The targeting of DNA methylation in cancer using DNA hypomethylating drugs has been well known to sensitize cancer cells to chemotherapy and immunotherapy by affecting multiple pathways. Herein, we investigated the combinational effects of DNA hypomethylating drugs and ionizing radiation (IR) in human sarcoma cell lines both in vitro and in vivo. Clonogenic assays were performed to determine the radiosensitizing properties of two DNA hypomethylating drugs on sarcoma cell lines we tested in this study with multiple doses of IR. We analyzed the effects of 5-aza-dC or SGI-110, as DNA hypomethylating drugs, in combination with IR in vitro on the proliferation, apoptosis, caspase-3/7 activity, migration/invasion, and Western blotting using apoptosis- or autophagy-related factors. To confirm the combined effect of DNA hypomethylating drugs and IR in our in vitro experiment, we generated the sarcoma cells in nude mouse xenograft models. Here, we found that the combination of DNA hypomethylating drugs and IR improved anticancer effects by inhibiting cell proliferation and by promoting synergistic cell death that is associated with both apoptosis and autophagy in vitro and in vivo. Our data demonstrated that the combination effects of DNA hypomethylating drugs with radiation exhibited greater cellular effects than the use of a single agent treatment, thus suggesting that the combination of DNA hypomethylating drugs and radiation may become a new radiotherapy to improve therapeutic efficacy for cancer treatment.

Malignant glioma cells invading surrounding normal brain are inoperable and resistant to radio- and chemotherapy, and eventually lead to tumor regrowth. Identification of genes related to motility is important for understanding the molecular biological behavior of invasive gliomas. According to our previous studies, Metallothionein 1E (MT1E) was identified to enhance migration of human malignant glioma cells. The purpose of this study was to confirm that MT1E could modulate glioma invasion in vivo. Firstly we established 2 cell lines; MTS23, overexpressed by MT1E complementary DNA construct and pV12 as control. The expression of matrix metalloproteinases (MMP)-2, -9 and a disintegrin and metalloproteinase 17 were increased in MTS23 compared with pV12. Furthermore it was confirmed that MT1E could modulate MMPs secretion and translocation of NFkB p50 and B-cell lymphoma-3 through small interfering ribonucleic acid knocked U87MG cells. Then MTS23 and pV12 were injected into intracranial region of 5 week old male nude mouse. After 4 weeks, for brain tissues of these two groups, histological analysis, and immunohistochemical stain of MMP-2, 9 and Nestin were performed. As results, the group injected with MTS23 showed irregular margin and tumor cells infiltrating the surrounding normal brain, while that of pV12 (control) had round and clear margin. And regrowth of tumor cells in MTS23 group was observed in another site apart from tumor cell inoculation. MT1E could enhance tumor proliferation and invasion of malignant glioma through regulation of activation and expression of MMPs.

O-2-$^{18}F$-fluoroethyl-l-tyrosine ($[^{18}F]FET$) has been widely used for glioblastomas (GBM) in clinical practice, although evaluation of its applicability in non-clinical research is still lacking. The objective of this study was to examine the value of $[^{18}F]FET$ for treatment evaluation and prognosis prediction of anti-angiogenic drug in an orthotopic mouse model of GBM. Human U87MG cells were implanted into nude mice and then bevacizumab, a representative anti-angiogenic drug, was administered. We monitored the effect of anti-angiogenic agents using multiple imaging modalities, including bioluminescence imaging (BLI), magnetic resonance imaging (MRI), and positron emission tomography-computed tomography (PET/CT). Among these imaging methods analyzed, only $[^{18}F]FET$ uptake showed a statistically significant decrease in the treatment group compared to the control group (P=0.02 and P=0.03 at 5 and 20 mg/kg, respectively). This indicates that $[^{18}F]FET$ PET is a sensitive method to monitor the response of GBM bearing mice to anti-angiogenic drug. Moreover, $[^{18}F]FET$ uptake was confirmed to be a significant parameter for predicting the prognosis of anti-angiogenic drug (P=0.041 and P=0.007, on Days 7 and 12, respectively, on Pearson's correlation; P=0.048 and P=0.030, on Days 7 and 12, respectively, on Cox regression analysis). However, results of BLI or MRI were not significantly associated with survival time. In conclusion, this study suggests that $[^{18}F]FET$ PET imaging is a pertinent imaging modality for sensitive monitoring and accurate prediction of treatment response to anti-angiogenic agents in an orthotopic model of GBM.
Apigenin은 과일과 야채에 들어있는 플라보노이드로 다양한 악성 세포에 항증식효과를 보여준다. 세포성장 저해효과를 확인하기 위하여 KB 구강암세포주를 96 well plate에 $6{\times}10^3$ cells/well로 분주하고 24시간 후에 apigenin을 24시간 동안 처치하여 MTT assay를 수행하였다. Apigenin은 배양 후 용량 의존적으로 세포사를 유도하였다. Apigenin 100 ${\mu}M$을 24시간 동안 처치하고 대조군과 세포성장을 비교하였을 때 유의적인 감소를 확인하였다. KB 구강암세포주에서의 apoptosis를 확인하기 위해 DAPI 염색을 수행하였다. Apigenin을 처치한 세포에서 핵의 응축이 존재함을 형광현미경으로 확인하였다. 우리는 누드마우스에 KB 구강암세포주를 이식하여 세포 성장 억제 효과를 알아보았다. Apigenin을 마우스에 25, 50 mg/kg을 0.2 ml의 PBS에 녹여 경구투여 하였다. 종양 사이즈는 대조군과 25, 50 mg/kg apigenin 투여군을 비교하였을 때 유의적으로 감소하였다. Apoptosis 분석을 위해 TUNEL염색을 수행하였다. 25 mg/kg apigenin 투여군과 대조군을 비교하였을 때 apoptosis의 유의적인 증가를 확인하였다. 육안적 소견을 위한 H&E 염색은 이상이 없었다. 본 연구는 apigenin이 구강암세포주 성장 억제를 apoptosis의 유도를 통하여 확인하였다.
목 적: 전립샘암 세포주 DU 145에서 엑스선과 중성자선에 의한 HIF-$1\alpha$와 아포프토시스 발현의 차이를 비교함으로써 엑스선과 중성자선의 방사선생물학적 차이의 기전을 알아보고자 한다. 대상 및 방법: 누드 마우스에 DU 145 전립샘암 세포주를 주입한 후 2 Gy 엑스선, 10 Gy 엑스선, 0.6 Gy 중성자선, 3.3 Gy 중성자선을 각각 조사했다. 엑스선을 조사한 군과 중성자선을 조사한 군에서 HIF-$1\alpha$, Bcl-2, Bax, 아포프토시스 발현 정도를 면역조직화학 염색과 western blotting을 이용하여 비교하였다. 아포토시스의 정도는 terminal deoxynucleotidyl biotin-dUTP nick end labeling (TUNEL) 염색을 이용하여 비교하였다. 결 과: 방사선 조사 1일째, X선을 조사한 군과 비교했을 때, 중성자선을 조사한 군에서 HIF-$1\alpha$와 Bcl-2의 발현은 감소하였고, Bax와 아포프토시스 세포의 수는 증가하였다. Bcl-2/Bax 비는 중성자선을 조사한 군에서 의미 있게 감소하였다. 이러한 HIF-$1\alpha$, Bcl-2, Bax, Bcl-2/Bax 비, 아포프토시스 발현의 차이는 방사선 조사 5일째에도 동일하게 유지되어 나타났다. 또한, HIF-$1\alpha$ 발현은 방사선 조사 5일째 Bcl-2 (p=0.031), Bax (p=0.037), TUNEL (p=0.016) 발현과 연관성을 보였다. 결 론: 중성자선 조사한 경우 엑스선에 비해 HIF-$1\alpha$와 Bcl-2 발현, Bcl-2/Bax 비가 감소하고, Bax 발현은 증가하였다. 중성자선 치료의 광자선과 다른 방사선생물학적인 반응은 HIF-$1\alpha$와 그로 인한 아포프토시스 관련 단백질 발현의 차이와 연관성이 있을 것으로 생각한다.
Aim of the study: An alternative source of adult stem cells that could be obtained in large quantities, under local anesthesia, with minimal discomfort would be advantageous. Adipose tissue could be processed to obtain a fibroblast-like population of cells or adipose tissue-derived stromal cells (ATSCs). This study was performed to confirm the availability of ATSCs in bone tissue engineering. Materials amp; Methods: In this study, adipose tissue-derived mesenchymal stem cell was extracted from the liposuctioned abdominal fat of 24-old human and cultivated, and the stem cell surface markers of CD 105 and SCF-R were confirmed by immunofluorescent staining. The proliferation of bone marrow mesenchymal stem cell and ATSCs were compared, and evaluated the osteogenic differentiation of ATSCs in a specific osteogenic induction medium. Osteogenic differentiation was assessed by von Kossa and alkaline phosphatase staining. Expression of osteocyte specific BMP-2, ALP, Cbfa-1, Osteopontin and osteocalcin were confirmed by RT-PCR. With differentiation of ATSCs, calcium concentration was assayed, and osteocalcin was evaluated by ELISA (Enzyme-linked immunosorbant assay). The bone formation by 5-week implantation of HA/TCP block loaded with bone marrow mesenchymal stem cells and ATSCs in the subcutaneous pocket of nude mouse was evaluated by histologic analysis. Results: ATSCs incubated in the osteogenic medium were stained positively for von Kossa and alkaline phosphatase staining. Expression of osteocyte specific genes was also detected. ATSCs could be easily identified through fluorescence microscopy, and bone formation in vivo was confirmed by using ATSC-loaded HA/TCP scaffold. Conclusions: The present results show that ATSCs have an ability to differentiate into osteoblasts and formed bone in vitro and in vivo. So ATSCs may be an ideal source for further experiments on stem cell biology and bone tissue engineering.
탈미네랄화된 골분(demineralized bone particle, DBP)은 골/연골 형성의 강력한 유도인자로 사용된다. 본 연구에서는 용매 캐스팅/염 추출법을 이용해 함량별 DBP와 PLGA가 하이브리드화된 다공성 지지체를 실제 디스크 형태와 유사하게 제조하였다. 제조된 지지체의 특성을 분석하기 위하여 다공도, 표면 젖음성 및 물 흡수성을 측정하였다. 디스크 세포인 섬유륜 및 수핵 세포는 토끼로부터 분리하여 제조된 지지체에 각각 파종한 후, 지지체를 재조합하여 배양하였다. 지지체에 파종된 디스크 세포의 생존율과 증식률은 MTT(3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium-bromide) 분석 방법을 이용하였고, 면역결핍 쥐의 피하에 삽입하여 이들의 디스크 조직 형성 정도를 확인하였다. 피하에 이식된 지지체를 적출하여 육안으로 관찰하고 모폴로지의 변화를 확인한 후, 조직을 파라핀으로 고정시켜 슬라이드를 제조하여 hematoxylin과 eosin 염색을 수행하였다. 천연/합성 하이브리드 담체로서의 DBP/PLGA 담체가 PLGA 단독으로 사용하였을 때와 비교하여 볼 때 디스크 조직의 형성이 우수하였으며, 특히 20, 40%의 DBP가 함유된 지지체가 세포의 성장과, 디스크 조직화에 유리함을 확인하였다.
종양영상과 치료에 이용할 수 있는 새로운 종양 친화성 물질을 발견하는 것은 핵의학 분야에서 중요하다. Vitamin $B_{12}$는 thymidylate 생합성과정에서 DNA 합성에 필수적인 조효소로서 분열속도가 빠른 세포와 조직 즉 태생기 세포와 일부 종양에 많은 양이 축적된다. 본 연구의 목적은 마우스에서 다양한 조직학적 형태의 종양에 의한 방사표지 vitamine $B_{12}$의 종양 친화성을 결정하여 $^{57}Co$-vitamine $B_{12}$가 종양영상 물질로서 가능한지를 규명하려고 하였다. 17 종류의 종양을 Balb/C와 Balb nu/nu 누드마우스에 피하주사하였다. 종양이 직경 약 $1.0cm^3$ 되었을 때 $^{57}Co$-methylcobalamin을 복강 내로 주사하였다. 24시간 후에 마우스를 희생하여 신장, 간, 종양, 비장, 뼈(우측 대퇴골), 장, 지방조직(복강내), 근육, 혈액(100 ul), 췌장을 제거하여 무게를 챈 후 gamma countcr로 방사능을 측정하였고 cpm/mg tissue로 표시하였다. 이때 마우스 문제에 따른 차이를 없게하기 위해 20g 몸무게로 표준화 하였다. 다양한 종류의 종양에서 $^{57}Co$-vitamine $B_{12}$가 의의 있게 축적되었고 종양대 정상조직의 섭취비도 대부분의 조직에서 높았으며 특히 배후 방사능에 중요한 영향을 미치는 뼈, 근육, 혈액에서 뚜렷하게 높았다. 본 연구의 결과 vitamin $B_{12}$는 종양친화성 물질로서 종양영상에 사용이 가능한 물질로 생각되며 앞으로 종양영상면에서 임상적 이용에 대한 더 많은 연구가 이루어져야겠다.

Background: Thyroid carcinoma is the most common malignancy of the endocrine organs. Although the majority of thyroid cancer patients experience positive outcomes, anaplastic thyroid carcinoma is considered one of the most aggressive malignancies. Current therapeutic regimens do not confer a significant survival benefit, and new therapies are urgently needed. Oncolytic herpes simplex virus (oHSV) may represent a promising therapy for cancer. In the present study, we investigated the therapeutic effects of a third-generation HSV vector, $G47{\Delta}$, on various human thyroid carcinoma cell lines in vitro. Two subcutaneous (s.c.) models of anaplastic thyroid carcinoma were also established to evaluate the in vivo anti-tumor efficacy of $G47{\Delta}$. Materials and Methods: The human thyroid carcinoma cell line ARO, FRO, WRO, and KAT-5, were infected with $G47{\Delta}$ at different multiplicities of infection (MOIs) in vitro. The survival rates of infected cells were calculated each day. Two s.c. tumor models were established using ARO and FRO cells in Balb/c nude mice, which were intratumorally (i.t.) treated with either $G47{\Delta}$ or mock. Tumor volumes and mouse survival times were documented. Results: $G47{\Delta}$ was highly cytotoxic to different types of thyroid carcinomas. For ARO, FRO, and KAT-5, greater than 30% and 80% of cells were killed at MOI=0.01 and MOI=0.1, respectively on day 5. WRO cells displayed modest sensitivity to $G47{\Delta}$, with only 21% and 38% of cells killed. In the s.c. tumor model, both of the anaplastic thyroid carcinoma cell lines (ARO and FRO) were highly sensitive to $G47{\Delta}$; $G47{\Delta}$ significantly inhibited tumor growth and prolonged the survival of mice bearing s.c. ARO and FRO tumors. Conclusions: The oHSV $G47{\Delta}$ can effectively kill different types of human thyroid carcinomas in vitro. $G47{\Delta}$ significantly inhibited growth of anaplastic thyroid carcinoma in vivo and prolonged animal survival. Therefore, $G47{\Delta}$ may hold great promise for thyroid cancer patients.
조직공학 기술은 in vitro와 in vivo에서 초기 세포 부착과 차후의 조직형성을 위해 3차원적인 지지체로서 다공성의 생분해성 담체의 사용이 필수적이다. 소장점막하조직(small intestinal submucosa, SIS)은 고유의 인장력과 생체적합성 때문에 생체물질로서 사용될 잠재력을 가지고 있는 콜라겐 조직이다. 근육유래 줄기세포는 배양조건에 따라 골세포, 연골세포, 및 근육세포 등으로 분화가 가능하다고 알려져 있다. 본 연구에서는 SIS를 함유한 락타이드-글리콜라이드 공중합체(PLGA) 다공성 지지체를 용매캐스팅/염추출법으로 제조하였고, 전자주사현미경 및 수은다공측정계를 이용하여 특성을 결정하였다 세포의 생존율과 성장률은 MTT(3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium-bromide) 분석 방법을 이용하였고 골로 분화된 세포를 알칼라인 포스파테이즈(ALP) 활성을 측정하여 확인하였다. SIS가 함유된 지지체와 SIS가 함유되지 않은 지지체를 면역결핍 쥐의 피하에 삽입하여 이들의 골형성 정도를 비교하여 보았다. 조직을 파라핀으로 고정시켜 슬라이드를 제조한 후 hematoxylin과 eosin, 트라이크롬 및 본쿠사 염색을 실시하였다. 천연/합성 하이브리드 담체로서의 SIS/PLGA 담체가 PLGA 단독으로 사용하였을 때와 비교하여 볼 때 골형성이 우수하였는데 이는 SIS 내에 함유하고 있는 여러 생체활성분자에 기인한 것으로 추측되었다.

④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
