Allelopathic and Autotoxic Effects of Alfalfa Plant and Soil Extracts

  • Alfalfa (Medicago sativa L.) plants have been reported to be autotoxic as well as allelopathic. Laboratory and greenhouse experiments through petri-dish and pot test were conducted to determine autotoxic effects of alfalfa leaf and soil extracts on the germination or early seedling growth of alfalfa, and to evaluate allelopathic effects of alfalfa leaf residues on alfalfa, barnyard grass, com, eclipta and soybean. Alfalfa seed germination was delayed depending on aqueous extract concentration, with no difference in final germination after 48 hours. Alfalfa root length was more sensitive to the autotoxic chemicals from leaf extracts than was germination or shoot length. Root growth of alfalfa was significantly inhibited at extract concentration of more than 1 g dry tissue/L (g $\textrm{L}^{-1}$). Hypocotyl growth, however, was not affected by all the concentrations of leaf extracts. Soil extracts from 4-yr-old alfalfa stand significantly reduced alfalfa root length by 66%, while soil extracts from 0,1, and 3yr-old stand stimulated root length up to 14-32% over the control. Residue incorporation with dry matters of alfalfa leaf at 100 g $\textrm{kg}^{-1}$ reduced seedling length of several crop and weed species, ranging from 53 to 87% inhibition. Addition of nutrient solution into alfalfa leaf extracts alleviated alfalfa autotoxic effect. This result indicates alfalfa leaf and soil extracts or residues could exert autotoxic as well as allelopathic substances into soil environments during and after establishment.

Analytical Electron Microscopy and Atomic Force Microscopy Reveal a Physical Mechanism of Silicon-Induced Rice Resistance to Blast

  • Locations of silicon accumulation in rice leaves and its possible association with resistance to rice blast were investigated by analytical electron microscopy and atomic force microscopy. A blast-susceptible cultivar, Jinmi, and partially resistant cultivars, Hwaseong and Suwon345, were grown under a hydroponic culture system with modified Yoshida's nutrient solution. Electron-dense silicon layers were frequently found beneath the cuticle in epidermal cell walls of silicon-treated plants. Increasing levels of silicon were detected in the outer regions of epidermal cell walls. Silicon was present mainly in epidermal cell walls, middle lamella, and Intercellular spaces within subepidermal tissues. Furthermore, silicon was prevalent throughout the leaf surface with relatively small deposition on stomatal guard cells in silicon-treated plants. Force-distance curve measurements revealed relative hardness and smaller adhesion force in silicon-treated plants (18.65 uN) than control plants (28.39 uN). Moreover, force modulation microscopy showed higher mean height values of elastic Images In silicon-treated plants(1.26 V) than in control plants (0.44 V), implying the increased leaf hardness by silicon treatment. These results strongly suggest that silicon-induced cell wall fortification of rice leaves may be closely associated with enhanced host resistance to blast.

  • In 1923, Dr. Warburg had observed that tumors acidified the Ringer solution when 13 mM glucose was added, which was identified as being due to lactate. When glucose is the only source of nutrient, it can serve for both biosynthesis and energy production. However, a series of studies revealed that the cancer cell consumes glucose for biosynthesis through fermentation, not for energy supply, under physiological conditions. Recently, a new observation was made that there is a metabolic symbiosis in which glycolytic and oxidative tumor cells mutually regulate their energy metabolism. Hypoxic cancer cells use glucose for glycolytic metabolism and release lactate which is used by oxygenated cancer cells. This study challenged the Warburg effect, because Warburg claimed that fermentation by irreversible damaging of mitochondria is a fundamental cause of cancer. However, recent studies revealed that mitochondria in cancer cell show active function of oxidative phosphorylation although TCA cycle is stalled. It was also shown that blocking cytosolic NADH production by aldehyde dehydrogenase inhibition, combined with oxidative phosphorylation inhibition, resulted in up to 80% decrease of ATP production, which resulted in a significant regression of tumor growth in the NSCLC model. This suggests a new theory that NADH production in the cytosol plays a key role of ATP production through the mitochondrial electron transport chain in cancer cells, while NADH production is mostly occupied inside mitochondria in normal cells.

Control of Environments in Greenhouse Using Programmable Logic Controller (PLC를 이용한 온실의 환경제어)

  • This study was carried out to develop the control system with PLC and its operating software and to investigate its control ability of greenhouse environments. Two experimental greenhouses were controlled by PLC and ON/OFF controller, respectively. In greenhouse controlled by PLC, target values of air temperature, relative humidity and $CO_2$ concentration were automatically changed. In warm-water heating, the variation of air temperature was reduced to $\pm$ $0.6^{\circ}C$ by the method of proportional-integration(PI) control with an inverter. In ventilation, the variation of air temperature was reduced, since windows open and close with multistage by mutual relation formula among the target, indoor, and outdoor temperature. Relative humidity at daytime was maintained with range of 35% to 55% by PLC controlled fogger. $CO_2$ concentration was automatically controlled from 300 to 800 $\mu$molㆍ$mol^{-1}$ according to amount of solar radiation. The suppling amount and frequency of nutrient solution were controlled by total integrated solar radiation. Difference in the yield of cucumber in the greenhouse controlled by PLC and by ON/OFF controller was not significant at the 5% level.

  • This study was carried out to develop a fully-controlled plant growth system for studying the effects of environmental factors on plant growth. The plant growth system consists of the growth chamber, the measurement system for sensing and signal conditioning the environmental factors and the control system which includes control algorithm required for optimum operation and on-line monitoring. The results of the study on the performance of the controlled plant growth system are summarized as follows : 1. Under the light intensity of 18,000~20,000 lux, the temperature was controlled in the range of $22{\pm}0.4^{\circ}C$, the relative humidity of $70{\pm}5%$ respectively. 2. The $CO_2$ concentration was controlled in the range of $1,000{\pm}40ppm$(from 1st to 4th day) and $1,500{\pm}40ppm$(from 4th to 9th day). 3. The electric conductivity(EC) and pH of the nutrient solution were controlled in the range of $1.9{\pm}0.2mS/cm$ and $6.8{\pm}0.5pH$ respectively. 4. Under the above environmental conditions, the average fresh weight of leaf lettuce increased from 10g to 74g in 9 days with 24 hour lighting.

The Selection of the Optimum Cherry Tomato Cultivar for Hydroponics (수경재배에 적합한 방울토마토의 품종선발)

  • Cherry tomatoes(Lycopersicon esculentum) were grown in circulating deep flow hydroponic system. The pH of the nutrient solution was increased and the EC was decreased with time. The clusters of ‘Mini Carol’ were branched severely, whereas those of ‘Chelsea Mini’ and ‘Popo’ were a little branched or not. The vegetative growth was similar between cultivars. But the yield of fruits was by far the largest in ‘Mini Carol’ compared to other cultivars, and not different between ‘Chelsea Mini’ and ‘Popo’. Internal qualities of fruits, like as sugar, sugar/organic acid, and vitamin C, are good in ‘Mini Carol’. As the result of panel test, ‘Mini Carol’ is good for softness, easeness of eating, toughness of skin and so on, so it is believed that ‘Mini Carol’ is the most acceptable to consumers in hydroponic culture.

  • Alternative subirrigation way, capillary wick system (CWS) was tested to reduce labor cost, waste water, contamination of ground water, and use of fungicide compared to overhead irrigation system (OIS). CWS helped reduce remarkably the working hours for watering from 4 hours in OSI to just 5 minutes. Labor cost was saved 98% in CWS compared to OIS. By the physical characteristics of various growing media, 1 coconut coir+2 perlite (v/v) mixture was selected because it had an ideal distribution of three phase, e.g. 1 solid: 1 liquid: 2 gas phase. Medium mixture containing scoria had so high bulk and particle density to hurt root. In bark-containing medium, the liquid phase and the percent saturation of liquid phase with time elapsed was lower than that of other mixture. It meant that the mixture contained very low level of water. Application of CWS for cyclamen pot production played an important role in reducing the incident of fusarium wilt symptom from 18% in conventional over watering system to 4%. Cyclamen pot irrigated by capillary wick had shorter petiole and more leaves than those by overhead watering. As a result, this system was highly beneficial to get uniform pot products with high quality. It improved water and nutrient solution efficiency relative to conventional overhead irrigation system (OIS).

Effects of $NO_3^-$ Gradients on Nitrogen Fixation, Nitrate Reduction and Ureide Content of Soybean (대두의 공소개정, 공산환원 및 Ureide함량에 미치는$NO_3^-$의 영향)

  • Soybean, inoculated with effective Rhizobium japonicum 110, were grown by sand culture with nutrient solution containing either of 0, 1, 3, 10 or 30mM NO3-/l, and analyzed growth characteristics, NR activity, N2-fixation activity, and changes of ureide contents during the growing period. The amount of nodule formation decreased abruptly by nitrate treatment, the maximum nodule dry weight was 1.59, 1.05, 0.78, 0.09 and 0.008 g plant-1, respectively for each treatment on the 98th day. Specfic activity of N2-fixation showed the maximum rates of 140, 101, 37, 5 and 2.2 nM dw.mg-1.hr-1, respectively for each treatment in the earlier growth period. The maximum acetylene reduction activity on the 98th day after sowing was 81.5, 35.3, 14.3, 0.1 and 0.0045 $\mu$M C2H4 plant-1.hr-1, respectively for 0, 1, 3, 10 and 30 mM of NO3- gradients. Nitrate reduction activity increased along with nitrate gradients, and decreased abruptly with age. Relative abundance of ureides in plant organs was high in reproductive growth, and showed the maximum value in fully symbiotic dependent plant. Relative abundance of ureides in stem is a useful indication for the evaluation of nitrogen fixation in nodules of symbiotic plant.

  • Corn (Zea may L.) and Algae (Botridium sp.) were grown in the nutrient solution to investestigate the effects of humic substances on the moderation of high salt toxicity. The results are as follows: 1) The addition of small amount of humic substances (5 ppm C) drastically decreased the high salt toxicity of corn growth. 2) The addition of 20ppm humic substances was also decreased the high salt toxicity in algaegrowth. Different concentration of humic substances, however, did not show significant difference in algae yield.

Primary metabolic responses in the leaves and roots of bell pepper plants subjected to microelements-deficient conditions

  • Plants need essential mineral elements to favorably develop and to complete their life cycle. Despite the irreplaceable roles of microelements, they are often ignored due to the relative importance of macroelements with their influence on crop growth and development. We focused on the changes in primary metabolites in the leaves and roots of bell pepper plants under 6 microelements-deficient conditions: Copper (Cu), Zinc (Zn), Iron (Fe), Manganese (Mn), Boron (B) and Molybdenum (Mo). Bell pepper plants were grown in hydroponic containers, and individual elements were adjusted to 1/10-strength of Hoagland nutrient solution. A remarkable perturbation in the abundance of the primary metabolites was observed for the Fe and B and the Mn and B deficiencies in the leaves and roots, respectively. The metabolites with up-accumulation in the Fe-deficient leaves were glucose, fructose, xylose, glutamine, asparagine and serine. In contrast, the Mn deficiency also resulted in a higher accumulation of glucose, fructose, xylose, galactose, serine, glycine, β-alanine, alanine and valine in the roots. The B deficiency noticeably accumulated alanine, valine and phenylalanine in the roots while it showed a substantial decrease in glucose, fructose and xylose. These results show that the primary metabolism could be seriously disturbed due to a microelement deficiency, and the alteration may be either the specific or adaptive responses of bell pepper plants.
