Karyological Variation of Callus-derived Regenerants in Allium victorialis var. platyphyllum

  • The calli obtained from the bulbs of A. victorialis var. platyphyllum on MS based medium containing 2 mg/L 2, 4-D and 1mg/L BA. Plants from calli were regenerated on MS basal media supplemented with combinations of NAA and four kinds of cytokinin, BA, zeatin, kinetin and 2iP, and with only each cytokinin. The good response of shoot regeneration was observed in the media with combinations of NAA and BA or zeatin, and with only BA or zeatin. Shoot regenerating response in the medium with combinations of NAA and BA or zeatin, and with only BA or zeatin. Shoot regerating response in the medium with combinations of NAA and BA or zeatin was about two times higher than that in the mediuim with only BA or zeatin. From the karyotypic analysis of the regenerated plants, abnormal diploids, aneuploids and mixoploid with structural aberrations were investigated. The somaclonal variants (AV 16-04, AV 13-03) were shown the considerable differences from normal diploid regenerant (AV 18-03) in the external morphology.

A Study on the Facade Image and Emotional Response considering Geographical Language of Railway Station - focused on Metropolitan Line of Chungcheong Railway - (철도 역명의 유래를 통하여 본 파사드 이미지 및 감성반응에 관한 연구 -충청권 광역 전철화 노선을 중심으로-)

  • This study is aimed to reflect the current status survey, analysis and response of 10 railway stations using geographical names in one of 19 stations located on Gyeongbu, Honam and Chungbuk line. Most railway stations in the study have shown normal images that are close to the standard type by selecting similar building materials rather than reflecting geographical features. Aside from the shape of the traditional Korean house in Cheongju, almost all newly constructed stations tend to incorporate light gray plating material on their modern exterior to produce Family look design as a railroad station. According to a survey of the sensitivity of the station's name, Osong Station was 73.6 percent, Gyeryong Station 67.3 percent, Heukseok-ri Station was 64.5 percent, Shintan-jin 62.6 percent, and Yeonsan Station 57 percent, more than half. Therefore, the results of the five stations were presented as a calculation and proposed design release focusing on the facades of each station. Each of these stations suggested a way to express the facade design image, considering the railway operation status of Korea Railroad, the area of the station and other surroundings.

Carboxymethyl Chitosan Promotes Migration and Inhibits Lipopolysaccharide-Induced Inflammatory Response in Canine Bone Marrow-Derived Mesenchymal Stem Cells

  • The study was conducted to evaluate the effects of carboxymethyl chitosan (CMC) on proliferation, migration, and lipopolysaccharide (LPS)-induced inflammatory response in canine bone marrow-derived mesenchymal stem cells (BMSCs). The proliferation and migration of BMSCs were examined after treatment with CMC. The effect of CMC on the mRNA expression of inflammatory cytokines, such as interleukin (IL)-1β, IL-6, tumor necrosis factor (TNF)-α, IL-10, and transforming growth factor (TGF)-β, was also evaluated by reverse transcription polymerase chain reaction (RT-PCR). In the proliferation assay, no significant changes were found at all CMC concentrations compared with controls. The migration assay showed that CMC dose-dependently stimulated the migration of BMSCs in normal and LPS-treated conditions. RT-PCR showed that TNF-α and IL-10 expressions were suppressed in the BMSCs after CMC treatment. However, other genes were not affected. Taken together, CMC promoted BMSC migration and inhibited TNF-α and IL-10. Therefore, CMC may be possible to regulate wound healing when mesenchymal stem cells are applied in inflammatory diseases.

Ethylene-Induced Auxin Sensitivity Changes in Petiole Epinasty of Tomato Mutant dgt

  • The tomato (Lycopersicon esculentum Mill.) mutant diageotropica (dgt) lacking normal gravitropic response is known to be less sensitive to auxin compared with its isogenic parent VFN8. Straight growth as well as ethylene production in response to added auxin in hypocotyl segments of dgt was negligible. However, there was no significant difference between the two genotypes in auxin transport in petiole segments and its inhibition by the phytotropin N-1-naphthylphthalamic acid(NPA). Kinetic parameters of NPA binding to microsomal membranes were also non-distinguishable between the two. Its petiolar explants treated with ethylene developed epinastic curvature with the magnitude of response increased about 3 folds over non-mutant wild type. Ethylene-induced epinasty in both dgt and VFN8 was nullified by treatment of explants with the ethylene autagonist 2,5-norbonadiene. Lateral transport of 3H-IAA toward the upper side of ethylene-treated petioles in dgt, however, was not significantly more pronounced than in VFN8, the implications being that auxin sensitivity in the mutant was restored, or even rised above the wild type, by ethylene.

Field Observation and Quasi-3D Numerical Modeling of Coastal Hydrodynamic Response to Submerged Structures

  • Even though submerged breakwater reduces incident wave energy, it redistributes the coastal area's wave-induced current, sediment transport, and morphological change. This study examines the coastal hydrodynamics and the morphological response of a wave-dominated beach with submerged breakwaters installed through field observation and quasi-3D numerical modeling. The pre-and post-storm bathymetry, water level, and offshore wave under storm forcing were collected in Bongpo Beach on the East coast of Korea and used to analyze the coastal hydrodynamic response. Four vertically equidistant layers were used in the numerical simulation, and the wave-induced current was examined using quasi-3D numerical modeling. The shore normal incident wave (east-northeast) generated strong cross-shore and longshore currents toward the hinterland of the submerged breakwater. However, the oblique incident wave (east-southeast) induced the southeastward longshore current and the sedimentation in the northeast area of the beach. The results suggested that the incident wave direction is a significant factor in determining the current and sediment transport patterns in the presence of the submerged breakwaters. Moreover, the quasi-3D numerical modeling is more appropriate for estimating the wave transformation, current, and sediment transport pattern in the coastal area with the submerged breakwater.

Dyssynergic Defecation in Chronically Constipated Children in Korea

  • Purpose: Dyssynergic defecation (DSD) is one of the important causes of chronic constipation in children. We aimed to analyze the clinical features, diagnostic test results, and treatments for DSD in children. Methods: Children diagnosed with DSD using fluoroscopic defecography were enrolled in this study. Clinical data, including the results of colon transit time (CTT) test and biofeedback (BF) therapy, were collected from medical records retrospectively. Results: Nineteen children were enrolled. The median age was 9 years (6-18 years), the median frequency of bowel movement was 1/7 days (1-10 days), the median duration of constipation was 7.0 years (2-18 years), the median age of onset of constipation was 2.5 years (1-11 years). In the CTT test, outlet obstruction type was noted in 10/18 (55.6%), slow transit type in 5/18 (27.8%), and normal transit in 1/18 (5.6%). The median CTT was 52 hours (40-142 hours). Initial medical therapy was performed with the polyethylene glycol 4000, and the response was good in 9/19 (47.4%), fair in 9/19 (47.4%), and poor in 1/19 (5.0%). BF was performed in 8/19, with good results in 6/8 (75.0%) children and failure in 2/8 (25.0%) children. After long-term medical therapy (11/19), 3/5 showed good response with medication alone, 6/8 showed good response with BF and medication combined. Conclusion: DSD should be considered as a cause of chronic constipation in children, especially in those with abnormal CTT test results. BF combined with medical therapy is effective even with age-limited cooperation.

TRAIP regulates Histone H2B monoubiquitination in DNA damage response pathways

  • Histone H2B monoubiquitination has been shown to play critical roles in diverse cellular processes including DNA damage response. Although recent data indicate that H2B monoubiquitination is strongly connected with tumor progression and regulation, the implications of this modification in lung adenocarcinoma are relatively unknown. In the present study, we demonstrated the clinical implication of H2B monoubiquitination and the potential role of tumor necrosis factor receptor-associated factor-interacting protein (TRAIP) in regulating its modification in lung adenocarcinoma. Immunohistochemical analysis showed that H2B monoubiquitination was significantly downregulated in 68 human lung adenocarcinoma patient samples compared to their normal adjacent tissues. Depletion of TRAIP by specific siRNA treatment markedly decreased ionizing radiation (IR)-induced H2B monoubiquitination. In addition, deletion mutants without RING domain or C-terminus of TRAIP diminished the ability to induce H2B monoubiquitination at lysine 120. Notably, the nuclear expression of TRAIP was positively related with H2B monoubiquitination levels in patients with lung adenocarcinoma. Furthermore, statistical analysis indicated that low levels of both TRAIP and H2B monoubiquitination, not each alone, in patients with lung adenocarcinoma were strongly correlated with poor survival. Taken together, these results suggest that TRAIP is a novel regulator of H2B monoubiquitination in DNA damage response and cancer development in lung adenocarcinoma.

  • This paper is concerned with the positive steady states of a diffusive Holling type II predator-prey system, in which two predators and one prey are involved. Under homogeneous Neumann boundary conditions, the local and global asymptotic stability of the spatially homogeneous positive steady state are discussed. Moreover, the large diffusion of predator is considered by proving the nonexistence of non-constant positive steady states, which gives some descriptions of the effect of diffusion on the pattern formation.

  • The effect of radio- and chemotherapy for cancer are excellent, but their toxicities to normal tissue and organ of the body is relatively strong, which leads secondary side effect to patients during therapies. Particularly, due to the response for bone marrow suppression such as agranulocytosis limits the therapy periods and dose of drugs, new drug development that reproduces lymphocytes has been focused. A note on Box-Cox transformation and application in microarray data

  • The Box-Cox transformation is a well known family of power transformations that brings a set of data into agreement with the normality assumption of the residuals and hence the response variable of a postulated model in regression analysis. Normalization (studentization) of the regressors is a common practice in analyzing microarray data. Here, we implement Box-Cox transformation in normalizing regressors in microarray data. Pridictabilty of the model can be improved using data transformation compared to studentization.
