The Effect of Fat and Oil Differently Composed of ${\omega}3\;and\;{\omega}6$ Polyunsaturated Fatty Acids on Lipid Metabolism of Rats -Centered on Brain, Kidney, Testis and Serum Lipoprotein- (${\omega}-3$계와 ${\omega}-6$계 고도 불포화 지방산의 혼합 비율이 다른 유지가 흰쥐의 지질대사에 미치는 영향(I) -뇌, 신장, 고환 및 혈청 lipoprotein을 중심으로-)

  • This research was intended to know the influence of ${\omega}3\;and\;{\omega}6$ polyunsaturated fatty acids on rat's brain, kidney, testis tissues and composition of serum lipoprotein. Different kinds of diets containing perilla oil (PO) and red pepper oil (RPO) were administered to the rats: group 1, 10% PO; group 2, 7.5% PO and 2.5% RPO ; group 3, 5% PO and 5% RPO ; group 4, 2.5% PO and 7.5% RPO ; group 5, 10% RPO. The following are the results. 1. No significance of cholesterol concentration in brain was noticed among the groups, but group 5 was the highest and other groups tended to decrease. 2. As for concentration of triglyceride and phospholipid, the groups with more percentage of RPO increased gradually while those with more PO decreased by degrees. 3. No significance of cholesterol level in kidney was found among the groups. About triglyceride level, group 5 was the highest and it was relatively low in groups2, 3 and 4. group 5, however, was the lowest in phospholipid level. 4. As to cholesterol concentration in testis, groups 1 to 4 were low. About triglyceride level, group 1 increased while groups 2 to 4 decreased. 5. Concerning lipoprotein pattern in serum, HDL increased in groups 1 to 3, while LDL decreased gradually with the increase of perilla oil.

The Effect of Acyl Chain Structure on the Hydrolysis of Fatty Acids from Fish Oil by Lipase-OF 360,000 (아실체인 구조가 효소 Lipase-OF 360,000에 의한 지방산의 가수분해 특성에 미치는 영향)

  • The hydrolysis characteristics of each fatty acid composing the fish oil by means of the lipase from Candida cylindracea was investigated. The saturated fatty acids, C14:0, C16:0 and C18:0, and the unsaturated fatty acids with one double bond, C16:1, C18:1(n-7), C18:(n-9), C20:1 and C22:1 were more easily hydrolyzed than the $\omega$-3 polyunsaturated fatty acids. when the number of carbon of the $\omega$-3 fatty acids was same but that of double bond was different, the hydrolysis of the $\omega$-3 fatty acids having lower number of double bond was more rapidly carried out. When the degree of polyunsaturation was same but the number of carbon was different, the lipase acted more rapidly upon the $\omega$-3 fatty acids with lower number of carbon. Docosahexaenoic acid(DHA) was most highly concentrated in the glyceride mixture after 120hr hydrolysis among the various $\omega$-3 polyunsaturated fatty acids. The weight percentage of DHA to the $\omega$-3 fatty acids in the fish oil was 31.87% but that in the glyceride mixture after 120hr hydrolysis was 51.89%.

  • This study was designed to examine the effect of ${\omega}-3$ fatty acid, linlenic acid, EPA, DHA on serum lipid and cytokines of male rats(Sprague-Dawley). Animals of 3 groups were administrated perilla oil, salmoon oil, and tuna oil of 0.4 $m{\ell}/day$ for 8 weeks respectively. These oils were used for a source of linolenic acid, EPA and DHA. ${\omega}-3$ polyunsaturated fatty acid decreases significantly body weight, serum $PGE_2$ content and serum cytokines content of the rat, and increases internal organs weight, specially liver weight and serum HDL-cholesterol level of the rat. In the results, authors propose to use perilla oil for source of effective ${\omega}-3$ poly-unsaturated fatty acid(linolenic acid) to Prevent cardiovascular and immune diseases.

  • Omega-3 fatty acids (eicosapentaenoic acid [EPA] and docosahexaenoic acid [DHA]) may be beneficial for the primary and secondary prevention of cardiovascular events (CVEs), especially in patients with myocardial infarction or heart failure with reduced ejection fraction. For this purpose, one to two seafood meals per week is preferentially recommended. Omega-3 fatty acids with a high-dose EPA formula (4 g/day) may be more effective than EPA+DHA mixed supplements for the secondary prevention of CVE. Krill oil also contains omega-3 fatty acids, but at a much lower dose compared to fish oil. Supplemental vitamins and minerals have not shown the preventive effects on CVE in prospective, and randomized clinical trials, except for one Chinese study showing the stroke prevention effects of folic acid. The clinical benefit of chelation therapy in reducing CVEs is uncertain.

Identification of Fatty Acids in the Pulp Oils of Jujube and Their Compsitional Changes in the Ripening Period (대추의 과육지질(果肉脂質)에 존재(存在)하는 지방산(脂肪酸)의 동정(同定)과 숙성(熟成)에 따른 그 조성(組成)의 변화(變化))

  • In search for several fatty acid with unusual structure in vegetable oils, we have found that unknown peaks were shown on GLC in the analysis of fatty acids of the lipids from the pulp of ripened jujube (Zizypus jujuba var. inermis) fruits. These fatty acids were identified as a series of cis-monoenoic acids with ${\omega}-5$ double bond system such as $C_{14:1{\omega}5}$, $C_{16:1{\omega}5}$ and $C_{18:1{\omega}5}$, including ${\omega}-7$ fatty acid as $C_{16:1{\omega}7}$ and $C_{18:1{\omega}7}$, by GLC, solid-phase extraction silver ion-column chromatographic, GLC-mass spectrometric and IR techniques. First of all, total fatty acid methyl esters were resolved into saturated and branched fatty acid, monoenoic acid, dienoic acid, and trienoic acid fraction, respectively, with 100% dichloromethane (DCM), DCM/acetone (9:1, v/v) 100% acetone, and acetone/ acetonitrile (97:3, v/v) solvent system. Unknown fatty acids were included in the monoenoic fraction and were confirmed to have cis-configuration by IR. Picolinyl esters of monoenoic fatty acids gave distinct molecular ion peak and dominant diagnostic peaks, for example, m/z 317, 220 and 260 fragment for $cis-C_{14:1{\omega}5}$, m/z 345, m/z 248 and 288 fragment for $cis-C_{16:1{\omega}5}$ and m/z 373, m/z 276 and 316 fragment for $cis-C_{18:1{\omega}5}$. In this way the occurrence of $cis-C_{16:1{\omega}7}$ and $cis-C_{18:1{\omega}7}$ could be deduced from the appearance of prominent fragments as m/z 345, 220 and 260, and m/z 373, 248 and 280. Level of total ${\omega}-5$ fatty acids amounted to about 30% in the fatty acid composition with the predominance of $C_{16:1{\omega}5}$ $ (18.7{\sim}25.0%)$, in the semi-ripened and/or ripened samples collected in September 14 ($C_{16:1{\omega}5}$ ; 18.7%, $C_{14:1{\omega}5}$ ; 3.6% and $C_{18:1{\omega}5}$ ; 3.0%), September 22 ($C_{16:1{\omega}5}$ ; 25.0%, $C_{14:1{\omega}5}$ ; 1.4% and $C_{18:1{\omega}5}$ ; 2.6%), and October $7 (C_{16:1{\omega}5}$ ; 24.7%, $C_{14:1{\omega}5}$ ; 7.7% and $C_{18:1{\omega}5}$ ; 2.5%). However, the lipids extracted from unripened jujube in July and August contain these unusual fatty acids as low as negligible. It could be observed that the level of ${\omega}-5$ fatty acids in the pulps increased sharply with an elapse of ripening time of jujube fruits. Other monoenoic fatty acids with ${\omega}-7$ series, $C_{16:1{\omega}7}$ (palmitoleic acid) and $C_{18:1{\omega}7}$ (cis-vaccenic acid) could be detected. And in the lipids of the kernel and leaf of jujube, none of ${\omega}-5$ fatty acids could be detected.

Identification of ${\omega}$-Aminotransferase from Caulobacter crescentus and Sitedirected Mutagenesis to Broaden Substrate Specificity

  • A putative ${\omega}$-aminotransferase gene, cc3143 (aptA), from Caulobacter crescentus was screened by bioinformatical tools and overexpressed in E. coli, and the substrate specificity of the ${\omega}$-aminotransferase was investigated. AptA showed high activity for short-chain ${\beta}$-amino acids. It showed the highest activity for 3-amino-n-butyric acid. It showed higher activity toward aromatic amines than aliphatic amines. The 3D model of the ${\omega}$-aminotransferase was constructed by homology modeling using a dialkylglycine decarboxylase (PDB ID: 1DGE) as a template. Then, the ${\omega}$-aminotransferase was rationally redesigned to increase the activity for 3-amino-3-phenylpropionic acid. The mutants N285A and V227G increased the relative activity for 3-amino-3-phenylpropionic acid to 3-amino-n-butyric acid by 11-fold and 3-fold, respectively, over that of wild type.

Effect of heat treatment on the electrical and optical properties of ZnO : Al thin films prepared by reactive magnetron sputtering method (반응성 sputtering법으로 제막된 ZnO : Al 박막의 전기.광학적 특성에 미치는 열처리의 영향)

  • AZO transparent conducting thin films were fabricated by reactive DC magnetron sputtering method using Zn metla target containing 2 wt% of Al, and electrical and optical properties were investigated after heattreatment. Electrical resistivity was reduced 50% and had reached $1{\times}10^{-3}~3.5{\times}10^{-4}\;{\Omega}cm$ by heat treatment. In the case of oxide AZO films, the resistivity of $10^{3}\;{\Omega}cm$ was also decreased to $2{\times}10^{-3}\;{\Omega}cm$ after heat treatment. The optical transmittance of AZO films deposited in the transition range was increased from 59.4 % to 77.4 % by $400^{\circ}C$, 30 min heat treatment.

Nonlinear Analysis of a Forced Circular Plate with Internal Resonance (내부공진을 가진 원판의 비선형 강제진동해석)

  • An analysis is presented for the combination resonance of a clamped circular plate, which occurs when the frequency of the excitation is near the combination of the natural frequencies, that is, when ohm.=2.0mega./sub 1/+omega./sub 2/. The internal resonance, Omega./sub 3/=omega./sub 1/+2.omega./sub 2/, is considered and its influence on the response is studied. The clamped circular plate experiencing mid-plane stretching is governed by a nonlinear partial differential equation. By using Galerkin's method the governing equation is reduced to a system of nonautonomous ordinary differential equations. The method of multiple scales is used to obtain steady-state responses of the system. Results of numerical investigations show that the increase of the excitation amplitude can reduce the amplitudes of steady-state responses. We can not find this kind of results in linear systems.

Omega 3 fatty acids as a host modulator in chronic periodontitis patients: a randomised, double-blind, palcebo-controlled, clinical trial

  • Purpose: Periodontitis is an infectious disease caused predominantly by gram-negative anerobes. The host inflammatory response to these bacteria causes alveolar bone loss that is characterized as periodontitis. Omega-3 fatty acids (${\omega}$-3 FAs) have anti-inflammatory properties, thus have been used to treat some chronic inflammatory diseases such as cardiovascular disease and rheumatoid arthritis. We aimed to evaluate the effect of dietary supplementation with ${\omega}$-3 FAs as a host modulating agent in patients with chronic periodontitis. Methods: Sixty otherwise healthy subjects with moderate and severe chronic periodontitis were enrolled in our randomised, double-blind, placebo-controlled trial. The control group (CG, n=30) was treated with scaling and root planing (SRP) and given a placebo; the treatment group (TG, n=30) was treated with SRP and dietary supplementation of ${\omega}$-3 FAs (one 300 mg tablet daily for 12 weeks). Periodontal clinical parameters and serum C-reactive protein (CRP) levels were evaluated in all patients at baseline, a 6-week and 12-week period after treatment. Results: A significant reduction in the gingival index, sulcus bleeding index, pocket depth, and clinical attachment level was found in the TG compared to the CG at a 12-week period. However, no statistically significant changes in serum CRP levels were found. Conclusions: Our findings suggest that ${\omega}$-3 FAs can successfully reduce gingival inflammation, pocket depth, and attachment level gain. Dietary supplementation with ${\omega}$-3 FAs may have potential benefits as a host modulatory agent in the prevention and/or C management of chronic periodontitis.
