The introduction of doubled haploid (DH) approach into breeding programs has reduced the times and population sizes required for the production of pure lines. We carried out the experiment for development on effective method of producing haploid in wheat. Emasculated spikelets of wheat were pollinated with maize pollen and cultured in the solution containing 40 g/L sucrose and 2,4-D, NAA, 2,4,5-T and dicamba 24 h after pollination, and then incubated until embryo rescue. Fourteen to sixteen days after pollination, the embryos are excised and cultured in half-strength MS basal medium supplemented with 20 g/L sucrose and 1 mg/L NAA. The type of plant growth regulators was found to be most significant in production of haploid plants. The application of synthetic auxins to pollinated florets, stimulates haploid embryo development to a stage where the embryos can be rescued onto nutrient media. The percentage of seeded florets was significantly affected by 100 mg/L 2,4-D, 150 mg/L 2,4,5-T and 50 mg/L dicamba. The percentage of embryos formed was significantly increased by treatment with 2,4-D and 2,4,5-T at 100 mg/L, and dicamba at 50 mg/L, but the treatments with 150 mg/L 2,4-D inhibited embryo development and plant regeneration. The optimum application time of plant growth regulators was 24 hrs after pollination.
This study was carried out to isolate soil bacteria with antimicrobial activity and evaluate antimicrobial substances produced by isolated bacteria. Among many isolates Bacillus subtilis DS660 and Paenibacillus polymyxa DS842 showed high antimicrobial activities against 6 species of microbial residents on human skin and 3 species of pathogenic bacteria. DS660 and DS842 showed 15.3~26.8 and 11.3~27.5 mm of inhibition zone diameter, respectively on nutrient agar medium against most target bacteria and fungi. DS660 and DS842 produced $57{\pm}8$ and $170{\pm}15{\mu}mol/ml$ of siderophore, respectively as an antimicrobial substance. Analysis of ethyl acetate extract of culture supernatants of DS660 and DS842 suggested production of glycolipid biosurfactant which reduced surface tension of culture supernatant of DS660 and DS842 from 60.0 to 40.3 and 30.3 mN/m, respectively. DS660 and DS842 also showed $169.2{\pm}9.9$ and $357.2{\pm}13.7nmol/min/mg$ protein of ${\beta}-1,3$-glucanase activity, respectively, and hydrolyzed cell wall components of 3 bacterial species. These results suggest that B. subtilis DS660 and P. polymyxa DS842 may be utilized as an environment-friendly biocontrol agent against some skin microbes and pathogenic bacteria.
Objective: The objectives of this study were to evaluate the effects of daily feed intake during the laying period on embryonic myogenic differentiation 1 (MYOD1), myogenic factor 5 (MYF5), and myogenic factor 6 (MYF6) gene expression in genetically fat and lean lines of chickens. Methods: An experiment in a 2×2 factorial design was conducted with two dietary intake levels (100% and 75% of nutrition recommendation) and two broiler chicken lines (fat and lean). Two lines of hens (n = 384 for each line) at 23th week of age were randomly divided into 4 treatments with 12 replicates of 16 birds. The experiment started at 27th week of age (5% egg rate) and ended at 54th week of age. Hatched eggs from the medium laying period were collected. Real time polymerase chain reaction analysis was used to analyse the MYOD1, MYF5, and MYF6 mRNA levels of E7, E9, E11, E13, and E15 body tissues and E17, E19, and E21 chest and thigh muscle samples. Results: The results indicated that there were significant effects of line, dietary intake, and interactions between them on MYOD1, MYF5, and MYF6 gene mRNA expression levels in embryonic tissues. Low daily feed intake did not change the expression trend of MYOD1 mRNA in either line, but changed the peak values, especially in lean line. Low daily feed intake altered the trend in MYF5 mRNA expression level in both lines and apparently delayed its onset. There was no apparent effect of low daily feed intake on the trends of MYF6 mRNA expression levels in either line, but it significantly changed the values on many embryonic days. Conclusion: Maternal nutrient restriction affects myogenesis and is manifested in the expression of embryonic MYOD1, MYF5, and MYF6 genes. Long term selection for fat deposition in broiler chickens changes the pattern and intensity of myogenesis.
This experiment was conducted to determine the maximum dietary energy levels on growth performance and carcass characteristics of White Pekin duck. the Six dietary treatments were formulated based on their apparent metabolizable energy (AME) concentrations from 2,700 to 3,200 kcal/kg with a 100 kcal/kg gap to evaluate the accurate dietary AME requirement to address current knowledge and further issues for fulfilling the genetic potential of meat-type white Pekin ducklings. A total of 432 one-day-old male White Pekin ducklings were randomly allocated into one of six dietary treatments with six replicates (12 birds per pen). The diets were formulated as corn-soybean meal-based diets to meet or exceed the Nutrient Requirement of Poultry specification for meat-type ducks. Growth performance indices (i.e. average daily gain [ADG], average daily feed intake, feed conversion ratio) were measured weekly. Medium body weight (BW) ducklings from each pen were sacrificed to analyze the carcass traits and abdominal fat content on day 21. Obtained data were analyzed to estimate significant effect using the one-way ANOVA of IBM SPSS Statistics (Version, 25). If the p-value of the results were significant, differences in means among treatments were separated by Tukey's post hoc test. Significant differences were then analyzed with a linear and quadratic broken model to estimate the accurate concentration of AME. Ducklings fed higher dietary AME diets increased (p < 0.05) BW, ADG. Ducklings fed higher AME than 2,900 kcal/kg diets increased abdominal fat accumulation and leg meat portion. The estimated requirement by linear plateau method showed from 3,000.00 kcal/kg to 3,173.03 kcal/kg whereas the requirement by quadratic plateau method indicated from 3,100.00 kcal/kg to 3,306.26 kcal/kg. Collectively, estimated dietary requirements exhibit diverse results based on the measured traits and analysis methods. All the estimated requirements in this experiment present higher than previous research, the maximum requirement for the next diet formulation should be selected by the purpose of the diet.
Purpose: This study investigates the necessary educational materials from the perspective of caregivers managing food allergies of preschoolers. Methods: An online survey enrolled 400 caregivers of preschoolers with food allergies. The study was conducted in July 2022, and data were analyzed using the IBM SPSS Statistics 25. Results: The caregivers of preschoolers with food allergies showed low food allergy knowledge. The highest percentage of incorrect answers were for items related to the cause (74.2%) and response (65.2%) to food allergies. Moreover, categorizing the levels of knowledge as high, medium, and low, we found that caregivers with younger children exhibited a better understanding of food allergies. Approximately 56.9% of subjects in the high class were caregivers with preschoolers under three years, whereas caregivers with preschoolers over four years comprised 43.2% of the high-class subjects. Regarding food allergy treatment, four questions measured how well the caregivers tended to the children. With higher levels of knowledge, caregivers were more likely to read nutrient information and check for food allergens. The caregivers of preschoolers who were clinically diagnosed with food allergies demonstrated a higher percentage of using separate ingredients and cooking utensils. Conclusion: This study shows how the caregivers' knowledge levels are critical in managing and treating food allergies among preschoolers. With caregivers' lower knowledge of food allergies and preschoolers younger than four years, self-diagnosis of food allergies and existing food allergies were not managed efficiently. Hence, customized education for caregivers is essential to improve their knowledge level regarding limiting foods that may cause allergies, reading food labels, and using ingredients and cooking utensils separately.

Volvariella bombycinais an edible mushroom commercially cultivated in Korea and other countries. In this study, the culture characteristics and optimal growth conditions of milky mushroom strains were determined. The growth temperature and pH range of V. bombycinawas extensively investigated between 15 and 35℃ and pH 3-11. For efficient cultivation, 20 types of nutrient sources were selected, specifically, 21 carbon sources, 6 organic nitrogen sources, 7 inorganic nitrogen sources, 13 amino acids, 9 organic acids, and 13 inorganic salts. The impact of each of the selected nutrition sources and their concentration on growth was investigated. The optimum temperature and pH were determined to be 30℃ and pH 8.0, respectively. The optimum concentration of medium elements required for mycelial growth of V. bombycinawas determined to be as follows: carbon source, 0.7% Soluble starch; organic nitrogen source, 0.7% Soytone; inorganic nitrogen source, 1.0% (NH4)2C4H4O6; amino acid, 0.1% cysteine; organic acid, 0.07% lactic acid; and inorganic salt, 0.07 mM CaCl2.

The objective of this research was to investigate the influence of various root media on the growth of mother and daughter plants during propagation and early yield after transplanting of 'Seolhyang' strawberry. To achieve this, daughter plants were fixed to connected small pots that contained expanded rice-hull (ERH), a strawberry-specialized commercial medium (SSCM), soil mother materials (SMM), or loamy sand (LS). Then, growth of daughter plants in above- and below-ground tissue as well as early yield after transplanting to plastic house soil were investigated. The growth of daughter plants in terms of plant height, leaf area and fresh weight were the highest in the SSCM treatment. Root growth in terms of the amount of primary roots and root dry weight were the highest in the treatments of ERH and SMM and the lowest in that of SSCM, among treatments tested. The ERH treatment also showed the highest values among treatments in root length, surface area and volume when roots with 0 to 0.4 mm in diameter were investigated. The flower bud differentiation of daughter plants began on Sept. 3 in the ERH treatments, earlier than the SMM (Sept. 5) and in SSCM (Sept. 7) treatments. The tissue N contents of daughter plants were in the range of 1.41 to 1.55% in all treatments, and no significant differences were observed among treatments. This indicates that the low moisture retention capacity of ERH and water stress, rather than tissue N contents, promote the flower differentiation of daughter plants. In the evaluation of early yield after transplant, the ERH treatment of showed the highest yield in the period from November to December, reaching 667 g fruit weight per 10 plants. The yields per 10 plants in the other treatments were 581 g in SMM, 475 g in SSCM and 295 g in LS. Above results imply that the various root media have different effects on the growth of daughter plants as well as flower bud differentiation. Therefore, improvement in early yield after transplant can be achieved through selection of proper root medium for daughter plant propagation.
A feeding trial was conducted with Euglena strains grown under different media. The effect of supplementation of Euglena on the performance, nutrient availability and fatty acid composition of breast muscle was studied. In experiment I, two hundred ten hatched broiler chicks (Ross) were assigned to seven dietary treatments for 5 weeks. Each treatment consisted of 3 replications with 10 birds each. Control diet was formulated to have $22\%$ CP and 3,150 kcal ME/kg for starter diet, $19\%$ CP and 3,200 kcal ME/kg for finisher diet. Euglena gracilis Z. (EG) was added to control diet at the plevel of 0.25, 0.5, $1.0\%$ and Euglena gracilis Z. bleached and DHA enriched (EGBD; a strain mutated by streptomycin and cultivated in DHA enriched medium) at the level of 0.5, 1.0, $2.0\%$ in the diet. In experiment 2, two hundred fifty hatched broiler chicks (Ross) were assigned to five dietary treatments: T1; Control, T2; T1 + Euglena gracilis Z. DHA enriched (EGD; cultivated in DHA enriched medium) $0.5\%$, T3; T1 + EGD $1.0\%$, T4; T1 + EGBD $0.5\%$, T5; T1 + EGBD $1.0\%$. The weight gain and feed consumption were measured weekly. Fatty acid composition of breast muscle was determined. In experiments I and 2, Euglena supplementation had no significant effects on weight gain, feed intake and feed conversion ratio. In experiment 1, EGBD treatments significantly increased DHA concentration but decreased concentration of linoleic acid and arachidonic acid in breast muscle. EGBD 2% treatment showed the highest DHA concentration (14.27%) which is 3.9 times of that of the control ($3.66\%$). In experiment 2, $1.0\%$ EGBD treatment showed highest EPA, lignoceric acid and DHA level in breast muscle (P<0.05). Also, EGD treatments significantly increased DHA and EPA concentration. It was concluded that EGBD and EGD can be supplemented to broiler diet to produce DHA enriched broiler meat.
Purpose: This study evaluated the dietary quality and nutritional status of elderly people using the Nutrition Quotient for Elderly (NQ-E). Methods: The participants were 204 elderly people over 65 years of age (38 men and, 166 women) in Seoul. The dietary information was analyzed using a questionnaire of NQ-E, which consisted of 19 checklist items, and 24-recall test data. The NQ-E scores and its four factors, including 'balance', 'moderation', 'diversity', and 'dietary behavior' factors, were calculated according to general characteristics of the subjects. The subjects were divided into the 'monitoring needed group' (62 > NQ-E score) and the 'good group' (62 ≤ NQ-E score) according to their NQ-E score. Results: The mean NQ-E score of the total subjects was 61.9, which was within the mediumhigh grade. The scores of balance, moderation, and dietary behavior factors were within the medium-high grade, while the score of the diversity factor was within the medium-low grade. The NQ-E score was 54.8 in the monitoring needed group and 69.3 in the good group. For the score of the diversity factor, the elderly living alone had a significantly lower score than the score for the elderly living with a spouse. The female subjects showed significantly higher scores of moderation and dietary behavior factors than did the male subjects. The daily intakes of vitamin A, thiamin, riboflavin, vitamin C and calcium in the monitoring needed group were significantly lower than those in the good group. The nutrient adequacy ratios (NAR) of vitamin A, thiamin, riboflavin, vitamin C and calcium in the monitoring needed group were significantly lower compared to the scores of the good group. The indexes of nutritional quality (INQ) of vitamin A, thiamin, riboflavin, vitamin C, calcium and potassium were less than 1 for all the subjects. The monitoring needed group had a significantly lower consumption of total foods, vegetables and mushrooms than the good group. As a result, the nutritional status of the monitoring needed group was significantly lower than that of the good group. Conclusion: The results of this study show that NQ-E would be a useful tool for assessing the dietary quality of the elderly. In conclusion, a focused-nutrition education program and a useful guideline are needed for promoting the health and nutritional status in elderly people.
In this study, three GRAS (generally recognized as safety) strain was isolated from Doenjang and Cheonggukjang and identified as a protease-producing microorganism, following the appearance of a clear zone around its colony when cultured on a medium containing skim milk. Based on an analysis of the nucleotide sequence of 16S ribosomal RNA, the strains wereas identified as Bacillus amyloliquefaciens and wereas therefore named Bacillus amyloliquefaciens CDD5, Bacillus amyloliquefaciens CPD4, and Bacillus amyloliquefaciens CGD3. Here, we analyzed the protease and ${\alpha}$-glucosidase inhibitory activities of the three B. amyloliquefaciens strains. Among the isolated strains, B. amyloliquefaciens CGD3 exhibited the highest protease activity (9.21 U/mL, 24 hr). The protease activities of B. amyloliquefaciens CDD5 and B. amyloliquefaciens CPD4 reached 1.14 U/mL and 8.02 U/mL, respectively, at 48 hr. The proteases from the three B. amyloliquefaciens strains showed the highest activities within a pH range of 8.0-9.0 at $50^{\circ}C$, and casein was found to be the preferred substrate on evaluating enzyme activity in the substrate specificity assay. The B. amyloliquefaciens strains exhibited maximal growth when the nutrient broth medium had an initial pH within the range of 5.0-10.0, 6-9% sodium chloride (NaCl), and 5% glucose. B. amyloliquefaciens CDD5 exhibited a low ${\alpha}$-glucosidase inhibition rate (5.32%), whereas B. amyloliquefaciens CPD4 and B. amyloliquefaciens CGD3 exhibited relatively higher inhibition rates of 96.89% and 97.55%, respectively.

제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
