A Study on the Effects of Nuclear Power Plant Structure-Component Interaction in Component Seismic Responses (원전 구조물-기기 상호작용이 기기 지진응답에 미치는 영향 연구)

  • Seismic design and analysis of nuclear power plant components are performed based on an decoupled model. However, this decoupled analysis has a limitation in that it generates inaccurate results compared to the coupled analysis because it cannot simulate actual phenomena such as the interaction between structures and components. Thus, this study performed seismic coupled and decoupled analysis on an existing nuclear containment structure and related components, considering the mass and natural frequency ratios. And based on these results, comparative analyses of responses of components were conducted. Consequently, the seismic coupled analysis result generally gave a smaller value than the decoupled analysis result. These results were similar to the analysis results for the simple coupled model, which was an existing study, but the difference in component responses was much more pronounced. Also, this was influenced by the installation location of the component rather than the influence of the input frequency of the input seismic motions. Finally, the difference between the decoupled and coupled seismic analysis occurred in the region where the mass ratio of the components was large, and the natural frequencies were almost similar due to the considerable dynamic interaction between the structure and the component in this realm.

  • The neutral beam injection generate ultra-high temperature energy in the tokamak of nuclear fusion. The neutral beam injection make up arc power supply, filament power supply and acceleration & deceleration power supply. The arc power supply has characteristics of low voltage and high current. Arc power supply generate arc through constant output of voltage and current. So this paper proposed suitable buck converter for low voltage and high current. The proposed buck converter used parallel switch because it can be increased capacity and decrease conduction loss. When an arc generated, the neutral beam injection chamber occur high voltage. And it will break output capacitor of buck converter. Therefore the output capacitor was removed in the proposed converter. Thus the proposed converter should be designed for the characteristics of low voltage and high current. Also, the arc power supply should be guaranteed for system stability. The proposed parallel buck converter enables the system stability of the divided low output voltage and high current. The proposed converter with constant output be the most important design of the output inductor. In this paper, designed arc power supply verified operation of system and stability through simulation and prototype. After it is applied to the 288[kW] arc power supply for neutral beam injection.

Subcellular Localization of Diacylglycerol-responsive Protein Kinase C Isoforms in HeLa Cells

  • Subcellular localization of protein kinase often plays an important role in determining its activity and specificity. Protein kinase C (PKC), a family of multi-gene protein kinases has long been known to be translocated to the particular cellular compartments in response to DAG or its analog phorbol esters. We used C-terminal green fluorescent protein (GFP) fusion proteins of PKC isoforms to visualize the subcellular distribution of individual PKC isoforms. Intracellular localization of PKC-GFP proteins was monitored by fluorescence microscopy after transient transfection of PKC-GFP expression vectors in the HeLa cells. In unstimulated HeLa cells, all PKC isoforms were found to be distributed throughout the cytoplasm with a few exceptions. PKC$\theta$ was mostly localized to the Golgi, and PKC$\gamma$, PKC$\delta$ and PKC$\eta$ showed cytoplasmic distribution with Golgi localization. DAG analog TPA induced translocation of PKC-GFP to the plasma membrane. PKC$\alpha$, PKC$\eta$ and PKC$\theta$ were also localized to the Golgi in response to TPA. Only PKC$\delta$ was found to be associated with the nuclear membrane after transient TPA treatment. These results suggest that specific PKC isoforms are translocated to different intracellular sites and exhibit distinct biological effects.

  • Klystron which is micro wave amplifier tube are mainly used in fields of science such as accelerator, nuclear fusion, broadcasting, communication fields, and defense industry fields, tract. The quality of Klystron anode and cavity are determined by form accuracy and roughness of the worked surface. Therefore anode and cavity are restricted the from accuracy strictly and the surface roughness be under Rmax 0.03S. As a work material of anode and cavity, the oxygen-free copper, that is used for optical pares of aerospace and laser mirror is selected. An outside diameter of material is $\Phi$100 mm and an inside diameter is $\Phi$30~33 mm. In this study, to find the optimum ultra precision cuffing condition of oxygen-free copper with diamond turning machine, the surface roughness is examined for various diamond toot nose radius, main spindle speed, fred rate and depth of cut. As a result of experiment, we could machined the anode and cavity with a surface roughness within Ra 3.2 nm, a form accuracy within 0.01 $\mu\textrm{m}$.

  • In conventional SCC susceptibility test, there are constant strain test, constant load test, slow strain rate test(SSRT) and K$_{ISCC}$ test. Among them, the SSRT method is much more aggressive in producing SCC than the other tests, so that the test time of it is considerably reduced. But this SSRT method has mostly been worked using the uniaxial tensile specimen untill now. Therefore, the SSRT method using the tensile specimen(Ten-SSRT) has much difficulty in SCC susceptibility evaluation of a localized region like weldment and the advantage material of high order. Recentely, the small punch(SP) test method using miniaturized small specimen is the very effective test method for fracture strength evaluation of a localized region like weldment and fusion reactor wall irradiated in the nuclear power plant. This paper investigated the possibility of SCC susceptibility evaluation by the SP-SSRT method using the miniaturized small specimen. Therefore, we obtained the result that the SP-SSRT had the possibility for the evaluations of SCC susceptibility for shorter time to corrosive environment compare to Ten-SSRT which was conventional method.

Identification of Positive and Negative Regulatory Elements of the Human Cytochrome P4501A2 (CYP1A2) Gene

  • We previously demonstrated an enhancer-like positive regulatory element within a 259-bp sequence (-2352 to-2094 bp) of the human CYP1A2 gene in HepG2 cells. Three protein binding sites were identified by DNase I footprint analyses within the 259-bp sequence: protected region A PRA ( -2283 to-2243 bp), PRB (-2218 to-2187 bp), and PRC (-2124 to-2098 bp) (I. Chung and E. Bresnick, Mol. Pharmacol. 47, 677-685, 1995). In the present study, the functional significance of those protected regions was examined. Transfection experiments with deletion and substitution mutants defined the PRB and PRC as containing positive and negative regulatory elements, respectively. Human breast carcinoma MCF-7 cells were cotransfected with a hepatocyte nuclear factor-1 (HNF-1) expression vector and CYP1A2 promoter-or thymidine kinase promoter-luciferase remoter gene constructs. HNF-1, which contributes to the liver specificity of genes, enhanced reporter gene activity in a PRC sequence-dependent manner. These results suggested that PRC could exist bound to a repressor which was displaceable by other transcription factors such as HNF-1. Results obtained by transfection of HepG2 hepatoma cells with various PRB substitution mutant-luciferase gene fusion constructs indicated that the entire sequence of PRB was necessary for promoter activity. Consequently, the regulation of CYP1A3 expression is very complex, requiring a number of both positive and negative regulatory factors.

The NBI consists of filament power supply acceleration and deceleration power supply and arc power supply(APS). The APS has characteristics of low voltage and high current. APS generate arc through constant output of voltage and current. So this paper proposed suitable buck converter for low voltage and high current. And it will break output capacitor of buck converter. Therefore the output capacitor was removed in the proposed converter. Thus buck converter with constant output is the most important design of the output inductor. In this paper, designed APS verified operation of system and stability through simulation and prototype.

Glycosylation of Flavonoids with E. coli Expressing Glycosyltransferase from Xanthomonas campestris

  • Glycosyltransferase family 1 (UOT) uses small chemicals including phenolics, antibiotics, and alkaloids as substrates to have an influence in biological activities. A glycosyltransferase (XcGT-2) from Xanthomonas campestris was cloned and consisted of a 1,257 bp open reading frame encoding a 45.5 kDa protein. In order to use this for the modification of phenolic compounds, XcGT-2 was expressed in Escherichia coli as a glutathione S-transferase fusion protein. With the E. coli transformant expressing XcGT-2, biotransformation of flavonoids was carried out. Flavonoids having a double bond between carbons 2 and 3, and hydroxyl groups at both C-3' and C-4', were glycosylated and the glycosylation position was determined to be at the hydroxyl group of C-3', using nuclear magnetic resonance spectroscopy. These results showed that XcGT-2 regiospecifically transferred a glucose molecule to the 3'-hydroxyl group of flavonoids containing both 3' and 4'-hydroxyl groups.

Effect of long-term thermal aging on the microstructural and mechanical characteristics of nickel-based alloy weldment (니켈계 합금 용접부의 미세조직 및 기계적 특성에 대한 장기 열적 시효의 영향)

  • To investigate the effect of long-term thermal aging on the microstructural and mechanical characteristics of weldment made of nickel base alloy and its weld metal, an accelerated heat treatment was applied to simulate the process of long-term thermal aging in the operating condition of nuclear power plant. A representative nickel-based weldment with Alloy 600 and Alloy 182 was fabricated and heat-treated at $400^{\circ}C$ for 1,713 h and 3,427 h to simulate the thermal aging for the period equivalent to 15 and 30 years in operating pressurized water reactors, respectively. The microstructural and mechanical characteristics were analyzed by using optical microscopy, scanning electron microscopy and Vickers microhardness measurement. Changes were observed in precipitation behavior and microhardness of each specimen, and these changes were mainly attributed to the change in precipitated morphology and residual stress across the weld during the thermal aging process.

Cryopreservation of Unfertilized Oocytes and Use as Recipient Oocyte for Nuclear Transplant in Rabbits (토끼에서 미수정난자의 동결보존과 핵이식을 위한 수핵난자로서의 이용에 관한 연구)

  • This study was conducted to investigate freezability of in vitro and in vitro matured rabbit oocytes, possibility of NT using frozen-thawed unfertilized oocytes, and NT efficiency by zona-slit micromanipulation. After freezing of in vitro matured oocytes, 33 to 49% of oocytes appeared normal morphology and 1.0M DMSO and 1.5M glycerol showed slightly high survival rate, but there was no difference in survival between two cryoprotectants. Freezability of in vitro matured oocytes was low in 1.5M glycerol and more sensitive to freezing. Efficiency of enucleation and fusion rate in method B was higher than that in method A and no difference in this efficiency was between 3 groups of oocytes in method B. Cleavage rate and developmental capacity to M+B stage of fused embryos derived from frozen oocytes was greatly lower than that from fresh oocytes, respectively(39.1% : 79.5% ; 3.1% : 19.3%) and there was no difference in cleavage rate between DC voltages in two group oocytes. Additional incubation in cytochalasin B after electrical stimulation did not affect embryo development. In conclusion, it is suggested that enucleation and nucelar transfer by slitting of zona is more effective method in rabbit and that further study on optimum freezing conditions for in vitro matured oocytes is necessary to use as recipient oocytes.
