Analysis of agricultural characteristics and qualities of wheat under high temperature

  • High temperature is one of major environmental stress. Heat in flowering season of wheat is able to effect negatively to fertilization and also heat effect to maturity. Therefore, Heat stress effects severely to qualities of wheat and yield productivity. In this study, we evaluated to agronomic characteristics and qualities by high temperature in 13 Korean wheat varieties. Weight of 1,000 grains decreased when wheat got the heat stress. In particular, heat stress during the heading dates were more damaged than after the end of heading dates except two varieties Keumkang and Jokyoung. Plant height of each cultivar under high temperature and normal field averaged 80.5 cm and 83.0cm, respectively. The length of spike and awn in each cultivar were similar to both condition. Flour yield and gluten contents of most heat damaged wheat decreased. Under the temperature, protein contents of six varieties like as Keumkang, Baekjoong, Hojoong, Yeonbaek, Joah and Shinmichal 1 decreased but the others increased. The sedimentation values (SDSF) of four varieties decreased under the high temperature. But SDSF of 7 varieties like as Baekjung, Suan, Hojoong, Jojoong, Uri, Shinmichal and Shinmichal 1 was increased. The lightness (L) of wheat flour derived from high temperature treated wheat was darker than non-treated wheat. As a result of this research, we confirmed that agricultural traits and qualities decreased in heat damaged wheat.

  • A low-cost and flexible potassium ion (K+) sensor was fabricated through a screen-printed process. Uniform and conformal coating of conductive inks was verified by scanning electron microscopy and optical microscopy measurements. The K+-sensors showed a high sensitivity, fast response time, and low detection limit. The sensitivity of K+-sensor was similar to that of both mechanically normal and bent states. The K+-sensor exhibited a good reproducibility with no hysteresis effect and excellent long term stability. In addition, the K+-sensor showed an excellent selectivity for K+ concentrations in the presence of other interfering cation ions. Successful measurements of K+ concentrations in sports drink samples were demonstrated by comparing K+ concentration values from K+-sensor to those of using a commercial K+-meter.

  • Blood pressure is possible to diagnose a disease associated with blood pressure and judgment the current health of patients. Automatic blood pressure monitor capable of measuring a blood pressure easily in hospital and at home have become spread. In this study, we developed the blood pressure simulator (BPS) that can test the arm-type automatic blood pressure monitor that is commonly used in hospital. BPS is to produce a pressure similar to the pressure wave generated in the human blood using a servo disk motor. Then, using the silicon tube, it implements the situations such as human blood vessels, and to output the generated pressure waveform. Simply the BPS's phantom put on the cuff and it is able to simulate blood pressure. So anyone can quickly test the blood pressure monitor within one minute and it is possible to shorten the test time required for the automatic blood pressure monitor. In Performance test, the trends and the standard deviation of the values measured in the BPS is similar to the value of the measured pressure from people with normal blood pressure. Thus, the development BPS showed a possibility of taking into account the actual blood pressure measurement environment simulator.

A Proposal of Digital Photo-Biometry of MRD1 and New Levator Function Test (디지털 사진을 이용한 MRD1의 계측과 새로운 위눈꺼풀 올림근 근력 측정법의 제안)

  • Purpose: The history of biometry dates back to ancient Greek. The ideal body ratio from biometry is used as a guideline in many works of art. Biometry is also used as a medical standard to determine normal or abnormal. Since the biometry of face is so complicated, many surgeons tend to regard preoperative evaluation as a bother and substitute medical records with some photographs. We introduce a new method to measure $MRD_1$ and levator function using digital photographs, which becomes widely used lately. Methods: $MRD_1$ can be measured with the primary-gaze-view photo which is magnified by PhotoshopR 7.0.1. The distance from coneal reflex of ring flash to upper eyelid margin is converted to mm-unit using the piece of ruler attached on the patient's face. Levator function can be measured with up-gaze-view photo and down-gaze-view photo which are superimposed on another. The excursion distance of upper eyelid margin (a) and the excursion distance of eyebrow (b) are measured respectively. The levator function can be assessed through subtraction of two values (a-b). This method is simple and precise and can be applied directly to patient without photos. Results: Using magnified digital photos and computer, this method can reduce personal error and instrumental error. Taking some digital photos doesn't take long time, so it can reduce the effort of preoperative evaluation and discomfort of patients. Conclusion: Digital photo-biometry is useful for retrospective study. Especially reducing personal error, it is useful when the number of specimens is huge. New levator function test is much more useful for Asian-specific eyes than Berke's method allowing frontalis muscle compensation.

The Study on Intraoral Pressure, Closure Duration, and VOT During Phonation of Korean Bilabial Stop Consonants (한국어 양순 파열음 발음시 구강내압과 폐쇄기, VOT에 대한 연구)

  • Acoustic analysis study was performed on 20 normal subjects by speaking nonsense syllables composed of Korean bilabial stops(/p, $p^{*}$/, ph/) and their Preceding and/or following vowel /a/(that is, [pa, $p^{*}a$, pha, apa, $ap^{*}a$, apha]) with an ultraminiature pressure sensor in their mouths. Speech materials were phonated twice, once with a moderate voice, another time with a loud voice. The acoustic signal and intraoral pressure were recorded simultaneously on computer. By these procedures, we were to measure the intraoral pressure, closure duration and VOT of Korean bilabial stops, and to compare the values one another according to the intensity of phonation and the position of the target consonants. Intraoral pressure was measured by the peak intraoral pressure value of its wave; closure duration by the time interval between the onset of intraoral pressure build-up and the burst meaning the release of closure; Voice onset time(VOT) by the time interval between the burst and the onset of glottal vibration. Heavily aspirated bilabial stop consonant /ph/ showed the highest intraoral pressure value, unaspirated /p$^{*}$/, the second, slightly aspirated /p/, the lowest. The syllable initial bilabial stops showed higher intraoral pressure than word initial stops, and the value of loudly phonated consonants were higher than moderate consonants. The longest closure duration period was that of /$p^{*}$/ and the shortest, /p/, and the duration was longer in word initial position and in the moderate voice. In VOT, the order of the longest to shortest was /ph/, /p/, /$p^{*}$/, and the value was shorter when the consonant was in intervocalic position and when it was phonated with a loud voice.

Changes of relative impulse of foot on carrying 5 types of schoolbag during level walking (가방 휴대 방법에 따른 보행 시 발의 비례추진력 변화)

  • Purpose: The purpose of this study was to analyze the changes of the relative impulse of the foot when carrying 5 types of schoolbag during level walking. Methods: Forty-four normal subjects were divided into 5 groups according to the method of carrying a bag: carrying no bag (condition 1), a backpack (condition 2), a shoulder bag (condition 3), a cross bag (condition 4) and a one-hand bag (condition 5). To evaluate the partial relative impulse, 7 areas were measured on the sole of a foot: the hallux, toe, 1st metatarsal head (Met1) and the 2nd & 3rd metatarsal heads (Met2/3), the 4th & 5th metatarsal heads (Met4/5), mid foot and heel. Repeated one-way analysis of variance (ANOVA) and independent t-tests were used to analyze the statistical data. Results: In the right foot, the relative impulses in every areas revealed no significant difference (p>0.05), however, the relative impulses at Met1, Met2/3, Met4/5 and heel were significantly different between the different conditions (p<0.05). There were significant differences on comparing the relative impulse between left and right foot within one condition, the mid foot in condition 2, as well as the hallux and Met1 in condition 3 (p<0.05). In condition 4, the toe, Met1, Met4/5 and mid foot were significantly different (p<0.05). The values at the hallux, toe and Met1 in condition 5 also were significantly different (p<0.05). Conclusion: These findings showed that the symmetrical relative impulse of the foot was present in the case of carrying a backpack, but there was an extra load on specific regions and asymmetrical relative impulses in the case of carrying a shoulder bag, a across bag and a one-hand bag. Subacute Toxicity of SKI306X, an Antiinflammatory Herbal Extracts, in Rats (생약복합제 SKI306X의 랫드에 대한 4주 경구 반복투여 독성연구)

  • This study was performed to determine the subacute toxicities of SKI306X, an antiinflammatory herbal extract, in rats. SKI306X was administered orally to rats once a day for 4 weeks at doses of 0.3, 1.0, and 3.0 g/kg/ day. Each group consisted of 20 male and 20 female rats, including 5 male and 5 female rats per group for an interim study at the end of 2-week administration and for a 2-week recovery study, respectively. Throughout the study, all rats survived and no adverse clinical signs were observed. Although male rats treated with high dose (3.0 g/kg/day) of SKI306X showed slight loss of body weight (approximately 5%) in comparison with control animals during the administration period, their body weight loss was normally restored during the recovery period. No significant change was found in all hematological parameters of SKI306X-treated groups except for the decreased number of red blood cells in all female groups at the interim study. Statistically significant changes were observed in several blood enzyme levels of SKI306X-treated groups; however, most of these significant changes were within normal range and statistically significant values did not show dose-related responses. In SKI306X-treated groups, the absolute and relative weights of liver, heart, and stomach were statistically different from those of control group, but these differences disappeared at the end of recovery period and also drug-related gross and histopathological findings in these organs were not found. No other drug-related gross and histopathological findings were observed. It is concluded from the results of this study that non-toxic dose of SKI306X was estimated to be between 0.3 and 1.0 g/kg/day and the maximum tolerated dose of SKI306X was assumed to be higher than 3.0 g/kg/day.

Transcriptome profiling of the coffee (C. arabica L.) seedlings under salt stress condition

  • This research was conducted to study the gene expression of coffee (Coffea arabica L.) seedlings under salt stress condition. A solution of five percent ($2.3dS\;m^{-1}$) deep sea water was used for the salt treatment, and it was thereby compared to normal irrigation water ($0.2dS\;m^{-1}$) used for the control treatment. The mRNA was extracted from the leaves of the coffee seedlings for a comprehensive analysis. In this study, a total of 19,581 genes were identified and aligned to the reference sequences available in the coffee genome database. The gene ontology analysis was performed to estimate the number of genes associated with the identified biological processes, cellular components and molecular functions. Among the 19,581 genes, 7369 (37.64%) were associated with biological processes, 5909 (30.18%) with cellular components, and 5325 (27.19%) with molecular functions. The remaining 978 (4.99%) genes were therefore grouped as unclassified. A differential gene expression analysis was performed using the DESeq2 package to identify the genes that were differentially expressed between the treatments based on fold changes and p-values. Namely, a total of 611 differentially expressed genes were identified (treatment/control) in that case. Among these, 336 genes were up-regulated while 275 of the genes were down-regulated. Of the differentially expressed genes, 60 genes showed statistically significant (p < 0.05) expression, 44 of which were up-regulated and 16 which were down-regulated. We also identified 11 differentially expressed transcription factor genes, 6 of which were up-regulated and rest 5 genes were down-regulated. The data generated from this study will help in the continued interest and understanding of the responses of coffee seedlings genes associated with salinity stress, in particular. This study will also provide important resources for further functional genomics studies.

Treatment Planning and Dosimetry of Small Radiation Fields for Stereotactic Radiosurgery (Stereotactic Radiosurgery를 위한 소형 조사면의 선량측정)

  • The treatment planning and dosimetry of small fields for stereotactic radiosurgery with 10 MV x-ray isocentrically mounted linear accelerator is presented. Special consideration in this study was given to the variation of absorbed dose with field size, the central axis percent depth doses and the combined moving beam dose distribution. The collimator scatter correction factors of small fields $(1\times1\~3\times3cm^2)$ were measured with ion chamber at a target chamber distance of 300cm where the projected fields were larger than the polystyrene buildup caps and it was calibrated with the tissue equivalent solid state detectors of small size (TLD, PLD, ESR and semiconductors). The central axis percent depth doses for $1\timesl\;and\;3\times3cm^2$ fields could be derived with the same acuracy by interpolating between measured values for larger fields and calculated zero area data, and it was also calibrated with semiconductor detectors. The agreement between experimental and calculated data was found to be under $2\%$ within the fields. The three dimensional dose planning of stereotactic focusing irradiation on small size tumor regions was performed with dose planning computer system (Therac 2300) and was verified with film dosimetry. The more the number of strips and the wider the angle of arc rotation, the larger were the dose delivered on tumor and the less the dose to surrounding the normal tissues. The circular cone, we designed, improves the alignment, minimizes the penumbra of the beam and formats ball shape of treatment area without stellate patterns. These dosimetric techniques can provide adequate physics background for stereotactic radiosurgery with small radiation fields and 10MV x-ray beam.

Detection of urinary trypsinogen-2 for diagnosis of canine acute pancreatitis (뇨중 trypsinogen-2 검출을 이용한 개의 급성 췌장염 진단)

  • We performed this study to evaluate the potential clinical marker of urinary trypsinogen-2 together with amylase, lipase and urinary amylase creatinine clearance ratio (ACCR) for the diagnosis of acute pancreatitis in dogs. In the experiment on daily changing patterns of amylase, lipase and ACCR measurements in experimentally induced pancreatitis dogs, compared to values measured in pre-induction state, significant difference was seen in amylase until 5th day of induction, and for lipase significant difference was found during the 7th day of observation period (p < 0.05). No significant difference was found in ACCR for the study period (p > 0.05). On SDS-PAGE analysis of urine from experimentally induced pancreatitis dog, The 26kd band was markedly increased compared with that of normal state and that band was confirmed trypsinogen-2 using substrate interaction and isoelectric focusing assay after being eluted. When assessing the appearance of 26kd band on urine SDS-PAGE 87.1% (range: 50~100%) of experimentally induced pancreatitis dogs showed positive results, whereas no corresponding band was seen in dog without pancreatic disorders. With this result, determination of urinary trypsinogen-2 assay was found to have a high diagnostic value with a 70% of sensitivity and 100% of specificity as a routine test for pancreatitis, although the detection of trypsinogen-2 in urine can be varied on the progression stage of pancreatitis at the initial visit to animal clinic. We therefore suggest that the promising results in this study be used for the development of dipstick test for detecting acute pancreatitis in the future research.
