Influence of Morinda citrifolia (Noni) on Expression of DNA Repair Genes in Cervical Cancer Cells

  • Background: Previous studies have suggested that Morinda citrifolia (Noni) has potential to reduce cancer risk. Objective: The purpose of this study was to investigate the effect of Noni, cisplatin, and their combination on DNA repair genes in the SiHa cervical cancer cell line. Materials and Methods: SiHa cells were cultured and treated with 10% Noni, $10{\mu}g/dl$ cisplatin or their combination for 24 hours. Post culturing, the cells were pelleted, RNA extracted, and processed for investigating DNA repair genes by real time PCR. Results: The expression of nucleotide excision repair genes ERCC1, ERCC2, and ERCC4 and base excision repair gene XRCC1 was increased 4 fold, 8.9 fold, 4 fold, and 5.5 fold, respectively, on treatment with Noni as compared to untreated controls (p<0.05). In contrast, expression was found to be decreased 22 fold, 13 fold, 16 fold, and 23 fold on treatment with cisplatin (p<0.05). However, the combination of Noni and cisplatin led to an increase of 2 fold, 1.6 fold, 3 fold, 1.2 fold, respectively (p<0.05). Conclusions: Noni enhanced the expression of DNA repair genes by itself and in combination with cisplatin. However, high expression of DNA repair genes at mRNA level only signifies efficient DNA transcription of the above mentioned genes; further investigations are needed to evaluate the DNA repair protein expression.

  • To determine the optimal mixing conditions of Noni jelly, samples were prepared with various compounding ratios of Noni juice(120, 160, 200, 240 and 280 g), gelatin(12, 16, 20, 24 and 28 g) and sucrose(80, 100, 120, 140 and 160 g) using a central composite design. Physical and sensory evaluations were performed and the results analyzed using response surface methodology. The optimum mixing rate satisfying sensory items was Noni juice 192 g, gelatin 18.25 g and sucrose 135 g.

Antioxidant and α-Amylase Inhibitory Activity of 70% Ethanolic Extract from Morinda citrifolia L. (Noni) (노니가루 70% 에탄올 추출물의 항산화 및 α-Amylase Inhibitory 활성)

  • In this study, polyphenol and flavonoid contents were measured, and DPPH, OH, H2O2 radical scavenging activity, and the α-amylase inhibitory activity were measured to study the antioxidant activity of 70% ethanol extract from Morinda citrifolia L. The polyphenol and flavonoid contents of noni 70% ethanol extract were 29.52 GAE/g and 12.48 CE/g, respectively. Also, the IC50 values of DPPH, hydroxyl radical, and hydrogen peroxide scavenging activity of 70% ethanol extract from noni were 18.70 mg/mL, 26.45 mg/mL, and 35.67 mg/mL, respectively. Measurement of the α-amylase inhibitory activity of 70% ethanol extract from noni showed 45% inhibitory activity at 10 mg/mL.

Physicochemical properties, bioactive composition and antioxidant activities of noni fruit juices from different regions of cultivation (재배지에 따른 노니열매 착즙액의 이화학적 특성, 생리활성 성분 및 항산화 활성)

  • The objectives of this study were to compare the physicochemical properties, functional components and antioxidant activities of noni fruit juices from six different regions of cultivation: Samoa, Indonesia, China (Hainan), Hawaii, Thailand and Tahiti. The pH values of noni fruit juices ranged from 3.63 to 3.83, and the soluble contents were $5.97-6.97^{\circ}Brix$. In regard to color, the L, a and b values of noni juices were in the ranges of 33.41-46.51, 3.44-7.98, and 2.42-22.20, respectively. The polyphenol content of noni fruit juice from Indonesia was significantly higher than other samples. The Thailand noni fruit juice contained the highest amounts of scopoletin (8.62 mg/100 mL) and rutin (2.03 mg/100 mL). The noni fruit juice from Indonesia showed the higher antioxidant activities (DPPH and ABTS radical scavenging activity) than others. In conclusion, These results suggest that noni fruit juice may be a good source of functional food with antioxidant activity and can serve as the basis data for the use of noni fruit in the food industry.

Extraction Procedures for Free Radical Scavenging Activity from Noni Fruit (Morinda citrifolia) (노니 과실에 함유된 항산화물의 추출 공정)

  • In this study, we investigated maximum yield of radical scavenging activity (RSA) using each of various extraction solvents and procedure from noni friut (Morinda citrifolia). Noni fruits were either sun-dried (FS) or hot-air dried (FO) at $60^{\circ}C$ after steam blanching. For optimum aqueous extraction, noni fruits should be sun dried and autoclave extraction time should not be over 30 min to produce extraction with high RSA with low cost. In case of 50% ethanol extraction, reflux extracts of FS and FO resulted in $IC_{50}$ of 1.92 mg/mL and 3.06 mg/mL at 8 hr. When $IC_{50}$ values were lower than 5 mg/mL, coefficient of correlation was 0.71 indicating that 71% of the phenolic antioxidants in noni fruits were accounted for the activity by scavenging free DPPH. However, coefficient of correlation significantly decreased to 0.63 over $IC_{50}$ values of 5 mg/mL. Autoclave extraction contained chlorogenic acid of $14.69 \;{\mu}g/mL$ and scopoletin of $3.86 \;{\mu}g/mL$. Reflux extraction showed all three compounds, chlorogenic acid ($26.19 \;{\mu}g/mL$), quercetin ($19.59 \;{\mu}g/mL$), and scopoletin ($17.4 \;{\mu}g/mL$). Therefore, the result of this study indicated that the potential antioxidant activities and functional values were obtained significantly with reflux extraction from noni fruit.

Formulation and Quality Characteristics of Noni Beverages Mixed with Red Ginseng, Rubus Coreanus and Pomegranate Extracts (홍삼, 복분자, 석류를 첨가한 노니 혼합음료의 개발과 품질특성)

  • The purpose of this study was to investigate the physicochemical and sensory characteristics of Noni beverages added with 10% of water(Noni 90), 1% of red ginseng(NR), 4% of rubus coreanus extract(NR), 5% of pomegranate extract(NP), and 4% of rubus coreanus extract and 5% of pomegranate extract(NPR). Calories, protein and total fiber contents were increased in NR, NP and NPR. Total sugar was significantly increased in NG and NPR. Fat, cholesterol and saturated fatty acid were not detected. Ca and Na contents were increased in NG while Fe content was increased in NG, NR, and NPR pH was significantly increased in NG and NP. Sugar degree was significantly increased in NP and NPR. L and b values were decreased in NG, NP, and NR and a value was increased in NG and NR General bacteria count was 0/mL, and Coliform, E. Coli and 0157:H7 were gram(-) in all beverages. In sensory evaluation, color was increased in NPR while taste and overall quality were increased in NG, NR and NP. Sour taste, sweet taste, and flavor were not different between Noni and the mixed beverages. Texture was significantly increased in NP and NPR. Therefore, mixed Noni beverages added with red ginseng, rubus coreanus extract, and pomegranate extract were better than Noni beverage.

  • Morinda citrifolia, commercially known as Noni, is a tree that grows widely throughout the Pacific, and is recognized as one of the most significant sources of traditional medicines among Pacific Island societies. All parts of the plant have traditional and/or modern uses, including roots and bark(dyes, medicine), trunks(firewood, tools), and leaves and fruits(food, medicine). The bark and the roots of the tree contain red and yellow pigments respectively, which are both used in the manufacture of dyes. Dyes from Noni are being used traditionally, to color clothing and fabrics. In this study, mordanting effects on the dyeing properties of Noni root extract on silk fabrics were investigated.

Comparison of bioactive composition, antioxidant activity, and nitric oxide inhibition effect of enzyme-treated and commercial noni juice (효소처리 노니주스와 시판 노니주스의 기능성 성분, 산화방지 활성 및 NO 생성 억제 효과)

  • This study was conducted to compare the functional component content, antioxidant activity, and nitric oxide (NO) inhibition effect of non-enzyme treated, enzyme-treated, and commercial noni juice samples. Enzyme-treated noni juice samples were prepared by cellulase (0.1%) and pectinase (0.1%) treatment of unripe fruits (EUN) and ripe fruits (ERN). Total polyphenol content of the noni juice samples ranged between 0.36 and 1.61 mg/mL. The EUN had the highest total polyphenol content (1.61 mg/mL), while the ERN had the highest scopoletin content (123.88 ㎍/mL) and the highest antioxidant activity. Nitric oxide levels in ERN and NJ5 were lower (55.95 and 60.14%, respectively) than those in other samples. Based on these observations, it was confirmed that enzyme treatment could improve the functional component content and physiological activity of noni juice. In particular, EUN prepared using the ripened noni fruit is expected to have greater utilization value due to its enhanced functionality.

Anti-wrinkle Effect of Morinda citrifolia (Noni) Extracts (노니 추출물의 주름개선 효과연구)

  • Wrinkle formation is mainly attributed to the environmental factors such as UV rays, air pollution, smoking and stress etc. Especially, UV rays induce premature skin aging which is characterized by deep wrinkle, leathery dryness etc. Recently, researches on the wrinkle formation and its prevention have been the main theme in cosmetics fields. We have studied the various plant extracts having anti-wrinkle effects and finally showed that Noni (Morinda citrifolia) extracts have the efficacy of promoting the type I collagen synthesis in normal human fibroblast, using PICP assay. We purified one active compound from Noni extracts and identified its structure. It was identified as 6,7-Dimethoxy-2H-1-Benzopyran-2-one; scopoletin by $^1H-NMR,\;^{13}C-NMR,$ IR, Mass analysis. Scopoletin increased collagen synthesis in a dose dependent manner (89.5% at $0.2{\mu}g/mL$). In order to verify the anti-aging effectiveness of the cream containing 3% noni extracsts, we performed the in vivo test with some female volunteers for 12 weeks. It reduced the signs of aging, especially face wrinkles. From these results, we conclude that the noni extracts could be used as an useful anti-wrinkle agent.

Fundamental Economic Feasibility Analysis on the Transition of Production Structure for a Forest Village in LAO PDR (라오스 산촌마을의 생산구조전환을 위한 투자 경제성 기초 분석)

  • This study analyzes the economic feasibility on the transition of production structure to increase income for a local forest village in Laos PDR. The study area was the Nongboua village in Sangthong district where the primary product is rice from rice paddy. Possible strategies were considered to increase the villagers' revenue, and Noni (Morinda citrifolia) was production in the short-term. We assumed that the project period was for 20 years for the analysis, and a total of 1,100 Noni tree was planted in 1 ha by $3m{\times}3m$ spacing. This study classified basic scenario one, scenario two, scenario three by the survival rate and purchase pirce of Noni. Generally Noni grows well. However, the seedlings' average survival rate (= production volume) was set up conservatively in this study to consider potential risks such as no production experience of Noni and tree disease. The scenario one assumed that the survival rate of Noni seedlings was 50% for 0-1 years, 60% for 0-2 years, and 70% for 3-20 years; the scenario two, 10% less, i.e., 40%, 50%, and 60%; and the scenario three, 10% less, i.e., 40%, 50%, 60% and purchase price 10% less, i.e., $0.29 to $0.26, respectively. Our analysis showed that all 3 scenarios resulted in economically-feasible IRR (internal rate of return) of 24.81%, 19.02%, and 16.30% of with a discounting rate of 10%. The B/C (benefit/cost) ratio for a unit area (1ha) was also analyzed for the three scenarios with a discounting rate of 10%, resutling in the B/C ratio of 1.71, 1.47, and 1.31. The study results showed that the Nongboua village would have a good opportunity to improve its low-income structure through planting and managing alternative crops such as Noni. Also the results can be used as useful decision-making information at a preliminary analysis level for planning other government and public investment projects for the Nonboua village.
