Fed-Batch Sorbose Fermentation Using Pulse and Multiple Feeding Strategies for Productivity Improvement

  • Microbial oxidation of D-sorbitol to L-sorbose by Acetobacter suboxydans is of commercial importance since it is the only biochemical process in vitamin C synthesis. The main bottleneck in the batch oxidation of sorbitol to sorbose is that the process is severely inhibited by sorbitol. Suitable fed-batch fermentation designs can eliminate the inherent substrate inhibition and improve sorbose productivity. Fed-batch sorbose fermentations were conducted by using two nutrient feeding strategies. For fed-batch fermentation with pulse feeding, highly concentrated sorbitor (600g/L) along with other nutrients were fed intermittently in four pulses of 0.5 liter in response to the increased DO signal. The fed-batch fermentation was over in 24h with a sorbose productivity of 13.40g/L/h and a final sorbose concentration of 320.48g/L. On the other hand, in fed-batch fermentation with multiple feeds, two pulse feeds of 0.5 liter nutrient medium containing 600g/L sorbitol was followed by the addition of 1.5 liter nutrient medium containing 600g/L sorbitol at a constant feed rate of 0.36L/h till the full working capacity of the reactor. The fermentation was completed in 24h with an enhanced sorbose productivity of 15.09g/L/h and a sorbose concentration of 332.60g/L. The sorbose concentration and productivity obtained by multiple feeding of nutrients was found to be higher than that obtained by pulse feeding and was therefore a better strategy for fed-batch sorbose fermentation.

Temporal Changes of Limiting Nutrients and Phytoplankton Growth Rate in Lake Paldang (팔당호 식물플랑크톤의 제한영양염과 성장률의 경시적 변화)

  • This study was conducted to determine limiting nutrients and the physiological characteristics of phytoplankton in response of nutrients in Lake Paldang from March 2002 to October 2002. A field research was conducted along with laboratory batch culture experiment to find the limiting nutrients and the growth kinetics. According the results of Chl. a TP relationship, TN/TP ratio, and nutrient addition bioassay, phosphorus appeared to be a major limiting nutrient in Lake Paldang and thus the lake productivity was greatly influenced by it. P limitation for the phytoplankton of Lake Paldang varied with season, and the possibility of limitation by nitrogen and silica also occurred. The degree of P limitation was greatest during spring when the concentration of dissolved phosphorus is relatively much lower than summer and autumn. The maximum growth rate (${\mu}_{max}$) and half saturation concentration ($K_u$) of Lake Paldang phytoplankton ranged from 0.8${\sim}$1.1$day^1$ and from 0.1${\sim}$O.8${\mu}M$, respectively. $K_u$ was highest during May ($0.8{\mu}M$) and the lowest during September ($0.1{\mu}M$). Such result may be induced by the phytoplankton cell quota that showed the lowest concentration ($0.13{\mu}gP/{\mu}gChl.$ a) during May. The growth kinetics showed that phytoplankton growth in Lake Paldang was faster during summer and autumn than spring, suggesting that the Potential of algal bloom is high after the summer monsoon season.

  • Streptomycetes are Gram-positive microorganisms producing secondary metabolites through unique physiological differentiation [4]. The microbes show unusual morphological differentiation to form substrate mycelia, aerial mycelia, and arthrospores on solid medium [19]. Substrate mycelium growth is sustaining with sufficient nutrients in the culture medium. The concentration of a specific individual substrate in the culture environment is the most important extracellular factor allowing vegetative mycelia growth, where extracellular hydrolytic enzymes participate in the utilization of waterinsoluble substrates. However, with starvation of nutrients in the culture medium, the vegetative mycelia differentiate to aerial mycelia and spores. It has been considered that shiftdown of essential nutrients for mycelia growth is the most important factor triggering morphological and physiological differentiation in Streptomyces spp. Since proteineous macromolecule compounds are the major cellular components, these are faced to endogenously metabolize following a severe depletion of nitrogen source in culture nutrients (Fig. 1). Various proteases were identified of which production was specifically related with the phase of mycelium growth and also morphological differentiation. The involvement of proteases and protease inhibitor is reviewed as a factor explaining the mycelium differentiation in Streptomyces spp.

  • There is a growing need to develop ion sensors for agriculture. As a result, several technologies have been developed, such as colorimetry, spectrophotometry, and ion-selective electrode (ISE). Among them, ISE has some advantages compared to others. First, it does not require pre-treatment processes and expensive equipment. Second, it is possible for the portable detection system by introducing small-sized electrodes. Finally, real-time and multiple detections of several ions are pursued. It is well-known that N, P, and K nutrients are critical for crop growth. With the development of agriculture techniques, the importance of soil nutrient analysis has attracted much attention for cost-effective and eco-friendly agriculture. Among several issues, minimizing the use of fertilizers is significant through quantitative analysis of soil nutrients. As a result, it is highly important to analyze certain nutrients, such as N (ammonium ion, nitrate ion, nitrite ion), P (dihydrogen phosphate ion, monohydrogen phosphate ion), and K (potassium ion). Therefore, developing sensors for accurate analysis of soil nutrients is highly desired. n this study, several ISEs have been fabricated to detect N, P, and K. Their performance has been intensively studied, such as sensitivity, selectivity coefficient, and concentration range, and compared with commercialized ISEs. In addition, preliminary tests on the in-situ N, P, and K monitoring have been conducted inside the soil.

Mass Loss and Changes of Mineral Nutrients during the Decomposition of Mushrooms, Russula alboareolata and Lactarius violascens

  • Mass loss and changes of mineral nutrients during the decomposition of mushrooms, Russula alboareolata and Lactarius violascens, were investigated for 7d from June 29 to July 5 in 1999 in an oak stand in Kongju, Korea. At 7d after installation of litterbags, the remaining mass of R. alboareolata and L. violaxcens was 9.4% and 25.9%, respectively. The mass loss rate of R. alboareolata was significantly higher than that of L. violascens. Concentration of N, P, K, Ca and Mg of R. alboareolata and L. violascens were 37.7 mg/g, 0.97mg/g, 38.25 mg/g, 0.04 mg/g, and 0.75 mg/g for R. alboareolata and 45.7 mg/g, 1.31mg/g, 24.0 mg/g, 0.06 mg/g, and 0.80 mg/g for L. violascens, respectively. Concentrations of nutrients in R. alboareolata and L. violascens were much higher than those in the surrounding leaf litter. N, P, Ca and Mg concentrations in the decomposing mushroom tissue were higher during the experimental period in both species than initial concentrations. Potassium increased during the first 3 d and then decreased in both species. Potassium contents in the mushroom were much greater than those of Ca and Mg. Except for Ca, there was no immobilization period in all the nutrients during decomposition. At 7 d after installation of litterbags, the remaining N, P, K, Ca and Mg of R. alboareolata and L. violascens were 9.8%, 8.9%, 2.7%, 47.7%, and 14.8% of the initial contents for R. alboareolata and 28.2%,30.5%, 19.6%, 199.9%, and 02.1% for L. violascens, respectively. Nutrients could be relocated spatially during the formation and decomposition of the Basidiomycetes fruiting body.

  • Dynamics of nutrients, non-polar, water solubles, acid solubles and acid insolubles (lignin) in decomposing litter were investigated for 2 years in the oak, Quercus acutissima, and the pitch pine, Pinus rigida, stands in the vicinity of Kongju, Chungnam Province. Nitrogen and phosphorus conetrations in decomposing litter increased with time elapsed, however, potassium decreased rapidly within three months and then remined constant with time elapsed. Calcium concentration in needle litter during experimental period was lower than that of initial concentration in needle litter during experimental period was lower than that of initial concentration, and showed no significant variation with time elapsed. Calcium concentration in oak litter during the experimental period, however, were higher than that of initial concentration. Magnesium concentration in oak litter decreased repidly during six months, and then remaines constant thereafter. Annual amount of nitrogen, phosphorus, potassium, calcium and magnesium which returned to soil via litter decomposition in the oak and the pitch pine stands was $3.3g/m^2$ and $0.9g/m^2$ for N, $0.03g/m^2$ and $0.01g/m^2$ for P, $1.3g/m^2$ and $0.7g/m^2$ for K, $0.7g/m^2$ and $1.2g/m^2$ for Ca, $0.9g/m^2$ and $0.4g/m^2$ for Mg, respectively. Non-polar, and water- and acid-soluble fractions in decomposing litter decreased and lignin increased with time.

A Study on the Influencing Factors of Macrountrient Concentrations in Human Milk (모유의 주요 영양소 농도에 영향을 주는 요인에 관한 연구)

  • This study was conducted longitudinally following the total nitrogen , total lipid, and lactose concentrations and their factors on concentrations of energy-yielding nutrients in human milk from 2-5 days to 12 weeks postpartum of 45 lactating Korean mothers. All samples were from well-defined subjects. And uniform collection procedures were used . Total nitrogen concentration of milk decreased significantly from 385mg/이 at 2-5 days to 201mg/dl at 12 weeks postpartum. Total lipid concentration increased from 1.98g/dl at 2-5 days to 3.09g/dl at 12 weeks postpartum. However, there was a large variation in the concentration of total lipids. Lactose concentration increased signifciantly from 6.53g/dl at 2-5 days to 7.48g/dl at 12 weeks. Total nitrogen concentration in milk was positively related to pregnancy weight gains at 2 and 6 weeks postpartum. Total lipid concentration was positively related to infant birth weight : however, it was negatively related to the maternal weight loss. In addition, total lipid concentration was positively related to parity and maternal percentage body fat at 12 weeks postpartum. Milk lactose concentration was positively related to parity at 2 weeks postpartum.

  • In order to elucidate the ecological responses of B. latissimum under global warming situation and prepare the conservation measures, we cultivated the plant within control(ambient $CO_2+$ambient temperature) and treatment(elevated $CO_2+$elevated temperature) from 2010 to 2011. Rising $CO_2$ concentration was treated with 2 times and temperature increased with $2.0^{\circ}C$ above than control. As a result, Growth response has received more the effect of $CO_2+$Temperature rising than light, moisture, nutrients, and it was grow well in $CO_2+$temperature rise sphere when many nutrients, and it was grow well in $CO_2+$temperature rise sphere when many nutrients. No. of leaves were many number in the treated group compared to the control at a low light gradient and high nutrient gradient, leaf width was narrow in the treatment compared to the control in the middle gradient of light and nutrients. Shoot length, petiole length, lamina length was no difference between control and $CO_2+$temperature rise sphere. Based on the results of this study, in order to preserve the B. latissimum in future climate, we should be supply of nutrients(containing ${NH_4}^+$, $NO_3$, $P_2O_5$, $K_2O$, etc.) in the habitat of the B. latissimum, and must ensure that the habitat of B. latissimum is not destroyed. Also, We should be find similar area with habitat of the B. latissimum, restoring, expanding.

Comparison of the Fertility of Stream Waters Depending on the Drainage Systems in the Lake Shihwa Watershed, Korea (시화호 유역에서 배수시스템별 하천수의 비옥도 비교)

  • The fertility of stream water in major streams of the Lake Shihwa Watershed was compared using water analyses and algal growth potential test (AGPT) in typical drought seasons from December 2001 to April 2002, The water quality varied considerably depending on streams. These streams were very rich in inorganic nutrients that the nutrient levels and characteristics of each stream could be easily determined. Through AGPT, 63.6% of growth was observed in the average values of each stream, with non-growth accounting for 36.4%. AGPT results showed that 40.9% of the 22 stations were in hypertrophic condition and 54.5% in eutrophic condition. AGPT values were significantly correlated with TIN, $NH_4$, and SRP (p <0.001); compared to other nutrients, however, they were more related to SRP and $NH_4$. Moreover, the values increased with high concentration of N and P and low N/P ratios. Nonetheless, the values were more dependent on P concentration than N concentration. This suggests that the effect of P on the water quality of lake situated in downstream may serve as a potential indicator of phytoplankton development. Depending on the drainage pattern of streams, the wastewaters of wastewater treatment plant (WwTP) and untreated wastewater (UTW) were found to have 53.4% and 46.6%, respevtively, of TIN, 51.9% and 48.1% of $NH_4$, 62.9% and 37.1% of $NO_3$, 62.6% and 37.4% of SRP, and 44.1% and 55.9% of SRSi. The AGPT value was 51.1% in WwTP wastewater and 48.9% in UTW wastewater, the concentration of WwTP wastewater was slightly higher. For untreated wastewaters flowing into the constructed wetland and into the lake, TIN accounts for 43.0% and 57.0%, respectively, of nitrogen components, $NH_4$ 44.4% and 55.6%, $NO_3$ 39.6% and 60.4%, SRP 53.5% and 46.5%, and SRSi 52.3% and 47.7%, respectively. The AGPT value was 58.0% in the constructed wetland and 42.0% in Lake Shihwa; the concentration in streams flowing into the wetland was slightly higher. Therefore, Persistent and large development of phytoplankton in Lake Shihwa cannot be prevented unless a measure tophytoplankton control is implemented. This is because the concentration of nutrients in specific streams flowing into the lake is very high, even though the inflow of water is low.

Removal of Heavy Metals in Wastewater Using Glycine max Merr (콩을 이용한 식물filter에 의한 중금속 제거에 관한 연구)

  • For the removal of heavy metals, Cd, Cu and Cr were used. The initial concentration of Cd and Cu were 1-10 ppm, the removal efficiency of Cd and Cu was 76.2-89.0% and 69.0-79.0%, respectively. The initial concentration of Cr were 1~5 ppm, and the-removal efficiency was low especially at high concentration. In general, the initial concentrations of heavy metals had no relation to the removal efficiency. At the beginning, the removal efficiency was very high, but it was maintained at constant concentration. The trends of accumulations of heavy metals in the stem increased in proportion to the initial concentration. The removal efficiency of heavy metals increased a little bit when nutrients existed in the solution. So that, the initial concentration of Cd and Cu were 1-10 ppm, the removal efficiency of Cd and Cu was 84.8-91.0% and 75.9-82.0%. The initial concentration of Cr were 1-5 ppm, the removal efficiency was 25.0-67.0%.
