Determination of Octylphenol, Nonylphenol, and Di(2-ethylhexyl)phthalate in water samples (물 시료 중 Octylphenol, Nonylphenol, Di(2-ethylhexyl)phthalate의 연구)

  • Determination were performed for the presence of three common environmental endocrine disrupting compounds (EDC), namely nonylphenol (NP), octylphenol (OP), di(2-ethylhexyl)phthalate (DEHP) in rainwater, distilled water, millipore water, groundwater, tap water, and bottle water in market, using OASIS cartrige extraction and GC/MS-SIM. Octylphenol was not detected in any water samples. The order of nonylphenol concentrations were increased in rainwater < millipore water < distilled water < groundwater < tap water < bottle water. The concentration of nonylphenol in bolltle water was $0.44{\mu}g/L$(ppb) at highest. Di(2-ethylhexyl)phthalates were detected in every water samples, and the concentrations were to be $1.70{\sim}2.92{\mu}g/L$ in rainwater, $8.7{\sim}31.7{\mu}g/L$ in distilled water, $0.6{\sim}5.6{\mu}g/L$(mean: $2.5{\pm}0.3{\mu}g/L$) in millipore water, and that were very stable values than other waters, $1.1{\sim}6.0{\mu}g/L$ in groundwater at near Jeonju, $3.1{\sim}5.8{\mu}g/L$ by means in tap water, $0.5{\sim}67.6{\mu}g/L$ in bottle water from market. The concentration of DEHP in bottle water samples from market were in higher than USEPA regulation at over 40%.

  • 4-nonylphenol (4-NP)이 해산어류인 조피볼락, Sebastes schlegeli의 혈장 vitellogenin (VTG), alkaline-labile protein phosphorus (ALPP), calcium(Ca), glutamate pyruvate transaminase (GPT) 및 hepatosomatic index (HSI)에 미치는 영향을 조사하였다. 실험어에 3일간격으로 $estradiol-17{\beta}$ ($E_2$, 5 mg/kg B.W.) 또는 4-NP(0, 10, 50, 100및 200 mg/kg B.W.)을 복강에 2번 주사한 후, 7일째에 채혈과 적출을 통해 혈장과 간장을 수집해 분석이 실시되었다. 대조 실험어에는 용매로 사용된 70% 에탄올만이 투여되었다. $E_2$ 투여 실험어의 혈장 단백질을 전기 영동상으로 분석한 결과 약 170 kDa의 위치에서 짙은 VTG 밴드가 관찰되었으나, 용매만 투여한 대조 실험어의 혈장에서는 동일 밴드가 관찰되지 않았다. 4-NP 투여한 모든 실험어의 혈장 단백질에서는 $E_2$ 투여 실험어와 동일한 VTG 밴드가 관찰되었다. 혈장 ALPP와 Ca 농도도 4-NP 투여 실험어에서 $E_2$ 투여 실험어와 유사하게 증가하였으며, 이들 농도 변화는 VTG 합성과 더불어 증가하는 경향을 나타냈다. 혈장 전위효소인 GPT와 HSI도 $E_2$ 투여 실험어와 유사하게 4-NP가 투여된 모든 실험어에서 급격히 증가하였다. 이상의 결과로부터 연안생태계 내에서 서식하는 어류가 4-NP과 같은 내분비 장애물질(Endocrine Disrupting compounds, EDCs)에 의해 영향을 받는지를 규명하기 위한 생물학적 지표로서 VTG와 더불어 혈장 ALPP와 Ca이 사용가능 할 것으로 판단된다. 또한, 조피볼락과 같은 해산어가 EDCs에 노출되어 VTG가 합성될 때 간장 기능의 손상으로 혈장 전위효소인 GPT가 일시적으로 종가하고 간장도 비대해져 HSI가 높아지는 것으로 판단된다.

Determination of alkylphenol ethoxylate in water by high performance liquid chromatography/electrospray ionization/mass spectrometry (HPLC/ESI/MS를 이용한 물 중의 알킬페놀에톡실레이트 분석)

  • A method is described for the analysis of short-chain alkylphenol ethoxylates (APEOs), 4-octylphenol-di-ethoxylate (OP2EO) and 4-nonylphenol-di-ethoxylate (NP2EO), in drinking water or wastewater using reversed phase high-performance liquid chromatography with electrospray ionization mass spectrometry. The solvent system was water and methanol containing $10{\mu}M$ trifluoroacetic acid as an ionization solvent. We acidified 1 L of water samples to less than pH 2 with concentrated $H_2SO_4$ and loaded onto Sep-Pak $C_{18}$, and eluted with acetone. The calibration of OP2EO and NP2EO was performed for the concentration range from 20 to 500 ng/L and the correlation coefficients were 0.999 and 0.990, respectively. The limits of detection were 20 ng/L (OP2EO) and 50 ng/L (NP2EO) at a signal-to-noise ratio of 3. Accuracy and precision of this analytical method were 85.8 ~ 122.1% and 8.2 ~ 18.8%, respectively. The proposed method allowed a sensitive and rapid detection of OP2EO and NP2EO and it could be applied for monitoring of APEOs from environmental samples.

Effects of 4-Nonlyphenol Exposure on $P.$ $olivaceus$ and $S.$ $schlegeli$ Vitellogenesis

  • The effects of the estrogenic compound 4-nonylphenol (4-NP) on vitellogenesis in juvenile olive flounder ($Paralichthys$ $olivaceus$) and rockfish ($Sebastes$ $schlegeli$) exposed continuously at 10, 50 and $100{\mu}g{\ell}^{-1}$ levels for 7 days were compared. The expression of VTG mRNA level and protein using specific probes were examined. The levels of plasma estradiol-17 ${\beta}(E_2)$ and testosterone (T) were assessed by radioimmunoassay (RIA). Plasma $E_2$ concentrations increased significantly in two female fish species exposed to $100{\mu}g{\ell}^{-1}$ of 4-NP over concentrations in control fish. Plasma T concentrations increased in $P.$ $olivaceus$. Four days after exposure, the level of VTG mRNA expression increased in $P.$ $olivaceus$ and $S.$ $schlegeli$ exposed to $20{\mu}g{\ell}^{-1}$ of 4-NP. In addition, plasma VTG protein expression was seen in $P.$ $olivaceus$ and $S.$ $schlegeli$. In $S.$ $schlegeli$ and $P.$ $olivaceus$ exposed to 4-NP, the changes were noticed mainly in hepatocytic vaculation after 7 days of exposure. Thus, 4-NP may disrupt vitellogenesis in immature fish both directly and indirectly via disrupted steroidogenesis and liver pathology. Immature $S.$ $schlegeli$ were the most sensitive to 4-NP exposure in vitellogenesis.

Chemical Contamination and Toxicity of Sediments from the Gunsan Coast, Korea

  • Polycyclic aromatic hydrocarbons (PAHs), butyltins (BTs), nonylphenol (NP), and fecal sterols concentrations in sediments were investigated from Gunsan coast of Korea to evaluate organic pollution from anthropogenic activities. Sediment toxicity was also examined by bacterial bioluminescence toxicity test (Vibrio fischeri). The concentrations of 16 PAHs in sediments ranged from 67.9 to 425 ng/g dry wt; BTs ranged from 2.79 to 14.1 ng Sn/g dry wt; NP ranged from 20.7 to 2171 ng/g dry wt; and coprostanol, a fecal sterol, ranged from 7.60 to 245 ng/g dry wt. Effective concentration 50% ($EC_{50}$) of sediments ranged from 0.38 to 23.8 mg/mL. Most of the chemicals were present at levels lower than or comparable to the previously reported values from Korea. However, NP levels in the present study were in the high range of levels reported from the Korean coast, and 40% of the measured samples exceeded screening and ecotoxicological values of NP suggested by the Netherlands and Canada. This suggests that an ongoing source of NP is a serious concern in the Gunsan coast. High levels of contaminants were found in the proximity of potential sources, such as the outfall of a wastewater treatment plant for NP, an anthracite-fired power plant for PAHs, and ports for BTs, fecal sterols, and sediment toxicity. This indicates that Gunsan coast has various potential sources of marine sediment contaminants.

  • Objectives Heat shock protein 90 (HSP90) is a highly conserved molecular chaperone important in the maturation of a broad spectrum of protein. In this study, an HSP90 gene was isolated from Asian paddle crab, Charybdis japonica, as a bio-indicator to monitor the marine ecosystem. Methods This work reports the responses of C. japonica HSP90 mRNA expression to cellular stress by endocrine disrupting chemicals (EDCs), such as bisphenol A (BPA) and 4-nonylphenol (NP) using real-time. reverse transcription polymerase chain reaction. Results The deduced amino acid sequence of HSP90 from C. japonica shared a high degree of homology with their homologues in other species. In a phylogenetic analysis, C. japonica HSP90 is evolutionally related with an ortholog of the other crustacean species. The expression of HSP90 gene was almost distributed in all the examined tissues of the C. japonica crab but expression levels varied among the different body parts of the crabs. We examined HSP90 mRNA expression pattern in C. japonica crabs exposed to EDCs for various exposure times. The expression of HSP90 transcripts was significantly increased in C. japonica crabs exposed to BPA and NP at different concentrations for 12, 24, 48 and 96 hours. The mRNA expression of HSP90 gene was significantly induced in a concentration- and time-dependent manner after BPA or NP exposures for 96 hours. Conclusions Taken together, expression analysis of Asian paddle crab HSP90 gene provided useful molecular information about crab responses in stress conditions and potential ways to monitor the EDCs stressors in marine environments.

Effects of Nonylphenol and 2,2', 4,6,6'-pentachlorobiphenyl on in vitro Sex Steroid Production in Maturing Oocytes of the Yellowfin Goby, Acanthogobius flavimanus

  • Several studies have reported that nonylphenol (NP) and 2,2', 4,6,6'-pentachlorobiphenyl (PCB104) exhibit estrogenic activity. To investigate the estrogenic potency of NP and PCB104 during oocyte maturation, fully vitellogenic oocytes (0.76 mm diameter in average) of yellow fin goby, Acanthogobius flavimanus, were exposed in vitro to these chemicals at different concentrations (0.1, 1, 10, 100, and 1,000 ng/mL) with the exogenous precursor $17\alpha$-hydroxyprogesterone ($17{\alpha}OHP$) 50 ng/mL in the presence or absence of human chorionic gonadotropin (HCG). The production of testosterone (T), estradiol-$17\beta$ (E2), and $17\alpha,20\beta$-dihydroxy-4-pregnen-3-one ($17\alpha20{\beta}OHP$) in response to NP or PCB104 were measured by radioimmunoassay. Steroid levels were also expressed as E2/T and E2/$17\alpha20{\beta}OHP$ ratios. In the absence of HCG, no significant differences in either NP or PCB104 treatment groups were observed. In the presence of HCG, NP treatment did not show significant differences in the production of T, E2, and $17\alpha20{\beta}OHP$ at any concentrations tested, but E2/T ratios were decreased at concentrations of 0.1, 1, 10, and 1,000 ng/mL compared with the control group. PCB104 decreased E2 production at concentrations of 0.1, 10, and 1000 ng/mL, but did not show significant differences in the production of T and $17\alpha20{\beta}OHP$ at any concentration tested. While E2/T ratios were decreased at PCB104 concentrations of 0.1, 1, 10, and 1,000 ng/mL, E2/$17\alpha20{\beta}OHP$ ratios were also decreased at 0.1, 10, and 1,000 ng/mL compared with the control. Results indicate that both NP and PCB104 appeared to have antiestrogenic effects during this phase.

Survival Rate, Growth and NP Accumulation of the Striped Bitterling, Acheilognathus yamatsutae Long-term Exposed to Nonylphenol (NP) (노닐페놀 (NP)에 장기간 노출된 줄납자루, Acheilognathus yamatsutae의 NP 체내 축적, 성장 및 생존율)

  • Striped bitterling, Acheilognathus yamatsutae (TL: 4.25 ㎝, TW: 0.73 g)were exposed to 0.3, 1.3, 17.6 and 33.7 ㎍ L-1 (nominal concentrations) of nonylphenol (NP) for a period of 20 weeks. We studied on accumulation, growth and survival rate. After 20 weeks of exposure, the accumulation of NP in the body of the female and male was increased with increasing dose-dependent and female was higher than male. 4 weeks of exposure, TL of female striped bitterling was found to be increased in all of the dose group compared to control and 12 weeks of exposure, similar or some decreased in all of the dose group compared to control. 20 weeks of exposure, increased in all of the dose group compared to control and TL at 33.7 ㎍ NP L-1 were significantly higher than those in control (p <0.05). 4 weeks of exposure, TL and TW of male striped bitterling was found to be increased in all of the dose group compared to control and significantly higher in TL of 33.7 ㎍ NP L-1 (p <0.05). 12 and 20 weeks of exposure, TL and TW decreased in all of the dose group compared to control and significantly lower in 1.3 ㎍ NP L-1 of 12 weeks. Survival rate in both groups decreased with increasing NP concentration and there was a significant difference between control group and experimental groups exposed to each NP concentration.

Chronic and Low Dose Exposure to Nonlyphenol or Di(2-Ethylhexyl) Phthalate Alters Cell Proliferation and the Localization of Steroid Hormone Receptors in Uterine Endometria in Mice

  • Based on our preliminary results, we examined the possible role of low-dose and chronic-exposing of the chemicals those are known as endocrine disrupting chemical (EDC), on the proliferation of uterine endometrium and the localization of steroid receptors. Immunohistochemical or immunofluorochemical methodology were employed to evaluate the localization of antigen identified by monoclonal antibody Ki 67 protein (MKI67), estrogen receptor 1 (ESR1), estrogen receptor 2 (ESR2), and progesterone receptor (PGR). In $133{\mu}g/L$ and $1,330{\mu}g/L$ di(2-ethylhexyl) phthalate (DEHP) and $50{\mu}g/L$ nonylphenol (NP) groups, the ratio of MKI67 positive stromal cells was significantly increased but not in $500{\mu}g/L$ NP group. The ratios of MKI67 positive glandular and luminal epithelial cells were also changed by the chronic administration of NP and DEHP in tissue with dose specific manner. ESR1 signals were localized in nucleus in glandular and luminal epithelia of control group but its localization was mainly in cytoplasm in DEHP and NP administered groups. On the other hand, it was decreased at nucleus of stromal cells in $1,330{\mu}g/L$ DEHP group. The colocalization patterns of these nuclear receptors were also modified by the administration of these chemicals. Such a tissue specific and dose specific localization of ESR2 and PGR were detected as ESR1 in all the uterine endometrial tissues. These results show that the chronic lows-dose exposing of NP or DEHP modify the localization and colocalization of ESRs and PGR, and of the proliferation patterns of the endometrial tissues.

These phenolic compounds are used in a number of commercial products and have been reported to be weakly estrogenic in previous studies.
