The Recycling Water Treatment of High Density Fish Culture System Using the Aerated Submerged Filter -1. Ammonia Removal Characteristics in Sea Water- (폭기식 잠수여상을 이용한 고밀도 양식장의 순환수 처리 -1. 해수중의 암모니아 제거 특성-)

  • Submerged filter process was used to evaluate the nitrifying efficiency of ammonia in the recycling water of marine aquatic culture system. The ammonia removal efficiency was achieved as high as $99\%$ at the hydraulic surface loading rate of up to $4.3{\ell}/m^2-day$. And the nitrite accumulation did not occur in the reactor even when the hydraulic surface loading rate of up to $36.8{\ell}/m^2day$ was applied. In the present study, the relationship between the effluent ammonia concentration and ammonia surface loading rate was formulated as an equation. The attachment rate of biofilm on the filter media at the ammonia surface loading rate of 62.3 and $311.7mg/m^2day$ was 15 and $55mg/m^2-day$, respectively, showing the linear relationship between the attachment rate and ammonia loading rates. Biofilm thickness and density of the filter media were found to be the function of the ammonia loading rate.

Purification Siderophore from Vibrio mimicus ATCC 33653 and its Effect to Bacterial Pathogenecity (Vibrio mimicus ATCC 33653이 생산하는 Siderophore의 정제와 균의 병원성에 미치는 영향)

  • Growth under conditions of iron-restriction and the production of siderophore was examined in Vibrio mimicus ATCC 33653. This strain grew and multiplied in the presence of the high-affinity iron chelators ethylenediamine-di (o-hydroxyphenylacetic acid). Chrome azurol S (CAS) agar and solution were used to detect the production of siderophore under these condition. Siderophore could be detected in the iron-restricted culture supernatants. The siderophore was extracted from iron-restricted culture supernatants by phenol-chloroform-ether method and purified by Dowex ion-exchange and Sephadex G-25 gel filtracton chromatography. The purified siderophore was confirmed by paper chromatography and HPLC. The Purified siderophore enhanced the growth of V. mimicus when the bacterium was grown in iron limited medium. Injection of both the siderohore and the bacteria to mice resulted in more rapid death than that of the only bacteria. However, the siderophore did not show lethality to mice and any toxicity to cell line like HeLa and U937.

  • Formalin, 37% formaldehyde, has been employed as a chemical for controlling ectoparasites and aquatic fungi responsible for infectious fish of diseases in hatcheries and culture facilities (Roberts, 1978; Schnick, 1991; Rach et al., 1997). Regarding the commercial usage of formaldehyde in the aquaculture industry, the U.S. Food and Drug Administration (FDA) approved three commercial products as parasiticides in a species-specific manner: Paracide-F and Formalin-F for bluegill, catfish, largemouth bass, salmon, and trout and Parasite-S for all finfish (FDA, 1998). Withdrawal time for these products was legally zero when used as permitted under the regulations. With the increased production of cultured fish in Korea, such as olive Hounder Paralichthys olivaceus and black rockfish Sebastes schlegeli, application of formalin to diseased fish has become more frequent. Moreover, there is still some concern about environmental exposures caused by effluents from fish culture facilities. The purpose of the present study was to evaluate residues in fish resulting from therapeutic usage of formalin in the aquaculture industry and to document the rate of disappearance of formaldehyde in seawater treated with formalin.   • With increase of people's leisure time for marine facilities it is expected to increase the demand for marine tourism. It is necessary for lighthouse to meet the demand for marine tourism. Now, the lighthouse is not only functioning as a traditional aids to navigation for vessel, but also it becomes the new facility and space for all civilians who might visit and experience the marine culture. This research suggests how to use the facilities of the lighthouse as marine tourism resources by reviewing the cases of domestic lighthouses.

Geographical Isolation and Root-Associated Fungi in the Marine Terrains: A Step Toward Establishing a Strategy for Acquiring Unique Microbial Resources

  • This study aimed to understand whether the geo-ecological segregation of native plant species affects the root-associated fungal community. Rhizoplane (RP) and rhizosphere (RS) fungal microbiota of Sedum takesimense native to three geographically segregated coastal regions (volcanic ocean islands) were analyzed using culture-independent methods: 568,507 quality sequences, 1399 operational taxonomic units, five phyla, and 181 genera were obtained. Across all regions, significant differences in the phyla distribution and ratio were confirmed. The Chao's richness value was greater for RS than for RP, and this variance coincided with the number of genera. In contrast, the dominance of specific genera in the RS (Simpson value) was lower than the RP at all sites. The taxonomic identity of most fungal species (95%) closely interacting with the common host plant was different. Meanwhile, a considerable number of RP only residing fungal genera were thought to have close interdependency on their host halophyte. Among these, Metarhizium was the sole genus common to all sites. These suggest that the relationship between potential symbiotic fungi and their host halophyte species evolved with a regional dependency, in the same halophyte species, and of the same natural habitat (volcanic islands); further, the fungal community differenced in distinct geographical regions. Importantly, geographical segregation should be accounted for in national culture collections, based on taxonomical uniqueness.

Characterization of an isolated reovirus from the paradise fish Macropodus opercularis imported from Southeast Asia (동남아시아로부터 수입된 paradise fish Macropodus opercularis로부터 분리한 reovirus의 특성)

  • In 2008, mass mortality was observed in paradise fish Macropodus opercularis which was imported from Indonesia. PCR of these fish found positive for megalocytivirus and Mycobacterium sp., while an unidentified virus was culture-isolated using CHSE-214 cells. In the present study, we investigated characterization of the unidentified virus and its pathogenicity to determine whether the virus was the causative agent of the mass mortality of paradise fish. The unidentified virus induced cytopathic effect (CPE) with syncytia in CHSE-214 and other fish cells, BF-2, GF, SSN-1, FSP and FFN. The virus was resistant against treatments with IUdR, chloroform, acidity at pH 3, basicity at pH 11 and high temperature at $56^{\circ}C$ for 3h. By electron microscopy, the viral particles were spherical having a double capsid structure with approximately 65 nm in external diameter. Viral genome was composed of at least 10-segmented RNA with sizes ranging from 0.7 kb to 3.6 kb. Based on these characters, this virus can be classified into family Reoviridae. This reovirus did not cause any mortality in an artificial experiment conducted by injecting the virus to paradise fish. This indicates that the reovirus is not only responsible for the mass mortality of paradise fish in 2008.

Rent-seeking Analysis of the Cultural Voucher from the Viewpoint of Culture and Arts Management (문화예술경영 관점으로 본 문화이용권사업의 지대추구론적 분석)

  • This study deals with the rent-seeking behavior that exist in cultural voucher from the viewpoint of culture and arts management. Art organizations open to consumers, producers and governments of the arts are dependent on the internal and external influence of an open system. Researcher has found rent-seeking in the course of introducing policies and legalization of the cultural voucher business which has been promoted in the direction of democratization of culture or cultural democracy. Cultural voucher business is a legal term. Although the government has increased the budget or tried to change the policies of the cultural voucher business, the implementation of the cultural voucher business has been opposed to the diversity of consumption and equity as the consumption of genre and the concentration of the capital region have increased. These results were structurally related to the process of legalization and rent-seeking behavior in bureaucracy. This study reaffirms that the efficient operation standard of the cultural voucher business is a balance between the choice of the beneficiary, the competition of the supplier, and access to the cultural voucher. And the theory of rent-seeking was applied as a criterion to analyze this balance. Thus, it is suggested that the criteria of evaluation and improvement to check the conservativeness of bureaucrats are needed to establish a legal system applied to the purpose of 'cultural democracy' and 'democratization of culture' ideology and to guarantee individual creativity and autonomy.

  • Characteristics of ${\beta}$-agarase production of Agarivorans sp, JA-1 isolated from north-eastern sea of Jeju marine environment was studied. Optimal cell growth was definite that the medium containing agar is 0.2%. The decreasing pattern of viscosity and agar concentration was same and they reached almost zero after 15 hours. Fed-batch culture was studied to improve agarase productivity by Agarivorans sp. JA-1 in marine broth containing 2.0 g/L agar with intermittent addition of 0.8 g agar two times. The hydrolysis products were identified oligosaccharide of degrees of polymerization 6.

Ethanol Production from Glycerol Using Immobilized Pachysolen tannophilus During Microaerated Repeated-Batch Fermentor Culture

  • Herein, we established a repeated-batch process for ethanol production from glycerol by immobilized Pachysolen tannophilus. The aim of this study was to develop a more practical and applicable ethanol production process for biofuel. In particular, using industrial-grade medium ingredients, the microaeration rate was optimized for maximization of the ethanol production, and the relevant metabolic parameters were then analyzed. The microaeration rate of 0.11 vvm, which is far lower than those occurring in a shaking flask culture, was found to be the optimal value for ethanol production from glycerol. In addition, it was found that, among those tested, Celite was a more appropriate carrier for the immobilization of P. tannophilus to induce production of ethanol from glycerol. Finally, through a repeated-batch culture, the ethanol yield (Ye/g) of 0.126 ± 0.017 g-ethanol/g-glycerol (n = 4) was obtained, and this value was remarkably comparable with a previous report. In the future, it is expected that the results of this study will be applied for the development of a more practical and profitable long-term ethanol production process, thanks to the industrial-grade medium preparation, simple immobilization method, and easy repeated-batch operation.

Water Quality Control in the Semiclosed Culture System Growing a Flounder, Paralichthys Olivaceus (반폐쇄 순환여과식 넙치양식장의 수질제어에 관한 연구)

  • A comparative evaluation of water quality in the existing semiclosed fish culture system was made to improve the system management. From the comparison of water quality between the flow-through mode and the recycle mode of the semiclosed system, the malfunctioning of the settling channel deteriorated water quality and reduce the nitrification rate in the recycle mode. The peak concentrations of COD, $PO_4-P$ and SS in the fish tanks appeared just after feeding, and then decreased to normal levels within two hours. However, the $NH_4-N$ concentration increased slightly after two hours of feeding in the recycle mode. The water exchange rate was directly related to the water quality in the semiclosed fish culture system.
