Antimicrobial Effect of Phytochemicals to Listeria monocytogenes Isolated from Slaughterhouses

  • The objective of this study was to investigate the inhibitory effect of natural antimicrobials on Listeria monocytogenes isolated from Korean slaughterhouses. A mixture of 15 strains of L. monocytogenes at low (3 Log CFU/mL) or high (7 Log CFU/mL) concentration was exposed to various extracts (grapefruit seed extract, citrus fruit extract, ginger extract, pear extract, Japanese apricot concentrate, balloon flower extract, jujube extract, and omija extract) at $0.001-4.0{\mu}g/mL$. Ginger extract, pear extract, Japanese apricot concentrate, balloon flower extract, jujube extract, or omija extract showed no antimicrobial effects on high-concentration of L. monocytogenes (7 Log CFU/mL). However, grapefruit seed extract and citrus fruit extract showed antibacterial effects against L. monocytogenes at 3 and 7 Log CFU/mL with MBCs of 0.001 and $0.002{\mu}g/mL$, respectively. These results indicate that grapefruit seed extract and citrus fruits extract can be used to control L. monocytogenes as natural antimicrobials.

Effects of Additive materials on the Quality Characteristics of Dasik (다식의 제조시 첨가하는 부재료와 품질특성)

  • This study was carried out to investigate the possibility of improving the texture and flavor of Dasik by adding various types of sugar (syrup, honey) and flavor ingredients (omija, chija, coffee, green tea extract) to rice powder. Dasik samples were prepared, and the sensory quality and physical characteristics of those were compared. The moisture content of Dasik added with syrup was higher than that of honey. Coffee Dasik with syrup was the highest (23.6) in moisture content. In sensory quality, the omija and coffee Dasik showed the highest score in flavor quality (p<.001). Omija Dasik with honey and coffee Dasik with syrup showed the highest scores in overall acceptability (6.4, 6.2). Green tea Dasik with syrup showed the highest value in the lightness (L) of color. Omija Dasik with syrup showed the highest value in the redness (a) of color Chija Dasik was the highest in the yellowness(b) of color. In physical characteristics, the hardness was negatively correlated with the moistness, tenderness, and texture acceptability in sensory quality(p〈0.001). The cohesiveness was positively correlated with the overall acceptability in sensory quality (p〈0.01). In the relation of texture characteristics and sensory quality, the higher the moisture content, the lower the hardness and springiness were, but the higher the brittleness and the cohesiveness were(p〈.001). Overall, omija and coffee Dasik appeared to have desirable flavor, taste and overall acceptability.

Protective Effects of Omija-tang on $H_2O_2$-induced apoptotic death of H9c2 cardiomyoblast cells (오미자탕(五味子湯)이 심근세포에 미치는 영향)

  • The water extract of Omija-tang(OMJT) has been traditionally used for treatment of ischemic heart and brain damage in oriental medicine. However, little is known about the mechanism by which the water extract of OMJT protects cells from such damage. Therefore, this study was conducted to investigate the protective mechanisms of OMJT on $H_2O_2$-induced toxicity in H9c2 cardiomyoblast cells. Treatment of $H_2O_2$ markedly induced death of H9c2 cardiomyoblast cells in a dose-dependent manner. The characteristics of $H_2O_2$-induced death of H9c2 showed apparent apoptotic features, such as DNA fragmentation. However, OMJT significantly reduced both $H_2O_2$-induced cell death and chromatin fragmentation. The decrease of Bcl-XL expression by $H_2O_2$ was inhibited by OMJT. In addition, the increase of Bcl-XS and Bax expression were also inhibited by OMJT. In particular, Fas expression, which is generally recognized as cell death inducing signal by Fas/FasL interaction, was markedly increased by $H_2O_2$ in a time-dependent manner, whereas this increase was completely prevented by OMJT. The combined treatment of OMJT and $H_2O_2$ in H9c2 cells also reduced activation of caspase-9 and caspase-3 like protease. Taken together, this study indicates that the protective effects of the water extract of OMJT against oxidative damage may be mediated by the modulation of BcI-XL/S and Bax expression by way of the regulation of mitochondrial membrane potential and caspase cascades.

Production of fermented Omija (Schizandra chinensis) beverage fortified with high content of gamma-amino butyric acid using Lactobacillus plantarum (오미자(Schizandra chinensis) 열매 추출물의 Lactobacillus plantarum 젖산발효를 통한 고농도 GABA 함유 발효음료 제조)

  • Omija (Schizandra chinensis) extract (OE) was fermented by using Lactobacillus plantarum EJ2014 to produce a beverage fortified with gamma-aminobutyric acid (GABA). After 2 days of fermentation in the presence of 2% monosodium glutamate (MSG) and 0.5% yeast extract (YE), the four-fold-diluted OE showed a higher viable cell count ($2.2{\times}10^9CFU/mL$) and lower acidity (1.2%) than that of the unfermented OE. In particular, addition of MSG as a precursor resulted in a small increase in the initial pH. MSG (2%) was completely converted to GABA (0.92%) during lactic acid bacteria fermentation for 3 days. Furthermore, the acidity of the fermented OE decreased from 1.74% to 0.56%. In addition, the original red color of the OE disappeared during LAB fermentation. However, when the fermented OE was mixed with 50% of the original OE, the original red color was recovered, with 19.56 and 13.92 for Hunter L and a values, respectively. The mixture of 50% original OE and 50% fermented OE showed the highest sensory score including the highest overall preference. In conclusion, the OE fortified with GABA and probiotics was produced by fermentation with a static culture, L. plantarum EJ2014.

The properties and extracting conditions of juice preperation from Schizandra nigra Max. (흑오미자(Schizandra nigra Max) 즙액의 추출조건과 추출물의 특성)

  • To determine the properties for juice preperation of Black Omija (Schizandra nigra Max.) and Omija (Schizandra chinensis), yield of extraction, chromaticity and lightness, pH and soluble solid of the extract were investigated. The rate of extract yield from Schizandra nigra was highest when extracted for 3 hours at 80$^{\circ}C$ in 20% ethanol solution. For the desirable chromaticity coordinates, the optimum extraction time and temperature of Schizandra nigra extract were 3 hours at 80$^{\circ}C$. The lightness of the extract was low of the value when extraction time and temperature was long and high. The sugar content of the extract of S. nigra was 2.0­2.6% Brix, lower than that of S. chinensis, but the difference was insignificant. The pH of the extract from S. nigra was 0.1­0.2 higher than that of S. chinensis. Although the pH of the extract from S. nigra was a little low when extracted by water, the pH range was enough to maintain the stability of color of extract from the S. chinensis.

Comparison of the Properties and Extracting Conditions of Juice Preperation from Schizandra nigra (흑오미자 즙액의 추출조건과 추출물의 특성)

Although the pH of the extract from Schizandra nigra was a low in comparison with that of water extract the pH range was proper to maintain the stability of color of extract from the Schizandra chinensis.

Development of Functional Misutkaru Drink Added with Several Medicinal Herbs Extracts (약용식물추출물을 첨가한 건강미숫가루음료 개발)

  • In order to develop of health-oriented convenience foods for rice consumption, functional Misutkaru drink was made by adding several medicinal herbs extracts to fundamental Misutkaru based cereal powder(rice, barley and soybean). Fundamental Misutkaru drink was prepared with the ratio of cereal mixed rice(5g), barley(5g) and soybean(5g) on water (100 mL) and several medicinal herbs extracts. Liriopis Tuber(50%), Omija(15%), Ginseng(10%) and Jujube(25%) were mixed to fundamental Misutkaru as functional Misutkaru. And then it was extracted to added water of 10times during 36hrs at 75$^{\circ}C$. For instant Misutkaru drink of health-oriented convenience foods, mixture of them was made with fundamental Misutkaru drink(80%), extract of several medicinal herbs(20%), and it's sensory score was high. Autoclaved functional Misutkaru drink for long storage period was changed rheological type badly. Overall acceptability and storage period of nonautoclaved functional Misutkaru drink to added extract of several medicinal herbs was higher than fundamental Misutkaru drink.

Antioxidant effect of Omija (Schizandra Chinesis Baillon) Extracts (오미자 추출물의 항산화 효과)

  • Antioxidative activities of the extracts from Schizandra Chinesis Baillon (Omija) with various solvent were compared with some commercial antioxidants. AS (antioxidative index; induction period of oil containing extract/induction period of control oil) of Omija extracts from five kinds of solvents (MeOH, EtOH, BtOH, EA, PE) and other antioxidants were shown as following orders: 0.02% BHT > 0.05% EA > 0.01% MeOH > 0.05% EtOH > 0.1% EA > 0.05% PE > 0.05% BuOH >> 0.02% alpha-tocopherol. Antioxidative effects of 0.05% EA and 0.05% MeOH extracts during autoxidation (60${\pm}$ 2$^{\circ}C$) were higher than those (If the other extracts but were not greater than that of 0.02% BHT. However, Al of EA and MeOH, EtOH extracts during thermal oxidation (180${\pm}$2$^{\circ}C$) were greater than that of BHT. The antioxidant effect of alpha-tocopherol showed no apparent difference or a prooxidant effect as compared with result of control.

Preparation and Characteristics of Drinkable Yoghurt Added Water Extract of Omija(Schizandra chinensis Baillon) (오미자(Schizandra chinensis Baillon) 추출물 첨가 Drinkable Yoghurt의 제조 및 특성)

  • A new type of drinkable yoghurt was prepared to develop a functional yoghurt. Skim milk containing 0.4-1.0%(w/v) water extract of omija(Schizandra chinensis Baillon) was fermented by the mixed strains of Lactobacillus acidophilus and Streptococcus thermophilus. Quality characteristics of the drinkable yoghurts were evaluated in terms of compositions, keeping-quality(pH, titratable acidity, number of viable cells) and sensory properties. The drinkable yoghurts added with water extract of Schizandra chinensis were composed of 2.92-3.03% protein, 9.98-10.23% lactose, 0.81-1.08% fat, 16.21-16.64% total solid, and 14.57-15.17% solid-not-fat. The L values(brightness) were significantly lower in the yoghurts containing water extract of Schizandra chinensis than that of the control which had no water extract of Schizandra chinensis, while the a values(redness) and b values(yellowness) were significantly higher than those of the control. The pH, titratable acidity and number of viable cells of the lactic acid bacteria of all yoghurts were not changed during the storage at 4$^{\circ}C$ for 15 days, while the pH and titratable acidity were remarkedly changed during the yoghurts stored at 20$^{\circ}C$ for 15 days. The drinkable yoghurts containing 0.4%(w/v) water extract of Schizandra chinensis added 15%(w/v) oligosaccharide, or 0.6%(w/v) water extract of Schizandra chinensis added 20%(w/v) oligosaccharide had the highest sensory score in taste and overall acceptability among the treatments. From the results, Schizandra chinensis was a useful natural additive with the bioactive effect by provision of a acceptable physicochemical and sensory properties in the drinkable yoghurt.

Effect of Omija(Schizandra chinensis) Extract on the Growth Inhibition of Food Borne Pathogens in Yoghurt (오미자 추출액 첨가 요구르트의 식중독균 증식 억제 효과)

  • Escherichia coli O157:H7, Staphylococcus aureus and Salmonella enteritidis are food borne pathogens involved in food poisoning in numerous countries. This study aimed to obtain knowledges on the survival of Esc coli O157:H7, Sta aureus and Sal. enteritidis in the yoghurt added with water extract of Omija(Schizandra chinensis). The growth inhibition of Schizandra chinensis extract on the food borne pathogens were measured by total microbial count and effect of growth inhibition was correspondent to the concentration of Schizandra chinensis extract. The highest growth inhibition effect of Schizandra chinensis extract was shown on the Sta aureus followed by Sal. enteritidis and Esc. coli O157:H7. The number of surviving Esc. coli O157:H7 cell(3.55${\times}$10$\^$5/ CFU/mL) was decreased to 1.00${\times}$10$^1$∼3.00${\times}$10$^1$ CFU/mL after 24 hours incubation by the addition of 0.4∼l.0% of Schizandra chinensis extract in the yoghurt. And also the viable cell counts of surviving Sta. aureus cells (initial inoculum 1.24${\times}$10$\^$5/ CFU/mL) were decreased gradually to 4.00${\times}$10$^2$∼8.50${\times}$10$^2$ CFU/mL after 48 hours of incubation, but the viable cells of Sal. enteritidis were not detected after 24 hours of incubation. Growth of the food borne pathogens was strongly inhibited by the addition and incubation of Schizandra chinensis extract for 48 hours in the yoghurt.
