Development of a continuous electrolytic system with an ion exchange membrane for pH-control with only one discharge of electrolytic solution and its characteristics (단일 전해액 배출만을 가지는 pH조절용 연속식 이온 교환막 전해 시스템의 개발과 그 특성)

  • In order to produce only a pH-controlled solution without discharging any unwanted solution, this work has developed a continuous electrolytic system with a pH-adjustment reservoir being placed before an ion exchange membrane-equipped electrolyzer, where as a target solution was fed into the pH-adjustment reservoir, some portion of the solution in the pH-adjustment reservoir was circulated through the cathodic or anodic chamber of the electrolyzer depending on the type of the ion exchange membrane used, and some other portion of the solution in the pH-adjustment reservoir was discharged from the electrolytic system through the other counter chamber with its pH being controlled. The internal circulation of the pH-adjustment reservoir solution through the anodic chamber in the case of using a cation exchange membrane and that through the cathodic chamber in the case of using an anion exchange membrane could make the solution discharged from the other counter chamber effectively acidic and basic, respectively. The phenomena of the pH being controlled in the system could be explained by the electro-migration of the ion species in the solution through the ion exchange membrane under a cell potential difference between anode and cathode and its consequently-occurring non-charge equilibriums and electrolytic water- split reactions in the anodic and cathodic chambers.

The Effects of Cynanchi wilfordii Radix Ethanol Extracts upon Irradiated Rat's Blood and Organ (백하수오 에탄올추출물이 방사선조사에 따른 흰쥐의 혈구 및 장기에 미치는 영향)

  • The development of radioprotector is being actively conducted in order to reduce the damage from over radiation exposure at radiation accident or radiation therapy. So this study was confirmed for radiation protective effects using the Cynanchi wilfordii Radix that has been known to be effective for antioxidant activity, anti-cancer, immune enhancing effects. The method of this study was administered orally Cynanchi wilfordii Radix ethanol extracts to Sprague Dawley Rat(SD Rat) for 14 days once a day, while measuring changed blood cell, spleen index, liver and uterus tissue along the change in time of 1, 4, 7 and 21 days after X-ray beam of 7 Gy irradiation. As the result of the experiment, the experimental group's rats which are administered with Cynanchi Wilfordii Radix ethanol extracts showed a rapid recovery in white blood cell count(p < 0.05) and spleen index(p < 0.05). In addition, condensation of nuclei, cytoplasmic swelling, and inflammatory cell infiltration in experimental group's liver cell was decreased more than in irradiation group's component. Further, experimental group's Uterine gland decreased the apoptosis more than irradiation group's components did. It is expected that Cynanchi Wilfordii Radix extracts will be useful as a new radioprotector. With above in mind, this paper may provide appropriate implications with the field of emergency management such as radiation accident.

Conceptual Source Design and Dosimetric Feasibility Study for Intravascular Treatment: A Proposal for Intensity Modulated Brachytherapy (혈관내 방사선치료를 위한 이론적 선원 설계 및 선량적 관점에서의 적합성 연구: 출력변조를 이용한 근접치료에 대한 제안)

  • Purpose: To propose a conceptual design of a novel source for intensity modulated brachytherapy. Materials and Methods: The source design incorporates both radioactive and shielding materials (stainless steel or tungsten), to provide an asymmetric dose intensity in the azimuthal direction. The intensity modulated intravascular brachytherapy was performed by combining a series of dwell positions and times, distributed along the azimuthal coordinates. Two simple designs for the beta-emitting sources, with similar physical dimensions to a $^{90}Sr/Y$ Novoste Beat-Cath source, were considered in the dosimetric feasibility study. In the first design, the radioactive and materials each occupy half of the cylinder and in the second, the radioactive material occupies only a quater of the cylinder. The radial and azimuthal dose distributions around each source were calculated using the MCNP Monte Carlo code. Results: The preliminary hypothetical simulation and optimization results demonstrated the 87$\%$ difference between the maximum and minimum doses to the lumen wall, due to off-centering of the radiation source, could be reduced to less than 7$\%$ by optimizing the azimuthal dwell positions and times of the partially shielded intravascular brachytherapy sources. Conclusion: The novel brachytherapy source design, and conceptual source delivery system, proposed in this study show promising dosimetric characteristics for the realization of intensity modulated brachytherapy in intravascular treatment. Further development of this concept will center on building a delivery system that can precisely control the angular motion of a radiation source in a small-diameter catheter.

A Foreign Cases Study of the Deep Borehole Disposal System for High-Level Radioactive Waste (고준위 방사성폐기물 심부시추공 처분시스템 개발 해외사례 분석)

  • If the spent fuels or the high-level radioactive wastes can be disposed of in the depth of 3~5 km and more stable rock formation, it has several advantages. For example, (1)significant fluid flow through basement rock is prevented, in part, by low permeability, poorly connected transport pathways, and (2)overburden self-sealing. (3)Deep fluids also resist vertical movement because they are density stratified and reducing conditions will sharply limit solubility of most dose-critical radionuclides at the depth. Finally, (4) high ionic strengths of deep fluids will prevent colloidal transport. Therefore, as an alternative disposal concept to the deep geological disposal concept(DGD), very deep borehole disposal(DBD) technology is under consideration in number of countries in terms of its outstanding safety and cost effectiveness. In this paper, for the preliminary applicability analyses of the DBD system for the spent fuels or high level wastes, the DBD concepts which have been developed by some countries according to the rapid advance in the development of drilling technology were reviewed. To do this, the general concept of DBD system was checked and the study cases of foreign countries were described and analyzed. These results will be used as an input for the analyses of applicability for DBD in Korea.

Overseas Review on the In-situ Demonstration of EBS for IN-DEBS Development (공학적방벽 현장실증 시스템 (IN-DEBS) 개발을 위한 해외 실증연구 현황 분석)

  • The worldwide Status-of-Art survey for the in-situ experiments of the engineered barrier system for HLW underground disposal was performed as a preliminary action for the design of the in-situ demonstration at KURT. Some nations, which have executed or is ongoing the in-situ experiments at their underground research facilities, were summarized in this review. The demonstration projects reviewed were TBT/Sweden-France, LOT/Sweden, HE-E/Switzerland, PRACLAY/Belgium, FEBEX/Spain, HORONOBE/Japan, and BCE/Canada. The investigated items for the projects were mainly their purposes, constitutional structures, test conditions, monitoring parameters and the measuring tools, and test results. In this review, the hardware design and the assembling of the test system were more concentrated rather than their experimental results, because the purpose of this review is to achieve the necessary information for the practical design of the in-situ experiment to be installed at KURT. A mid scale in-situ demonstration of EBS at KURT, that is called IN-DBES, will be launched right after the completion the expanding project of KURT in 2015. It is hoped that the structural design, installing methods, hardware equipments required in the establishment of IN-DEBS will be referred on this review.

The Oogenesis of Tiger Barb, Cyprinidae, Teleostei (경골어류 잉어과 Tiger barb의 난자형성과정)

  • Tiger barb (Puntius tetrazona Bleeker, 1855) is a teleost belonging to Cyprinidae. The oogenesis of tiger barb was investigated by light microscope. The ovary was of white and ellipsoidal shape with the major axis 2 cm and the minor axis 1 cm. Cytoplasm of oogonia was basophilic and many nucleoli were located at inside of nuclear membrane. In early stage of primary oocyte, yolk vesicles were distributed only in the marginal area. In secondary oocyte, the egg envelope was formed and yolk vesicles was formed in the cytoplasm. The basophilic substance of cytoplasm was changed to acidic. In case of matured egg, thickness of egg envelope and size of egg were increased. The yolk vesicles were changed to yolk mass in accordance with development. Also, there is not the formation of oil droplets in cytoplasm. In conclusion, the oogenesis of tiger barb was characterized by the increase in cell size, the formation of yolk, the decrease of basophilic substance in the cytoplasm, and formation of yolk mass.

Effect of Electric Stimulation on Parthenogenesis of In Vitro Matured Oocytes from Korean Native Cows (한우 체외성숙란의 단위발생에 대한 전기자극의 효과)

  • The suitable electric stimulation is essential for activation and fusion of oocytes before or after nuclear transplantation The present study was undertaken to determine the optirnal condition for the parthenogenetic activation of in vitro rnatured(IVM) bovine oocytes by electric stimulation. Different direct current(DC) electric voltage of 1.0, 1.5 and 2.0 kV/cm and pulse duration of 30, 60 and 120 $\mu$sec were applied to the JVM nocytes in 0.3 M mannitol solution containing each 100 $\mu$M CaCl$_2$ and MgCl$_2$. IVM occytes at 24, 28 and 32 hours Post-maturation(hpm) were also electrically stimulated at 1.5 kV /cm, for 60 $\mu$ sec. The stimulated nocytes were then co-cultured in TCM-199 solution containing 10% fetal calf serum with bovine oviductal epithelial cells for 7~9 days in a 5% $CO_2$ incubator at 39$^{\circ}C$ ~ Their activation and in vitro development to morula and blastocyst were assessed under an inverted microscope. The higher activation rates 62.8 and 63.4% and in vitro de- velopment rates to morula and blastocyst 5.1 and 10.9% were shown in the oocytes stimulated at the voltage of 1.0 and 1.5 kV/cm than 2.0 kV/cm, respectively. No signifi- cantly(P<0.05) different activation rate was shown in JVM oocytes stimulated for 30, 60 and 120 $\mu$sec, but developmental rates to morula and blastocyst was significantly(P<0.05) higher in the oocytes stimulated for 30 $\mu$sec(6~3%) and 60 $\mu$sec(10~0%) than 120 $\mu$sec(0~ 0%). The aged oocytes at 28 and 30 hpm showed significantly(P<0.05) higher activation rates(72~7 and 79.7%) than the oocytes at 24 hpm(50~9%)~ Also, their developmental rates to morula and blastocyst were significantly(P<0.05) higher in the nocytes at 28(14.3%) and 32 hpm(15.9%) than 24 hpm(3.6%). From these results, it can be suggested that the optimal electric stimulation for IVM bovine occytes is a DC voltage between 1.0 and 1.5 kV/cm, pulse duration of 30 or 60 $\mu$sec, and the optimal age of IVM oocytes for electric activation is at 32 hpm.

The Imprinted Messenger RNA Expression in Cloned Porcine Pre-implantation Embryos

  • The objective of this study was to determine the mRNA expression patterns of several putative imprinted genes in in vivo and in vitro fertilized, parthenogenetic, and cloned porcine preimplantation embryos. Both maternally (Dlk1, IGF2, Peg1/Mest and Ndn) and paternally (IGF2r, H19 and Xist) imprinted genes were selected. We have used reverse transcription polymerase chain reaction (RT-PCR) to investigate gene expression patterns in the porcine embryos. IGF2 transcripts were detected in the most of embryos. In nuclear transfer (NT), Peg1/MEST transcripts showed fluctuating pattern. Dlk1 was only expressed partially from the morula and blastocyst stage of NT embryos. Ndn gene expression was started somewhat early for in vivo embryos. However, the expressions of maternally imprinted genes were similar in all types of blastocysts (NT, in vivo and in vitro fertilized, and parthenogenetic embryos). The IGF2R gene expression level was somewhat irregular and varied among samples. However, for the majority samples of all types of embryos, IGF2R expression was diminished after one- to two-cell stages and reappeared at the morulae or blastocyst stage embryos. H19 gene was only expressed early in parthenogenetic and in vivo embryos. For NT embryos, H19 was only expressed in blastocysts. Xist expression was detected in all blastocysts with the earliest being in vivo 8-cell stage embryos and the last one being NT blastocysts. These putative imprinted genes appeared to have stage specific expression patterns with a fluctuating pattern for some genes (Peg/Mest, IGF2r, H19). These results suggest that stage specific presence of imprinted genes can affect the embryo implantation and fetal development.

Effects of Transverse Reinforcement on Headed Bars with Large Diameter at Cut-off Points (컷오프 구간에 정착된 대구경 확대머리철근에 대한 횡보강근의 효과)

  • The nuclear structures are composed of large diameter bars over No.36. If the hooked bars are used for anchorage of large diameter bars, too long length of the tail extension of the hook plus bend create congestion and make an element difficult to construct. To address those problems, headed bars were developed. Provisions of ACI 318-08 specify the development length of headed bars and ignore the effect of transverse reinforcement based on the background researches. However, if headed bars are used at the cut-off or lap splice, longitudinal reinforcements, which are deformed in flexural members, induce tensile stress in cover concrete and increase the tensile force in the transverse reinforcement. The object of this research is to evaluate the effects of transverse reinforcement on the anchorage capacity of headed bar so anchorage test with variable of transverse rebar spacing was conducted. Specimens, which can consider the behavior at the cut-off, were tested. Test results show that failure of specimen without transverse reinforcement was sudden and brittle with concrete cover lifted and developed stress of headed bars was less than half of yield strength of headed bars. On the other hand, in the specimen with transverse reinforcement, transverse rebar directly resist the load of free-end so capacity of specimens highly increased.

Effects of Di-(2-ethylhexyl) phthalate (DEHP) on Ultrastructure of Rat Testis (흰쥐 정소의 미세구조에 미치는 Di-(2-ethylhexyl) phthalate (DEHP)의 영향)

  • Di-(2-ethylhexyl) phthalate (DEHP) is a plasticize. known as one of endocrine disruptors. The present study was carried out to investigate the ultrastructural changes of prepubertal rat testis after oral administration of DEHP in dosages of 1 g/kg, 3 g/kg or 5g/kg in 0.5 ml of corn oil daily for a week. This study revealed the DEHP inhibited the development of seminiferous tubules and induced structural changes on various cell types of the rat testis. Leydig cells, Sertoli cells and the developing germ cells seemed to be impaired their differentiations in terms of the structural changes of cell organelles. The increase of heterochromatin in amount were common features in all 3 cell types. In addition, the Leydig cells were characterized by the increases in number and size of lysosomes and the scantiness of smooth endoplasmic reticulum. The Sertoli cells became irregular in nuclear envelope and the cytoplasm decreased, but the number of lysosomes and vacuoles seemed to be increased. There were some indications of necrosis of the germ cells, such as vacuolized nucleus and segregated nucleolus. These detrimental effects of DEHP on the rat testis were dose dependent and suppressed spermatogenesis decreasing developing germ cells in number and appearances. The effect of DEHP on ultrastructure of rat testis, as its known physiological functions, seems come from the decreased level of testosterone by Leydig cells, followed by the abnomalities of Sertoli cells and the germ cells.
