Extraction and Characteristics of Purple Sweet Potato Pigment (자색고구마 색소의 추출과 특성)

  • Studies on extraction and color characteristics of purple sweet potato (PSP) pigment were performed to provide the basic information for the utilization of PSP as a new source of natural food colorant. PSP pigment was extracted well with the polar solvents such as distilled water, ethanol, and methanol. but hardly extracted with the non-polar solvents. Among the tested solvents, 20% ethanol solution containing 0.1% citric acid was found to be the most efficient for extraction of the pigment from PSP. PSP contained high amount of pigment not only in the epidermis but also in the flesh of the potato. The PSP pigment was heat stable even under pretreatments such as autoclaving and blanching of the potato before extraction. The optimum temperature of the extraction for the PSP Pigment was decided to be $30^{\circ}C$ by considering the stability and the rate of extraction. The pigment was markedly influenced by the change of pH. The color of the pigment solution was red at the pH range of $1.0{\sim}3.0$, became blue at $7.0{\sim}8.0$, then turned green at $9.0{\sim}10.0$. A characteristic batho-chromic shift of the pigment solution was observed as the pH of the solution increased.

A Study of Improvement of Skin Condition and Sensory Efficacy by Periodic Application of L-α-Amino Acid (L-α-아미노산의 주기적 도포에 의한 피부개선 효능과 소비자 체감 효능 연구)

  • In this study, $L-{\alpha}-amino$ acid, which is a constituent of natural moisturizing factor, was applied to skin in periodic cycle to improve skin tone and texture roughness. Based on the polarity of the alkyl group (R) of the $L-{\alpha}-amino$ acid, the acid was categorized into two groups and their efficacy was studied. As a result, it was found that the improvement rate of $L-{\alpha}-amino$ acid complex with polar alkyl group ($L-{\alpha}-AAC-1$) is 21% higher than that of $L-{\alpha}-amino$ acid complex with non-polar alkyl group ($L-{\alpha}-AAC-2$). For clinical trials, emulsions containing $L-{\alpha}-amino$ acid complex ($L-{\alpha}-AAC-1$) were applied to the randomly selected 20 to 40 year old female participants, as an experimental group, on the right facial cheek once per day for 8 weeks, and emulsions without $L-{\alpha}-amino$ acid complex, as a control group, were applied to the left facial cheek in the same way. Improvements in skin tone were measured using $JANUS^{(R)}$ equipment and analyzed using image analysis software. Skin texture improvement was measured and analyzed mechanically using the phaseshift rapid in-vivo measurement of the skin (PRIMOS) equipment. As a result, improvements of skin tone and skin texture were 11.7% and 6.7%, respectively. In addition, a questionnaire survey was conducted to the participants on the aesthetic improvement and the degree of the feeling of skin improvement. The results suggest that $L-{\alpha}-amino$ acid can be used as a cosmetic substance that can provide aesthetic satisfaction through physiological skin tone and texture roughness.

Cloning and Expression of the Cathepsin F-like Cysteine Protease Gene in Escherichia coli and Its Characterization

  • In this study, we have cloned a novel cDNA encoding for a papain-family cysteine protease from the Uni-ZAP XR cDNA library of the polychaete, Periserrula leucophryna. This gene was expressed in Escherichia coli using the T7 promoter system, and the protease was characterized after partial purification. First, the partial DNA fragment (498 bp) was amplified from the total RNA via RT-PCR using degenerated primers derived from the conserved region of cysteine protease. The full-length cDNA of cysteine protease (PLCP) was prepared via the screening of the Uni-ZAP XR cDNA library using the $^{32}P-labeled$ partial DNA fragment. As a result, the PLCP gene was determined to consist of a 2591 bp nucleotide sequence (CDS: 173-1024 bp) which encodes for a 283-amino acid polypeptide, which is itself composed of an 59-residue signal sequence, a 6-residue propeptide, a 218-residue mature protein, and a long 3'-noncoding region encompassing 1564 bp. The predicted molecular weights of the preproprotein and the mature protein were calculated as 31.8 kDa and 25 kDa, respectively. The results of sequence analysis and alignment revealed a significant degree of sequence similarity with other eukaryotic cysteine proteases, including the conserved catalytic triad of the $Cys^{90},\;His^{226},\;and\;Asn^{250}$ residues which characterize the C1 family of papain-like cysteine protease. The nucleotide and amino acid sequences of the novel gene were deposited into the GenBank database under the accession numbers, AY390282 and AAR27011, respectively. The results of Northern blot analysis revealed the 2.5 kb size of the transcript and ubiquitous expression throughout the entirety of the body, head, gut, and skin, which suggested that the PLCP may be grouped within the cathepsin F-like proteases. The region encoding for the mature form of the protease was then subcloned into the pT7-7 expression vector following PCR amplification using the designed primers, including the initiation and termination codons. The recombinant cysteine proteases were generated in a range of 6.3 % to 12.5 % of the total cell proteins in the E. coli BL21(DE3) strain for 8 transformants. The results of SDS-PAGE and Western blot analysis indicated that a cysteine protease of approximately 25 kDa (mature form) was generated. The optimal pH and temperature of the enzyme were determined to be approximately 9.5 and $35^{\circ}C$, respectively, thereby indicating that the cysteine protease is a member of the alkaline protease group. The evaluation of substrate specificity indicated that the purified protease was more active towards Arg-X or Lys-X and did not efficiently cleave the substrates with non-polar amino acids at the P1 site. The PLCP evidenced fibrinolytic activity on the plasminogen-free fibrin plate test.

Antioxidant Activity of Solvent Fraction from Black Garlic (흑마늘 용매 분획물의 항산화 활성)

  • To confirm antioxidant activity of black garlic, methanol extract of black garlic was fractioned by hexane, chloroform, ethyl acetate, buthanol and water. Antioxidant activities of solvent fractions were assayed in 100, 250, 500 and $1,000\;{\mu}g/mL$ concentrations. The contents of total phenol and flavonoids were significantly higher 5.5~11.6 times in chloroform, ethyl acetate and hexane fraction than other fractions. Antioxidant activities of solvent fractions were increased by higher sample concentrations and their activities were significantly higher in chloroform and ethyl acetate fractions than others. DPPH radical scavenging activity was over 50% in $1,000\;{\mu}g/mL$ concentration, except butanol and water fraction. In the same concentration, reducing power was also significantly lower in butanol and water fraction. ABTS radical scavenging activity was higher in hexane, chloroform and ethyl acetate fractions and was over 70% at $1,000\;{\mu}g/mL$ concentration. In $1,000\;{\mu}g/mL$ concentration, the range of hydroxy radical scavenging activity was 50.27~81.02% and SOD-like ability was 26.73~47.64%. Antioxidant activity in linoleic acid reaction system was significantly higher when storage time was longer and sample concentration was higher in non-polar solvent fractions. Nitrite scavenging activity was relatively higher than antioxidant activity and the activity in $100\;{\mu}g/mL$ concentration was over than 50%, except butanol fraction.

Determination of Liquid Paraffins in Foods by Using GC-FID (GC-FID를 이용한 식품 중 유동파라핀 함량 분석)

  • Liquid paraffin is a mixture of heavier alkanes derived from petroleum. It can be used as a lubricant in processing machinery, as a coating agent, or as a releasing agent. The purpose of this study was to analyze liquid paraffins in foods by using a gas chromatography-flame ionized detector (GC-FID). Liquid paraffin was extracted from the food samples using n-hexane. Non-polar aromatic or olefinic co-extractives were removed by alkaline permanganate oxidation followed by clean up on an aluminium oxide SPE cartridge before the GC-FID analysis. The results of recovery tests were 91.5-103.2%. Based on this optimized method, we investigated the amount of liquid paraffin in various food samples purchased from domestic markets. The levels of liquid paraffin in bread were $95.5{\pm}156.0$ mg/kg (0.008%), those in capsules were $40.2{\pm}54.5$ mg/kg (0.001%), and those in dried fruits and vegetables were $3.0{\pm}18.1$ mg/kg (0.0001%). No liquid paraffin was detected in fresh fruits and vegetables. We propose that our method can be used to monitor and detect liquid paraffin in foods for food safety management.

Bioavailability Soil-aged Residues of the Herbicide Bentazon to Rice Plants (토양중(土壤中) 신생(新生) 및 숙성(熟成) Bentazon 잔유물(殘油物)의 벼에 의(依)한 흡수(吸收))

  • The amounts of $^{14}CO_2$ evolved during the $^{14}C-bentazon$ aging in soil for 3 and 6 months were 6.1 and 14.8% of the original radioactivity, respectively. The presence of earthworms in soil tended to increase the uptake of $^{14}C-bentazon$ by the roots of rice plants, even if it was not statistically significant. The evolution of $^{14}CO_2$ from $^{14}C-bentazon$ in soil increased in the presence of rice plants and earthworms compared with in the absence of them. The uptake of $^{14}C-bentazon$ residues by rice plants decreased remarkably with increasing the aging period within the limit of 3 months both in the absence and presence of earthworms, but there is not much difference between 3-month-aging and 6-month-aging. Much larger amounts of $^{14}C-labelled$ compounds were translocated to the shoots, compared with the data from a previous investigation using maize plants. The amount of non-extractable bound residue increased remarkably with the aging period up to 3 months. The polarity of the compounds extracted from soil increased with the aging and the growing of rice plants, indicating the formation of some polar metabolites.

Fabrication and characterization of tilted R-plane sapphire wafer for nonpolar a-plane GaN (경사각을 갖는 비극성 a-GaN용 R-면 사파이어 기판의 제조 및 특성)

  • Tilt angle of r-plane wafer is a one of the important factors related with the quality of the GaN epi, so the fine control of the tilt angle is important for the growing of high quality non-polar a-GaN epi. We prepared the R-plane sapphire wafers with slight tilt angles for nonpolar a-plane GaN. The target tilt angles of ${\alpha}$ and ${\beta}$ were 0, -0.1, -0.15, -0.2, -0.4, $-0.6^{\circ}$ and -0.1, 0, $0.1^{\circ}$, respectively. The tilt angles of sliced R-plane sapphire wafers were measured by x-ray and the statistical evaluation of reliability of tilt angles of wafers were performed. The tolerance of the tilt angle was ${\pm}0.03^{\circ}$. R-plane sapphire wafers have relatively large distributions of BOW and TTV data than c-plane sapphire wafers due to the large anisotropy of R-plane. As the tilt angle ${\alpha}$ was increased from -0.1 to $-0.6^{\circ}$, the step widths and heights were decreased from 156 nm to 26 nm and 0.4 nm to 0.2 nm, respectively. The growth and qualities of GaN epi seems to be largely affected by the change of step structure of R-plane sapphire wafers with tilt angle.

A Study on Decisions on Investment Factors in the Development of Local Fishing Harbors (지방어항개발의 투자결정요인 분석 - 충청남도 어항 중심으로 -)

  • In the recent WTO/DDA negotiations, discussions are progressing to forbid fishery subsidies which have negative effect on the natural environment and the fishery resources and to lower or to eliminate the tariff/non-tariff barriers that distort trade liberalization. As a result, linking to the weak structural environment of Korea's fishing villages; such a scheme has jeopardized the livelihood of fishermen who settle down in local fishing harbors. Against this backdrop, the government is attempting to positively respond to changes in the fishing environment in a number of different ways: promotion of fishing tourism, fishing harbor reinforcement in function, developing fishing villages, and harbors. With respect to investment by priority in fishing harbor development, it is very important to appropriately select investment targets based on objective criteria. A small number of harbors are chosen and investment plans by harbor are systematically devised, economic feasibility and the effects of investment are analyzed and reviewed in relation to each target harbor. This paper is designed to review and assess objective evaluation methods and the degree of importance for the designation of target harbors, dealing with local harbors in Chung Nam Do as a model. Each local government has tried to reorganize existing harbors and develop new harbors, considering a fall in the number of fishing vessels and the expansion of maritime leisure activities. In order to overcome the limit of existing harbors in function, to raise the income of fishermen, and to activate tourism, they have shifted their focus from simple functions such as vessel evacuation and anchoring to harbor beautification, which can work as a basis for fishing tourism. This paper points out that the AHP analysis method for prioritizing local harbors in Chung Nam Do should help to be chosen target harbors in other cities and provinces. The Study on the Improvement of Piezoelectric and Electrical Characteristics of Bi0.5(Na0.78K0.22)0.5TiO3 Ceramics Modified by the La-based ABO3 Pervskite Structure (La 기반의 ABO3 구조를 갖는 첨가물에 따른 Bi0.5(Na0.78K0.22)0.5TiO3의 압전 및 전기적인 특성 향상 연구)

  • The $0.99Bi_{0.5}(Na_{0.78}K_{0.22})_{0.5}TiO_3-0.01LaAlO_3$, $0.01LaMnO_3$ or $0.01LaFeO_3$ (0.99BNKT-0.01LA, 0.01LM or 0.01LF) ceramics were prepared by a conventional mixed mothod. The structure and morphology of the lead free ceramics were characterized by XRD (X-ray diffraction) and FE-SEM (field emission scanning electron microscopy). XRD results indicated that the BNKT ceramics modified by LA, LM or LF induced a transition from a ferroelectric tetragonal to a non-polar pseudo-cubic phase, leading to decrease in the remnant polarization ($P_r$) and coercive field ($E_c$) in the P-E hysterisis loops. The effects of the BNKT ceramics modified by La-based $ABO_3$ pervskite structure on the electric-field induced strain were investigated, and the largest normalized unipolar strain ($S_{max}/E_{max}$) was found in BNKT-0.01LF ceramic.

Deinococcus rubrus sp. nov., a Bacterium Isolated from Antarctic Coastal Sea Water

  • Two Gram-staining-negative, red-pinkish, coccus-shaped, non-motile, and aerobic bacterial strains, designated $Ant21^T$ and Ant22, were isolated from the Antarctic coastal sea water. Strains $Ant21^T$ and Ant22 showed UVC and gamma radiation resistance. Phylogenetic analyses based on 16S rRNA gene sequences determined that these strains belong to the genus Deinococcus. Through the analyses of the 16S rRNA gene sequences, strains $Ant21^T$ and Ant22 were found to have 97.7% and 97.8% similarity to Deinococcus marmoris DSM $12784^T$ and 97.0% and 97.2% similarity to Deinococcus saxicola AA-$1444^T$, respectively. The sequence similarity with the type strains of other Deinococcus species was less than 96.9% for both strains. Strains $Ant21^T$ and Ant22 shared relatively high 16S rRNA gene sequence similarity (99.3%) and had a closely related DNA reassociation value of $84{\pm}0.5%$. Meanwhile, they showed a low level of DNA-DNA hybridization (<30%) with other closely related species of the genus Deinococcus. The two strains also showed typical chemotaxonomic features for the genus Deinococcus, in terms of the major polar lipid (phosphoglycolipid) and the major fatty acids ($C_{16:0}$, $C_{16:1}$ ${\omega}6c/{\omega}7c$, $iso-C_{17:0}$, and $iso-C_{15:0}$). They grew at temperatures between $4^{\circ}C$ and $30^{\circ}C$ and at pH values of 6.0-8.0. Based on the physiological characteristics, the 16S rRNA gene sequence analysis results, and the low DNA-DNA reassociation level with Deionococcus marmoris, strains $Ant21^T$ ($=KEMB\;9004-167^T$ $=JCM\;31436^T$) and Ant22 (KEMB 9004-168 =JCM 31437) represent novel species belonging to the genus Deinococcus, for which the name Deinococcus rubrus is proposed.
