식이지방의 수준과 P/S 비율이 철쥐의 중성스베로이드 및 담즙산 배설에 미치는 영향 (Effect of Level and P/S Ratio of Dietary Fat on Fecal Neutral Steroids and Bile Acids Excretion of Rats)

  • Cholesterol의 주요한 대사산물인 중성steroids와 담즙산의 배설에 관한 내용은 cholesterol 대사를 이해하는데 있어 중요한 부분이다. 식이지방이 steroids의 배설에 미치는 효과에 대하여 아직 일치된 견해가 없어 본 실험에서는 식이지방의 수준(식이에너지의 15, 30 및 45% 과 P/S 비율(0.5 및 1.0)을 달리한 실험식이를 흰쥐에 4주간 급여하여 분변을 통한 총지질, 중성steroids 및 총담즙산배설에 미치는 영향을 조사하였다. 그 결과는 다음과 같다. 체중증가량, 식이섭취량 및 식이변전율은 cholesterol이 섭취되지 않은 경우나 섭취된 경우 모두 P/S 비율에 관계없이 식이지방의 수준에 유의적인 영향을 받았다. 지질소화율은 cholesterol이 섭취되지 않은 경우나 섭취된 경우 모두 P/S 비율에 관계없이 식이지방의 수준에 비례하여 유의적으로 증가하였다. 분변량, 총지질배설량, 중성steroids배설량과 그 조성 및 총담즙산배설량은 식이지방의 수준이나 P/S 비율에 따른 일관성 있는 영향을 나타내지 않았다. Cholesterol의 섭취는 체중증가량과 식이변전율을 유의적으로 향상시켰으며, 총지질배설량을 유의적으로 감소시켰고, 지질소화율을 증가시키고 경향을 나타내었다. 또한 중성 steroids배설량을 증가시키는 경향을 나타내면서, coprostanol의 조성비율을 감소시켰고 coprostanone의 조성비율을 증가시켰다. 반면에 총담즙산배설량은 감소시켰다.

The Concept of Standardized Ileal Amino Acid Digestibilities: Principles and Application in Feed Ingredients for Piglets

  • In this review, the terminology that is used to describe ileal amino acid (AA) digestibilities in piglet feed ingredients is defined. If one accepts that the determination of AA digestibilities should be based on the ileal analysis method, one should consider that ileal digesta contains variable amounts of endogenous crude protein (CP), which originates mainly from digestive secretions, sloughedoff epithelial cells and mucins. The ileal endogenous CP and AA losses are separated into basal ileal endogenous CP and AA losses ($IAAL_{B}$), which are not influenced by the feed ingredient composition, and specific ileal endogenous CP and AA losses ($IAAL_{S}$), which are induced by feed ingredient characteristics such as level and type of fiber and anti-nutritional factors (ANF). Depending how ileal endogenous CP and AA losses are considered in the measurement of CP and AA digestibilities, digestibility values are expressed as apparent (AID), standardized (SID), or true (TID) ileal digestibilities of CP and AA. The main concern associated with the use of AID values in diet formulation for pigs is that they are not additive in mixtures of feed ingredients. Consequently, the concept of standardized ileal CP and AA digestibilities was introduced by correcting AID values for basal ileal endogenous CP and AA losses ($IAAL_{B}$). The correction for both $IAAL_{B}$ and $IAAL_{S}$ yields TID values, however, routine procedures to measure $IAAL_{S}$ are not yet available. In principle, SID values should be preferred, because they represent the fundamental properties of the feed ingredient. There exist only few reports on SID of CP and AA in feedstuffs frequently used in piglet nutrition. These include soybeans (SB), soybean meal (SBM), soy proteins (SP), soy protein concentrate (SPC), soy protein isolate (SPI), corn gluten (CG), wheat gluten (WG), pea protein (PeaP), potato protein (PotP), fish meal (FM) and whey proteins (WP), but the results obtained are inconsistent. Differences in SID values within feed ingredients may, at least in part, be attributed to different processing conditions or inherent differences of the assay feed ingredients. Moreover, there is some evidence that the determination of SID values and $IAAL_{B}$ in piglets may be confounded by the dietary CP level of the assay diet, age and (or) body weight (BW), the level of feed intake or the methodological approach used to determine $IAAL_{B}$.

Effects of inclusion level and amino acid supplementation on energy values of soybean oil determined with difference or regression methods in growing pigs

  • Objective: This study was conducted to evaluate the effects of inclusion level and amino acid (AA) supplementation on energy values of soybean oil (SO) as determined by difference method or regression method when fed to growing pigs. Methods: Thirty-six barrows (initial body weight: 28.0±1.3 kg) were randomly assigned to one of 6 dietary treatments, which included 2 control diets formulated using a basal diet with or without AA supplementation, and 4 experimental diets with 5% or 10% SO addition in the 2 control diets, respectively. All pigs were individually housed in metabolism crates for 19 d, and during the last 5 d, total urine and feces production were collected. The nutrient digestibility in diets and the digestible energy (DE) and metabolizable energy (ME) values of SO were determined using the difference method and the regression method, respectively. Results: Our results showed that there were no interaction effects (p>0.05) between AA supplementation and SO inclusion levels on energy values of SO and dietary nutrient digestibility. The DE and ME values of SO determined by the difference method were not affected (p>0.05) by AA supplementation, however, the ME value of SO increased (p<0.05) as the inclusion level of SO increased. Moreover, the energy values of SO determined using the regression method were close to those determined using difference method with 10% SO inclusion, but were greater than those obtained using difference method with 5% SO inclusion. Conclusion: We concluded that the DE and ME values of SO increased with the inclusion level but were not affected by AA supplementation in the range of 0% to 10%. The difference method can substitute for the regression method to determine the DE and ME values of SO when the inclusion level is 10%, but not at 5% inclusion level.

Influence of Dietary Supplementation of Condensed Tannins through Leaf Meal Mixture on Intake, Nutrient Utilization and Performance of Haemonchus contortus Infected Sheep

  • The study assessed the effect of dietary supplementation of leaf meal mixture (LMM) containing condensed tannins (CT) on feed intake, nutrient utilization and performance of sheep infected with Haemonchus contortus. Eighteen adult sheep of similar age and body weight ($25.03{\pm}1.52$) were included in this study and out of these, 12 sheep were infected with single dose of infective third stage larvae of H. contortus at 2,000 larvae per sheep. The experimental sheep were allocated in three different groups' i.e. negative control (NC; no infection), control (C; H. contortus infected) and treatment (T; H. contortus infected+CT at 1.5% of the DM through LMM) and the experiment was conducted for a period of 90 d. The intake of dry matter (DM), organic matter (OM) and digestibility of DM, OM, neutral detergent fibre (NDF) and acid detergent fibre (ADF) were comparable among three animal groups. However, digestibility of crude protein (CP) and ether extract (EE) were significantly (p<0.05) higher in NC group as compared to both C and T groups. Nitrogen (N) retention (g/d or % of N intake) was significantly (p = 0.038) lower in C group as compared to T and NC groups. Daily intake (g/kg $W^{0.75}$) of digestible crude protein (DCP), digestible organic matter (DOM) and total digestible nutrient (TDN) did not differ significantly (p<0.05) in the three groups. Haemoglobin (Hb) and packed cell volume (PCV) were significantly (p<0.001) higher in treatment group as compared to control. The level of Hb and PCV reduced (p<0.001) after 30 days of experimental feeding. CT significantly (p<0.001) reduced serum urea in T group as compared to NC and C groups. Serum proteins differed significantly (p<0.01) among the three groups. The activity of serum enzymes AST, ALT, ALP and LDH were also statistically non significant (p<0.05) among treatments. The weight of abomasal lymph nodes (ALN) in T group was higher (p<0.05) than in C group. Treatment group had lower (p<0.05) total worms and fecal egg count compared to control group. It may be concluded that dietary supplementation of CT through LMM significantly improved the N retention, and inhibited the different developmental stages of Haemonchus contortus in experimental sheep.

Growth and Nutrient Utilization in Buffalo Calves Fed Ammoniated Wheat Straw Supplemented with Sodium Sulphate

  • Ten male buffalo calves (aged 6-8 months, average body wt. $88.5{\pm}0.5kg$) were divided into two groups of five animals in each. All the animals were fed on urea-ammoniated wheat straw (4% urea, 50% moisture) along with concentrate mixture (50:50 on DM basis). In addition animals in group II were given sodium sulphate to see the effect of sulphur on the utilization of nitrogen added through urea-ammoniation. This feeding practice continued for a period of 120 d, during which fortnightly body weights were taken to assess their growth rate. A metabolism trial was conducted after 90 days of feeding to know the digestibility of nutrients and their balance. Results revealed no significant difference in the intake of DM and other nutrients in two groups. The digestibility of DM, OM, EE, NDF, ADF and cellulose was alike in animals fed ammoniated straw and ammoniated straw+sodium sulphate supplemented group, whereas the digestibilities of CP and hemicellulose was significantly (p<0.01) more in group I and II respectively. There was no significant difference in intake of nitrogen, calcium and phosphorus in 2 groups. Similarly, the balance of these 3 nutrients was positive and statistically alike in two groups. Intake and excretion of sulphur through faeces and urine was significantly (p<0.01) more in group II than in group I. Inspite of higher excretion of sulphur through faeces and urine in group II, the sulphur balance was significantly (p<0.05) more in group II than in group I, probably due to significantly (p<0.01) higher intake of sulphur in this group. There was no significant difference in total body weight gain or average daily gain between two groups, indicating that addition of sodium sulphate did not have any positive effect on these parameters. Similarly the intake of DM, DCP and TDN were also alike in two groups. The DCP and TDN values of the two diets were 8.0, 60.4 and 6.8, 56.6% respectively. Feeding cost/unit gain was alike in both the groups.

Effect of Feeding Different Ratios of Green Fodder and Straw Supplemented with Wheat Bran on the Performance of Male Crossbred Calves

  • Twenty male crossbred calves of about one year of age (average body weight, 196 kg) were distributed in four equal groups following complete randomized design. Wheat bran was supplemented to four different combinations of wheat straw and green fodder (Sorghum vulgare) at 40:60, 30:70, 20:80 and 10:90 ratios (on as fed basis) for the feeding of animals in Group 1, 2, 3 and 4, respectively. The feeding trial was continued for a period of 70 days including one metabolism trial of 6 days collection of feed, faeces and urine sample to determine the intake and utilization of nutrients. The intakes (g/kg $W^{0.75}$) of DM, TDN and CP were $93.0{\pm}1.8$, $55.5{\pm}1.1$ and $9.51{\pm}0.18$ in Group 1; $98.0{\pm}1.8$, $59.6{\pm}1.1$ and $10.33{\pm}0.19$ in Group 2; $98.1{\pm}2.4$, $60.5{\pm}1.5$ and $10.79{\pm}0.26$ in Group 3; and $97.7{\pm}1.7$, $59.1{\pm}1.0$ and $10.78{\pm}0.19$ in Group 4, respectively. The digestibility of nutrients did not differ significantly among the groups. Relatively higher nutrient intake and balances of nitrogen reflected non-significantly high her live weight gain in the later three groups (436, 439 and 464 g, respectively) as compared to Group 1 (400 g). The DM intake remained unchanged by increasing the proportion of green fodder beyond 20:80 ratio and thus was assessed to be satisfactory for optimum productivity in animals.

Effect of Modified Casein to Whey Protein Ratio on Dispersion Stability, Protein Quality and Body Composition in Rats

  • The present study was designed to investigate the effects of protein formula with different casein (C) to whey protein (W) ratios on dispersion stability, protein quality and body composition in rats. Modification of the casein to whey protein (CW) ratio affected the extent of protein aggregation, and heated CW-2:8 showed a significantly increased larger particle (>100 ㎛) size distribution. The largest protein aggregates were formed by whey protein self-aggregation. There were no significant differences in protein aggregation when the CW ratios changed from 10:0 to 5:5. Based on the protein quality assessment (CW-10:0, CW-8:2, CW-5:5, and CW-2:8) for four weeks, CW-10:0 showed a significantly higher feed intake (p<0.05), but the high proportion of whey protein in the diet (CW-5:5 and CW-2:8) increased the feed efficiency ratio, protein efficiency ratio, and net protein ratio compared to other groups. Similarly, CW-2:8 showed greater true digestibility compared to other groups. No significant differences in fat mass and lean mass analyzed by dual-energy x-ray absorptiometry were observed. A significant difference was found in the bone mineral density between the CW-10:0 and CW-2:8 groups (p<0.05), but no difference was observed among the other groups. Based on the results, CW-5:5 improved protein quality without causing protein instability problems in the dispersion.

Animal protein hydrolysate reduces visceral fat and inhibits insulin resistance and hepatic steatosis in aged mice

  • BACKGROUND/OBJECTIVES: An increasing life expectancy in society has burdened healthcare systems substantially because of the rising prevalence of age-related metabolic diseases. This study compared the effects of animal protein hydrolysate (APH) and casein on metabolic diseases using aged mice. MATERIALS/METHODS: Eight-week-old and 50-week-old C57BL/6J mice were used as the non-aged (YC group) and aged controls (NC group), respectively. The aged mice were divided randomly into 3 groups (NC, low-APH [LP], and high-APH [HP] and fed each experimental diet for 12 weeks. In the LP and HP groups, casein in the AIN-93G diet was substituted with 16 kcal% and 24 kcal% APH, respectively. The mice were sacrificed when they were 63-week-old, and plasma and hepatic lipid, white adipose tissue weight, hepatic glucose, lipid, and antioxidant enzyme activities, immunohistochemistry staining, and mRNA expression related to the glucose metabolism on liver and muscle were analyzed. RESULTS: Supplementation of APH in aging mice resulted in a significant decrease in visceral fat (epididymal, perirenal, retroperitoneal, and mesenteric fat) compared to the negative control (NC) group. The intraperitoneal glucose tolerance test and area under the curve analysis revealed insulin resistance in the NC group, which was alleviated by APH supplementation. APH supplementation reduced hepatic gluconeogenesis and increased glucose utilization in the liver and muscle. Furthermore, APH supplementation improved hepatic steatosis by reducing the hepatic fatty acid and phosphatidate phosphatase activity while increasing the hepatic carnitine palmitoyltransferase activity. Furthermore, in the APH supplementation groups, the red blood cell (RBC) thiobarbituric acid reactive substances and hepatic H2O2 levels decreased, and the RBC glutathione, hepatic catalase, and glutathione peroxidase activities increased. CONCLUSIONS: APH supplementation reduced visceral fat accumulation and alleviated obesity-related metabolic diseases, including insulin resistance and hepatic steatosis, in aged mice. Therefore, high-quality animal protein APH that reduces the molecular weight and enhances the protein digestibility-corrected amino acid score has potential as a dietary supplement for healthy aging.

해양식량자원의 가공조건별 영양적 품질평가 -2. 명태연육 단백질품질에 미치는 냉동변성방지제의 영향- (Effects of Processing Conditions on Nutritional Qualities of Seafood -2. Effects of Cryoprotectants on the Protein Qualities of Pollock Surimi-)

  • 명태연육 냉동변성을 억제할 수 있는 냉동변성방지제의 적정량을 알아보기 위하여 crystalline sorbitol을 비롯한 세가지의 변성방지제 및 sorbitol/sucrose 혼합제제를 첨가하 여 $-25^{\circ}C$에서 16주간 저장했을 때의 단백질품질 변화를 실험하였다. $8\%$ 수준의 sucrose-sorbitol 혼합제제(1:1, w/w)를 $0.2\%$ Na-pyrophosphate/ Na-triphosphate(1:1)와 병용하였을 때 drip loss가 가장 적었으며, 염용성단백질량은 가장 많았다. 또한 혼합제제를 사용했을 때는 trypsin inhibitor량, 단백질소화율 및 단백효율비의 변화가 거의 없어 단백질품질 보전에 효과적이었으나, 냉동변성방지제를 처리하지 않았을 경우에는 단백효율비와 소화율은 급격히 떨어졌었다. Polyphosphate나 maltodextrin의 경우에는 처리 농도가 증가하여도 소화율저하 방지에는 별효과가 없었으나, $4{\sim}6\%$의 sorbitol, 또는 $10\%$수준의 sucrose는 polyphosphate를 병용하지 않아도 소화율저하는 효과적으로 막을 수 있었다. $8\%$ 수준의 sorbitol/sucrose 혼합제제(5:3, w/w)처리가 소화율 보전효과로 볼 때 냉동연육의 단백질품질저하 방지에 가장 효과적이었다.

In vitro에서 메밀 다당분획이 casein 가수분해에 미치는 영향 (Effects of Buckwheat Polysaccharides on the Digestibilty of Casein in vitro)

  • 본 실험에서는 메밀에서 추출한 다당분획이 in vitro에서 casein 가수분해에 미치는 영향을 관찰하였다. 처리방법(날것, 볶은것, 찐것)을 달리한 메밀로부터 추출한 다당중 분자량 10,000 이상의 고분자 가용성다당(HMS-P)과 저분자 불용성다당(LMI-P)이 in vitro에서 trypsin에 의한 casein의 가수분해율을 각각 $80{\sim}89%$, $69{\sim}99%$ 감소시켰으며, 분자량 10,000 이하의 저분자 가용성다당(LMS-P)은 $5{\sim}7%$ 감소시켰다. Chymotrypsin에 의한 가수분해율은 고분자 가용성다당(HMS-P)과 저분자 불용성다당에서 각각 $63{\sim}88%$, $71{\sim}79%$이였으며 저분자 가용성다당은 $2{\sim}5%$였다. Casein 가수분해율을 현저히 감소시킨 볶은 메밀로부터 추출한 다당을 casein과 흔합하여 trypsin과 chymotrypsin으_로 각각 가수분해시킨 후 가수분해산물을 Sephdex G-100 column에 통과시켰다 Casein만 가수분해시켰을 때와 저분자 가용성다방(LMS-P)을 첨가했을 때에 가수분해물은 total volumne에서 1개의 peak가 용출되었으나 고분자 가용성다당(HMS-P)과 저분자 불용성다당(LMI-P)에서는 void volumne에서 고분자량의 peptide peak가 1개 더 용출되었다. Casein 가수분해를 억제하는 고분자 가용성다당(HMS-P)의 구조를 분식한 결과 주로 1,4결합의 glucose로 구성되어 있으며 소량의 1,6결합의 glucose와 단일 glucose도 함유되어 있었다. 따라서 메밀로부터 추출한 고분자 가용성다당분획에 의한 casein 가수분해율의 감소는 다당에 의한 것이라기 보다는 괌자량 10,000이상의 수용성물질에 의한 것으로 추측된다.
