Zinc upregulates bone-specific transcription factor Runx2 expression via BMP-2 signaling and Smad-1 phosphorylation in osteoblasts

  • Purpose: Runx2 (runt-related transcription factor 2), a bone-specific transcription factor, is a key regulator of osteoblast differentiation and its expression is induced by the activation of BMP-2 signaling. This study examined whether zinc modulates BMP-2 signaling and therefore stimulates Runx2 and osteoblast differentiation gene expression. Methods: Two osteoblastic MC3T3-E1 cell lines (subclones 4 as a high osteoblast differentiation and subclone 24 as a low osteoblastic differentiation) were cultured in an osteogenic medium (OSM) as the normal control, Zn-($1{\mu}M$ Zn) or Zn+($15{\mu}M$ Zn) for 24 h. The genes and proteins for BMP-2 signaling (BMP-2, Smad-1/p-Smad-1), transcription factors (Runx2, osterix), and osteoblast differentiation marker proteins were assessed. Results: In both cell lines, BMP-2 mRAN and protein expression and extracellular BMP-2 secretion all decreased in Zn-. The expression of Smad-1 (downstream regulator of BMP-2 signaling) and p-Smad-1 (phosphorylated Smad-1) also downregulated in Zn-. Furthermore, the expression of the bone-specific transcription factors, Runx2 and osterix, decreased in Zn-, which might be due to the decreased BMP-2 expression and Smad-1 activation (p-Smad-1) by Zn-, because Runx2 and osterix both are downstream in BMP-2 signaling. Bone marker gene expression, such as alkaline phosphatase (ALP), collagen type I (COLI), osteocalcin, and osteopontin were also downregulated in Zn-. Conclusion: The results suggest that a zinc deficiency in osteoblasts suppresses the BMP-2 signaling pathway via the suppression of Smad-1 activation, and this suppressed BMP-2 signaling can cause poor osteoblast differentiation.

  • 근년 일본, 캐나다, 호주, 미국, EU(주로 노르웨이, 영국) 등에서 인공위성을 이용한 해양 안전의 확보를 위한 연구개발이 진행되고 있으며, 일부 실해역 적용의 분야도 도출되고 있는 실정이다. 9.11테러 이후, 국제해사기구에서도 해상보안의 문제는 주요 이슈로 대두되어, 해상보안에의 활용 기술 개발이 먼저 시작되었다. 그 외, 밀입국 선박 감시 덴 해양오염 모니터링이 주요 활용분야이다. 간단하게 요약하면 다음과 같다. -노르웨이: Norwegian Defence Hesearch Establishment(NDRE)에서 주도적으로 선박 탐지 실험 및 기술 개발을 실시. 주로, ESA의 위성을 활용. 국가 보안의 목적으로는 적용을 하고 있음. -캐나다: 캐나다에서 소유하고 있는 RADARSAT을 이용하여 가장 많은 실험을 실시함. 영상을 처리하고 결과에 대한 평가를 수행하기 위한 시스템(Ocean Monitoring Workstation, OSM)을 개발하여 보급에 주력. -호주: 주로 캐나다의 위성 및 시스템의 적용을 하고 있음 영해 및 환경 감시의 역할을 수행. Coastwatch조직을 만들어 해상 감시활동을 하고 있음. -영국: 데이터 취득 후, 2.5시간 이내에 선박의 위치를 전송하는 인터페이스를 개발함. 일본의 경우, 다른 선진국에 비해서는 다소 늦게 시작되었다. 2003년 발간된 '재해 등에 대응한 인공위성이용기술에 관한 종합보고서'를 시작으로 정보수집위성 4기 및 지구관측위성을 이용한 해양 감시 활동이 시작되었다. 또한, 제 3기 과학기술기본계획(2006-2012)내에 해양 불법침입 탐지 기술 개발 항목이 반영되어 있다. 유럽의 해상보안서비스(MARISS)의 사용자 워크숍이 ESA ESRIN(이탈리아 프라스카티)에서 2008년 1월 22일 열렸다. 실질적인 내용은, '해상보안을 위한 우주 시스템'에 관한 것으로 인공위성 이용하는데 있어 설계안 및 데이터 이용 컨셉을 제시하는 것이었다. 여기서 중요한 것은 국가간의 협력이 절대적으로 필요하며, 기존의 시스템과의 통합에 있어 신뢰성을 어떻게 확보하는가에 있다고 할 수 있다. 또한, 보안과 환경모니터링의 기능이 분리되어 진행되고 있는 부분에 대한 정보 통합 방향도 제기되었다. 국내에서도 AIS와 SAR정보의 결합에 관한 검토는 이루어졌으며, 이를 바탕으로 EU와 같은 시스템의 구축(조직과 연구개발)을 위한 실질적인 검토가 필요하다.

Effects of 3 mg·kg-1 Caffeine Ingestion during Exercise on Fluid-Electrolyte Balance and Tympanic temperature changes in the Heat (고온 환경에서 3 mg·kg-1의 카페인 섭취가 운동 시 체액, 전해질 균형 및 외이온의 변화에 미치는 영향)

  • This study aims to find out the effect of ingestion of 3 mg·kg-1 caffeine on fluid-electrolyte balance and Tty change during exercise under heat environment of 30~32℃ (40-50% humidity). Five trained males who routinely trained for approximately 60 min·d-1, 3-6 d·wk-1 (age; 28.20±3.56yrs, height; 174.56±5.46 cm, body mass; 76.13±9.02 kg, body fat(%); 14.24±3.99, VO2max; 54.00±4.30 mL·kg-1·min-1, exercise career; 4.20±1.95yrs) performed 40min of treadmill running in heat chamber. The study was a double-blind, randomized, crossover design. Body mass change following exercise was higher for the PLAC (Placebo) and CAFF (Caffeine) in comparison to the CON (Control), there was no significant difference between the CAFF, PLAC, CON (p= .997). The Usg not significant differences (p= .731) and Osmurine not significant differences between the CAFF, PLAC, CON (p= .901). There also were not significant between the CAFF, PLAC, CON for [Na+]urine and [K+]urine (p= .928, p= .469). In the case of Tty, although the increase rate of Tty was the highest for the CAFF on exercise early, exercise the second half in comparison to the CON and PLAC, there was not significant interaction effect between the CAFF, PLAC, and CON of Tty (p= .067), In conclusion, it was confirmed that the 3 mg·kg-1 caffeine ingestion prior to exercising in heat environment does not impart negative effect on body fluid, electrolyte balance and changes in Tty.

Polygonum cuspidatum stem extract (PSE) ameliorates dry eye disease by inhibiting inflammation and apoptosis

  • [Purpose] Here, we aimed to determine the effect of Polygonum cuspidatum stem extract (PSE) on exorbital lacrimal gland-excised rat models and hyperosmotic stress-stimulated human conjunctival cells (HCCs). [Methods] Seven week old male Wistar rats were divided into six groups. Only the rats in the control group (NOR, n=5) did not undergo surgery. Three days after the surgery, the exorbital lacrimal gland-excised rats were randomly allocated to five groups: (1) vehicle-treated dry-eyed rats (DED, n=5); (2) PSE (10 mg/kg) treated DED rats (PSE-10, n=5); (3) PSE (100 mg/kg) treated DED rats (PSE-100, n=5); and (4) PSE (250 mg/kg) treated DED rats (PSE-250, n=5). In addition, the HCC line was co-treated with hyperosmolar media (528 mOsm) and PSE (1-100 μg/ml). [Results] PSE treatment restored the tear volume and goblet cell density by inhibiting severe corneal irregularities and damage. The treatment with PSE significantly attenuated the hyperosmolar stress-induced inflammation and cell death through the suppression of mRNA expression levels of Tumor necrosis factor-α (TNF-α), Interleukin-6 (IL-6), Interleukin-1β (IL-1β), and Interferon-γ (IFN-γ), and the expression of Bcl-2-associated X protein (Bax) as well as the activation of caspase-3 in vitro. [Conclusion] The inhibitory effects of PSE treatment on dry eye disease indicate the potential of nutritional intervention by PES against inflammatory diseases without adverse effects.

  • Background : Alcoholic ketoacidosis(AKA) is a metabolic disturbance that is caused by prolonged and excessive alcohol consumption. Though the prognosis is reportedly good, its outcome is unclear in some cases that are combined with multi-organ failure. There are few reports of an analysis of cases admitted to an intensive care unit(ICU) Method: Cases of AKA admitted to the ICU over the last 5 years were retrospectively analyzed. Severe AKA was characterized by multi-organ failure that required treatment in an ICU Results : All patients were males with a history of excessive alcohol consumption. Five of them (50%) mainly complained of gastrointestinal symptoms (nausea, vomiting, diarrhea), showing metabolic acidosis with an increased asmolar and anion gap. Rhabdomyolysis with acute renal failure was the most common combined organ failure. Mechanical ventilation was performed in 80%. Six patients died and 4 patients survived. In the surviving patients, the arterial blood gas analysis(ABGA) was normalized within 12 hours after admission. Conclusion : In severe AKA patients, rhabdomyolysis with acute renal failure was the most common complication. The mortality rate was high and death from shock occurred within 3 days.

Osmoregulation Capability of Juvenile Grey Mullets (Mugil cephalus) with the Different Salinities (어린 숭어 (Mugil cephalus)의 염분별 삼투조절 능력)

  • In order to investigate the osmoregulation capability of grey mullet, Mugil cephalus with the different salinities, juvenile fish $(13.6{\pm}0.2\;TL)$ stocked in seawater (SW) were abruptly transferred to each experimental group $0\%SW(0\%_{\circ}),\;25\%SW(7.7\%_{\circ}),\;50\%SW(16.1\%_{\circ})\;and \;100\%SW(32.8\%_{\circ})$ and reared for 60 days. Blood samples were taken by the time schedule after the transfer. Plasma $Na^{+},\;K^{+},\;Cl^{-}$ and osmolality, muscle water content, and the electron microscopical observations of chloride cells were analyzed and made by the time schedule. In $100\%SW$, the maintainable levels of plasma $Na^{+},\;K^{+},\;Cl^{-}$ and osmolality were $167.1{\pm}7.7mM/l,\;9.1{\pm}2.1mM/l,\;137.8{\pm}5.6mM/l\;and\;351{\pm}18\;mOsm/kg$, respectively. These values were significantly changed at $6h\~1\;day$ after the beginning of the experiment with four different salinities. Fish from $0\%\;and\;25\%SW$ had lower osmolalities than those of fish from $50\%\;and\;100\%SW$, and showed the hyposmotic regulation pattern. At the end of the experiment (60 days after transfer), however, no significant difference was found in the concentrations of plasma $Na^{+},\;K^{+}\;and\;Cl^{-}$ among four experimental groups. Hematocrit was increased with salinity (P<0.01). After 10 days, fish from $0\%\;and\;25\%SW$ showed the hypertrophy, fusion and edema of epithelial layer in gill lamella. However, at the 15th day, epithelial layer in gill lamella was back to the normal status. On gill of fish from $0\%SW$, one apical pit held two or three chloride cells in common. Muscle water content was subsequently regulated to near the normal levels within 4 days, and there was no significant difference among four different salinities at the end of the experiment.

Dynamic Characteristics of Water Column Properties based on the Behavior of Water Mass and Inorganic Nutrients in the Western Pacific Seamount Area (서태평양 해저산 해역에서 수괴와 무기영양염 거동에 기초한 동적 수층환경 특성)

  • In order to understand the dynamic characteristics of water column environments in the Western Pacific seamount area (approximately $150.2^{\circ}E$, $20^{\circ}N$), we investigated the water mass and the behavior of water column parameters such as dissolved oxygen, inorganic nutrients (N, P), and chlorophyll-a. Physico-chemical properties of water column were obtained by CTD system at the nine stations which were selected along the east-west and south-north direction around the seamount (OSM14-2) in October 2014. From the temperature-salinity diagram, the main water masses were separated into North Pacific Tropical Water and Thermocline Water in the surface layer, North Pacific Intermediate Water in the intermediate layer, and North Pacific Deep Water in the bottom layer, respectively. Oxygen minimum zone (OMZ, mean $O_2$ $73.26{\mu}M$), known as dysoxic condition ($O_2<90{\mu}M$), was distributed in the depth range of 700~1,200 m throughout the study area. Inorganic nutrients typified by nitrite + nitrate and phosphate showed the lowest concentration in the surface mixed layer and then gradually increased downward with representing the maximum concentration in the OMZ, with lower N:P ratio (13.7), indicating that the nitrogen is regarded as limiting factor for primary production. Vertical distribution of water column parameters along the east-west and south-north station line around the seamount showed the effect of bottom water inflowing at around 500 m deep in the western and southern region, and concentrations of water column parameters in the bottom layer (below 2,500 m deep) of the western and southern region were differently distributed comparing to those of the other side regions (eastern and northern). The value of Excess N calculated from Redfield ratio (N:P=16:1) represented the negative value throughout the study area, which indicated the nitrogen sink dominant environments, and relative higher value of Excess N observed in the bottom layer of western and southern region. These observations suggest that the topographic features of a seamount influence the circulation of bottom current and its effects play a significant role in determining the behavior of water column environmental parameters.

Therapeutic Effect of Nocturnal Water Restriction in Children with Primary Nocturnal Enuresis (일차성 야뇨증 아동에서 야간 수분 제한의 치료 효과)

  • Purpose : Treatment of primary nocturnal enuresis (PNE) includs folk remedies and various treatments based on pathogenesis. We assessed the therapeutic effect of nocturnal water restrict ion as the primary treatment of PNE. Materials and methods : From October 1998 to June 1999, 41 children with PNE (>3 wet nights per week) who visited Ewha Womans University Mokdong hospital and who had good compliances to nocturnal water restriction for 2 months were included. Before and during nocturnal water restriction, daily fluid intake and urine volume were recorded for 2 days every 2 weeks. Responses to nocturnal water restriction were classified according to the decrease of wet nights as complete (>90$\%$), partial (50-90$\%$) and no (<50$\%$) response. Predictors such as age, sex, daytime voiding dysfunction, fluid intake, urine volume, maximum urine volume per void and fasting urine osmolality were evaluated. Results . The response rate to nocturnal water restriction fir 2 month was 82.9$\%$(34/41) [complete response 39.0$\%$(16/41), partial response : 43.9$\%$(18/41)]. The response rate to nocturnal water restriction was significantly higher in monosymptomatic PNE than polysymptomatic PNE and more effective in PNE with or nocturnal fluid intake, nocturnal urine volume, and maximum urine volume than lower nocturnal fluid intake, nocturnal urine volume and maximum urine volume per void (P<0.05). Nocturnal urine volume, maximum urine volume per void and luting urine osmolality after nocturnal water restriction has significantly increased higher in complete response and partial response group than in no response group (P<0.05). Conclusion : The nocturnal water restriction was effective in monosymptomatic PNE with nocturnal polydypsia, nocturnal polyuria and high bladder capacity. Comparison of Blood and Urine Renal Indices Between Hypercalciuric and Non-hypercalciuric Hematuria Patients (혈뇨 환아에서 고칼슘뇨군과 비고칼슘뇨군의 혈액 및 소변화학검사와 신기능 지표들의 비교)

  • Purpose : The purpose of this study was to investigate whether hypercalciuria patients with hematuria show different renal indices compared to non-hypercalciuria patients with hematuria. Methods : We retrospectively reviewed the medical records of patients with gross or microscopic hematuria whose blood chemistry and 24 hour urine chemistry were examined. After excluding the patients with more than $4 mg/m^2/day$ proteinuria or the patients with urinary calcium excretion between 3 and 4 mg/kg/day, we divided the patients into two groups: a hypercalciuria group whose calcium excretion was more than 4 mg/kg/day(n=30) and a non hypercalciuria group whose calcium excretion was less than 3 mg/kg/day(n=41). The urinary excretion, clearance, and fractional excretion(FE) of Na, K, Cl, Ca, P, urea, and creatinine were calculated and compared between the two groups. Results : The hypercalciuria group had more calcium excretion($6.1{\pm}2.9$ vs $1.5{\pm}0.9 mg/kg/day$), more urea excretion($341{\pm}102$ vs $233{\pm}123 mg/kg/day$), greater glomerular filtration rate(GFR) ($93.7{\pm}31.1$ vs $79.5{\pm}32.0 mL/min$) but lower FENa($1.0{\pm}0.4%$ vs $1.3{\pm}0.6%$) than the nonhyper-calciuria group, although the urinary sodium excretion was similar between the two groups. Conclusion : The greater urea excretion and GFR in hypercalciuric patients suggest that they might be on a higher protein diet than the non-hypercalciuria group. The increased glomerular filtration of sodium and calcium induced by the higher GFR in hypercalciuria would have increased their delivery to the distal tubule, where sodium is effectively reabsorbed but calcium is not, which is suggested by the lower FENa but higher FECa in hyercalciuria. It is recommended that the diet of hematuria patients be reviewed in detail at initial presentation and during treatment.

Effect of Changing Amniotic Fluid Osmolarity on the $Li^+$ Transport Through the Membrane Surrounding Amniotic Fluid in the Rabbit

  • To study the regulation of amniotic fluid volume and electrolyte concentration by the Membranes surrounding the amniotic fluid, the rate of $Li^+$ disappearance from amniotic sac of expired fetuses were examined while increasing the amniotic volume and osmolarity in rabbits. After intraamniotic injection of 1 ml isosmotic saline (about 20% of the amniotic fluid volume) containing 15 mM LiCl and 0.5 g/L Censored, the time courses of $Li^+$ and Censored disappearance were determined. From there the $Li^+$ clearance through the extrafetal routes was estimated and compared with that obtained from living fetuses. The volume, $Na^+$ concentration and osmolarity of amniotic fluid were measured and their relationships with $Li^+$ disappearance were evaluated. The fellowing results were obtained: 1. The rate of disappearance from amniotic fluid of living fetuses during the first 30 minutes was strikingly higher for $Li^+$ than for Censored, suggesting that extrafetal routes exist. At 60 and 90 minutes, however, the disappearance rate of $Li^+$ was less than that of Censored, suggesting the possibility of $Li^+$ reentry through fetal urination. 2. The disappearance of $Li^+$ from the amniotic fluid of the expired fetus was substantial, although lower than that of living fetuses, throughout the experimental period. 3. The $Na^+$ concentration and the osmolarity of the amniotic fluid of expired fetus measured 30 minutes after an intraamniotic injection of isoosmotic saline showed wide variation, but thereafter they changed gradually towards the normal extracellular fluid level. 4. When the amniotic fluid was iso- or hyposmolar, the rate of $Li^+$ disappearance from the amniotic fluid of the expired fetuses showed little variation. However, when the amniotic fluid was hyperosmolar, the rate at 30 minutes was markedly lower than those of isosmotic or hyposmotic amniotic fluid. At 90 minutes, the rate of $Li^+$ disappearance in hyperosmolar fluid reached a similar level to the rate in isosmolar fluid. 5. The intraamniotic injection of 400 mOsm/L saline solution decreased the disappearance rate of $Li^+$ from expired fetuses, while the injection of mannitol into the maternal vein induced no significant change. From these results it is concluded that: 1) a significant amount of $Li^+$ may leave the amniotic fluid via filtration through the membranes surrounding the amniotic fluid, 2) during hyperosmolar challenge to amniotic fluid, osmotic bulk flow might counteract the filterable loss, and 3) $Li^+$ disappearance might continue even after the volume and osmolarity of the amniotic fluid have recovered to control values.
