데이터 마이닝을 이용한 제주 양식 넙치(Paralichthys olivaceus)의 스쿠티카증 발생 패턴 분석 (Data Mining for Scuticociliatosis Outbreak Patterns in Cultured Olive Flounder Paralichthys olivaceus in Jeju, Korea)

  • In the aquaculture industry, few studies are analyzing big data for intrinsic meaning. Fishcare Laboratory (www.fishcare.kr) diagnostic data from 2016-2018 was analyzed for scuticociliatosis (caused by Miamiensis avidus) outbreak patterns in cultured olive flounder Paralichthys olivaceus in Jeju, Korea. The scuticociliatosis monthly occurrence ratio is reported in the summary table after preparing and filtering the basic dataset model. Nonparametric test results suggest differences in the water temperature, body length, and weight between groups with and without scuticociliatosis. Data distribution visualization revealed that shorter body length and lighter weight increased the occurrence of scuticociliatosis. The association rule mining technique was applied to determine the primary clinical signs of mixed scuticociliatosis and bacterial infections. Venn diagrams were used to report clinical signs and suggest commonalities. These results may help diagnose and treat fish and provide a decision-making reference.

  • Effect of mercury (Hg) toxicity on survival, growth, feed efficiency and oxygen consumption were examined in the juvenile olive flounder. Paralichthys olivaceus. Fishes were exposed to sublethal concentrations of Hg ranging from 0 to 0.13mg/L for 6 weeks. Hg reduced survival rate in a concentration and exposure period-dependent way and suddenly reduction occurred at Hg concentrations greater than 0.05mg/L after 6 weeks. Growth rate and feed efficiency also significantly decreased at greater than 0.028 and 0.05 mg/L respectively. Oxygen consumption rate was significantly decreased to 25 and 32% than that of the control at the Hg concentration of 0.05 and 0.13 mg/L respectively. These results suggest that Hg toxicity inhibit physiological function including growth, feed efficiency and oxygen consumption in the juvenile olive flounder, resulting in survival failure at high concentration.

  • We determined the optimal culture conditions for obtaining the maximum number of intestinal bacteria from the olive flounder Paralichthys olivaceus, and studied bacterial diversity using both culture-dependent and culture-independent methods. Using six culture conditions, mean bacterial numbers were greater than $10^6$ per gram of gut mucus, regardless of the medium. However, the bacterial diversity, based on colony morphology, appeared much higher on Marine agar (MA) and Zobell 2216 agar than on other media. We found eight and 17 cultured bacterial phylotypes with 99% minimum similarity in gut mucus grown on MA and tryptic soy agar, respectively. Furthermore, we used genomic DNA extracted from gut mucus to generate 78 random clones, which were grouped into 25 phylotypes. Of these, six were affiliated with Firmicutes, Actinobacteria, and Verrucomicrobia, and were not found using our culture-dependent methods. Consequently, we believe that Marine agar and Zobell 2216 agar are optimal media for culturing diverse intestinal microbes; we also discovered several novel sequences not previously recognized as part of the gut microbiota of olive flounder.

Cu Accumulation and Elimination in the Tissues of the Olive Flounder Paralichthys olivaceus

  • Olive flounder, Paralichthys olivaceus were exposed to waterborne Cu (control, 50, 80, 150, and 320 ${\mu}g$/L) for 30 days and then depurated for 20 days to investigate the effects of waterborne Cu exposure on growth, accumulation, and elimination. The weight-specific growth rate was significantly negatively related to waterborne Cu concentrations at 150 and 320 ${\mu}g$/L. The order of Cu accumulation in different tissues of exposed fish was liver>intestines>gills>kidneys>muscle, suggesting that the liver is more important than other tissues for the storage of Cu in olive flounder. The accumulation factor for the gills, intestines, liver, and muscle increased with increasing exposure time, and accumulation was negatively related to exposure concentration for the gills, kidneys, and muscle. Cu concentrations in the gills, intestines, and liver continuously decreased for 20 days of depuration. The fastest elimination rate occurred in the intestines at all exposure concentrations, and the order of Cu elimination in the different tissues was intestines>liver>gills.

  • This study was investigated the survival and hatching rate of fertilized eggs in olive flounder Paralichthys olivaceus at water temperature, salinity, duration of transport and different stock densities during commercial transport. The observed optimal temperature during transport was 17.5℃ similar to the natural environmental conditions. The proper salinity was observed to be at 33 psu (practical salinity unit) in both surviving and hatching rate. In terms of the duration of transport, there were no differences until 12 hours between survival and hatching rate, but shorter time of transport would be better. With the best conditions during transport, 20,000 eggs/L were handled in both survival and hatching rate. These results can be essential in the production and transport of healthy olive flounder seeds.

Expressed Sequence Tag Analysis of Olive Flounder (Paralichthys olivaceus): Genes and Expression Profile from the Liver

  • Expressed sequence tag (EST) analysis was conducted using a cDNA library made from the liver mRNA of olive flounder (Paralichthys olivaceus). In the survey of 421 ESTs, 362 showed significant homology to previously described genes while 59 were unidentified or novel. Comparative analysis of the identified ESTs showed that 69 $(19.0\%)$ clones were identified as homologous to the previously reported olive flounder ESTs, and 279 $(77.1\%)$ clones were identified as orthologs of known genes from other organisms. The remaining 14 $(3.9\%)$ clones were identified as orthologs of known sequences with unknown functions. These tagged cDNA clones, identified and unidentified, could provide fundamental baseline data for genomic studies of this species.

  • Streptococcal diseases are known as serious problems in fresh water and marine fish culture industry worldwide. Recently, the importance of the infection of Streptococcus parauberis has been increased among the streptococcicosis since severe outbreaks in cultured olive flounder (Paralichthys olivaceus) were recorded in Korea. The aims of the present study were to investigate immunomodulatory differences between S. parauberis and S. iniae. In this study, S. parauberis isolated from diseased olive flounder was investigated on the characteristics of morphological and immune responses. Immune response of the fish to the pathogen was characterized by bactericidal activity in the serum, phagocytic activity and reactive oxygen intermediaters (ROIs) production of olive flounder. The capsules of both bacteria, S. parauberis and S. iniae seem to be associated with the ability to resist killing activity of normal sera. Also, capsulated strains could survive in the phagocytes and induce to lower ROIs production.

  • We determined the resistance rates of pathogenic bacteria isolated from cultured olive flounder Paralichthys olivaceus to erythromycin (Em), antibiotic typically used in aquaculture and analyzed the genotypes of resistant bacteria using polymerase chain reaction (PCR). We isolated and utilized 160 isolates of Streptococcus parauberis, 1 of S. iniae, 66 of Edwardsiella tarda, 56 of Vibrio sp. and 23 of unidentified bacteria from presumed infected olive flounder from Jeju Island from March 2016 to October 2017. Of the 306 isolated strains, Em-resistant strains included 33 of S. parauberis, 39 of E. tarda and 2 of Vibrio sp. We conducted PCR to assess the resistance determination of Em-resistant strains. Five different types of Em-resistance genes were detected in the 74 Em-resistant strains: erm (A), erm (B), erm (C), mef (A) and mef (E); erm (A) and erm (B) were detected in 1 (3%) and 24 (72.7%) S. parauberis isolates, respectively. In E. tarda, erm (B) was detected in five isolates (12.8 %) and no Em-resistance genes were detected in the two Vibrio sp. isolates.

  • 넙치의 생존, 성장 및 대사에 미치는 납의 만성적 독성을 파악하기 위하여 6주 동안 납 0, 80, 150, 230, 420 ${\mu}g/L$농도에 노출시켜 실험을 실시하였다. 넙치의 생존율은 납 농도 230 ${\mu}g/L$이상에서 유의한 감소를 나타냈다. 넙치의 성장률과 사료효율은 납 농도가 증가할수록 감소를 하여 150 ${\mu}g/L$이상의 농도에서는 대조구에 비하여 유의한 감소를 나타냈다. 넙치의 대사율은 납 농도 80, 150, 230, 420 ${\mu}g/L$에서 대조구보다 각각 21.4, 23.8, 36.8, 44.9 %가 저하했으며, 80 ${\mu}g$/L이상의 농도에서는 유의한 감소를 나타냈다.

Identification of Vibrio species isolated from cultured olive flounder (Paralichthys olivaceus) in Jeju Island, South Korea

  • Olive flounder (Paralichthys olivaceus) is the major species developed for aquaculture in South Korea. Over the long history of olive flounder aquaculture, complex and diverse diseases have been a major problem, negatively impacting industrial production. Vibriosis is a prolific disease which continuously damages olive flounder aquaculture. A bacterial disease survey was performed from January to June 2017 on 20 olive flounder farms on Jeju Island. A total of 1710 fish were sampled, and bacteria from the external and internal organs of 560 fish were collected. Bacterial strains were identified using 16 s rRNA sequencing. Twenty-seven species and 184 strains of Vibrio were isolated during this survey, and phylogenetic analysis was performed. Bacterial isolates were investigated for the distribution of pathogenic and non-pathogenic species, as well as bacterial presence in tested organs was characterized. V. gigantis and V. scophthalmi were the dominant non-pathogenic and pathogenic strains isolated during this survey, respectively. This study provides data on specific Vibrio spp. isolated from cultured olive flounder in an effort to provide direction for future research and inform aquaculture management practices.
