Invasion and metastasis is the major cause of tumor recurrence, difficulty for cure and low survival rate. Excavating key transcription factors, which can regulate tumor invasion and metastasis, are crucial to the development of therapeutic strategies for cancers. PU.1 is a master hematopoietic transcription factor and a vital regulator in life. Here, we report that, compared to adjacent non-cancerous tissues, expression of PU.1 mRNA in metastatic hepatocellular carcinoma (HCC), but not primary HCC, was significantly down-regulated. In addition, levels of PU.1 mRNA in metastatic hepatoma cell lines MHCC97L and MHCC97H were much lower than in non-metastatic Hep3B cells. Transwell invasion assays after PU.1 siRNA transfection showed that the invasion of hepatoma cell lines was increased markedly by PU.1 knockdown. Oppositely, overexpression of PU.1 suppressed the invasion of these cells. However, knockdown and overexpression of PU.1 did not influence proliferation. Finally, we tried to explore the potential mechanism of PU.1 suppressing hepatoma cell invasion. ChIP-qPCR analysis showed that PU.1 exhibited a high binding capacity with miR-615-5p promoter sequence. Overexpression of PU.1 caused a dramatic increase of pri-, pre- and mature miR-615-5p, as well as a marked decrease of miR-615-5p target gene IGF2. These data indicate that PU.1 inhibits invasion of human HCC through promoting miR-615-5p and suppressing IGF2. These findings improve our understanding of PU.1 regulatory roles and provided a potential target for metastatic HCC diagnosis and therapy.
Background: High risk human papillomaviruses (HPVs) are the leading cause of cervical cancer (CC) and Pap smear screening has not been successful in preventing CC in Tunisia. HPV vaccination that targets HPV16 and 18 offers a new efficient prevention tool. Identification of HPV types in CC is thus essential to determine the impact of HPV vaccine implementation. The aim of this study is to provide specific data from Tunisia. Materials and Methods: A total of 89 histological confirmed paraffin embedded samples isolated from patients with CC diagnosed between 2001 and 2011 were collected from five medical centres from Northern and Southern Tunisia. HPV DNA was detected using a nested PCR (MY09/MY11-GP5+/GP6+) and genotyping was assessed using a reverse blot line hybridisation assay that enables the detection of 32 HPV types. Results: HPV DNA was detected in all samples. Twelve high risk types were detected; HPV16 and/or 18 were predominant, accounting together for 92.1% of all the CC cases (HPV16: 83.1%). Single infections accounted for 48.8% of the cases and were mostly linked to HPV 16 (32.6%) and less frequently to HPV 18 (2.4%). The other high risk HPV single infections were linked to HPV 35 (4.6%), 45 (4.6%), 58 (2.3%) and 59 (2.3%). Multiple infections with mixing of 2 to 4 genotypes predominately featrued HPV16 and/or 18 with HPV 35 and 45 (96.6 %) and less frequently with HPV 59, 40, 66, 73 and 58. There was no statistically significant variation in the relative distribution of HPV types with age. Conclusions: These results strongly indicate that prophylactic HPV vaccines can have a major impact in preventing CC in Tunisia.

1D PCA utilized in the face appearance-based face recognition methods such as eigenface-based face recognition method may lead to less face representative power and more computational cost due to the resulting 1D face appearance data vector of high dimensionality. To resolve such problems of 1D PCA, 2D PCA-based face recognition methods had been developed. However, the face representation model obtained by direct application of 2D PCA to a face image set includes both face common features and face distinctive identity features. Face common features not only prevent face recognizability but also cause more computational cost. In this paper, we first develope a model of a face distinctive identity feature subspace separated from the effects of face common features in the face feature space obtained by application of 2D PCA analysis. Then, a novel robust face recognition based on the face distinctive identity feature subspace model is proposed. The proposed face recognition method based on the face distinctive identity feature subspace shows better performance than the conventional PCA-based methods (1D PCA-based one and 2D PCA-based one) with respect to recognition rate and processing time since it depends only on the face distinctive identity features. This is verified through various experiments using Yale A and IMM face database consisting of face images with various face poses under various illumination conditions.

Prosthetic replacement is one of the most common methods of reconstruction after resection of malignant tumor around the knee. Gait analysis provides a relative objective data about the gait function of patients with prosthesis. The purpose of this study was to compare the gait pattern of the patients who underwent limb salvage surgery with prosthesis for distal femur and that of patients with prosthesis for proximal tibia. This study included ten patients (4 males, 6 females, mean age 22.7 years, range 14-36) who underwent a wide resection and Kotz hinged modular reconstruction prosthesis replacement and six normal adult(Control). The site of bone tumor was the distal femur (Group 1) in six patients and proximal tibia (Group 2) in 4 patients. The follow-up period ranged from 15 to 82 months (mean : 33 months). The evaluation consisted of clinical assessment, radiographic assessment, gait analysis using VICON 370 Motion Analysis System. The gait analysis included the linear parameters such as, walking velocity, cadence, step length, stride length, stance time, swing time, single support and double support time and the three-dimensional kinematics (joint rotation angle, velocity of joint rotation) of ankle, knee, hip and pelvis in sagittal, coronal and transverse plane. For the kinetic evaluation, the moment of force (unit: Nm/kg) and power (unit: Watt/kg) of ankle, knee and hip joint in sagittal, coronal and transverse plane. In the linear parameters, cadence, velocity, step time and single support were decreased in both group 1 and group 2 compared with control. Double support decreased in group 2 compared with control significantly(p<.05). In contrast to our hypothesis, there was no significant difference between group 1 and group 2. In Long Term Evolution-Advanced (LTE-A), small cell enhancement(SCE) has been developed as a cost-effective way of supporting exponentially increasing demand of wireless data services and satisfying the user quality of service(QoS). However, due to the dense and irregular distribution of a large number of small cells, the offloading scheme should be applied in the small cell network. In this paper, we propose an user offloading scheme for SCE in LTE-Advanced system. We divide the small cells into different clusters according to the reference signal received power(RSRP) from user equipment(UE). Within a cluster, We apply the user offloading scheme with the consideration of the number of users and interference conditions. Simulation results show that proposed scheme can improve the throughput, and spectral efficiency of small cell users. Eventually, proposed scheme can improve overall cell performance.

In this paper, we propose a clustering based interference management scheme in dense small cell network. We divide the small cells into different clusters according to the reference signal received power(RSRP) from user equipment(UE). Within a cluster, an almost blank subframe(ABS) is implemented to mitigate interference between the small cells. In addition, we apply the power control to reduce the interference between the clusters. Simulation results show that proposed scheme can improve Signal to Interference plus Noise Ratio(SINR), throughput, and spectral efficiency of small cell users. Eventually, proposed scheme can improve overall cell performance.

Numerous platforms have been developed for ZigBee-based network embedded systems. Also, operating systems like TinyOS have been installed to facilitate efficient implementation of wireless sensor network applications which collect data, and/or execute commands. First of all, porting an operating system on a new platform may need invention of a substitute for a required but unsupported hardware component. This paper presents a multiplexed virtual system timer for a platform without a counter comparator which we have contrived to emulate by using an extra counter. Such porting also injects unexpected faults which cause a variety of painful failures. Unfortunately, TinyOS requires to handle a lot of asynchronous hardware interrupts which are hard to trace during debugging. Besides, simulators are not available for a new platform since the models of hardware on the platform are not usually developed, yet. We propose novel instrumentation techniques which can be used to effectively trace the bugs in such lack of debugging environment. These techniques are used to identify and fix a great deal of nasty issues in porting TinyOS 2.0 on MG2400 and MG2455 platforms made by RadioPulse Inc.

This paper proposes a novel reduced-reference assessment method for video quality assessment, in which one-dimensional (1-D) histograms of motion vectors (MVs) are used as features of videos. The proposed method is more efficient than the conventional methods in view of computation time, because the proposed quality metric decodes MVs directly from video stream in the parsing process instead of reconstructing the distorted video at the receiver. Moreover, in view of data size, the propose method is efficient because a sender transmits 1-D histograms of MVs accumulated over whole input video sequences. Here, we use 1-D histograms of MVs accumulated over the whole video sequences, which is different from the conventional methods that assessed each image independently. For testing the similarity between histograms, we use histogram intersection and histogram difference methods. We compare the proposed method with the conventional methods for 52 video clips, which are coded under varying bit rate, image size, and frame rate. Experimental results show that the proposed method is more efficient than the conventional methods and that the proposed method is more similar to the mean opinion score (MOS) than conventional algorithms.

Purpose: The endoscopic management of a fully covered self-expandable metal stent (SEMS) has been suggested for the primary treatment of patients with anastomotic leaks after total gastrectomy. Embedded stents due to tissue ingrowth and migration are the main obstacles in endoscopic stent management. Materials and Methods: The effectiveness and safety of endoscopic management were evaluated for anastomotic leaks when using a benign fully covered SEMS with an anchoring thread and thick silicone covering the membrane to prevent stent embedding and migration. We retrospectively reviewed the data of 14 consecutive patients with gastric cancer and anastomotic leaks after total gastrectomy treated from January 2009 to December 2016. Results: The technical success rate of endoscopic stent replacement was 100%, and the rate of complete leaks closure was 85.7% (n=12). The mean size of leaks was 13.1 mm (range, 3-30 mm). The time interval from operation to stent replacement was 10.7 days (range, 3-35 days) and the interval from stent replacement to extraction was 32.3 days (range, 18-49 days). The complication rate was 14.1%, and included a single jejunal ulcer and delayed stricture at the site of leakage. No embedded stent or migration occurred. Two patients died due to progression of pneumonia and septic shock 2 weeks after stent replacement. Conclusions: A benign fully covered SEMS with an anchoring thread and thick membrane is an effective and safe stent in patients with anastomotic leaks after total gastrectomy. The novelty of this stent is that it provides complete prevention of stent migration and embedding, compared with conventional fully covered SEMS.

Objective: The current research was aimed to profile the transcriptomic picture of the peripheral blood lymphocytes (PBLs) associated with immunity in Chinese Holsteins supplemented orally with coated folic acid during the periparturient period. Methods: The total of 123 perinatal cows were selected for this study and divided into three groups; group A (n = 41, 240 mg/500 kg cow/d), group B (n = 40, 120 mg/500 kg cow/d) and group C (n = 42, 0 mg/cow/d) based on the quantity of folic acid fed. Three samples of PBLs were selected from each folic acid treated group (high, low, and control) and RNA sequencing method was carried out for transcriptomic analysis. Results: The analysis revealed that a higher number of genes and pathways were regulated in response to high and low folic acid supplementation compared to the controls. We reported the novel pathways tumor necrosis factor (TNF) signaling, antigen processing and presentation, Staphylococcus aureus infection and nuclear factor (NF)-kappa B signaling pathways) and the key genes (e.g. C-X-C motif chemokine ligand 10, TNF receptor superfamily member 1A, cluster difference 4, major histocompatibility complex, class II, DQ beta, NF-kappa-B inhibitor alpha, and TNF superfamily 13) having great importance in immunity and anti-inflammation in the periparturient cows in response to coated folic acid treatment. Conclusion: Collectively, our study profiled first-time transcriptomic analysis of bovine lymphocytes and compared the involved cytokines, genes, and pathways between high vs control and low vs control. Our data suggest that the low folic acid supplementation (120 mg/500 kg) could be a good choice to boost appropriate immunity and anti-inflammation as well as might being applied to the health improvement of perinatal dairy cows.
