Frost Heave Force of Ground and Countermeasure for Damage of Structures (지반의 동상력과 구조물의 피해대책)

  • Frost action may cause extensive damage to building, structures, roads, railways and utility lines in seasonal frost. The research about frost heave of natural ground has been considerably performed. In late years various structures have become complicated with the development of social infrastructure maintenance. Therefore countermeasure to frost heave becomes a matter of great importance from a new viewpoint. This study was aimed at catching natural ground frost heaving force quantitatively. Frost heaving forces on circular steel plates which were set on ground surface were measured in field test. The frost heaving forces arise at freezing front propagates to the structures through frozen soil layer. Besides, a full scale model of multi-anchored retaining wall was installed in field, and the freezing lines, frost heave pressure to act on a wall block, and so on were measured. Finally, the position and shape of frost line were estimated by using numerical simulation and a method to determine replacement range was suggested with soil properties and weather data.

Case Study on the Overseas Locally-Led Community Design Guidelines (주민참여형 커뮤니티정원 조성 가이드라인 마련을 위한 국외 사례 비교 연구)

  • With the changing of social needs and paradigms, citizens are concerned about aesthetic and cultural community gardens from productive urban farms in Korea. It is still difficult, however, to cultivate a beautiful garden with a community to design, install and manage it. Therefore, this study analyzed the community garden design guidelines of other countries to derive the characteristics and implications. The research method analyzed six guidelines for community garden design in the UK, Canada, the USA, Australia and Japan. As a result of the study, most community garden design guidelines are opened freely via on-line service. The guidelines are composed with chronological processes such as intro, site selection, design, construction and maintenance. The introduction section treats definition, purpose, meaning, efficiency and success factors for community gardens. Site selection emphasizes site conditions (soil, light, shadow, water, etc.), landholding and insurance. The design section, however, lacks adequate drawings and case images. These guidelines offer little explanation with few illustrations and sentences. Construction sections explain about plantings, facilities, expense and details. The maintenance guidelines of community gardens contain how to control members, funding, harvesting, pests and plants. In addition, some guidelines include vandalism, organic cultivation methods, recording and advertisement. Lastly, the appendix contains a variety of checklists, administrative documents, guidelines, drawings, and contacts. This study will help understand the trends and characteristics of overseas community gardens design guidelines so as to also supply directions for guidelines to be introduced in Korea.

Efficiently Development Plan from the User's Need Analysis of the Army Tactical C4I(ATCIS) System (지상전술 C4I(ATCIS)체계 운용자 요구분석을 통한 효율적 발전 방안)

  • This study was to minimize the trial and error in the primary step of the C4I system(ATCIS) of the each army corps on the front line, and test the economy and efficiency was tested by reviewing related papers and the system characteristics of other countries. The relationship was researched by analyzing the collected survey data and survey data related to the user's requirement level such as the army standards, that is, commonality, timeliness, simplification, automaticity, field availability and viability, multi-stage security and interoperability, unification. The result showed that the C4I system was efficiently operated through the system reliability for the specification of the system and operation manual, maneuverability and security, adaptability of the war field and system support and management, and good education and training about system operation, and less system maintenance and supplementary element. As a result, the development plan confirmed that the continuous operator education and the construction of the maintenance, and the upgrade digitalization(C4ISR+D) with the korean characteristics based on IT of network systems, and system development of the measurement model of the operator performance must be continuously supplemented in the near future.

Molecular Aspects of Japanese Encephalitis Virus Persistent Infection in Mammalian Cells

  • Japanese encephalitis virus (JEV) is the causative agent of a mosquito-borne encephalitis and is transmitted to human via persistently infected mosquito vectors. Although the virus is known to cause only acute infection, there were reports that showed neurological sequelae, latent infection in peripheral mononuclear cells, and recurrence of the disease after acute encephalitis. Innate resistance of certain cell lines, abnormal SN1 expression of the virus, and anti-apoptotic effect of cullular bcl-2 have been suggested as probable causes of JEV persistence even in the absence of defective interfering (DI) particles. Although possible involvement of DI particles in JEV persistence was suggested, neither has a direct evidence for DI presence nor its molecular characterization been made. Two questions asked in this study are whether the DI virus plays any role in JEV persistent infection if it is associated with and what type of change(s) can be made in persistently infected cells to avoid apoptosis even with the continuous virus replication, DI-free standard stock of JEV was infected in BHK-21, Vero, and SW13 cells and serial high multiplicity passages were performed in order to generate DI particles. There different-sized DI RNA species which were defective in both structural and nonstructural protein coding genes. Rescued ORFs of the DI genome maintained in-frame and the presence of replicative intermediate or replicative form RNA of the DI particles confirmed their replication competence. On the other hand, several clones with JEV persistent infection were established from the cells survived acute infections during the passages. Timing of the DI virus generation during the passages seemed coincide to the appearance of persistently infected cells. The DI RNAs were identified in most of persistently infected cells and were observed throughout the cell maintenance. One of the cloned cell line maintained the viral persistence without DI RNA coreplication. The cells with viral persistence released the reduced but continuous infectious JEV particle for up to 9 months and were refractory to homologous virus superinfection but not to heterologous challenges. Unlike the cells with acute infection these cells were devoid of characteristic DNA fragmentation and JEV-induced apoptosis with or without homologous superinfection. Therefore, the DI RNA generated during JEV undiluted serial passage on mammalian cells was shown to be biologically active and it seemed to be responsible, at least in part, for the establishment and maintenance of the JEV persistence in mammalian cells. Viral persistence without DI RNA coreplication, as in one of the cell clones, supports that JEV persistent infection could be maintained with or without the presence of DI particles. In addition, the fact that the cells with JEV persistence were resistant against homologous virus superinfection, but not against heterologous one, suggests that different viruses have their own and independent pathway for cytopathogenesis even if viral cytopathic effect could be converged to an apoptosis after all.

Walnut phenolic extracts reduce telomere length and telomerase activity in a colon cancer stem cell model

  • BACKGROUND/OBJECTIVES: Telomeres are located at the chromosomal ends and progressively shortened during each cell cycle. Telomerase, which is regulated by hTERT and c-MYC, maintains telomeric DNA sequences. Especially, telomerase is active in cancer and stem cells to maintain telomere length for replicative immortality. Recently we reported that walnut phenolic extract (WPE) can reduce cell viability in a colon cancer stem cell (CSC) model. We, therefore, investigated the effect of WPE on telomere maintenance in the same model. MATERIALS AND METHODS: $CD133^+CD44^+$ cells from HCT116, a human colon cancer cell line, were sorted by Fluorescence-activated cell sorting (FACS) and treated with WPE at the concentrations of 0, 10, 20, and $40{\mu}g/mL$ for 6 days. Telomere lengths were assessed by quantitative real-time PCR (qRT-PCR) using telomere specific primers and DNA extracted from the cells, which was further adjusted with single-copy gene and reference DNA ($ddC_t$). Telomerase activity was also measured by qRT-PCR after incubating the PCR mixture with cell protein extracts, which was adjusted with reference DNA ($dC_t$). Transcriptions of hTERT and c-MYC were determined using conventional RT-PCR. RESULTS: Telomere length of WPE-treated cells was significantly decreased in a dose-dependent manner ($5.16{\pm}0.13$ at $0{\mu}g/mL$, $4.79{\pm}0.12$ at $10{\mu}g/mL$, $3.24{\pm}0.08$ at $20{\mu}g/mL$ and $3.99{\pm}0.09$ at $40{\mu}g/mL$; P = 0.0276). Telomerase activities concurrently decreased with telomere length ($1.47{\pm}0.04$, $1.09{\pm}0.01$, $0.76{\pm}0.08$, and $0.88{\pm}0.06$; P = 0.0067). There was a positive correlation between telomere length and telomerase activity (r = 0.9090; P < 0.0001). Transcriptions of both hTERT and c-MYC were also significantly decreased in the same manner. CONCLUSION: In the present cell culture model, WPE reduced telomere maintenance, which may provide a mechanistic link to the effect of walnuts on the viability of colon CSCs.

Numerical Analysis for Dynamic Characteristics of Next-Generation High-Speed Railway Bridge (차세대 고속철 통과 교량의 동적특성에 대한 수치해석)

  • To take into account of the increasing speed of next generation high-speed trains, a new design code for the traffic safety of railway bridges is required. To solve dynamic responses of the bridge, this research offers a numerical analyses of PSC (Pre-stressed Concrete) box girder bridge, which is most representative of all the bridges on Gyungbu high-speed train line. This model takes into account of the inertial mass forces by the 38-degree-of-freedom and interaction forces as well as track irregularities. Our numerical analyses analyze the maximum vertical deflection and DAF (Dynamic Amplification Factor) between simple span and two-span continuous bridges to show the dynamic stability of the bridge. The third-order polynomial regression equations we use predict the maximum vertical deflections depending on varying running speeds of the train. We also compare the vertical deflections at several cross-sectional positions to check the influence of running speeds and the maximum irregularity at a longitudinal level. Moreover, our model analyzes the influence lines of vertical deflection accelerations of the bridge to evaluate traffic safety.

Effect of Culture Medium Containing Chicken Serum on Growth and Differentiation of QM7 Quail Muscle Cells (닭 혈청을 포함한 배양액 조성 변화가 QM7 메추리 근육세포의 성장 및 분화에 미치는 영향 분석)

  • QM7 cell, a quail muscle cell line, has been used in various studies. In cell culture, it is well known that the culture medium has a significant influence on cell growth and maintenance of cell characteristics. This study aimed to adjust the culture medium to make it more suitable for QM7 muscle cells. A culture medium was prepared by adding 2% chicken serum (CS) instead of 10% tryptose phosphate broth (TPB) to the conventional culture medium. In the culture medium prepared with CS, the QM7 muscle cells changed from a pointed, thin to a broader shape. In addition, they grew and divided more rapidly in the new culture medium than in the conventional culture medium. The number of cells increased faster in the CS-containing culture medium from day two after passaging, and significantly increased from day three. The muscle cells grown in the medium containing CS maintained their undifferentiated state prior to differentiation. Myotubes formed well when the cells were maintained in CS-containing medium, resulting in a longer length and uniformity. According to the above results, it is clear that the culture of QM7 myocytes using a medium containing CS rather than TPB helps obtain better results in cell maintenance and differentiation.

  • The mediator-based system integrates heterogeneous information systems with the flexible manner. But it does not give much attention on the query optimization issues, especially for the query reusing. The other thing is that it does not use standardized metadata for schema matching. To improve this two issues, we propose mediator-based Distributed Information Integration System (DIIS) which uses query caching regarding performance and uses ISO/IEC 11179 metadata registry in terms of standardization. The DIIS is designed to provide decision-making support, which logically integrates the distributed heterogeneous business information systems based on the Web environment. We designed the system in the aspect of three-layer expression formula architecture using the layered pattern to improve the system reusability and to facilitate the system maintenance. The functionality and flow of core components of three-layer architecture are expressed in terms of process line diagrams and assembly line diagrams of Eriksson Penker Extension Model (EPEM), a methodology of an extension of UML. For the implementation, Supply Chain Management (SCM) domain is used. And we used the Web-based environment for user interface. The DIIS supports functions of query caching and query reusability through Query Function Manager (QFM) and Query Function Repository (QFR) such that it enhances the query processing speed and query reusability by caching the frequently used queries and optimizing the query cost. The DIIS solves the diverse heterogeneity problems by mapping MetaData Registry (MDR) based on ISO/IEC 11179 and Schema Repository (SCR).

Effect of Induction Heat Bending Process on the Properties of ASME SA106 Gr. C Carbon Steel Pipes

  • Recently, the bending process is greatly applied to fabricate the pipe line. Bending process can reduce welding joints and then decrease the number of inspection. Thus, the maintenance cost will be reduced. Induction heat bending process is composed of bending deformation by repeated local heat and cooling. By this thermal process, corrosion properties and microstructure can be affected. This work focused on the effect of induction heating bending process on the properties of ASME SA106 Gr. C low carbon steel pipes. Microstructure analysis, hardness measurements, and immersion corrosion test were performed for base metal and bended area including extrados, intrados, crown up, and down parts. Microstructure was analyzed using an optical microscope and SEM. Hardness was measured using a Rockwell B scale. Induction heat bending process has influenced upon the size and distribution of ferrite and pearlite phases which were transformed into finer structure than those of base metal. Even though the fine microstructure, every bent area showed a little lower hardness than that of base metal. It is considered that softening by the bending process may be arisen. Except of I2, intrados area, the others showed a similar corrosion rate to that of base metal. But even relatively high rate of intrados area was very low and acceptable. Therefore, it is judged that induction heat bending process didn't affect boric acid corrosion behaviour of carbon steel.

Optimization of Ballast Depth of Ballasted Track Bridges to Improve Ride Comfort (승차감 향상을 위한 유도상교량의 도상두께 최적화)

  • The ballast abrasion occurs on the ballasted track upon bridges more than soil roadbed because the track vibration occurs a lot in the ballasted track upon bridges due to girder vibration when a train's weight is loaded onto track even though the identical ballast is used. The phenomena of mud pumping especially, which occurs when drainage is not properly secured for heavy rain, leads to the increase of maintenance work load and the decline of ride comfort. The ballast thickness range in domestic railroad construction rule is uniformly set up according to the design speed of railroad and passing tonnage of train without considering field conditions which is considered in foreign railroad companies. The purpose of this study is to verify the effect of vibration decrease by measuring the acceleration, displacement and ride comfort of ballasted track with the change of ballast thickness on the ballast tracked bridges and to suggest the optimal height of ballast on the Yocheon Bridge built for the test in Honam Line.
