Microbial diversity in the soil is responsive to changes in soil composition. However, the impact of soil amendments on the diversity and structure of rare and abundant sub-communities in agricultural systems is poorly understood. We investigated the effects of different Chinese herb residue (CHR) soil amendments and cropping systems on bacterial rare and abundant sub-communities. Our results showed that the bacterial diversity and structure of these sub-communities in soil had a specific distribution under the application of different soil amendments. The CHR soil amendments with high nitrogen and organic matter additives significantly increased the relative abundance and stability of rare taxa, which increased the structural and functional redundancy of soil bacterial communities. Rare and abundant sub-communities also showed different preferences in terms of bacterial community composition, as the former was enriched with Bacteroidetes while the latter had more Alphaproteobacteria and Betaproteobacteria. All applications of soil amendments significantly improved soil quality of newly created farmlands in whole maize cropping system. Rare sub-communitiy genera Niastella and Ohtaekwangia were enriched during the maize cropping process, and Nitrososphaera was enriched under the application of simple amendment group soil. Thus, Chinese medicine residue soil amendments with appropriate additives could affect soil rare and abundant sub-communities and enhance physicochemical properties. These findings suggest that applying soil composite amendments based on CHR in the field could improve soil microbial diversity, microbial redundancy, and soil fertility for sustainable agriculture on the Loess Plateau.
A study was conducted to investigate the effect of ACAMS(Autoclaved-cooled amylomaize VII) and ACNMS(Autoclaved-cooled normal maize starch) containing resistant starch(RS) on ASW (Australian wheat flour) rheology and noodle quality. The water absorption in farinograph increased with the addition of ACAMS and ACNMS, but the dough stability decreased with the addition. The ACNMS added flours showed the highest initial pasting temperature and the lowest peak viscosity in RVA. The addition of ACAMS and ACNMS were not effective on the weight and volume of cooked noodles during cooking time for 5 min. However, as the cooking time increased, noodle weight and volume were the highest in control(no RS added flour) and the lowest in ACNMS added flours. Noodle texture was evaluated using rheometer. The hardness of RS(ACAMS, ACNMS) added noodles was higher than that of control. Cohesiveness was significantly different between control and ACAMS added noodles, but the cohesiveness of ACNMS added noodles was similar to other noodles. The elasticity of ACNMS added noodles in sensory test was lower than that of control but the smoothness and overall acceptibility were higher.
The effects of high pressure homogenization (microfluidization) on physicochemical properties of normal maize and oxidized maize starch film were studied. The molecular dispersibility of amlyose and amylopectin and the disintegration of granular structure had a marked effect on the physicochemical properties of starch films. The high pressure homogenized starch films showed increased solubility and transmittance due to the absence of gelatinized starch granules. The tensile strength of starch film increased significantly with decreasing oxygen permeability after high pressure homogenization, indicating that starch molecules were more uniformly and fully dispersed during the film formation. As a result, a clear starch film with improved mechanical properties was obtained after high pressure homogenization due to the increased interactions between the uniformly dispersed starch molecules.
Effects of native and RS-3 type resistant starches prepared from autoclaved-cooled amylomaize VII(AVII) and normal maize starches(NMS) on the rheological and baking properties of wheat flour dough and quality of breads were investigated. In farinogram, water absorption and dough development time were increased, but stability was reduced by the addition of RS. The addition of native starches or resistant starch made from AVII to wheat flour improved the total volume and specific loaf volume of bread compared with the control(no addition), but the addition of NMS-RS reduced those. During the storage of bread at room temperature, the moisture content of control was decreased but those of native starch- or RS-added breads remained constantly. AVII-RS- or NMS-RS-added bread was evaluated to have good overall acceptability compared with control by elementary school students. The addition of enzyme-resistant starch to bread regardless of botanical sources of starch not only improved the overall acceptability and nutritional benefits but also improved the sensory acceptability.
Aleurone layers of normal and vp1 mutant maize kernels were extracted and centrifuged at 100,000g to yield a cytosol fraction. Binding of [3H]ABA cis, trans (+)ABA to a soluble macromolecular components present in the cytosol was demonstrated by Sephadex chromatography and non-denaturing PAGE. The binding component was of high molecular weight and seems to be an aggregate of proteins. A rapid DEAE-cellulose filter method for assaying bound [3H]ABA to a soluble protein was adapted. Binding assays were performed with cytosol that had been preheated or incubated with several enzymes, indicating that heat and protease treatments disrupted the binding. This suggested that binding occurred to proteins. Some properties of the ABA binding proteins were described. The [3H]ABA binding were reduced dramatically when unlabeled ABA was added as a competitor, suggesting a specific binding of [3H]ABA. Gel filtration profiles and autoradiogram of [3H]ABA binding showed no difference in the binding components of Vp1 and vp1/vp1 mutant cytosol, indicating that Vp1 protein is not a sole ABA binding protein.
The R-r:standard (R-r:std) allele of maize R gene complex consists of S subcomplex and P component; the S subcomplex regulates anthocyanin pigmentation of seed aleurone layer, and the P component confers pigmentation of the other plant parts. The S subcomplex contains two functional genes, S1 and S2 components. In the presence of Pl gene some alleles of R gene induce anthocyanin pigmentation of pericarp. In the present study, the effects of different R alleles on the anthocyanin pigmentation of pericarp in the presence of Pl gene were analyzed in order to identify the R gene component responsible for pericarp pigmentation. The results show that R-ch and r-ch alleles condition similar degrees of pericarp pigmentation, and that R-r:Ecuador (R-r:Ec) conditions stronger pigmentation. The r-ch allele, which is inferred that its S subcomplex has lost function but the P component is normal, induces pericarp pigmentation in the presence of Pl gene. On the contrary, the R-g:g1111 allele, derived from R-r:Ec and inferred that its S subcomplex functions normal but the P component has lost its function, did not induce pericarp pigmentation in the presence of Pl gene. Moreover, PCR analysis of genomic DNA's of R-ch and r-ch indicate that R-ch maintains both P and S1 components, whereas r-ch lacks for the S1 component. Taken together, The results suggest that the P components of R alleles inducing pericarp pigmentation in the presence of Pl gene are responsible for pericarp pigmentation.
In Korea, production of super sweet corn has been economically feasible and is substituting for traditional sweet corn due to better flavor in recent years. Major limiting factors for super sweet corn production are low field emergence and low seedling vigor. The optimum water potential (WP) for the priming of normal and aged seeds of dent, sweet (su) and super sweet (sh2) corns was studied to improve low seed quality. Seeds were primed at 0, -0.3, -0.6, -0.9, and -1.2 MPa of polyethylene glycol (PEG) 8000 solution at $15^{\circ}C$ for 2 days. Priming effects differed depending on the type of corn, seed quality, and WP of PEG solution. Although WP of priming solution did not influence the emergence rate of extremely high quality normal dent corn seeds, it reduced time to $50\%$ emergence (T50) and increased plumule weight. In contrast, the emergence rate of aged field corn was improved by seed priming at 0 MPa and plumule weight and $\alpha-amylase$ activity was enhanced. The optimum WP for both normal and aged sweet and super sweet corn seeds was between -0.3 and -0.6 Mpa. At the optimum WP emergence rate, $\alpha-amylase$ activity, and content of DNA and soluble protein increased, while T50 and leakage of total sugars and electrolytes reduced.

The objective of this study was conducted to calculate the damage of whole crop maize in accordance with abnormal climate using the forage yield prediction model through machine learning. The forage yield prediction model was developed through 8 machine learning by processing after collecting whole crop maize and climate data, and the experimental area was selected as Gyeonggi-do. The forage yield prediction model was developed using the DeepCrossing (R2=0.5442, RMSE=0.1769) technique of the highest accuracy among machine learning techniques. The damage was calculated as the difference between the predicted dry matter yield of normal and abnormal climate. In normal climate, the predicted dry matter yield varies depending on the region, it was found in the range of 15,003~17,517 kg/ha. In abnormal temperature, precipitation, and wind speed, the predicted dry matter yield differed according to region and abnormal climate level, and ranged from 14,947 to 17,571, 14,986 to 17,525, and 14,920 to 17,557 kg/ha, respectively. In abnormal temperature, precipitation, and wind speed, the damage was in the range of -68 to 89 kg/ha, -17 to 17 kg/ha, and -112 to 121 kg/ha, respectively, which could not be judged as damage. In order to accurately calculate the damage of whole crop maize need to increase the number of abnormal climate data used in the forage yield prediction model.
Glycine betaine has been reported as an osmoprotectant compound conferring tolerance to salinity and osmotic stresses in plants. We previously found that the expression of betaine aldehyde dehydrogenase 1 gene (OsBADH1), encoding a key enzyme for glycine betaine biosynthesis pathway, showed close correlation with salt tolerance of rice. In this study, the expression of the OsBADH1 gene in transgenic tobacco was investigated in response to salt stress using a transgenic approach. Transgenic tobacco plants expressing the OsBADH1 gene were generated under the control of a promoter from the maize ubiquitin gene. Three homozygous lines of $T_2$ progenies with single transgene insert were chosen for gene expression analysis. RT-PCR and western blot analysis results indicated that the OsBADH1 gene was effectively expressed in transgenic tobacco leading to the accumulation of glycine betaine. Transgenic lines demonstrated normal seed germination and morphology, and normal growth rates of seedlings under salt stress conditions. These results suggest that the OsBADH1 gene could be an excellent candidate for producing plants with osmotic stress tolerance.
To explore the possibility of introducing Zea mays transposable element Ac(activator) which can be used as a mutagen and gene tag in tobacco plants other than maitre, we tried to introduce a cloned Ac element into tobacco cells by an Agrobacterium tumefaciens binary vector system. Transformation of N. babacum cv. Burley 21 tissues and regeneration to whole plant were carried out. The frequency of the transformed callus induced in shoot induction media was higher than that of transformed callus induced in callus induction media. However, the calli were not grown in the second selection media, and became yellow senescent calli. Regenerated tobacco plantlets with foreign gene were also obtained in shoot induction media containing 100 $\mu\textrm{g}$/ml kanamycin and 100$\mu\textrm{g}$/ml carbenicillin. The leaf tissues of transformant was also resistant to 1000 $\mu\textrm{g}$/ml kanamycin. The chromosomal DNAs of transformant and normal plant of N. tabacum were digested by EcoR I and Hind III but not by Pst I.
