재배 조건에 따른 바위솔의 Flavonol Glycoside 함량 변화 (Changes in Flavonol Glycoside Contents of Orostachys Japonicus a. Berger according to Cultivation Conditions)

  • Night-break, 일장조절, 월동가온 실험으로부터 얻어진 바위솔에 포함된 flavonol glycoside, $kaempferol-3-O-{\beta}-D-glucoside$(1), $kaempferol-3-O-{\beta}-D-glactoside$ (2), $kaempferol-3-O-{\beta}-D-rhamnoside$ (3), $quercetin-3-O-{\beta}-D-glucoside$ (4) and $quercetin-3-O-{\alpha}-D-rhamnoside$ (5)의 함량이 분석되었으며 자연산 바위솔의 함량과 비교되었다. 월동가온 시험에서 얻은 flavonol glycoside 1-5의 함량은 자연산보다 감소하였으며 일장조절 실험 및 night-break 시험에서 얻은 flavonol glycoside 함량은 조사한 빛의 양이 증가함에 따라 증가하였다. 월동가온은 flavonol glycoside의 생성에 부정적으로 작용하는 반면 조사되는 빛의 양의 증가와 같은 경작조건은 flayonol glycoside의 생성에 유리하다.

옥돔(Branchiostegus japonicus) 비늘 유래 젤라틴의 가수분해 및 가수분해물의 기능성 (Processing and Biological Activity of Gelatin Hydrolysate from Branchiostegus japonicus Scales)

  • The potential utility of fish scales to the functional food industry has been investigated due to its antioxidant and antihypertensive characteristics. In this study, we report on the reactive oxygen species (ROS) scavenging and angiotensin I converting enzyme (ACE) inhibitory activities of gelatin hydrolysates processed from Branchiostegus japonicus scales, which are also high in protein content (about 46.1%). We prepared the enzymatic gelatin hydrolysates with four proteases (${\alpha}$-chymotrypsin, Alcalase, Neutrase and trypsin) from B. japonicus scale gelatin, which was prepared according to different reaction times, substrate/enzyme ratios and substrate concentrations. The enzymatic hydrolytic degrees of the gelatin increased time-dependently up to 6 hrs, while the Alcalase gelatin hydrolysates showed the highest hydrolysis degrees compared to the others. Furthermore, gelatin hydrolysates of Neutrase and ${\alpha}$-chymotrypsin showed the highest DPPH radical and $H_2O_2$ scavenging activities ($IC_{50}$ value; 9.18 mg/mL and 9.74 mg/mL), respectively. However, the activities were not significant (P<0.05). We also observed that the four gelatin hydrolysates significantly increased ACE inhibitory activities from approximately 20% to 60% (P<0.05), Among them, the Alcalase gelatin hydrolysates showed the higher ACE inhibitory activity ($IC_{50}$ value; 0.73 mg/mL) compared to the others. These results suggest that the enzymatic gelatin hydrolysates prepared from B. japonicus scales may possess a potentially useful function as an ACE inhibitory agent. As such, the utility of B. japonicus scales should be given due consideration for application in the functional food industry.

생약재와 와송 추출물의 복합조성물이 Streptozotocin 유발 당뇨성 흰쥐의 혈당강하 및 혈중 지질성분에 미치는 영향 (Hypoglycemic and Hypolipidemic Effects of Orostachys japonicus with Medicinal Herbs in Streptozotocin-Induced Diabetic Rats)

  • 생약재(백복령, 사인 및 창출) 추출물을 동량으로 혼합한 것에 와송 추출물을 첨가하여 만든 복합물이 당뇨쥐의 혈당강하 및 혈중 지질 성분 변화에 미치는 영향을 실험하였다. 정상군, 당뇨 대조군, 생약재 혼합물 급여군(D-MH), 생약재 혼합물과 와송 추출물의 1:1 복합물 급여군(D-MHOA) 및 1:3 복합물 급여군(D-MHOB)으로 구분하여 4주간 실험 사육하였다. 실험 사육 4주 후 체중의 증가량은 모든 실험군이 당뇨 대조군에 비해 증가되었으나, D-MHOB군에서만 유의적인 증가를 보였다. 실험 사육 기간 동안 공복 혈당은 실험 식이의 급여 2주 후부터 모든 실험군에서 대조군에 비해 유의적인 감소를 보였으며, 2~3주 동안에는 D-MH군과 D-MHOA군 간에 비슷한 수준이었다. 4주 후에는 D-MHOB군의 공복 혈당이 가장 낮았다. 내당능 측정 결과, 포도당 급여 60분에 모든 실험군에서 최고 혈당치를 보였으나, 생약재 혼합물 및 와송의 복합물 급여군이 대조군에 비해 유의적으로 개선되었다. 당화 헤모글로빈 함량은 정상군(3.9%)에 비해 대조군이 약 1.9배 증가되었으며, D-MH군에 비해 D-MHOA 및 D-MHOB군에서 유의적으로 감소되었다. 생약재에 대한 와송의 첨가량이 많을수록 혈청 총 지질 및 총 콜레스테롤 함량은 대조군에 비해 유의적으로 감소되었으나, HDL-C 함량은 D-MHOB군이 대조군에 비해 유의적으로 높았다. GPT 활성은 D-MHOB군이 여타의 실험군에 비해 유의적으로 낮았다. 생약재 혼합물 급여군에 비해서 와송 추출물의 첨가군에서 지질과산화물 함량은 감소되었으나 와송의 첨가량에 따른 유의차는 작았다. 항산화 활성은 와송의 첨가량에 따라 유의적으로 증가하였다. 따라서 와송은 생약재 혼합물에 첨가됨으로써 혈당 저하 및 지질 개선 효과를 증가시키는 것으로 확인되었다.

  • 본 연구에서는 와송(Orostachys japonicus A. Berger)의 산화방지 효과와 산화스트레스로부터의 신경세포 손상에 대한 보호효과 및 주요 생리활성물질을 분석하였다. 와송 아세트산에틸 분획물(ethylacetate fraction from Orostachys japonicus A. Berger extract: EFOJ)은 다른 분획물보다 높은 총 페놀화합물 함량을 나타냈으며, 와송 아세트산에틸 분획물(EFOJ)로 ABTS, DPPH 라디칼 소거 활성과 FRAP 검정 및 지방질과산화물 억제력 분석을 실시한 결과, 우수한 라디칼 소거 활성, 총산화방지력과 과산화 지방질 생성물의 억제력을 확인하였다. 또한 와송 아세트산에틸 분획물(EFOJ)은 과산화수소로 인한 신경세포 사멸에 대한 세포 생존율을 향상시켰으며, 신경 세포막 손상을 보호하는 능력이 우수한 것을 확인하였다. 이러한 생리 활성을 가진 와송 아세트산에틸 분획물(EFOJ)의 주요물질을 분석하기 위하여 $LC-MS^e$를 실시하였으며, 주요 생리활성 물질은 퀘세틴 배당체와 캠페롤 배당체로 추정되었다. 본 연구 결과들을 바탕으로, 와송 추출물은 우수한 산화방지력을 가지고 있을 뿐만 아니라 산화적 스트레스로부터의 신경세포 보호효과를 통해 퇴행성 신경질환과 같은 질병을 예방 할 수 있는 건강기능성식품의 천연산화방지제 소재로써 고부가 가치가 있다고 판단된다.

동충하초의 면역조절 및 항암효과 (Immuno-modulatory and Anti-carcinogenic Property of Cordyceps militaris and Paecilomyces japonicus)

  • The purpose of this research was to investigate the immuno-modulatory effect and anti-carcinogenic property of Cordyceps militaris(CM) and/or Paecilomyces japonicus (PJ). The proliferation of cultured splenocytes and thymocytes were enhanced by the addition of 10 ㎍/ml of CM and/or PJ. B lymphocytes subpopulation in splenocytes were increased both CM and/or PJ administered(p.o. for 7 days)-mice. Thymic T lymphocytes, especially TH cells were significantly increased in CM-administered mice. CM and/or PJ treatment inhibited the cell viability of L 1210 mouse leukemia and HL60 human leukemia cells and induced the apoptosis of L1210 and HL60 cells. In addition, CM and/or PJ increased the hemaggutination(HA) titer against SRBC. These results suggest that CM and/or PJ have an immuno-modulatory action and anti-carcinogenic property.

Temporal and Spatial Variation in Fish Larvae in Gamak Bay and Yeoja Bay, South Sea of Korea

  • Fish larvae were collected monthly with an ichthyoplankton net from 18 stations (including four stations located in eelgrass beds) in Gamak Bay and Yeoja Bay, southern Korea, in 2007. In total, 33 species of fish larvae were collected, of which Engraulis japonicus (48.5%), Tridentiger trigonocephalus (21.5%), and Omobranchus elegans (9.2%) were dominant. Dominant species varied seasonally: Hexagrammos otakii during December and January, Pholis nebulosa during January and March, Acanthogobius flavimanus in April, T. trigonocephalus in May, E. japonicus during June, July, September, and November, and Sillago japonica in August. Dominant species also differed between sites inside and outside the bays. Leiognathus nuchalis, O. elegans, and T. trigonocephalus were more abundant inside, while H. otakii was more abundant outside. From cluster analysis, three groups were identified according to sampling months (January-April, May-September, and October-December) and two groups according to station (inside and outside bays). The occurrence of small larvae of almost all major fish species indicated that the bays were used as spawning and nursery grounds. An exception was Lateolabrax japonicus, whose specimens were relatively large (>19 mm TL), suggesting that this fish may spawn offshore, with its juveniles approaching the bays with growth.

  • 한국에 자생하는 22지역의 바위솔속 식물들의 유전적 근연관계를 구명하기 위하여 RAPD분석을 실시하였다. 18종의 바위솔속 식물은 3개의 군(A, B, C 군)으로 집괴되었으며, 나머지 4종은 어떤 분류군에도 속하지 않았다. A군인 연화바위솔(No.18-No.22)은 유사도가 66.3-73.9%로 낮게 나타났으며, B군인 둥근바위솔(No.12-No.17)도 유사도 지수가 69.7-83.7%로 비교적 낮게 나타났다. C군인 바위솔(No.3-No.9)은 안면도 수집종(No.9)을 제외한 유사도 지수가 84.2-92.3%로 높아, 지역종 간에 변이가 적은 비교적 유전적으로 안정된 종으로 생각되었다.

와송약침액이 Oxidant에 의한 신장세포손상에 미치는 영향 (Beneficial effect of Orostachys japonicus A. berger herbal acupuncture on oxidant-induced cell injury in renal epithelial cell)

  • Objectives : This study was performed to determine if Orostachys japonicus A. Berger herbal acupuncture (OjB) provides the protective effect against the loss of cell viability and DNA damage induced by oxidant in renal proximal tubular cells. Methods : The cell viability was evaluated by a MTT reduction assay and DNA damage was estimated by measuring double stranded DNA breaks in opossum kidney (OK) cells, an established proximal tubular cell line. Lipid peroxidation was determined by measuring malondialdehyde (MDA), a product of lipid peroxidation. Results : H2O2 increased the loss of cell viability in a time-dependent manner, which were prevented by 0.1% OjB. The protective effect of OjB was dose-dependent over concentration range of 0.05-0.5%. H2O2 caused ATP depletion and DNA damage, which were prevented by OjB and the hydrogen peroxide scavenger catalase. The loss of cell viability by H2O2 was not affected by the antioxidant DPPD, but lipid peroxidation by the oxidant was completely inhibited by DPPD. Generation of superoxide and H2O2 in neutrophils activated by phorbol-12,13-dibutyrate was inhibited by OjB in a dose-dependent manner. OjB inhibited generation of H2O2 in OK cells treated with antimycin A and exerted a direct H2O2 scavenging effect. Exposure of OK cells to 1 mM tBHP caused a significant depletion of glutathione which was prevented by OjB. OjB accelerated the recovery in cells cultured for 20 hr in normal medium without oxidant following oxidative stress. Conclusions : These results suggest that OjB exerts the protective effect against oxidant-induced cell injury and its protective effect was resulted from radical scavenging and antioxidant activities.

  • 머위 잎을 80% MeOH수용액으로 추출하고, 얻어진 추출물을 EtOAc, n-BuOH 및 $H_2O$로 용매 분획하였다. EtOAc 분획에 대하여 silica gel과 ODS column chromatography를 반복하여 2종의 triterpenoid 화합물을 분리하였다. 각 화합물은 $^1H-{^1H}$ gCOSY, gHMBC 및 gHSQC와 같은 2D-NMR기법을 포함한 스펙트럼 데이터를 해석하여 rosamutin(1) 및 peduncloside(2)로 구조를 결정하였다.

Antibacterial effect of ethylacetate fraction of Orostachys japonicus on Enterococcus faecalis causing Endophthalmitis

  • Endophthalmitis is a disease that causes ocular inflammation and has a catastrophic effect on eyesight. Recent studies show that Enterococcus faecalis is rapidly increasing causative bacterium of endophthalmitis. It is predicted that the increased endophthalmitis by E. faecalis is presumable due to the high resistance of E. faecalis to moxifloxacin (MFX), which is a common antibiotic used for eye drop. Because of the need for therapeutic agents to overcome this problem, this study sought to explore the feasibility of developing a combination therapy using Orostachys japonicus. The ethylacetate fraction of O. japonicus (OJA) used in this study. Antimicrobial activity was tested 13 E. faecalis strains including one E. faecalis standard strain, eight clinically isolated E. faecalis strains and four quinolone resistant E. faecalis strains using CLSI antibiotic susceptibility test method. Minimal Inhibitory Concentration (MIC) of OJA was confirmed to be $500{\mu}g/ml$ for all 13 strains. Then we tested for the synergistic effect of OJA to MFX using checkboard test method. The MIC of MFX was $0.25{\mu}g/ml$ for the standard strain and 8 for the clinical isolates, and $16{\sim}64{\mu}g/ml$ for the quinolone - resistant strains. When OJA was mixed with MFX, no synergistic effect was observed in all strains, but the antibacterial activity of OJA remained unchanged. Most ocular other strains can be removed by MFX except the MFX resistant E. faecalis, which can be removed by OJA in combination therapy. Therefore, OJA can be a potential candidate for the combined treatment endophthalmitis.
