Euclidean Genetic Distances of Four Manila Clam (Ruditapes philippinarum) Populations analyzed by PCR Research

  • The PCR analysis was performed on DNA samples extracted from a total of 20 individuals using six oligonucleotides primers. The author accomplished clustering analyses to reveal the Euclidean genetic distances among four clam populations from Gochang, Seocheon, Taean and Anmyeon of the Korean peninsula. The oligonucleotides primer OPA-08 generated 5 unique loci to each population, approximately 550 bp and 600 bp, respectively, in the MCS population. Especially, the primer OPA-20 generated 15 unique loci to each population, which were identifying each population, approximately 400 bp, 750 bp and 800 bp, in the MCT population. Individuals from MCG clam population ($0.637{\pm}0.227$) exhibited higher band-sharing values than did individuals from MCG clam population ($0.402{\pm}0.115$) (P<0.05). The dendrogram obtained by the six oligonucleotides primers indicates four genetic clusters: cluster 1 (MCG 01, 02, 04 and 05), cluster 2 (MCS 06, 07, 08, 09 and 10), cluster 3 (MCT 11, 12, 13, 14 and 15) and cluster 4 (MCA 16, 17, 18, 19, 20 and MCG 03). Among the twenty clam individuals, the shortest genetic distance that displayed significant molecular differences was between individuals 14 and 15 from the MCT population (genetic distance = 0.094), while the longest genetic distance among the twenty individuals that displayed significant molecular differences was between individuals MCG no. 01 and MCG no. 02 (genetic distance = 0.687). Comparatively, individuals of MCS clam population were fairly closely related to that of MCT clam population, as shown in the hierarchical dendrogram of Euclidean genetic distances.

Morphological, Cytological and Molecular Evidence for Intersubgeneric F1 Hybrid between Glycine max x G. tomentella (콩 Glycine max와 G. tomentella의 종간교잡으로부터 얻은 Fl식물체 검증을 위한 형태적 · 세포학적 · 분자유전학적 연구)

  • This study was carried out to demonstrate morphological, cytological and molecular evidence for intersubgeneric $F_1$ hybrid between Glycine tomentella and G. max cv. ‘Baemkong’. Morphological features of $F_1$ plant for pistil and stamen, flower color and growth habit showed intermediate type between G. tomentella and G. max cv. ‘Baemkong’. Chromosome number of $F_1$ plant was 2n=39, which explained the evidence of $F_1$ hybrid between G. tomentella (2n=38) and G. max cv. ‘Baemkong’ (2n=40). Polyacrylamide isoelectric focusing pattern for esterase and peroxidase also showed that the $F_1$ plant was true $F_1$ hybrid between G. tomentella and G. max cv. ‘Baemkong’. From RAPD analysis, we identified that 62 primers showing bands in $F_1$ hybrid had both bands from G. tomentella and G. max cv. ‘Baemkong’, which suggested that this was true $F_1$ hybrid. Based on our results from morphological, cytological and molecular analyses, we suggest that the $F_1$ plant was true intersubgeneric hybrid between G. tomentella and G. max cv. ‘Baemkong’. Our results still remain us further study to recover fertility of $F_1$ hybrids. The occurrence of maternal and/or paternal inheritance in $F_1$ hybrid from intersubgeneric cross between G. tomentella and G. max cv. ‘Baemkong’ need to be explained.

  • Decamer RAPD primers were tested on dioeceious and hermaphrodite plants of Commiphora wightii to identify sex-specific molecular markers. Sixty different random decamer primers were screened out of which only three primers were found to be associated with sex expression. A ~1,280-bp fragment from the primer OPN06 was found to be present in all the female individuals. Another primer OPN 16 produced a unique ~400-bp amplification product in only hermaphrodite individuals. The third marker, OPA20 amplified a ~1,140-bp fragment from female and hermaphrodite DNAs, but failed to do so from the male plant DNAs.

Spectrofluorimetric determination of Trimethoprim in pharmaceutical preparations

  • The development of a spectrofluorimetric method for the determination of trimethoprim according to the reaction between O-phthalaldehyde (OPA) in highly alkaline media, and 2-mercaptoethanol (2ME) and this reaction gives fluorescent product measured at (458) nm when excited at (342) nm. The optimization of the analytical parameters that influence intensity was investigated. The intensity of fluorescence of the formed product was linearly related to the concentration of trimethoprim in the (100-1200) ng mL-1 range. The limit of detection and limit of quantification were estimated to be (22.54) ng mL-1 and (75.15) ng mL-1 respectively. The utility of the proposed methods was successfully verified by analysis of trimethoprim in pure and real pharmaceutical preparations with high accuracy, the recovery percentages Re%, were found to be (100.5) % and (99.76) % for pure drug and pharmaceutical preparations respectively.

Morphology and Molecular Characterization of a Fungus from the Alternaria alternata Species Complex Causing Black Spots on Pyrus sinkiangensis (Koerle pear)

  • A small-spored Alternaria was found from black spots of storaged Koerle pear (Pyrus sinkiangensis), one of the economically important fruit in Xinjiang province, China. The morphology is similar to A. limoniasperae but obviously different in secondary conidiophores and conidial septa. A phylogenetic analysis using sequence datasets of ITS, GAPDH, TEF1, RPB2, Alt a1, OPA10-2, and EndoPG genes revealed that it belonged to the Alternaria alternata complex group. Pathogenicity tests illustrated that the fungus was the causal pathogen of black spot on Koerle pear fruit.

  • The enactment of OPA90 (Oil Pollution Act of 1990) in the USA and the consequent moves by IMO(International Maritime Organization) to introduce new Regulations for the design of oil tankers led the oil transportation industry to undergo a period of big change. This resulted in the introduction of double hull tankers. This paper introduces the design for the 300, 000 DWT double hull VLCC of World-Wide Shipping Agency Pte Ltd. in Hong Kong, which is the first of this type constructed by Daewoo Shipbuilding & Heavy Machinery Ltd.(DSHM). The characteristics of the compartment and structural arrangement of this vessel are briefly described, and the scope of structural analysis is illustrated. In addition, the merits/demerits of different crosstie arrangements are described in the appendices.

Preparation of in situ Patterned ZnO Thin Films by Microcontact Printing (Microcontact Printing을 이용한 미세패턴 ZnO 박막 제조)

  • In situ patterned zinc oxide thin films were prepared by precipitation of Zn(NO$_3$)$_2$ aqueous solution containing urea and by microcontact printing using Self-Assembled Monolayers(SAMs) on A1/SiO$_2$/Si substrates. The visible precipitation of Zn(OH)$_2$ that was formed in the Zn(NO$_3$)$_2$ aqueous solution containing urea was enhanced with an increase of the reaction temperature and the amount of urea. As the reaction time of Zn(NO$_3$)$_2$ with urea was prolonged, the thickness and grain size of Zn(OH)$_2$ thin layers were increased, respectively. The optimum precipitation condition was at 80$\^{C}$ for 1 h for the solution with the ratio of Zn(NO$_3$)$_2$ to urea of 1 : 8. Homogeneous ZnO thin films were fabricated by the heat treatment of 600$\^{C}$ for 1 h of Zn(OH)$_2$ precipitation on Al/SiO$_2$/Si substrate. This was available to the in-situ patterned ZnO thin films with uniform grain size. Hydrophobic SAM, Octadecylphosphonic Acid(OPA) and hydrophilic SAM, 2-Carboxyethylphosphonic Acid(CPA) were applied on the Al/SiO$_2$/Si substrate by microcontact printing method. In situ patterned ZnO thin film was successfully prepared by the heat treatment of Zn(OH)$_2$ precipitated on the surface of hydrophilic SAM, CPA.

  • A 2-step process for the safe destruction of chemical wafare agents(agent hydrolysis followed by supercritical water oxidation) was studied to obtain kinetic data for the pilot plant design. This process is simple to operate by using commercial equipments and could be applied as an alternative technology to incineration. Sarin(GB) and sulfur mustard(HD) were hydrolysed in sodium hydroxide and water respectively and their hydrolysates and OPA, which is binary agent for GB were oxidized in a continuous flow supercritical water oxidation system. Destruction efficiencies of the materials were above 99.99% in supercritical water.

RAPD Loci for Seed Protein and Oil Content in Soybean (Glycine max)

  • Seed protein and oil content is important trait in the soybean. Both seed protein and oil content in this plant species is inherited quantitatively. A 68-plant $F_2$ segregation population derived from a mating between Mercury and PI 467.468 was evaluated with random amplified polymorphic DNA (RAPD) markers to identify QTL related to seed protein and oil content. Marker OPB12 was found to be associated with differences in seed protein content. Four markers, OPA09b, OPM07b, OPC14, and OPN11b had highly significant effects on seed oil content. By interval mapping, the interval between marker OPK3c and OPQ1b on linkage group 13 contained a QTL that explained 25.7% variation for seed oil content.

  • RAPD analysis was performed from four host-specific toxin (HST) producing Alternaria, i.e., A. kikuchiana, A. mali, a. longipes and A. Longipes and A. alternata f. sp. lycopersici, nonpathogenic A. alternata and A. brassicicola to assess their phylogenetic relationship. DNA polymorphism was detected among species (pathotypes) of HST producing Alternaria by PCR amplification and differentiation of the species was recognized by RAPD analysis. Primer OPA-02 was the most profitable among 7 notificated primers for differentiation of the HST producing Alternaria species. UPGMA analysis of the RAPD bands from alternaria spp. revealed that HST producing Alternaria and nonpathogenic a. alternata are closely related.
