건강기능성 식품소재로서 pinitol의 특성 (Characteristics of Pinitol as a Functional Food Biomaterial)

  • D-pinitol, another chemical structure of 3-O-methyl-D-chiro-inositol, is an important insulin-sensitizer. The purpose of this review is to examine the characteristics of pinitol and other analogs as functional food biomaterials which were well known to reduce blood glucose levels. Pinitol can be converted to chiro-inositol in normal humans, while diabetic patients can not use the molecule, resulting in exhibiting low level of chiro-inositol in their urine. Recently, it is reported that pinitol can trigger phospholipase C/D, thus the rate of glucose metabolism accelerates to use as fuel for human body. To not only reduce insulin resistance of diabetic patients, but also alleviate the symptoms of diabetes, obesity, and muscle contraction, pinitol and its dietary supplementation is needed.

Inhibition of Red Ginseng on 5-Hydroxyeicosatetraenoic Acid (5-HETE) Biosynthesis from Arachidonic Acid in Helicobacter Pylori-infected Gastric Cells

  • Helicobacter pylori (H. pylori) infection rapidly stimulated either COX-2 or 5-LOX and released arachidonic acid metabolites that have been considered as pivotal mediators in H. pylori-induced inflammatory responses. To determine whether red ginseng extract (RGE) can suppress the biosynthesis of 5(S)-hydroxyeicosatetraenoic acids (HETE), a precursor metabolite of leukotrienes B4 (LTB4) in H. pylori-provoked inflammatory responses in gastric epithelial cells, the biosynthesis of monohydroxy fatty acids was measured using radioactive arachidonic acid and validated by RP-HPLC using non-radioactive AA as substrate in AGS cells cocultured with H. pylori (ATCC 43504) with or without pretreatment of RGE. Among three known major HETEs, H. pylori infection specifically induced the biosynthesis of $^{14}C-5(S)-HETE$ rather than the complex of $^{14}C-15S-/^{14}C-12(S)-HETE$ from $^{14}C-AA$, concomitantly obtained by HPLC(p<0.01). RGE, 1 to $100{\mu}g/ml$, selectively suppressed H. pylori-stimulated $^{14}C-5(S)-HETE$ production implying the attenuation of 5-lipoxygenase activity, of which was similar to known LOX inhibitor NDGA $(10{\mu}M)$ (p<0.01). However, the amount of 5(S)-HETE was significantly reduced by higher dose of RGE $(100{\mu}g/ml)$ (p<0.05). These results indicated that LOX pathway might be one of principle pathogenic mechanisms of H. pylori and red ginseng could be a nutraceutical against H. pylori infection through inhibiting action of LOX activity.

Intercalation of Functional Organic Molecules with Pharmaceutical, Cosmeceutical and Nutraceutical Functions into Layered Double Hydroxides and Zinc Basic Salts

  • Negatively charged functional organic molecules such as retinoic acid, ascorbic acid, indole acetic acid, citric acid, salicylic acid, acidic dye (indigo carmine, Food Blue 1) are intercalatively encapsulated by zinc basic salt (hydrozincite) and layered double hydroxide. Such functional organic-inorganic nanohybrids are realized via coprecipitation reaction involving simultaneous formation of layered inorganic lattice and intercalation of anionic species. The heterostructural nature of these nanohybrids, their particle morphology and textural characterizations are mainly discussed on the basis of Powder X-ray Diffraction and Field Emission Scanning Electron Microscopy results.

In vitro and Cellular Antioxidant Activity of Arginyl-fructose and Arginyl-fructosyl-glucose

  • Arginyl-fructose (AF) and arginyl-fructosyl-glucose (AFG) were chemically synthesized and purified. Their in vitro and cellular antioxidant activity was investigated using oxygen radical absorbance capacity (ORAC) and cellular antioxidant activity assay, respectively. The peroxyl radical scavenging activity of AF was much higher than that of AFG, which was in good agreement with their reduction capacity to donate electrons or hydrogen atoms. On the other hand, the hydroxyl radical scavenging activity of AF was weaker than that of AFG, which was consistent with their metal chelating activity, suggesting that AFG-$Cu^{2+}$ complex may be less redox-active than AF-$Cu^{2+}$ complex due to 1 glucose molecule attached. The cellular antioxidant activity of AF and AFG appeared to depend on both their permeability into cell membrane and the scavenging activity on peroxyl or hydroxyl radicals. These results indicate that AF and AFG, Maillard reaction products, may have a high potential as a material for the development of nutraceutical food with antioxidant activity.

  • Recent developments in the application of molecular biology to food grade lactic acid bacteria (LAB) have shown that it could be feasible to engineer metabolic pathways to either enhance specific metabolic fluxes or to divert metabolites for the production of different or new end products. This engineering requires detailed knowledge of enzymes involved in metabolism and regulation within the targeted organism but little works have been done in this area. During biochemical and molecular characterisation of lactic bacterial enzymes, some of probiotic Lactobacillus and Bifidobacterium species were found to be very useful for food, nutraceutical and pharmaceutical industries. The enzymes are usually intracellular and the yields are very low to be useful for industrial applications. Among many enzymes and proteins of lactic bacteria studied, some of our gene cloning achievements have contributed to overproduction of lactic bacterial enzymes such as peptidases, esterases, lactases, bile salt hydrolases and linoleate isomerases for foods and nutraceuticals.

Polymethoxylated Flavone Extracts from Citrus Peels for Use in the Functional Food and Nutraceutical Industry

  • Polymethoxylated flavones (PMFs) extracted from Citrus sinensis 'Jincheng' peel were characterized by chromatographic and spectroscopic techniques. Seven individual PMF were identified. 3, 3', 4', 5, 6, 7-hexamethoxyflavone (HEX), nobiletin (NOB), heptamethoxyflavone (HEP), 5-demethylnobiletin (DN), and tangeretin (TAN) were characterized through electrospray ionization mass spectrometry (ESI-MS) in positive mode of protonated molecular ions $[M+H]^+$, the diagnostic fragment ions, together with the UV-Vis spectra and high performance liquid chromatography (HPLC) elution order from literature data. Sinensetin (SIN) and tetramethyl-O-scutellarein (SCU) were isolated and identified through their MS, $^1H$ nuclear magnetic resonance (NMR) and UV-Vis spectral studies. The levels of PMFs in peels from different cultivars of citrus fruits grown in China were determined for the first time. The results showed that C. aurantium 'Bitter orange' peel was the most promising variety for HEP. C. sinensis peel was a good source for SIN and SCU.

Anti-diabetic Effects of Ethanol Extract from Bitter Melon in Mice Fed a High-fat Diet

  • Present study aimed to determine the effect of 'bitter melon', a popularly used fruit in Bangladesh and several other Asian countries, on high-fat-diet-induced type 2 diabetes. To investigate the effect, ethanol extract from bitter melon (BME) as a dietary supplement with mouse chow was used. BME was found to significantly attenuate the high-fat diet (HFD) -induced body weight and total fat mass. BME also effectively reduced the insulin resistance induced by the HFD. Furthermore, dietary supplementation of BME was highly effective in increasing insulin sensitivity and reducing hepatic fat and obesity. These results indicate that BME could be effective in attenuating type 2 diabetes and could therefore be a preventive measure against type 2 diabetes.

천연소재 흔합물의 보충급여가 부고환 지방세포 크기에 미치는 영향 (Effects of Natural Plants Supplementation on Adipocyte Size of the Epididymal Fat Pads in Rats)

  • 본 연구에서는 천연소재 혼합물이 혈중 지질 함량 및 부고환 지방세포의 크기에 미치는 영향을 측정하였다. 그 결과 혈액 중 총 콜레스테롤과 LDL 콜레스테롤은 천연소재 섭취에 의한 유의적인 감소 효과가 나타나지 않았지만, 중성지방은 정상식이군에서 감소하였고 HDL 콜레스테롤은 정 상식이군에서 증가하였다. 부고환 지방조직은 고지방식이 섭취에 의해 지방세포 크기가 유의적으로 증가되었으나 천연소재 섭취로 인하여 유의적으로 감소하였다. 이는 천연소재섭취에 의해 체지방 축적이 억제되어 지방세포의 hypertrophy를 억제했기 때문으로 사료된다.

Hot Water Extract of Wheat Bran Attenuates White Matter Injury in a Rat Model of Vascular Dementia

  • Vascular dementia is characterized by white matter lesions involving the demyelination and activation of astrocytes and microglia. In a previous study, we showed that the supernatant of a laboratory-scale, hot water extract of ground whole wheat (TALE) attenuated white matter injury and astrocytic activation in a rat model of bilateral common carotid artery occlusion (BCCAO). In the present study, we made several modifications to the hot water extraction process to remove starch and enable large-scale production. We used wheat bran (WB), which contains less starch, instead of ground whole wheat. In addition, we removed starch granules with a decanter before hot water extraction. The final product, wheat bran extract (WBE), contained 2.42% arabinose, a surrogate marker of arabinoxylan, which is an active constituent of WBE. Supplementation of the rat model of BCCAO with WBE (400 mg/kg/day) for 33 days attenuated white matter injury, which was assessed by Luxol Fast Blue staining, in the corpus callosum (cc) and optic tract (opt) regions. Attenuation of white matter injury in the opt region was accompanied by improvement of the pupillary light reflex. Immunochemical staining revealed that supplementation with WBE reduced astrocytic activation in the cc and opt regions and reduced microglial activation in the opt region. These findings indicate that supplementation with WBE is effective at attenuating white matter injury accompanied by the inhibition of astrocytic and microglial activation. Therefore, extracts from WB, a cheap by-product of wheat milling, can be developed as a nutraceutical to prevent vascular dementia, a disease for which there is no approved pharmaceutical treatment.

피부 미용을 위한 기능성 식이 소재의 이해 (Understanding of Functional Foods for Nutritional Skin Care)

  • 현대를 살고 있는 우리들은 건강함과 더불어 아름다운 피부를 갖기를 원한다. 산업화에 의한 대기 오염의 증가 및 사회가 복잡해지면서 증가하는 스트레스는 피부에 부담을 증가시키고 건강한 피부를 가졌다 하더라도 꾸준히 관리를 해야 만이 유지되기에 피부 미용에 대한 관심은 지대하다고 할 수 있다. 이에 따라 피부 미용을 위한 다양한 제품이 개발되고 있고, 특히 피부 미용은 건강 차원에서 영양과 밀접한 관련이 있다는 인식 변화에 따라 특정 영양소 성분을 이용한 먹는 화장품이나 건강 기능 식품의 개발이 진행되고 있다. 이를 위한 주된 소재로 비타민 A, C, E, Bl, B2, 나이아신, 엽산 등의 비타민류, 셀레늄, 황 등의 무기질류, 단백질 및 아미노산, 불포화 지방산, 약초 추출물 등이 이용되고 있으며 이외에도 콜라겐 단백질 분해물, 대두 추출물, 석류추출물, 포도씨 추출물, 화분 등과 수용성 키틴 등의 기능성 단일 물질을 함유한 제품도 있다. 이와 같이 다양한 식이 소재가 건강한 피부 유지를 위한 기능성 식품 소재로 이용되고 있으나 이 효과는 확실한 검증을 필요로 한다. 본 논문에서는 피부 미용의 관점에서 이들 소재들의 효능을 살펴 피부 미용을 위한 기능성 식이 소재에 대한 이해를 도모하고자 한다.
