermeasures for reducing $^{99}Tc$ concentrations in rice seeds after $^{99}Tc$ contamination of paddy fields, pot experiments were performed for two different paddy soils in a greenhouse. The upper soils for a depth of about 20 cm were treated with the agricultural materials and $^{99}Tc$ 15 d before transplanting. The effects were compared using the transfer factor (TF) defined as the ratio of the plant concentration to the soil concentration. In the case of control plants, TF values for brown rice in the two soils were $4.1{\times}10^{-4}$ and $4.3{\times}10^{-4}$. Of various types of the application, only the application of slaked lime at a lower dose (about 0.6 kg $m^{-2}$), which led to a 60% reduction in the TF value for one soil, seemed to be worth using as a countermeasure. Little effect of the same application was found in the other soil so it is important to determine the effect averaged for a number of soils. Organic fertilizer applications at both of two different doses increased the TF value. It is considered necessary to perform experiments for slake lime applications at doses lower than the above.

  • This study was conducted to examine the mesh selecivity and optimum mesh size of spring frame trap for conger eel. Conger myriaster. It was carried out on 25th Sep. 2002 at the coastal sea of Nung-po, Geojedo. The experimental fishing gear was used in five kinds of different mesh size spring frame trap that were 15mm, 20mm, 25mm, 30mm and 35mm, and one plastic pot as control fishing gear that was 6.7mm hole diameter. The mesh size 15mm, 20mm are the current gears, 25mm, 30mm are used in experiment, and 35mm is the legal mesh. These were made 50 traps, respectively. The mesh selectivity curve was analysed by the Kitahara's method(1968) and the optimum mesh size was estimated by relationship between the total length and diameter of conger eel and by the mesh selectivity master curve. The results obtained are summarized follows : 1. The total number of catch by the trap for conger eel was 835, it was consisted of 537 conger eel(64.4), 225 crabs(2639%) and 73 others(8.7%). 2. The value of maximum 1/m on the mesh selectivity curve was estimated at about 23.9. 3. The optimum mesh size of spring frame trap for conger eel was estimated 34.0mm in 50% selection range of the mesh selectivity master curve.

Simple, Rapid and Sensitive Portable Molecular Diagnosis of SFTS Virus Using Reverse Transcriptional Loop-Mediated Isothermal Amplification (RT-LAMP)

  • Recently, human infections caused by severe fever with thrombocytopenia syndrome virus (SFTSV), which can lead to fatality, have dramatically increased in East Asia. With the unavailability of vaccines or antiviral drugs to prevent and/or treat SFTSV infection, early rapid diagnosis is critical for prevention and control of the disease. Here, we report the development of a simple, rapid and sensitive portable detection method for SFTSV infection applying reverse transcription-loop mediated isothermal amplification (RT-LAMP) combined with one-pot colorimetric visualization and electro-free reaction platform. This method utilizes a pocket warmer to facilitate diagnosis in a resource-limited setting. Specific primers were designed to target the highly-conserved region of L gene of SFTSV. The detection limit of the RT-LAMP assay was approximately $10^0$ viral genome copies from three different SFTSV strains. This assay exhibited comparable sensitivity to qRT-PCR and 10-fold more sensitivity than conventional RT-PCR, with a rapid detection time of 30 to 60 minutes. The RT-LAMP assay using SFTSV clinical specimens has demonstrated a similar detection rate to qRT-PCR and a higher detection rate compared to conventional RT-PCR. Moreover, there was no observed cross-reactive amplification of other human infectious viruses including Japanese Encephalitis Virus (JEV), Dengue, Enterovirus, Zika, Influenza and Middle East Respiratory Syndrome Coronavirus (MERS-CoV). This highly sensitive, electro- and equipment-free rapid colorimetric visualization method is feasible for resource-limited SFTSV field diagnosis.

Development of a Drainage System to Mitigate Moisture Damage for Bridge Deck Pavements (교면포장의 수분손상 저감을 위한 체류수 배수공법 개발)

  • A major purpose of this study is to develop a drainage system that can quickly drain water penetrated into pavement layers to mitigate pot holes which is one of the major distress types in bridge deck pavements. This system can be established by applying a thin drainage layer between waterproof and pavement layers. The most important elements for this system are the performance of waterproof layer and construction technique for the thin drainage layer. The porous asphalt mix with the maximum aggregate size of 10mm is first developed based on the porous asphalt mix design guide proposed by NCAT, and various physical and mechanical tests are performed to confirm that the porous mix satisfies all the specification requirements. In addition, a series of laboratory tests including low-temperature bending and bonding strength tests for the MMA(Methyl Methacrylate) type of waterproofing material. It is observed from the tests that the MMA material satisfies all the specification requirements. To evaluate the Reld performance of the drainage system, a field study has been conducted on a relatively small size bridge. The QC/QA tests are conducted on the both waterproofing and pavement materials. It has been found that the drainage system works well to drain the water penetrated into the pavement layers.

  • In order to observe the effect of a continuous application of herbicides on the chemical properties of upland soil, samples were collected and analysed from a field experiment continuously applied with herbicides and from two pot experiments, one with a heavy dose and the other with a normal dose of herbicides, applied continuously. The soil samples were collected after three harvests of radish. The results of these analysis were summarized as follows; 1. Of herbicides, M.O., Lorox, 2, 4-D, Lasso, and P.C.P., the Lorox decreased soil pH and organic carbon content in soil and increased exchange and hydrolytic acids, and exchangeable potassium. The herbicide also showed an indication to decrease the exchangeable calcium, magnessium and sodium in soil. It seemed that such changes are mainly due to the indirect effect of Lorox or its metabolites, such as stimulating soil microorganisms, rather than direct chemical nature of them. 2. The other herbicides investigated in this study seemed to increase the organic carbon content in soil. 3. In addition, the continuous application of herbicides may result in an alteration of weed species in the field. All such facts might affect the properties of soil, which may call for a further detailed investigation having a set up of a long term experiment.

  • Recently, sludge disposal becomes one of the most serious environmental problems. Because the landfilling and ocean dumping of sludge materials will be prohibited in the near future, the proper treatment?disposal methods should be investigated. Also, oyster shells, piled at the coast, cause adverse effects in coastal fishery, public water surface, natural landscape, public health and so on. Thus, the purpose of this study is to evaluate the dissolution characteristic of metal compounds during soil application experiment using sewage treatment sludge mixed with oyster shells. The dissolution experiment conducted 100days under artificial rainfall and farming soil, mixed with sewage treatment sludge and oyster shells, was put into the pots(approx. 0.5L). The results from dissolution experiment as follows. 1. K, Na was $5{\sim}20mg/{\ell}$, and Ca was less than $90mg/{\ell}$. 2. Heavy metals such as Cd, Cu, As, Pb, Cr, Hg are dissoluted far less than the soil pollution guideline. The application of sewage sludge mixed with oyster shells increases pH(soil acidity)and buffer capacity(CEC) of farming soil, and heavy metals are thought to be attached to soil as insoluble forms.

Modelling The Population Dynamics of Laodelphax striatellus Fallén on Rice (벼에서 애멸구(Laodelphax striatellus Fallén) 개체군 밀도 변동 예측 모델 구축)

  • Temperature-dependent traits of Laodelphax striatellus, rice stripe virus vector, were investigated at 10 constant temperatures (12.5, 15.0, 17.5, 20.0, 22.5, 25.0, 27.5, 30.0, 32.5, and 35.0 ± 1℃) under a fixed photoperiod (14/10-hr light/dark cycle). Unit functions for the oviposition model were estimated and implemented into a population dynamics model using DYMEX. The longevity of L. striatellus adults decreased with increasing temperature (56.0 days at 15.0℃ and 17.7 days at 35.0℃). The highest total fecundity (515.9 eggs/female) was observed at 22.5℃, while the lowest (18.6 eggs/female) was observed at 35.0℃. Adult developmental rates, temperature-dependent fecundity, age-specific mortality rates, and age-specific cumulative oviposition rates were estimated. All unit equations described adult performances of L. striatellus accurately (r2 =0.94~0.97). After inoculating adults, the constructed model was tested under pot and field conditions using the rice-plant hopper system. The model output and observed data were similar up to 30 days after inoculation; however, there were large discrepancies between observed and estimated population density after 30 days, especially for 1st and 2nd instar nymph densities. Model estimates were one or two nymphal stages faster than was observed. Further refinement of the model created in this study could provide realistic forecasting of this important rice pest.
