Study on the Subacute Toxicity of Complex of Pine Needle Oil and Korean Medicinal Herbs against Rats

  • Pine needle oil and Korean medicinal herbs (KMH) are known as effective therapeutic agents on various blood vessel disease. We have already reported the ameliorative effect of complex of pine needle oil and Korean medicinal herbs against hyperlipidemia. But safety and non - toxicity of pine needle oil and Korean medicinal herbs to normal animal cells have not been studied clearly. In this study, we investigated whether pine needle oil and Korean medicinal herbs show side effects on rat or not. These materials were administered to rats, and subacute toxicity was examined by measuring the hematological values, CBC differentiation, biochemical levels of blood (TP, total protein; albumin; ALP, alkaline phosphotase; AST, aspatate aminotrans- ferase; ALT, alanine aminotransferase; T-Chol., total cholesterol; T-Bil., total bilirubin) and urine analysis, suggesting that the sample have no side effects and cytotoxicity. These results indicate that the complex of pine needle oil and Korean medicinal herbs may effective non- toxic, safety therapeutic agents on hepatocytes and hyperlipidemia.

Safety Evaluation of Chrysanthemum indicum L. Flower Oil by Assessing Acute Oral Toxicity, Micronucleus Abnormalities, and Mutagenicity

  • Chrysanthemum indicum is widely used to treat immune-related and infectious disorders in East Asia. C. indicum flower oil contains 1,8-cineole, germacrene D, camphor, ${\alpha}$-cadinol, camphene, pinocarvone, ${\beta}$-caryophyllene, 3-cyclohexen- 1-ol, and ${\gamma}$-curcumene. We evaluated the safety of C. indicum flower oil by conducting acute oral toxicity, bone marrow micronucleus, and bacterial reverse mutation tests. Mortality, clinical signs and gross findings of mice were measured for 15 days after the oral single gavage administration of C. indicum flower oil. There were no mortality and clinical signs of toxicity at 2,000 mg/kg body weight/day of C. indicum flower oil throughout the 15 day period. Micronucleated erythrocyte cell counts for all treated groups were not significantly different between test and control groups. Levels of 15.63~500 ${\mu}g$ C. indicum flower oil/plate did not induce mutagenicity in S. Typhimurium and E. coli, with or without the introduction of a metabolic activation system. These results indicate that ingesting C. indicum flower oil produces no acute oral toxicity, bone marrow micronucleus, and bacterial reverse mutation.

랏트에 대한 Polychlorinated Biphenyl의 독성에 미치는 $\alpha$-Tocopherol과 Perilla oil의 효과 (Effects of $\alpha$-Tocopherol and Perilla oil on the Toxicity of Polychlorinated biphenyl in Rat)

  • Effects of ${\alpha}$-tocopherol and perilla oil on the toxicity of polychlorinated biphenyls (PCB) in male rat were studied. Rats were fed ad libitum for 6 weeks with the animal diet which contains PCB 30 ppm and 100 ppm. Perilla oil (0.5 g/kg body weight) and ${\alpha}$-tocopherol (30 mg/kg body weight) were administered intraperitoneally twice a week for 6 weeks. Rats fed with PCB showed enlargement of liver and spleen, increase in aspartate aminotransferase, alkaline phosphatase, sereum lipid and cytochrome P 450 and decrease in body weight and glutathione. When perilla oil was administered to rats fed with PCB increase in aspartate aminotransferase, alkaline phosphatase, serum lipid and cytochrome P45O and decrease in body weight and glutathione were significantly augmented, compared to rats fed with PCB alone. This means that perilla oil potentiates the toxicity of PCB. On the other hand when ${\alpha}$-tocopherol was administered to rats fed with PCB increase in aspartate aminotransferase, alkaline phosphatase, serum lipid and cytochrome P45O and decrease in body weight and glutathione were signigicantly reduced, compared to rats fed with PCB alone. This means that u-tocopherol reduces the toxicity of PCB. From the above results, it may be concluded that PCB is metabolized by microsomal mixed function oxidase and the metabolite causes the toxicity and microsomal glutathione plays a role of protection on the toxicity of PCB.

Effects of ultraviolet radiation on the toxicity of water-accommodated fraction and chemically enhanced water-accommodated fraction of Hebei Spirit crude oil to the embryonic development of the Manila clam, Ruditapes philippinarum

  • The purpose of this study is to evaluate the effects of oil dispersant and ultraviolet (UV) radiation on the toxicity of crude oil. The toxicity of water-accommodated fraction (WAF) and chemically enhanced water-accommodated fraction (CEWAF) of Hebei Spirit crude oil was investigated in the embryo of the Manila clam, Ruditapes philippinarum with- and without ultraviolet radiation. The WAF and CEWAF with- and without UV radiation affected significantly the embryonic development of R. philippinarum. The EC50s of WAF without UV, WAF with UV, CEWAF without UV, and CEWAF with UV were 2.82, 0.79, 1.60, and 0.45 g/L, respectively. CEWAF was 1.6 times more toxic than WAF. UV radiation increased crude oil toxicity to 3.6 times for both WAF and CEWAF. The oil dispersant and UV radiation did not affect the acute toxicity to the embryo but retarded the period of embryonic development up to 26%. R. philippinarum proved to be a sensitive species to reflect the toxic effects of oil spill combined with oil dispersant and UV radiation. It is suggested that the chemical analyses on the WAF and CEWAF is important for the identification and quantitative explanation of the phototoxic compounds in crude oil.

홍화자약침(紅花子藥鍼)의 피부자극시험 및 안점막자극시험 (The study of irritation and toxicity of Carthami oil aquapuncture solution applied topically to the skin and the eye mucous membrane)

  • Carthami tinctorii Fructus is known for its good effect on diseases such as fracture, menorrhalgia, menoschesis, puerperium aneilema and so on. The study of irritation and toxicity of Carthami oil aquapuncture solution applied topically to the skin and the eye mucous membrane were carried out to prove the safety of Carthami oil aquapuncture solution in clinical use. Animal for the research was the rabbit and the solution for the test was made from Carthami semen. 6 animals were used for the skin test and 9 animals were used for the eye mucous membrane test. In results, both tests proved that Carthami oil aquapuncture solution makes no irritable reaction on skin and eye mucous membrane of rabbit. We consider that this result is helpful for saying about the safety of Carthami oil aquapuncture solution in clinical use.

혼합 용매계 저독성 농축형 유분산제의 제조와 광유에 대한 분산효율 특성 (Preparation and Properties of Mixed Solvent Type Low Toxic Concentrated Oil Dispersant on Oil)

  • Low toxic concentrated oil dispersant using n-Paraffin and Di(ethylene glycol)mono butylether mixed solvent was prepared, and tested by oil dispersant performance test method, and oil dispersant efficiency was measured using vertical shaking method to 3 kinds of Crude oil, Bunker oil and W/O emulsions with different physical properties by appling the prepared dispersant. Although toxicity test was performed with Flat fish and Rock fish by appling the mixed oils emulsified using prepared oil dispersant, couldn't find the toxicity to them. Concentrated oil dispersant prepared has a good dispersion efficiency of 97.2% after 0.5min settling time and 28.3% after 10min settling time to Bunker B oil with 10% water solution. Especially, the concentrated oil dispersant showing the low toxicity to Oryzias Latipes(24hr, TLm) was 54,000 ppm and to Brine Shrimp Artemia(24hr, TLm) was 51,000ppm, and also, it was completely biodegradated to 99.1% after $7{\sim}8$days.

Toxicity on Laboratory Grown Plankton by the Oils Released from the Hebei Spirit Spill with Emphasis on a Dispersant Used in the Aftermath

  • The in vitro toxicities of three crude oils of the Hebei Spirit were examined on laboratory grown plankton, with a focus on the effects of a dispersant. The specific growth rate of phytoplankton and the mortalities of two zooplankton were measured in response to exposure to various concentrations of water accommodated oil, dispersant or both. The effects of the oils varied among the plankton, but were generally low within the range of the oil concentrations used, with little difference in toxicity among the three oils. Such low toxicity appeared to be associated with weathering of the crude oils. Exposure to the dispersant, however, dramatically increased the mortality of zooplankton, with complete inhibition of phytoplankton growth. No synergistic toxic effect was observed with the crude oil and dispersant combination. A better decision making process could be crafted for future application of dispersant in the event of an oil spill in Korean waters to better protect the marine plankton community from the excessive use of dispersant.

편백 정유의 마우스에 대한 급성경구독성 (Acute Oral Toxicity Test of Chamaecyparis obtusa Essential Oil on ICR Mice)

  • 본 연구는 세균과 진균에 대한 항균작용, 살충작용, 항아토피 활성, 항염증 효과, 혈압강하 및 스트레스 완화 효과 등이 있는 것으로 알려진 편백 정유를 이용하여, 마우스에서의 급성경구독성시험을 수행하였다. 편백 정유를 마우스에 0, 125, 250, 500, 1,000, 2,000 mg/kg body weight의 농도로 각각 1회 경구투여한 결과, 마우스 암 수 모두 2,000 mg/kg에서 모두 생존하였으며, 모든 투여군의 체중 및 모든 혈액학적 혈액생화학적 지표값들이 대조군과 비교하여 통계적으로 유의적인 차이를 보이지 않았다. 따라서 편백 정유의 마우스에서의 $LD_{50}$은 2,000 mg/kg body weight 이상으로 확인되었다.

  • 털두꺼비하늘소(Moechotypa diphysis) 성충에 대한 18종 식물정유의 훈증독성을 조사하였다. 그들 중 10 $\mu\ell/\ell$ (공기)의 농도에서 eucalyptus oil, rosemary oil, 그리고 pennyroyal oil은 100%, sage oil은 85%의 살충활성을 나타내었다. 4종 정유의 치사속도를 조사한 결과, eucalyptus oil과 rosemary oil이 처리 후 6시간 이내에 100%의 살충률을 나타내었다. 각 정유에 대한 주요 구성성분을 GC와 GC/MS로 분석한 결과, eucalyptus oil, rosemary oil, sage oil의 주성분인 1,8-cineole과 sage oil의 또 다른 주성분인 thujone과 pennyroyal의 주성분인 pulegone 모두 20 $\mu\ell/\ell$(공기)의 농도에서 100%의 살충효과를 나타내었다.

Colorado Potato Beetle (Leptinotarsa decemlineata Say) Control Potential of Essential Oil Isolated from Iranian Cymbopogon citratus Stapf

  • Colorado potato beetle is a most destructive insect pest of potato throughout the world. Although utilization of chemical insecticides is a main method for management of this pest, their negative side-effects such as threat to humans and the environmental pollution prompted researchers to search for natural alternatives. Recently plant essential oils with low or without side-effects against noun-targeted organisms and with high availability were considered as safe bio-pesticides. In the present study, toxicity of essential oil of Iranian lemongrass, Cymbopogon citratus Stapf, was evaluated against 3th instar larvae and adults of Colorado potato beetle by a leaf dipping method. Results displayed essential oil had notable toxicity against both larvae and adults after 24 and 48 h exposure times. Probit analysis revealed $LC_{50}$ values (lethal concentration to kill 50% of population) with 95% confidence limits were 10.32 (9.17 - 11.72) and 7.76 (6.80 - 8.74) ${\mu}l/ml$ for larvae and 6.27 (4.82 - 8.15) and 4.35 (3.24 - 5.62) ${\mu}l/ml$ for adults after 24 and 48 h, respectively. Based on regression analysis, a positive correlation between log concentration of essential oil and insect mortality was achieved. Results indicated C. citratus essential oil can be candidate as a natural alternative to the harmful chemical insecticides in the management of Colorado potato beetle.
