The purpose of this study was to investigate the anti-obesity effects of crude saponin (CS) in Korean red ginseng (KRG) on rats fed a high fat (HF) diet. Methods: Male Sprague-Dawley (SD) rats were divided into a normal diet group and a high-fat (HF) diet group. The HF diet group became obese from being fed a HF diet over five weeks. The normal diet group were fed a normal diet. Each CS group of the normal diet group and HF diet group was administered CS (200 mg/kg, i.p.) for three weeks, the control group of both types was administered normal saline (1ml/kg, i.p.) instead of CS. Each group had 5 rats. Results: After administration of CS, the body weight, food consumption, adipose tissues, and expression of appetite peptides such as leptin and neuropeptide-Y (NPY) were investigated in the HF diet group as well as the normal diet group. Administration of CS reduced body weight, food intake, and fat content in the HF and normal diet groups. After CS administration, NPY expression and leptin were lower in the HF diet group. Conclusions: Our results suggest that CS may be useful in the treatment of obesity, especially of type qixu (氣虛).
The purpose of this study was to investigate the effects of dietary chungguajang powder on blood glucose level and hepatic activities of antioxidant enzymes in normal and streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male ratt ($200{\sim}220g$) of six groups including normal group fed normal diet (N), diabetic group fed normal diet (C), diabetic groups fed chunggugjang powder diet (DC-1%, DC-5%, DC-10%, DC-20%) were used for the experiments. Diabetes was induced by single intraperitoneal injection of streptozotocin as a dose of 70 mg/kg body weight. After 3 weeks the animals were sacrificed and hepatic activities of antioxidant enzymes, serum level of glucose and organ weight were evaluated. Food and water intakes were higher in diabetic groups than normal group. Body weight gain and food efficiency ratio were lower in diabetic groups. However, they were higher in chunggugjang diet groups (DC) than normal diet group (C). The serum glucose levels were significantly lower (p<0.05) in the diabetic groups fed chunggugjang diet (DC-10%, DC-20%) than diabetic group fed normal diet (C). Hepatic activities of superoxide dismutase, catalase, glutathione peroxidase and glutathione S-transferase were lower in diabetic groups than normal group and they were significantly lower in diabetic groups fed chunggugjang diet (DC-20%) compared to diabetic rats fed normal diet (p<0.05). In conclusion, these results indicated that chunggugjang powder would have antidiabetic and antioxidant effects in STZ-induced diabetic rats.
The purposes of this study were to : (a) analyze the portion size & plate waste of normal & soft diets provided by dietetic departments in hospital, (b) investigate the factors affecting plate waste, and (c) determine the nutritional & monetary value of plate waste. A questionnaire for food preference and sensory evaluation was developed and a survey was carried out for the 46 normal diet and 56 soft diet patients in Sanggye Paik hospital in Seoul. Serving size and plate waste were weighed by the electric scale, and CAN-Pro program was used to evaluate the nutrition value. The data were analyzed using the SAS package program for descriptive analysis, t-test, ANOVA, and Pearson correlation. The average plate waste rate for normal diet and soft diet were 30.3% and 49.6%. More plate waste amount occurred on female patients'diet than male patients'diets regardless of the kind of diet. The plate waste percentage of male patients were higher than those of female on normal diet. On soft diet, patients who hospitalized in surgery or pediatrics department were higher than in other wards. Plate waste of normal diet and amount were correlated positively, but wastage and preference were correlated negatively Negative correlation was between taste and plate waste, and between temperature and plate waste, too. On the other hand, plate waste caused the deficiency of some nutrients such as Ca, Fe, Vit. B$_2$and a great monetary loss.
This study was carried out to investigate the anti-oxidant effect of Wasabia japonica(WJ). The anti-oxidant activities of various extracts from WJ were investigated. The water and ethanol extracts of WJ leaf were found to cause significant free radical scavenging effects on DPPH and nitric oxide(NO). Sprague-Dawley male rats were randomly assigned to one normal diet and three high cholesterol diet groups which contained 1%(w/w) cholesterol. After 4weeks cholesterol diet, the groups of high cholesterol diet were classified to control(normal diet only), WJR5(normal diet and 5% WJ root) and WJL5(normal diet and 5% WJ leaf). Liver xanthine oxidase(XO) activity in the rats of high cholesterol diet was decreased up to the levels of normal diet group according to WJ diet. The results indicated that WJ possesses antioxidant effects through free radical scavenging effects on DPPH, NO and the decreasing of XO activity. In view of these results, WJ is expected to be an effective material for the anti-oxidant.
The thymus weight of the mouse was 54.1% in protein malnutritive diet group and 39.2% in group treated with saponin fraction of Panax ginseng in comparison to normal diet group. This decreasing effects of protein malnutritive diet and saponin fraction on the thymus weight practically disappeared after four weeks. The saponin fraction showed no effect on the spleen weight of the mouse. The supplement of the saponin fraction enhanced total peritoneal exudate cells, content of total serum protein and albumin content of the mouse, each 45, 8 and 10% respectively in comparision to that of normal diet group. And these values in protein malnutritive diet group were 61.2, 83.6 and 87.0% respectively in comparision to that of normal diet group, and recovered to the level of normal diet group by the supplement of the saponin fraction. The electrophoregram of the serum protein of the mouse fed with protein malnutritive diet was different from that of the mouse fed with normal diet, but this difference practically disappeared by the supplement of the saponin fraction.
This study examined the effect of excess loading of calcium (Ca)and iron(Fe) on the bioavailability of minerals in both normal and Ca-and Fe-deficient rats. Three-week-old male rats were divided into four groups and fed experimental diets for six weeks, containing either normal (0.5%) or high(1.5%) Ca and normal (35ppm) or high (350ppm)Fe. Likewise, three-week-old male rats were first fed a Ca-and Fe-deficient diet for three weeks, and then fed one of four experimental diets for additional three weeks. In both normal and Ca-and Fe-deficient rats, ca contents of serum, liver, kidney and femur were not significantly affected by dietary Ca and Fe levels. Apparent Ca absorption(%) decreased in rats fed a high Ca diet regardless of dietary Fe levels. Magnesium(Mg) contents of serum, liver and femur significantly decreased in rats fed a high Ca diet. Fe contents of serum and liver significantly increased in rats fed a high-Fe diet, but decreased in rats fed a high Ca diet. Fe content of serum and liver significantly increased in rate fed a high-Fe diet, decreased in rats fed a high-Ca diet. Apparent Fe absorption increased in rats fed a high-Fe diet, and decreased in rats fed a high-Ca diet in Ca-and Fe-deficient rats, but dietary Ca did not seem to affect Fe absorption in normal rats. Phosphorus(P) contents of serum and femur were not significantly affected by dietary Ca and Fe levels in both normal and Ca-and Fe-deficient rats. Serum copper(Cu) decreased in rats fed a high-Fe diet, while serum zinc(Zn) decreased in rats fed a high-Ca diet in normal rats. Cu contents of liver, and Zn contents of serum and liver decreased in rats fed a high-Fe diet in Ca-and Fe-deficient rats. There results suggest that a dietary overload of Ca and Fe in both normal and Ca-and Fe-deficient rats may decrease mineral bioavailability leading to potential health problems.
Objectives : This experimental study was designed to investigate the effect of SBY-III extract on the weight, cell size of epididymal fat-pad, fat accumulation area in liver, serum lipid level and UCP1 mRNA in brown adipose tissue of high fat diet-fed obese rats continued by high fat diet and regulated by normal Diet. Methods : The body weight gain, weight of the internal organs(epididymis, liver, brown adipose tissue), insulin, triglyceride, total cholesterol, total lopod, free fatty acid, expression of UCP1 mRNA were measured in high fat diet-fed obese rats continued by high fat diet and regulated by normal diet. The experimental study are divided into exp-I and exp-II. Each study was administered normal diet, high fat diet and SBY-III according to each situation. Normal group is normal diet for 8 weeks. Exp-I are divided into control group(high fat diet for 8 weeks) and sample group(high fat diet for 8 weeks and SBY-III for last 2 weeks). Exp-II are divided into control group(high fat diet for 6 weeks and normal diet for 2 weeks) and sample group(high fat diet for 6 weeks and normal diet with SBY-III for 2 weeks). These were then compared mutually. Results : 1. Irrespective of diet control, sample group taken SBY-III showed the more effective decrease of weight gain than control group and diet control-fed sample group with SBY-III showed the more effective decrease of weight loss including weight gain than control group. 2. Irrespective of diet control, sample group taken SBY-III showed the more effective decrease cell size of epididymal fat-pad, fat accumulation area in liver than control group. 3. Non diet control-fed sample group taken SBY-III showed the more effective decrease of serum triglyceride, total lipid, free fatty acid than control group and diet control-fed sample group taken SBY-III showed the decrease of serum triglyceride, free fatty acid than control group. 4. Only diet control-fed sample group taken SBY-III showed the decrease of UCP1 volume. Conclusions : These results shows that SBY-III has effects on anti-obesity, especially keeping pace with diet control.
The Ca and P balance studies were carried out with two different diets varying Ca levels : the current Korean RDA level(normal Ca diet) and the high level (High Ca diet) which was determined by adding 2SD to mean Ca intake of all subjects. The subjects were sever healthy adult woman, aged from 25 to 32 years old. The metabolic studies were conducted for two weeks with a days recess in between : during the fist week with normal Ca diet and during the second week with high Ca diet. The composition of nutrients excepting Ca of both diets was similar to subjects usual intake. The result were summarized as follows: Mean daily Ca intake was 728.8mg from norma Ca diet and 945.5mg from high Ca diet. Fecal excretion of Ca increased significantly(p<0.05) on high Ca diet, but urinary excretion of Ca did not show any differences between the two diet periods. There were also no significant differences in Ca retention between the two diet periods but it tended to be greater during high Ca diet period: 112.1mg/day during normal Ca diet period vs 208.2mg/day during high Ca diet period. Mean apparent Ca absorption was 41.2% on normal Ca diet and 42.1% on high Ca diet, indicating it was not affected by high Ca intake level used in this study. On the contrary, P retention was significantly increased up to 109.4mg/day with high Ca diet as compared to- 41mg/day with normal Ca diet. There were no significant differences in fecal and urinary excretion of P but those to be lower during high Ca diet period. The above results showed that higher Ca intake more than current RDA(700mg/day), in the level of 945.5mg/day, could increase Ca retention through Ca absorption comparable to the rate appeared on RDA level intake. P retention was also improved by high Ca intake. Therefore, higher Ca intake than the current RDA level seemed to produce favorable effects on bone health in adult women. However, the current RDA level seemed to be relatively appropriate, considering the results that all the subjects but one maintained positive Ca balance with normal Ca diet. The effects of indigestible dextrin on lipids in serum, tissue, and feces were investigated in two experiments carried out with 64 male Sprague-Dawley rats for eight weeks. Experiment I diet(normal fat diet) containing 5% corn oil and experiment II diet(high fat diet) containing 18% fat(13% beef tallow and 5% corn oil) and 1% cholesterol were divided into four groups with 0.5% cellulose(0.5CL : control), 10% cellulose(10CL), 10% Indigestible dextrin I(ID-I) and 10% indigestible dextrin II(ID-II), respectively. ID-I and ID-II diets produced a decrease in serum total lipid and cholesterol concentrations in both normal and high fat diets but more significantly in the groups fed high fat diet. ID-I, ID-II, and 10CL of high fat diet decreased serum glucose concentration. ID-I and ID-II diets also decreased liver cholesterol and triglyceride concentrations in rats fed normal and high fit diets. The fecal excretion of cholesterol in ID-II/high fat diet was significantly higher than that of 0.5CL diet. Bile acid excretion was increased in the rats fed 10CL and ID-II diets of normal and high fat diets. These observations indicat the hypolipidemic and hypocholesterolemic effects of indigestible dextrin on the rats. This study was performed to investigate the effects of liquid culture of Agaricus blazei Murill on the weight gains, food efficiency ratios, serum protein and mineral levels, and serum enzyme activities in growing male rats. Sprague-Dawley rats (7 weeks old) were given four different types of diets for a succeeding period of 6 weeks, respectively: a normal diet group, a control diet group (normal diet + 15% lard + 0.5% cholesterol), a 30% or 40% A. blazei diet groups (control diet + 30% or 40% A. blazei in water) according to the levels of A. blazei supplementation. The body weight gains, food efficiency ratios, and the liver of the rats fed control diet, 30% or 40% A. blazei diets were significantly increased compared to rats fed the normal diet, but those of rats fed the 30% and 40% A. blazei diets were similar to those of rats fed the control diet. The concentrations of total protein, albumin, glucose, and hematocrit value in serum of rats fed the control diet, 30% or 40% A. blazei diets were similar to those of rats fed the normal diet. The concentrations of urea and creatinine in serum of rats fed the 30% or 40% A. blazei diets were similar to those of rats fed the control diet. but the urea of rats fed the 30% and 40% A. blazei diets were significantly decreased compared to rats fed the normal diet. The concentrations of Ca, P, Fe and Mg, and $Ca^{++}$, $K^+$, $Na^+$ and $Cl^-$ of rats fed the control diet, 30% or 40% A. blazei diets were similar to those of rats fed the normal diet. There were no differences in the activities of aspartate aminotransferase, alanine aminotransferase, $\gamma$-glutamyltranspeptidase and alkaline phosphatase in the serum among the experimental groups. In conclusion, the rats fed the A. blazei maintained normal protein and mineral levels, and enzyme activities of serum. But the A. blazei feeding could not decrease the body and liver weights in the rats fed high cholesterol diets.
