A very reliable and specific method for the identification of fungi in ectotrophic mycorrhizal symbiosis was developed using a specific PCR assay based on the amplification of the ITS1 region. To obtain specific data, an ITS-diagnostic assay was carried out that reveals genera and species specific sequences. Here, an application of one method is presented, which covers the identification of pure mycelia, basidiocarps as well as mixed samples such as ectomycorrhizal roots that were mingled with remains of the host plant. For this purpose a protocol was established that allowed the extraction of DNA from single mycorrhizal roots. In order to perform a specific ITS analysis we generated a new ITS-primer(ITS8) by a multiple alignment of five different genera and species of mycorrhizal fungi. The utilization of ITS1 and ITS8 resulted in specific PCR amplicons, which were characterized by sequencing without purification steps, even when the template DNA was associated with roots.
Currently, cancer is the primary cause of death and 50% of cancer patients are incurable by surgery, radiotherapy and chemotherapy. Therefore, immunotherpy is interested as the fourth remedy. Biological response modifier (BRM), such as organometallic compounds, glycoproteins, polysaccharides and other natural products. Is the one which can enhance the immune response against cancer cell. To develop new BRM from natural sources, we investigated 63 species Korean traditional medicines by observing the mitogenic activity to splenocytes, generation of activated killer cells and activation of macrophages. Finally, we selected 9 species including Angelicae gigantis Radix, Mori Cortex Radicis, Arisaematis Tuber, Salviae Radix, Cnidii Rhizoma, Ligusti Fructus, Pasoraliae Semen, Loranthi Ramulus, Ginseng Radix. Bioassay-guided fractionation and purification is undergoing.
One specimen (103.7 mm SL) of Lactoria fornasini, belonging to the family Ostraciidae, collected from the coastal waters of Jeju Island, marks the first record of the species from Korea. It is characterized by having a strongly curved spine on the dorsal ridge, a pair of short spines in front of the eyes, a pair of spines projecting posteriorly from the rear end of the ventrolateral ridge, and wavy blue lines on the body. We add this species to the Korean fish fauna and propose its new Korean name, "Jul-mu-nui-ppul-bok."
Cymbidium is horticulturally important and has been one of the most commercially successful orchid plants as well as cut flowers around the world including Korea. Up to now, a huge number of elite Cymbidium cultivars have been released on the commercial market via cross-hybridization, mutation and polyploidization breeding techniques. To investigate on breeding system in Cymbidium, we inquired the brief history and techniques of breeding and the current status on Cymbidium breeding in Korea. Also, the general propagation process of elite Cymbidium lines via tissue culture should be presented. However, the slow process of conventional breeding and the lack of useful genes in Cymbidium species delays the introduction of new cultivars to the commercial market. To solve these limitations, efficient regeneration and genetic transformation systems should be established in the improvement of Cymbidium breeding program. During the last several decades, some progress has been made in tissue culture and genetic transformation in Cymbidium species. We review the recent status of tissue culture and genetic transformation systems in Cymbidium plants.
Ascomycota is the largest phylum of the Fungi, including approximately 6,600 genera. They are often isolated from soils, indoor air, and freshwater environments, but also from plants as pathogens or endophytes. In this study, four species of Ascomycota (two of Cladosporium and one of each Daldinia and Nigrospora) were collected from the leaves of four woody plants (Camellia japonica, Ginkgo biloba, Quercus sp., Vitis vinifera). Their cultural characteristics were investigated on five different media (PDA, V8A, CMA, MEA, CZA) at 3 days after incubation at $25^{\circ}C$ in darkness. BLASTn search and phylogenetic analysis were performed using the internal transcribed spacer (ITS) rDNA sequences, in addition to tef1 gene sequences for Cladosporium species. Based on the cultural, morphological, and phylogenetic data, the isolates were identified as Cladosporium anthropophilum, Cladosporium pseudocladosporioides, Daldinia eschscholtzii, and Nigrospora chinensis. Previously, some members of Cladosporium and Nigrospora have been recorded as endophytes inhabiting the leaves and stems of various plants, whereas Daldinia eschscholtzii is a wood-inhabiting endophyte or wood-decaying fungus. To our knowledge, this is the first report of these four ascomycetes in Korea.
This is the first report of Scomberoides lysan (Perciformes: Carangidae) from Korea. Two specimens (334 and 354 mm standard length) were collected by a set net from the coastal waters of Jejudo Island on 30 September, 2010. This species is characterized and distinguishable from a morphologically similar Scomberoides tol as follows. It has the posterior end of maxilla reaching to the posterior margin of eye (vs. not reaching in S. tol) and two rows of 5~8 small blotches on both sides of body (vs. one row of blotches). We add S. lysan to the Korean fish fauna and propose a new Korean name, "Du-jeom-jul-ga-si-jeon-gaeng-i" for the species because it has two series of dusk blotches on both sides of body.

A newly recorded chaetodontid species, Chaetodon speculum Cuvier, 1831 was collected in July, 2021 from Pohang, Korea. Its body color is yellow with black stripe across the eyes and a large spot occupying one fourth of the upper part of the body. C. speculum is very similar to congeneric species, C. zanzibarensis, but it can be distinguished by the size of the spot, shape of the lateral line, number of longitudinal line scales and geographic distribution. C. speculum is readily identified between post-larval stage and adult stage because of its similar external shape. Molecular analysis based on mitochondrial DNA COI sequences showed our specimen perfectly matched C. speculum. Therefore, we propose the new Korean name, "Heug-jeom-na-bi-go-gi" for C. speculum.

Owing to the rapid development of sequencing technologies, more than 1,000 plant genomes have been sequenced and released. Among them, 69 Korean plant taxa (85 genome sequences) contain at least one whole-genome sequence despite the fact that some samples were not collected in Korea. The sequencing-by-synthesis method (next-generation sequencing) and the PacBio (third-generation sequencing) method were the most commonly used in studies appearing in 65 publications. Several scaffolding methods, such as the Hi-C and 10x types, have also been used for pseudo-chromosomal assembly. The most abundant families among the 69 taxa are Rosaceae (10 taxa), Brassicaceae (7 taxa), Fabaceae (7 taxa), and Poaceae (7 taxa). Due to the rapid release of plant genomes, it is necessary to assemble the current understanding of Korean plant species not only to understand their whole genomes as our own plant resources but also to establish new tools for utilizing plant resources efficiently with various analysis pipelines, including AI-based engines.
Two new and one unrecorded hybrids of the genus Asplenium were newly found in Korea. They were proved to be hybrid based on intermediate morphology between putative parents. The first hybrid is $A.{\times}uiryeongse$ C.S. Lee & K. Lee (Aspleniaceae), nom. nov. (vernacular name: geo-mi-sa-cheol-go-sa-ri). This new hydrid is a cross between the Asian walking fern, A. ruprechtii and A. pekinens in Uiryeong-gil, Mt. Bukansan, Seoul, Korea. The other new one occurs in Mt. Seongsan in Yeoncheon-gun, Gyeonggi-do, Korea, $A.{\times}montanus$ C.S. Lee & K. Lee, nom. nov. (vernacular name: san-kko-ri-go-sa-ri) which might have experienced multiple hybridization events between A. ruprechtii ${\times}$ A. trichomanes and A. incisum following the hybrid between A. ruprechtii and A. trichomanes. In addition, one unrecorded hybrid, $A.{\times}kitazawae$ Kurata & Hutoh (vernacular name: geo-mi-dol-dam-go-sa-ri), reported first from Japan, is found in natural habitats in Do-dong, Daegu-si, and is a hybrid, between the Asian walking fern and A. sarelli. These hybrid taxa grow in sympatric regions with both putative parental species. The diagnostic characters for each taxon and evidence for their hybridization have been suggested. Descriptions, illustrations, and photographs of these hybrid taxa in their habitats are provided from Korea.

In this study, to obtain basic data for searching potential resources as new natural fibers, we investigated morphological and classificatory characteristics of 21 weed seed fibers. According to classification keys in this study, the collected weed seed fibers could be classified into total 13 types, showing their diversity. Seven species among them belonged to BOT3 type. Two species belonged to B2N0 and DOS3 type, respectively. Many of weed seed fibers had not branched. However, three species had various branched fibers at one main fibers on the seed. Three species had various branched fibers at several main fibers on the seed. Eight species had a smooth fiber surface but 13 species had a weakly or significantly developed-corniculum on the fiber surface. In the fiber cell shape, fiber cells of eight weed species were composed of one long cell without septum. But others had a fiber cell shape composed of a bunch of several long cells. Based on the easiness of harvesting, productivity of fibers, and morphological characteristics of seed fiber, it seemed that five seed fibers (TYPLA, METJA, HEMLY, IMPCK, and EREHI) should be additionally investigated if they are practically applicable as renewable resources for new natural fibers.
