A comparative study on the osteoporotic animal models in ddY mouse: ovariectomized and neurectomized models (ddY 마우스를 이용한 골다공증 모델에 대한 비교연구: 난소적출 및 신경절단 모델)

  • In order to compare the induced time of osteoporosis between ovariectomized and neurectomized models in ddY mice. Experimental groups were divided into Sham, ovariectomized (OVX group) and neurectomized (NX group) group. The changes of body weight, tibia weight and histomorphometry of epiphyseal regions of tibia that were generally used as criteria index in osteoporosis, were evaluated at 2 and 4 weeks after operations with other generally used index-changes of serum osteocalcin. Also, calcium and phosphorus levels in the ash tibia were demonstrated with their ratio (Ca/P ratio). From the result of this study, evidences which reflect osteoporotic states of animals such as decrease of absolute and relative tibia weight, histomorphometrical index of epiphyseal region of tibia including trabecular bone volume %, and calcium and phosphorous contents in tibia, were generally detected from 4 weeks after ovariectomy and 2 weeks after neurectomy with increase of serum osteocalcin levels. In conclusion, it is considered that more rapid and favorable osteoporosis was induced in neurectomized model compared to that of ovariectomized model.

Effects of Herbal-acupuncture with $Artemisiae$ $Vulgaris$ $Folium$ Extract at $KI_{10}$($Eumgok$) on Osteoporosis in Ovariectomized Mice (음곡 애엽약침이 난소적출 흰쥐의 골다공증에 미치는 영향)

  • Objectives & Methods : The purpose of this study is to observe the effects of herbal-acupuncture with $Artemisiae$ $Vulgaris$ $Folium$ extract(AaH-HA) at $KI_{10}$($Eumgok$) on osteoporosis in ovariectomized(OVX) ddy mice. We carried out several experimental items to analyze the changes in body weight, urine, weight, uterus index, tibial length, the ash bone weight, tibial BMD, serum ALP, serum osteocalcin, serum Ca, and the levels of Ca, P, Ca/P ratio in tibia, and we performed histological and histomorphological analysis as well. Results : 1. Herbal-acupuncture with AaH-HA at $KI_{10}$ significantly inhibited the reduction phosphorus level in serum in ovariectomized mice. 2. Herbal-acupuncture with AaH-HA at $KI_{10}$ significantly inhibited the reduction creatinine level in serum in ovariectomized mice. 3. Herbal-acupuncture with AaH-HA at $KI_{10}$ significantly inhibited the increase of tibial osteoclast cells in ovariectomized mice. 4. Herbal-acupuncture with AaH-HA at $KI_{10}$ significantly inhibited the reduction of TBV(trabecular bone volume) and TBT(trabecular bone thickness) in ovariectomized mice. 5. Herbal-acupuncture with AaH-HA at $KI_{10}$ significantly inhibited the overgrowth of tibial growth plate length(GPL) in ovariectomized mice. Conclusions : In conclusion, our study suggested that Herbal-acupuncturing with AaH-HA at $KI_{10}$ can be effective treatment for osteoporosis.

Estrogen Mediates Ischemic Damage and the Migration of Human Umbilical Cord Blood Cells

  • Human umbilical cord blood cells(HUCBC) are rich in mesenchymal progenitor cells, endothelial cell precursors and hematopoietic cells. HUCBC have been used as a source of transplantable stem and progenitor cells. However, little is known about survival and development of HUCBC transplantation in the CNS. Estrogen has a neuroprotective potential against oxidative stress-induced cell death so has an effect on reducing infarct size of ischemic brain. We investigated the potential use of HUCBC as donor cells and tested whether estrogen mediates intravenously infused HUCBC enter and survive in ischemic brain. PKH26 labeled mononuclear fraction of HUCBC were injected into the tail vein of ischemic OVX rat brain with or without $17\beta$-estradiol valerate(EV). Under fluorescence microscopy, labeled cells were observed in the brain section. Significantly more cells were found in the ischemic brain than in the non-ischemic brain. HUCBC transplanted into ischemic brain could migrate and survive. Some of cells have shown neuronal like cells in hippocampus, striatum and cortex tissues. These result suggest that estrogen reduces ischemic damage and increases the migration of human umbilical cord blood cells. Effects of Herbal-Acupuncture with Rosae Laevigatae Fructus Extract at KI10(Umgok) on Osteoporosis in Ovariectomized Mice (음곡(KI10) 금앵자 약침이 난소적출 생쥐의 골다공증에 미치는 영향)

Herbal-acupuncture with Rosae Laevigatae Fructus at KI10(Umgok) significantly inhibited the reduction of tibial trabecular bone thickness(TBT) in ovariectomized mice. 4. Herbal-acupuncture with Rosae Laevigatae Fructus at KI10(Umgok) significantly inhibited the overgrowth of tibial growth plate length(GPL) in ovariectomized mice. Conclusion : These results suggested that herbal-acupuncture with Rosae Laevigatae Fructus at KI10(Umgok) has a therapeutic effect on osteoporosis, and to be put to practical use in the future osteoporosis clinic.

Histomorphometric Analysis of the Spine and Femur in Ovariectomized Rats Using Micro-Computed Tomographic Scan

  • Objective : The purpose of this study was to evaluate the different patterns of bone loss between the lumbar spine and the femur after ovariectomy in rats. Methods : Twenty-four female Sprague-Dawley rats underwent a sham operation (the sham group) or bilateral ovariectomy (the ovariectomized group). Four and eight weeks after operation, six rats from each of the two groups were euthanized. Serum biochemical markers of bone turnover including osteocalcin and alkaline phosphatase (ALP), which are sensitive biochemical markers of bone formation, and the telopeptide fragment of type I collagen C-terminus (CTX), which is a sensitive biochemical marker of bone resorption, were analyzed. Bone histomorphometric parameters of the 4th lumbar vertebrae and femur were determined by micro-computed tomography. Results : Ovariectomized rats were found to have higher osteocalcin, ALP and CTX levels than sham controls. Additionally, 8 weeks after ovariectomy in the OVX group, serum levels of osteocalcin, ALP and CTX were significantly higher than those of 4 weeks after ovariectomy. Bone loss after ovariectomy was more extensive in the 4th lumbar spine compared to the femur. Bone loss in the 4th lumbar spine was mainly caused by trabecular thinning, but in the femur, it was mainly caused by trabecular elimination. Conclusion : The present study demonstrates different patterns of bone loss between the 4th lumbar spine and the femur in ovariectomized rats. Therefore, when considering animal models of osteoporosis, it is important that bone sites should be taken into account.

Effect of Dietary Lipids and Guar Gum on Lipid Metabolism in Ovariectomized Rats (식이지방의 종류와 Guar Gum의 첨가가 난소절제한 흰쥐의 지질대사에 미치는 영향)

  • This study examined the effect of dietary lipids and guar gum on lipid metabolism in ovariectomized rats. The experimental animals received ovarietomy (OVX). Lipids sources were divided into 3 groups (soybean oil(SB), beef tallow(BT)) and fish oil(FO)) and guar gum was supplemented to each lipid diet (SBG, BTG, FOG). Experimental diets were fed to therats for 16 weeks. Serum triglyceride (TG) levels were higher in the BT group and lower in the FO group as compared to the SB group .Serum total cholesterol (Tc) and HDL-C levels were lower in the FO group as compared to the SB group. Serum LDL-C and phospholipid levels were lower in the FO group as compared to the SB group. Serum lipids levels were lowered by guar gum supplement. Serum SFA(saturated fatty acids) contents were not significantly influenced by dietary lipids and guar gum. Serum MUFA(monounsaturated fatty acids) contents were the lowest in the SB group. Fecal weight was highest in the beef tallow group and lowest in the fish oil group. Fecal weight was increased by guar gum supplement in all lipid groups. Total bile acid content in feces was increased by guar gum supplement in the soybean oil and beef tallow groups. The endothelial cells of the beef tallow group changed from a flat shape to distorted round and enlarged shapes. The subendothelial layer was the thickness the thickest in the beef tallow group ; the interspace between elastic lamina was widened and elastic lamina was straightened and partly disrupted . The fish oil group showed more porminient endothelial cells and subendothelial layer. Internal elastic membrane and elastic lamina exhibited regularly wavy shapes. Guar gum supplement showed positive effects in all lipids groups. Based on the above results , it is suggested that beef tallow increased serum TG levels and injured the wall of the aorta. On the other hand, fish oil , which decreased serum lipid levels, has a positive effect on the walls of the aorta. Guar gum protects the aorta from injury by reducing the serum lipid levels. Therefore, it is suggested that soybean oil and beef tallow consumed with guar gum is beneficial.

Anti-oxidant Effect and Enhancement of Menopausal Signs by Sophorae fructus-derived Isoflavones in Ovariectomized Rats and Cytoprotective Effect in Neuro-2a (난소절제 랫드에서 회화나무 이소플라본의 갱년기증상 개선효과 및 신경세포 보호효과)

  • Isoflavones are reported to playa role in menopausal women as a phytoestrogen, which can replace estradiols in hormone replacement therapy (HRT). Recently; due to the risk of breast cancer by HRT, phytoestrogens (e.g. isoflavones) have been focused as an alternative therapy in menopause. In the study, we investigated whether isoflavones derived from Sophorae fructus (SISO) have more benefit than that of soybean isoflavones in estrogen deficient rats. We found that SISO effectively controled $H_2O_2$ comparing with the baseline (p<0.01 vs. post value of OVX-Cont), and the blood sugar and weight were also controlled with decreasing patterns. Additionally, in LDH assay for cytoprotective effect in Neuro-2a cell line, SISO protected cells from the damage by SNAP (p<0.05). In conclusion, SISO may have more beneficial effect in enhancing the menopausal signs than that of soybean isoflavones and the cytoprotective effect in neuron cells suggests that SISO can play a certain role in neuroprotection after menopause.

Antiosteoporotic activity of ‘Dae-Bo-Won-Chun’ in the ovariectomized rats

  • The preventive effect of herbal formulation, ‘Dae-Bo-Won-Chun’(DBWC), on the progress of bone loss induced by ovariectomy (OVX) was studied in rats. From light microseopic analyses, a porous or erosive appearances were observed on the surface of trabecular bone of tibia in ovariectomized rats. Whereas those of the same bone in sham rats were composed of fine particles. The trabecular bone area and trabecular thickness in ovariectomized rats decreased by 50% from those in sham-operated rats, these decreases were completely inhibited by administration of DBWC at concentration of 10mg/kg per day for 7 weeks. The mechanical strength in femur neck was decreased by ovariectomy, and this was significantly suppressed by the administration of DBWC. Serum phosphorus, alkaline phosphatase, and thyroxine levels in ovariectomized rats were increased compared to those in sham-operated rats, and increases were completely inhibited by the administration of DBWC. These results strongly suggest that DBWC is effective in preventing the development of bone loss induced by ovariectomy in rats.

Effects of Herbal-Acupuncture with Evodiae Fructus at KI10 on Osteoporosis in Ovariectomized Mice (음곡(陰谷) 오수유(吳茱萸) 약침(藥鍼)이 난소적출(卵巢摘出)생쥐의 골다공증(骨多孔症)에 미치는 영향(影響))

  • Objective : This study is aimed to evaluate the effects of Evodiae Fructus herbal-acupuncture (EF-HA) at KI10 on osteoporosis induced by ovariectomy in mice. Method : Mice underwent bilateral ovariectomy. After recovering, the ovariectomized (OVX) mice were treated by needle prick, saline injection, herbal acupuncture with Evodiae Fructus (EF-HA) at KI10 for 8 weeks. Result : 1. EF-HA at KI10 significantly inhibited the overgrowth of tibia in ovariectomized mice. 2. NP at KI10 significantly restored the tibial BMD (bone mineral density) in ovariectomized mice. 3. EF-HA at KI10 significantly restored the phosphorus and creatinine levels in ovariectomized mice serum. 4. EF-HA at KI10 significantly restored the tibial Ca and P levels in ovariectomized mice. 5. EF-HA at KI10 significantly reduced the tibial osteoclast-like cells in ovariectomized mice. 6. EF-HA at KI10 significantly inhibited the overgrowth of tibial GPL (growth plate length) in ovariectomized mice. Conclusion : EF-HA at KI10 has protective and therapeutic effect for osteoporosis in ovariectomized mice. Thus, it is suggested that EF-HA can be an useful therapeutics in clinical field after further researches.

Effects of the Herbal-Acupuncture with Corni Fructus Extract at Eumgok(KI10) on Osteoporosis in Ovariectomized Mice (음곡(陰谷)(KI10) 산수유 약침이 난소적출 생쥐의 골다공증에 미치는 영향)

  • Objective & Method: The purpose of this study is to investigate the effects of herbal-acupuncture with Corni Fructus extract (CF-HA) at Eumgok(KI10) on osteoporosis in ovariectomized (OVX) ddy mice. Result: 1. CF-HA at Eumgok(KI10) significantly did not increase the level of bone mineral density in overiectomized mice. 2. CF-HA at Eumgok(KI10) significantly decreased the level of serum phosphorus in ovariectomized mice and sinificantly increased the level of serum calcium in ovariectomized mice. 3. CF-HA at Eumgok(KI10) significantly increased the levels of tibial calcium and phosphorus in ovariectomized mice. 4. CF-HA at Eumgok(KI10) significantly decreased the number of tibial osteoclast like cell in ovariectomized mice. 5. In the histomorphometric analysis of tibia, GPL(growth plate length) was significantly decreased by CF-HA at Eumgok(KI10) in ovariectomized mice. Conclusions: These results suggest that CF-HA at Eumgok(KI10) may have useful therapy effects on osteoporosis in ovariectomized mice.
