Analysis of Fatty Acid in Rice Bran Oil by Gas Chromatography (Gas Chromatography에 의(依)한 미강유(米糠油)의 지방산분석(脂肪酸分析))

  • Through an experiment with gas chromatography carried out using diethylene glycol succinate(DEGS) as the packing material of the column, we have obtained the correction factor between the weight ratio and the peak dimension of the saturated fatty acid methyl esters of C10, C12, C14, C16, and C18 and unsaturated fatty acid methyl esters of oleic acid, linoloic acid, and linolenic acid, employing the detector of thermal conductivity type. Quantitative analysis of the fatty acids contained in rice Bran oil was performed with the above correction factor and the results are as follows; 1. Main components were found to be palmitic acid, oleic acid sand linolenic acid. No trases of capric acid (C10) lauric acid (C10) were found. 2. It was confirmed that there were straight line relation between the logarism retention time of each fatty acid and the number of carbon of saturated fatty acid or the number of double bond of other fatty acids having the same number of carbon. 3. The correction factor became larger as to the number of carbon increased up to C18 in case of saturated fatty acids, end as for other fatty acids, and as for other fatty acids of the same carbon number, it became larger according as the number of double bond increased.

  • Enzymatic synthesis and purification of sugar esters using supercritical $CO_2$ were investigated. The observed yield of suagr ester produced by transesterification of methyl glucoside and oleic acid using a lipase(Novozym 435) was 67% at 24 hours of reaction in the supercritical $CO_2$. The solubility of the fatty acids in the supercritical $CO_2$ was measured to find the conditions of supercritical separation of the fatty acids from the reaction mixture. The solubility of capric and oleic acid was 5 ${\times}$ $10^{-4}$, 1.7 ${\times}$ $10^{-4}$ mol/mol $CO_2$, respectively, varying at $35{\sim}50^{\circ}C$ and 80${\sim}$120 atm.

  • Quantitative analysis of the fatty acids contained in Duck meat was carried out by the Gas Chromatography with Flame ionization Detector, The general components and chemical constants have been performed with A.O.A.C. methods. The results art summarized as follows : 1. General composition of Duck meat come out to be 64.87% moisture, 19.06% protein, 17.05% fat, and 1.02% ash. 2. It was investigated that extraction of lipids were performed by Soxhlet extractor for 12 hours. Amounts of lipids were extracted 79.57% in ethylether, 70.15% in chloroform, and 72.35% in n-hexane. 3. Chemical constants of lipids in Duck meat were obtained as follows : Saponification number 201.5, Acid number 5.01, Iodine number 50.1 and Carbonyl number 4.5 4. It was investigated that the fatty acid component were quantitatively determined by the gas chromatography : Linolenic acid 1.6%, Linoleic acid 19.9%, Oleic acid 45.9%, Stearic acid 3.1% Palmitic acid 17.2% and Myristic acid 0.12% in leg portion. Linolenic acid 1.7% Linoleic acid 17.2%, Oleic acid 51.2%, Stearic acid 3.3%, Palmitic acid 17.1% and Myristic acid 0.17% in breast portion. 5. Cholesterol of blood, breast and leg portion fat in Duck were obtained as follows : Total cholesterol 200 mg%, 260 mg% , and 400 mg% respectively; cholesterol ester 120mg%, 151 mg%, and 240mg% respectively.

Fatty Acid and Volatile Oil Compositions of Allomyrina dichotoma Larvae

  • Thirty-two different volatile oils were identified from Allomyrina dichotoma (A. dichotoma) larvae by gas chromatography/mass spectrometry (GC/MS). The major volatile components were 2,2,4-trimethyl-3-carboxyisopropyl pentanoic acid isobutyl ester (5.83%), phenol,2,6-bis(a,a-dimethyl ethyl)-4-(1-methyl-1-phenylethyl) (5.72%), heptacosane (5.49%) and phenol,2,4-bis(1-methyl-1-phenylethyl) (5.47%). The composition of the fatty acids in A. dichotoma larvae was also determined by gas chromatography (GC) and fourteen constituents were identified. Oleic acid (19.13%) was the most abundant fatty acid followed by palmitic acid (12.52%), palmitoleic acid (3.71%) and linoleic acid (2.08%) in 100 g of A. dichotoma larvae on a dry weight basis. The quantity of unsaturated fatty acids (64.00%) were higher than that of saturated ones (36.00%). The predominant fatty acids in A. dichotoma consist of monounsaturated fatty acid (MUFA, 57.70%) such as oleic acid, myristoleic acid and palmitoleic acid, followed by saturated fatty acids (36.00%) and polyunsaturated fatty acids (PUFA, 6.50%). In particular, the presence of essential fatty acids, such as linoleic (5.30%) and linolenic acid (0.40%) give A. dichotoma larvae considerable nutritional and functional value and it may be a useful source for food and/or industrial utilization.

Evaluation of Contact Lens Care Regimens in Removing Tear Lipids on Silicone Hydrogel Lens (콘택트렌즈 관리방법에 의한 실리콘하이드로겔렌즈의 지방침전물 제거효과)

  • Purpose: An in vitro study was conducted to compare the lipid cleaning efficacy of care solutions on balafilcon A silicone hydrogel (SiHy) lens. Methods: Lipid spoilation was performed by incubating balafilcon A SiHy lenses in phosphate buffered saline (PBS) containing oleic acid, oleic acid methyl ester and cholesterol. Spoiled contact lenses rinsed with PBS were cleaned with surfactant cleaner, alcohol containing cleaner and multipurpose solution (MPS) respectively and repetitive spoilation and cleaning was conducted up to 14 times. To observe the cleaning effect of ultrasonic wave on the lipid deposit, each spoiled lens was ultrasonicated and then compared with non-sonicated lens. Lipids deposit on the contact lenses was extracted by methanol:chloroform (1:1, v/v) solution. High performance liquid chromatography was used to analyze and quantify lipid deposit extracts. Results: The effectiveness of alcohol containing surfactant cleaner on the lipid deposits was better than that of surfactant cleaner and MPS, and the cleaning efficacy was significantly higher in the ultrasonic wave treated group. Lipid deposits were not removed completely by contact lens care solutions so that lipid deposits increased continuously and cumulatively. Conclusions: The cleaning efficacy of contact lens care solutions was not satisfactory to remove lipid deposits on the SiHy lens that new cleaning products specially designed for SiHy lenses are needed to develop.

Analysis of Fatty Acid Compositions and Biodiesel Properties of Seeds of Woody Oil Plants in Korea (국내 목본 유지식물 종자의 지방산 조성 및 바이오디젤 특성 분석)

  • In order to evaluate their potential as sources of biodiesel, oil content and fatty acid composition of seeds and fatty acid methyl ester (FAME) properties from seven woody oil plants in Korea were analysed. The oil content of seed of all woody plant species ranged from 15.1 (Ligustrum lucidum) to 70.3% (Camellia japonica) by dry weight. Fatty acid composition consisted mainly of oleic acid, linoleic acid, linolenic acid, palmitic acid and stearic acid, with oleic acid being the most abundant. The content of unsaturated fatty acids of all species was higher than saturated fatty acids. Oxidation stability of seed oils of all woody plants ranged from 2.25 to 8.62 hours/$110^{\circ}C$. Fatty acid methyl ester of Styrax japonica has been found to have the highest iodine value, indicating that unsaturated fatty acid content is higher than other seed oils. Cold filter plug point(CFPP) was varied over a wide range from $0^{\circ}C$ to $-13^{\circ}C$. The cold fluidity of FAME of Chionanthus retusa were excellent.

Effect of Supplement of Korean Pinenut Oil on Plasma Total Fatty Acid Composition in Cholesterol-fed Rabbits (한국산 잣기름이 콜레스테롤 첨가식이로 사육한 토끼의 혈장 총지방산 조정에 미치는 영향)

  • The present study was carried out in atheroscleorotic New Zealend white(NZW) rabbits. to evaluate the effect of dietary supplementation with Korean pinenut oil, on plasma total fatty acid composition. In study I, NZW rabbits were fed 10 weeks on a commercial chow diet supplemented with 5% of energy as fats(soybean oil or pinenut oil) or 10% of energy as fats(soybean oil or pinenut oil) with the addition of 1% cholesterol to the diet. Nineteen fatty acids ranged from myristic acid (14:0) to cervonic acid (22:6 ${\omega}3$) were identified in all the samples. The c5, c9, $c12{\sim}18$ : 3 acid was not reported in the fatty acid methyl ester profiles of each group because it was included in the linoleic acid peak. The major constitutent fatty acids in the chow diet group were linoleic acid, oleic acid, palmitic acid and ${\alpha}$-linolenic acid. In the cholesterol group, oleic acid, linoleic acid and palmitic acid were the major fatty acids. In plasma of cholesterol-fed animals, the levels of 16:1 ${\omega}$ 7 and 18:1 1 ${\omega}$ 9 were increased. Linoleic acid was the major fatty acid in soybean oil/cholesterol and pinenut oil/cholesterol groups. Plasma linoleic acid levels were significantly incresed from 4 to 6% by the supplementation of 5% soybean or 5% pinenut oil in the cholesterol diet for 5 weeks, compared to cholesterol group. Plasma 16 : 1 ${\omega}$ 7 levels in animals fed with 5 or 10% pinenut oils were significantly lower than in those fed cholesterol for 5 weeks. After 10 weeks on the soybean oil and pinenut oil diet there were no significant differences in the fatty acid composition. In study II, the fatty acid composition was not affected by the types or levels of oils supplemented for 5 weeks. After 10 weeks on the oil diets 16:1 ${\omega}$ 7 and 18:1 ${\omega}$ 9 were decreased in 10% soybean in oil/cholesterol and 10% pinenut oil/cholesterol groups, compared to cholesterol group.

The influence of the way of fat recovery from fleshing scrap on the acid value and fatty acid composition (플레싱 스크랩으로부터 유지 회수 방법이 산가 및 지방산 조성에 미치는 영향)

  • Fleshing scrap is a kind of wastes produced during leather making process and used in the test of manufacturing biodiesel. The early step of manufacturing biodiesel is fat recovery from fleshing scrap. Hence, we investigated the influence of the way of fat recovery on the fatty acid composition. We used three different recovery ways, that is chemical method by protein decomposition with acid/fat recovering, physical method by protein denaturalization with heat and vacuum/fat pressing, and biodiesel method by protein decomposition/fat recovering. The biological method yielded the best results in terms of appearance transparency. It was most effective to lower acid value. Also the recovered fat by biological method would be favorable methyl-ester reaction raw material for biodiesel because it contains more than 5% of oleic acid among unsaturated fatty acid.

A Kinetic Study on the Esterification of Oleic Acid with Methanol in the Presence of Amberlyst-15 (Amberlyst-15 촉매의 존재 하에서 올레산과 메탄올의 에스테르화 반응 속도식 연구)

  • The esterification reaction of free fatty acid with methanol was investigated in the presence of catalyst, Amberlyst-15, producing fatty acid methyl ester, namely, biodiesel. In this paper, the effects of the reaction parameters such as reaction temperature, mole ratio of alcohol to oleic acid and mass of catalyst on the catalytic activity have been examined. The results showed that the reaction rate increased about twice as the temperature increased every $20^{\circ}C$ in the reaction temperature range from 333 K to 373 K. The equilibrium conversion rate of oleic acid increased with the feed mole ratio of alcohol to acid ranging from 6:1 to 44:1. When the feed mole ratio was higher than 44:1, all the results were similar to that of 44:1. As for the influence of the mass of catalyst, the initial reaction rate increased from 1.2 to 1.3 times as the mass of catalyst doubles in the range of the catalyst weight from 5 to 20 wt%. The experiment data obtained were well described by the second reaction rate using a pseudo-homogeneous model.

Comparison of Promoters Suitable for Regulated Overexpression of $\beta$-Galactosidase in the Alkane-Utilizing Yeast Yarrowia lipolytica

  • Promoters of the genes G3P, ICL1, POT1, POX1, POX2 and POX5 of the yeast Y. lipolytica were studied in respect to their regulations and activities during growth on different carbon sources. The aim of this study was to select suitable promoters for high expression of heterologous genes in this yeast. For this purpose the promoters were fused with the reporter gene lacZ of E. coli and integrated as single copies into the genome of Y. lipolytica strain PO1d. The measurement of expressed activities of ${\beta}$-galactosidase revealed that pICL1, pPOX2 and pPOT1 are the strongest regulable promoters available for Y. lipolytica, at present. pPOX2 and pPOT1 were highly induced during growth on oleic acid and were completely repressed by glucose and glycerol. pICL1 was strongly inducible by ethanol besides alkanes and fatty acids, however, not completely repressible by glucose or glycerol. Ricinoleic acid methyl ester appeared as a very strong inducer for pPOT1 and pPOX2, in spite of that it inhibited growth of Y. lipolytica transformants.
