Determination of Radical Scavenging Activity of Aster yomena (Kitam.) Honda (쑥부쟁이 추출물의 라디칼 소거활성 평가)

  • In this study, we investigated the antioxidative effects of AY by measuring 1.1-Diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical ($^{\cdot}OH$), and superoxide radical ($O_2{^-}$) scavenging activities in vitro. AY was extracted with ethanol and then partitioned with n-hexane, methylene chloride ($CH_2Cl_2$), ethylacetate (EtOAc) and n-butanol (n-BuOH). In the DPPH radical scavenging assay, AY at concentrations of 10 to $100{\mu}g/mL$ dose-dependently increased inhibition of DPPH oxidation, with the EtOAc fraction of AY showing the highest DPPH radical scavenging activity fractions. The $^{\cdot}OH$ radical scavenging activities of the extract and four fractions of AY increased by over 80% at a concentration of $50{\mu}g/mL$. In particular, the IC50 value of the EtOAc fraction was $0.03{\mu}g/mL$, which was the lowest value among all fractions. We also found that the EtOAc fraction of AY was better at $O_2{^-}$ radical scavenging than other fractions. Taken together, these results suggest that AY, especially the EtOAc fraction, can be used as a natural antioxidant against free radicals.

Free Radical Scavenging Activity of Butanol Fraction from the Fruit of Citrus junos

  • In this study, we investigated the free radical [1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical (${\cdot}OH$) and superoxide anion (${O_2}^-$)] scavenging activity of MeOH extract and 3 fractions of Citrus junos. Of the tested fractions, the BuOH fraction showed the strongest DPPH scavenging activity, showing the $IC_{50}$ values of 63.4 mg/mL. Therefore, we continuously carried out DPPH, ${\cdot}OH$ and ${O_2}^-$ scavenging activity tests of BuOH fraction of Citrus junos. The BuOH fraction of Citrus junos inhibited DPPH radical to 97.5% at a concentration of 1000 mg/mL and the scavenging activities were increased concentration-dependently. In addition, BuOH fraction from Citrus junos also scavenged ${\cdot}OH$ in a concentration dependent manner from 5 to 1000 mg/mL. Furthermore, BuOH fraction showed about 56% ${O_2}^-$-scavengimg activity at 25 mg/mL concentration but, the scavenging activities were not enhanced in a dose dependent manner. The present results suggest that BuOH fraction of Citrus junos would have the protective potential from oxidative stress induced by free radicals.

Screening of Effective Extraction Conditions for Increasing Antioxidant Activities from Fronds of Osmunda japonica (고비의 항산화활성 증가를 위한 효율적인 추출조건 탐색)

  • This study was conducted to investigate the optimum condition of extraction from fronds of Osmunda japonica to increase antioxidant compounds and antioxidant activity. Powder (1 g) of lyophilized fronds were mixed with three different solvents (MeOH, 80% EtOH and water). Extraction was carried out using not only by immersion (room temp.), heating ($60^{\circ}C$) and stirring (200 rpm) for 6 h, but also by sonication in 42 kHz ultrasonic bath for 15, 30 and 45 min. Extracts were filtered, and adjusted up to 50 mL to determine contents of soluble solids, total polyphenols and total flavonoids. Antioxidant capacity was measured by radical scavenging activity of 0.15 mM DPPH (2,2-diphenyl-1-picrylhydrazyl) and 7.4 mM ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)] radical. Among the solvents, MeOH and 80% EtOH appeared to be effective for extraction. Extract obtained from sonication in MeOH for 15 min resulted high polyphenol contents (45.15 $mg{\cdot}g^{-1}$ db) and DPPH radical scavenging activity ($RC_{50}$= 0.35 $mg{\cdot}mL^{-1}$). The highest flavonoid contents was obtained from immersion or heating extraction with MeOH (38.10~38.10 $mg{\cdot}g^{-1}$ db). ABTS radical scavenging was high in same extraction with 80% EtOH ($RC_{50}$= 0.21~0.22 $mg{\cdot}mL^{-1}$). Altogether, our results indicate that the extraction using ultrasonic bath with MeOH as a solvent (for 15~30 minutes) was the most effective way not only for increasing various antioxidant activities but also for saving labor and time in case of fronds of Osmunda japonica.

  • Characteristics of the lifted flame which is generated by issuing of the fuel through the miniature nozzle, d=0.164 mm, are studied using the planar laser induced fluorescence technique. OH radical is excited on the $Q_1$(8) line of the $A^2{\Sigma}\ ^+{\leftarrow}\ X^2{\prod}$ (1,0) band transition(283.55 nm) and LIF signals are captured at the bands of (0,0) and (1,1) transition(306-326 nm) using the filters and ICCD camera. Hydroxyl radical(OH) profile for nozzle attached flame shows that OH radical populations at the flame sides and flame tip are larger than those at the base. But for the lifted flame (tribrachial flame) case, those are larger at the flame base than at the flame tip and flame sides. The OH radical is more dense near the center line of flame base at the blowing out. This fact proves the Chung and Lee's blowout theory - blowout occurs when the flame is anchored at the flame axis.

Polyphenol and Anti-oxidant Effects of Kalopanax septemlobus Koidz. Leaf Extracts (음나무(Kalopanax septemlobus Koidz.) 잎 추출물의 폴리페놀 및 항산화효과)

  • The aim of the study was to assess the cosmeceutical activity of Kalopanax septemlobus leaf and it is possible that can be used as a cosmetic ingredient for applicaiton of cosmetic industries. The concentration of total phenolic compound of hot water and 70% EtOH extracts of K. septemlobus leaf showed 104 mg/L and 125 mg/L respectively. In the result of DPPH(1,1- diphenyl-2-picryl -hydrazyl) scavenging radical activity, 70% EtOH extracts of K. septemlobus leaf showed 93.1% and it was similar to BHA(butylated hydroxyanisole) effect at 1,000ppm concentration. Xanthine oxidase inhibition of hot water extracts and 70% EtOH extracts of K. septemlobus leaf were 46.6% and 60.4% at 1,000ppm, respectively. In these results, K. septemlobus leaf has a great potential as a cosmeceutical ingredient with a natural anti-oxidant source.

Effects of OH Radical Density from Atmospheric Plasma to Induce Cell Death in Lung Cancer and Normal Cells

  • Atmospheric plasma's electron temperature is less than thermal plasma, so it is useful at bio experiment. We have investigated the optical emission spectroscopy (OES) lines by spectrometer during Atmospheric plasma bombardment onto the PBS surface by using an Ar gas flow. Also we have measured the OH radical density inside the solution induced by the Atmospheric plasma bombardment. OH radical species are appeared at 308 nm and 309 nm. Densities of OH radical species has been found to be significantly decreased versus depth of the solution from 2 mm to 6 mm. OH radical density inside the PBS is measured to be about $1.87{\times}1016cm-3$ downstream at 2 mm from the surface under optimized Ar gas flow of 200 sccm in Atmospheric plasma. Also we have investigated cell viability of lung cancer and normal cell after Atmospheric plasma treatment for fixed exposure time in 60 seconds, but different depths. We used SEM, we observed change of cell morphorogy, did experiment about FDA & PI Staining method. It is found that there is selectivity between the lung cancer and lung normal cell, in which cancer cell definitely has higher cell death ratio more than normal cell. We have investigated change of bond structure in FT-IR spectroscopy, the following peaks were observed: and intense O-H peak at 3422 cm-1 and at 2925 cm-1 corresponds to C-H stretch vibrations of methylene group.

An Investigative Study on the Characterization of Cefaclor Decomposition in UV/H$_2$O$_2$ Process (UV/H$_2$O$_2$공정에 의한 Cefaclor 분해 특성에 관한 기초연구)

  • The combining process of UV irradiation and H$_2$O$_2$ was used to investigate characteristics of cefaclor decomposition in the aquatic environment. The separate mixing tank was used to minimize the decreasing effective of contact area caused by sampling. Four baffles were installed inside the UV reactor for the complete mixing of the sample and outside of the reactor was wrapped with aluminum foil to protect the emission of photon energy. Production of OH radical was measured using pCBA(p-Chlorobenzoic acid) indirectly and rate constants were withdrawn pseudo-frist order reaction. Optimum condition for the maximum production of OH radical was found to be pH 3, hydrogen peroxide of 5 mmol/L and recirculation rate of 400 mL/min. Pseudo-frist order reaction rate constant was 0.1051 min$^{-1}$. In the optimum condition, cefaclor was completely decomposed within 40 min and rate constant was 0.093 min$^{-1}$. Decomposition by OH radical producted intermediate anions such as chloride, nitrate, sulfite and acetic acid and phenylglycine. After 6 hr most cefaclor was decomposed by UV/H$_2$O$_2$ process and converted to CO$_2$ and H$_2$O, resulting of operation in the decrease of TOC and acetic acid and the disappearance of phenylglycine.

Measurement of Hydroxyl Radical Density at Bio-Solutions Generated from the Atmospheric Pressure Non-Thermal Plasma Jet

  • Atmospheric pressure non-thermal plasma of the needle-typed interaction with aqueous solutions has received increasing attention for their biomedical applications [1]. In this context, surface discharges at bio-solutions were investigated experimentally. We have generated the non-thermal plasma jet bombarding the bio-solution surface by using an Ar gas flow and investigated the emission lines by OES (optical emission spectroscopy) [2]. Moreover, The non-thermal plasma interaction with bio-solutions has received increasing attention for their biomedical applications. So we researched, the OH radical density of various biological solutions in the surface by non-thermal plasma were investigated by Ar gases. The OH radical density of DI water; deionized water, DMEM Dulbecco's modified eagle medium, and PBS; 1x phosphate buffered saline by non-thermal plasma jet. It is noted that the OH radical density of DI water and DMEM are measured to be about $4.33{\times}1016cm-3$ and $2.18{\times}1016cm-3$, respectively, under Ar gas flow 250 sccm (standard cubic centimeter per minute) in this experiment. The OH radical density of buffer solution such as PBS has also been investigated and measured to be value of about $2.18{\times}1016cm-3$ by the ultraviolet optical absorption spectroscopy.

Study on the Temporal Density Variation of Chemical Species in the Atmospheric Pressure Plasma Process (대기압 플라즈마 프로세스에 있어서 시간에 따른 화학종의 밀도변화 연구)

  • This study is to discuss simulation results with 51 principal chemical reactions in non-thermal plasma space under atmospheric pressure, and the ambient gas was mainly composed of oxygen and nitrogen molecules. The initial density of O and OH radicals under the ambient temperature of 300K is largely generated in comparison with other higher temperature, and the density of O radical decreased from $20{\mu}s$ according to increase the temperature. The initial density of OH radical seemed to decrease steeply at the initial stage. By increasing the initial density of $H_2O$ molecules, O radical's effect was few and the density of OH radical was largely generated about 2 times. In addition, ozone density was increased as increasing the density of O radical, but it was decreased as increasing the density of $H_2O$. In case of the temperature more than 300K, $NO_2$ tend to be removed, but NO was increased than the initial density.

Scavenging Effects of Tea Catechins on Superoxide and Hydroxy Radical

  • Tea catechins, the most important compounds in tea polyphenols, can efficiently scavenge superoxide anion free-radical ($O_2$.), hydroxyl radical. (.OH) The mechanism of scavenging active oxygen free radicals was investigated by ESR spin trapping technique and Chemiluminescence. Results showed that various tea catechins constitute an antioxidant cycle in accordance with the decreasing order of the first reductive potential, and produce the effect of cooperative strength each other. Esterificated catechins could scavenge active oxygen free radicals more effectively than the non-esterificated ones. When.OH and $O_2$.- were scavenged by (-)-epigallocatechin gallate [(-)- EGCG], the stoichiometric factors were 6, and the rate constants of scavenging reaction reached $7.71{\times}10^6$ and $3.52{\times}10^{11}$ L $mmol^{-1}s^{-1}$, respectively. In the mean time, tea catechins could scavenge superoxide anion fiee radical ($O_2$-.) and hydroxyl radical (.OH) in a dose dependent manner. But at higher concentration or pH value, tea catechins can induce the prooxidant.
