The Distribution and Migration Boundary Lines of Oncorhynchus keta in the Milyang River (연어(Oncorhynchus keta)의 밀양강 분포 및 소상 한계선 파악)

  • In this study, we carried out a distribution and migration boundary lines of chum salmon (Oncorhynchus keta) in the Milyang River. We measured the total length of dead chum salmons. As a results, 40 chum salmons were found during the study period, 25 dead salmons and 15 alive individuals. The ratio of female was 64%. We verified the migration boundary lines of chum salmons based on a sighting survey until detecting a structure that chum salmons are not able to migrate upstream. We discovered that chum salmons were not able to migrate up to 13 km in a mainstream of the Milyang River and up to 12 km in the Danjang stream (a tributary of the Milayang River) from upstream of the Yerim Bridge. Therefore, our results indicate that Milyang River should improve a river connectivity by demolishing weirs that disturb chum salmon's migration or installing appropriate fishways. Also, effective legislations are needed to retain naturality in spawning ground and micro-habitats to increase the survival rate of parrs and smolts.

The Expression of Leptin, Estrogen Receptors, and Vitellogenin mRNAs in Migrating Female Chum Salmon, Oncorhynchus keta: The Effects of Hypo-osmotic Environmental Changes

  • Leptin plays an important role in energy homeostasis and reproductive function in fish, especially in reproduction. Migrating fish, such as salmonoids, are affected by external environmental factors, and salinity changes are a particularly important influence on spawning migrations. The aim of this study was to test whether changes in salinity affect the expression of leptin, estrogen receptors (ERs), and vitellogenin (VTG) in chum salmon (Oncorhynchus keta). The expression and activity of leptin, the expression of ERs and VTG, and the levels of estradiol-$17{\beta}$ and cortisol increased after the fish were transferred to FW, demonstrating that changes in salinity stimulate the HPG axis in migrating female chum salmon. These findings reveal details about the role of elevated leptin levels and sex steroid hormones in stimulating sexual maturation and reproduction in response to salinity changes in chum salmon.

Genetic Analysis of Asian Chum Salmon Populations Based on Microsatellite DNA Variation

  • We examined the genetic variability of Asian chum salmon (Oncorhynchus keta) populations using nuclear microsatellite (ms) DNA analysis with four polymorphic loci (OKM4, OKM5, OKM7, and OKM8) in 397 individuals from nine populations, including one in Korea, seven in Japan, and one in Russia. The msDNA gene diversity was highest in the Japanese populations, suggesting greater genetic variation in the populations in Japan than in populations in Korea and Russia. The pairwise $F_{ST}$ estimates based on our msDNA data showed that the Korean population was genetically different from the Japanese and Russian populations, and there were higher $F_{ST}$ estimates between Hokkaido and Honshu populations than between other population pairs. A neighbor-joining tree showed that the Korean population was distinct from two other clusters, representing the populations in Honshu and the populations in Hokkaido and Russia. These results suggest that the observed population genetic patterns of Asian chum salmon might be influenced by low or restricted gene flow.

Investigation of Hygiene Indicator Organisms and Listeria monocytogenes Contamination in Smoked Salmon Oncorhynchus keta Distributed in Korea (국내 유통 중인 훈제 연어(Oncorhynchus keta)의 위생지표세균 및 Listeria monocytogenes 오염도 조사)

  • This study was conducted to evaluate the level of microbial contamination in smoked salmon products sold in hypermarkets in major metropolitan cities in Korea. Listeria monocytogenes is the primary cause of smoked and raw salmon product recalls. Here, we used L. monocytogenes as a bacterial hygiene indicator and investigated the microbial contamination level of frozen/refrigerated smoked salmon products collected from hyper markets. Contamination levels were analyzed by seasons, manufacturers, and consumption regions. For hygiene indicator bacteria, total bacteria count, coliforms, and Escherichia coli were examined according to the food code established by the Ministry of Food and Drug Safety, and polymerase chain reaction (PCR) was performed to detect Listeria sp. The highest contamination level was observed in spring in Busan among five consumption regions. Listeria was detected at a level of 2.1% among all samples collected. And especially L. monocytogenes was detected in two cases from the samples collected from Daegu and Daejeon. Sanitary standards and specifications should be established according to the contamination level of smoked salmon products investigated in this study, and continuous monitoring is necessary.

Genetic Similarity-dissimilarity Among Korea Chum Salmons of Each Stream and Their Relationship with Japan salmons (한국 연어의 소상하천간 유전적 유사성과 차이점 및 일본 연어와 유전적 관계)

  • Analysis of population structure of Oncorhynchus keta, the most abundant salmon in the East Sea of Korea, has not been much carried out despite its importance as a fishery resource in the North Pacific. Currently, molecular methods are being applied to stock identification and a method of using single nucleotide polymorphisms (SNPs) is getting more popular. In this study, we analyzed the 720 bp long sequence of the mtDNA COIII-ND3-ND4L region in order to examine genetic similarity-dissimilarity among the Korea chum salmons of each stream and their relationship with the Japan chum salmons. A total of 152 individuals were analyzed, 108 from 3 locations of Korea and 44 from 2 locations of japan, which resulted in as many as 29 different haplotypes. Pairwise $F_{ST}$ and AMOVA tests of the populations show that there is no significant population-level genetic difference among the chum salmons analyzed ($F_{ST}<0.07$). On the other hand, haplotype relationships among the individuals reveal that approximately 25% of the Korea salmons consist genetic lineages independent of Japan salmons and also that a genetic lineage exists in the Puk river and the Namdae river salmons independent of the Wangpi river salmons of Korea.

Utilization of Hypothalamic Hormones for Maturational Induction in Seawater Chum Salmon, Oncorhynchus keta (해면연어의 성숙을 유도하기 위한 시상하부호르몬의 이용)

  • Freshwater-adapted maturing chum salmon (Oncorhynchus keta) involuntarily captured by stationary nets in Yang-yang seashore areas were transferred to freshwater in an outdoor raceway tank at Yeongdong Inland Fisheries Research Institute, NFRDI, Yang-yang, Gangwon, Korea and kept over 1 day until the start of the experiments. The freshwater-adapted females were single-injected intraperitoneally with gonadotropin-releasing hormone analogue, (GnRH-a: $70\;{\mu}g/kg$ body weight, BW) alone or combined with a dopamine receptor antagonist, pimozide($700\;{\mu}g/kg$ BW). Although gonadosomatic indices [GSI, (gonad weight/BW)${\times}100$] did not show significant changes in both 2004 and 2005, GSI of GnRH-a-injected fish during the 2005 trial slightly increased on the 5th and 7th days post-injection compared to those of vehicle treated fish. Hepatosomatic indices [HSI, (liver weight/BW)${\times}100$] of fish injected with GnRH-a alone and combined with pimozide decreased significantly on the 7th day post-injection in 2004(P<0.05). In 2005 trials, HSI was significantly reduced in GnRH-a treated fish on the 7th day post-injection (P<0.05). Pimozide-injected fish showed a pattern with increase of GSI and decrease of HSI, without significant differences. Taken together, these results suggest that at least in part hypothalamic hormones and dopamine receptor antagonist may induce sexual maturation in freshwater-adapted maturing chum salmon. It remains to evaluate these preliminary results by further researches.

이석 내 미량원소 함량비율을 이용한 한국산 연어(Oncorhynchus keta)의 계군분리

  • 최근, 기계의 분석 정밀도가 높아짐에 따라 어류 이석 내 미량원소를 이용한 연구가 활발히 진행되고 있다. 이석은 균형감각기관으로서 칼슘과 단백질의 복합구조로 이루어져 있으며, 어류의 전 생활사에 걸쳐서 성장한다. 이렇게 형성된 어류의 이석내 화학조성은 형성 후엔 변화하지 않는다는 특징을 가지고 있다(Campana,1999). 위와 같은 특징을 바탕으로 이석 내 미량원소 또는 동위원소의 함량비율을 분석하여 어류에 대한 계군분리, 어류의 서식환경 추적, 오염 정도, 그리고 어류의 회유 유무와 경로, 연령사정 등의 연구를 하고 있다(Campana, 1999).   • 연어는 북태평양 연안의 냉수역에 서식하는 소하성 어류이다. 한국을 비롯한 북태평양 연안국들은 연어 치어를 생산하여 방류한다. 각 나라에서 방류된 연어의 회유경로가 서로 겹치고 인접국의 배타적 경제수역을 지나게 되는 경우가 많아 연어를 방류하는 나라들 간에 자원분쟁의 가능성이 높다. 이러한 분쟁을 조정하고, 연어자원에 대한 합리적인 관리를 위해 연어 계군에 대한 명확한 구분이 필요하다. (중략)
