Establishment and application of a qualitative real-time polymerase chain reaction method for detecting genetically modified papaya line 55-1 in papaya products (RT-PCR을 이용한 유전자변형파파야(55-1)검사법 확립 및 파파야가공식품의 적용 연구)

  • Genetically modified (GM) papaya line 55-1, which is resistant to PRSV infection, has been marketed globally. Prompt and sensitive protocols for specific detections are essential for the traceability of this line. Here, an event- and construct-specific real-time polymerase chain reaction (RT-PCR) method was established to detect 55-1. Qualitative detection was possible for fresh papaya fruit up to dilutions of 0.005% and 0.01% for the homozygous SunUp and heterozygous Rainbow cultivars, respectively, in non-GM papaya. The method was applied in the qualitative detection of 55-1 in eight types of commercially processed papaya products. Additionally, papaya products were monitored to distinguish GM papaya using the P35S and T-nos RT-PCR detection methods. As expected, detection capacity was improved via modified sample preparation and the established RT-PCR detection method. Taking these results together, it can be suggested that a suitable method for the extraction and purification of DNA from processed papaya products was established for the detection of GM papaya.

Phenolics Content and Browning Capacity during the White Winemaking (백포도주 양조중 페놀류의 함량과 갈변도)

  • Juices of Golden Queen, Neo-Muscat and Seibell-9110 were prepared with the addition of $SO_2(100ppm)$ in must and juice, of $SO_2(100ppm)$ and pectinase (40mg/l) in must, respectively and then 300mg/1 of Polyclar AT were added on different white wines from grape juices. Phenolics content in grape juices and white wines were determined. and also changes of browning capacity of white wines during the storage periods at $45^{\circ}C$ were investigated by accelerated method. Extraction of total phenol into grape juices was increased significantly in $SO_2-pectinase$ addition lot and content of total phenol of Seibell-9110 juice was two or three times higher than the others. Yield of grape juices was highest in $SO_2-pectinase$ addition lot. Ethanol content of white wines were 11.4-12.0 v/v% , and total acid, fusel oil and methanol content of Neo-Muscat white wine was lower than that of the other white wines. Total phenol content of white wines was 25-50% lower than that of grape juices and 50-80% lower in colour. Browning capacity of white wines was decreased by 25-40% for 18 days at $45^{\circ}C$ with addition of Polyclar AT, but Seibell-9110 white wine prepared from $SO_2-pectinase$ addition lot was not available commercially because of severe browning.

DISCRIMINATION BETWEEN VIRGIN OLIVE OILS FROM CRETE AND THE PELOPONESE USING NEAR INFRARED TRANSFLECTANCE SPECTROSCOPY

  • Food adulteration is a serious consumer fraud and a potentially dangerous practice. Regulatory authorities and food processors require a rapid, non-destructive test to accurately confirm authenticity in a range of food products and raw materials. Olive oil is prime target for adulteration either on the basis of the processing treatments used for its extraction (extra virgin vs virgin vs ordinary oil) or its geographical origin (e.g. Greek vs Italian vs Spanish). As part of an investigation into this problem, some preliminary work focused on the ability of near infrared spectroscopy to discriminate between virgin olive oils from separate regions of the Mediterranean i. e. Crete and the Peloponese. A total of 46 oils were collected: 18 originated in Crete and 28 in the Peloponese. Oils were stored in a temperature-controlled room at 2$0^{\circ}C$ prior to spectral collection at room temperature (15-18$^{\circ}C$). Samples (approximately 0.5$m\ell$) were placed in the centre of the quartz window in a camlock reflectance cell; the gold-plated baking plate was then gently placed into the cell against the glass so as to minimize the formation of air bubbles. The rear of the camlock cell was then screwed into place producing a sample thickness of 0.5mm. Spectra were recorded between 400 and 2498nm at 2nm intervals on a NIR Systems 6500 scanning monochromator. Spectral collection took place over 2-3 days. Data were analysed using both WINISI and The Unscrambler software to investigate the possibility of discriminating between the oils from Crete and the Peloponese. A number of data pre-treatments were used and discriminant models were developed using discriminant PLS (WINISI & Unscrambler) and SIMCA (Unscrambler). Despite the small number of samples involved, a satisfactory discrimination between these two oil types was achieved. Graphical examination of principal component scores for each oil type also holds out the possibility of separating oils from either Crete and the Peloponese on the basis of districts within each region. These preliminary data suggest the potential of near infrared spectroscopy to act as a screening technique for the confirmation of geographic origin of extra virgin olive oils. The sample presentation strategy adopted uses only small volumes of material and produces high quality spectra.

  • Malaria infection is sensitively influenced by regional meteorological conditions along with global climate change. Remote sensing techniques have become an important tool for extraction of climatic and environmental factors, including rainfall, temperature, surface water, soil moisture, and land use, which are directly linked to the habitat qualities of malaria mosquitoes. Improvement of sensor fidelity with higher spatial and spectral resolution, new multinational sensor development, and decreased data cost have nurtured diverse remote sensing applications in malaria research. In 1984, eradication of endemic malaria was declared in Korea, but reemergence of malaria was reported in mid-1990s. Considering constant changes in malaria cases since 2000, the epidemiological management of the disease needs careful monitoring. Geographically, northmost counties neighboring North Korea have been ranked high in the number of malaria cases. High infection rates in these areas drew special attention and led to a hypothesis that malaria dispersion in these border counties might be caused by north-origin, malaria-bearing adult mosquitoes. Habitat conditions of malaria mosquitoes are important parameters for prediction of the vector abundance. However, it should be realized that malaria infection and transmission is a complex mechanism, where non-environmental factors, including human behavior, demographic structure, landscape structure, and spatial relationships between human residence and the vector habitats, are also significant considerations in the framework of medical geography.

  • Methane, as a greenhouse gas, is some 21~25 times more detrimental to the environmental than carbon dioxide. Landfills generally constitute the most important anthropogenic source, and methane emission from landfill was estimated as 35~73 Tg per year. Biological approaches using biocover (open system) and biofilter (closed system) can be a promising solution for older and/or smaller landfills where the methane production is too low for energy recovery or flaring and installation of a gas extraction system is inefficient. Methanotrophic bacteria, utilizing methane as a sole carbon and energy source, are responsible for the aerobic degradation (oxidation) of methane in the biological systems. Many bench-scale studies have demonstrated a high oxidation capacity in diverse filter bed materials such as soil, compost, earthworm cast and etc. Compost had been most often employed in the biological systems, and the methane oxidation rates in compost biocovers/boifilters ranged from 50 to $700\;g-CH_4\;m^{-2}\;d^{-1}$. Some preliminary field trials have showed the suitability of biocovers/biofilters for practical application and their satisfactory performance in mitigation methane emissions. Since the reduction of landfill methane emissions has been linked to carbon credits and trading schemes, the verified quantification of mitigated emissions through biocovers/biofilters is very important. Therefore, the assessment of in situ biocovers/biofilters performance should be standardized, and the reliable quantification methods of methane reduction is necessary.

A Study on the Development of Dynamic Models under Inter Port Competition (항만의 경쟁상황을 고려한 동적모형 개발에 관한 연구)

  • Although many studies on modelling of port competitive situation have been conducted, both theoretical frame and methodology are still very weak. In this study, therefore, a new algorithm called ESD (Extensional System Dynamics) for the evaluation of port competition was presented, and applied to simulate port systems in northeast asia. The detailed objectives of this paper are to develop Unit fort Model by using SD(System Dynamics) method; to develop Competitive Port Model by ESD method; to perform sensitivity analysis by altering parameters, and to propose port development strategies. For these the algorithm for the evaluation of part's competition was developed in two steps. Firstly, SD method was adopted to develop the Unit Port models, and secondly HFP(Hierarchical Fuzzy Process) method was introduced to expand previous SD method. The proposed models were then developed and applied to the five ports - Pusan, Kobe, Yokohama, Kaoshiung, Keelung - with real data on each ports, and several findings were derived. Firstly, the extraction of factors for Unit Port was accomplished by consultation of experts such as research worker, professor, research fellows related to harbor, and expert group, and finally, five factor groups - location, facility, service, cargo volumes, and port charge - were obtained. Secondly, system's structure consisting of feedback loop was found easily by location of representative and detailed factors on keyword network of STGB map. Using these keyword network, feedback loop was found. Thirdly, for the target year of 2003, the simulation for Pusan port revealed that liner's number would be increased from 829 ships to 1,450 ships and container cargo volumes increased from 4.56 million TEU to 7.74 million TEU. It also revealed that because of increased liners and container cargo volumes, length of berth should be expanded from 2,162m to 4,729m. This berth expansion was resulted in the decrease of congested ship's number from 97 to 11. It was also found that port's charge had a fluctuation. Results of simulation for Kobe, Yokohama, Kaoshiung, Keelung in northeast asia were also acquired. Finally, the inter port competition models developed by ESB method were used to simulate container cargo volumes for Pusan port. The results revealed that under competitive situation container cargo volume was smaller than non-competitive situation, which means Pusan port is lack of competitive power to other ports. Developed models in this study were then applied to estimate change of container cargo volumes in competitive relation by altering several parameters. And, the results were found to be very helpful for port mangers who are in charge of planning of port development.

Assessment of Pollution Level and Contamination Status on Mine Tailings and Soil in the Vicinity of Disused Metal Mines in Kangwon Province (강원도 폐금속광산지역의 광미와 주변토양의 중금속 오염현황 및 오염도 평가)

  • The objectives of this study was to assess pollution level and contamination status on tailings and soil in the vicinity of four disused metal mines in Kangwon province. As the result of total metal concentrations analysis, the pollution degree of tailings and soil decreased in the order of Wondong > Second Yeonhwa > Sinyemi ${\fallingdotseq}$ Sangdong mines. Total metal concentrations of mine tailings in this study were $1.2{\sim}78.2$ and $1.1{\sim}80.6$ times higher than those in the background soil and the tolerable levels suggested by Kloke, respectively. From these results, we found that tailings served as contamination source of nearby soil. According to sequential extraction of metals, large proportion of heavy metals in all mine tailings existed in the form of a residual fraction, and heavy metals in non-residual form was mainly associated with Fe-Mn oxide fraction and sulfidic-organic fraction. Fe-Mn oxide fraction and sulfidic-organic fraction of heavy metals may be released into and contaminated the nearby environment under the oxidation or reduction condition in long-term. In particular, the proportions of the exchangeable and carbonate fraction of Cd in mine tailings from Second Yeonhwa mine were relatively high. This suggests that Cd may be easily released into and contaminated the nearby environment in the near time. Concentrations of heavy metals in mine tailings and the nearby soil exceeded the standard (agricultural area) of Soil Environment Conservation Law. So it was thought that remediation for mine tailings and the nearby soil is needed. The pollution indices of the samples in this study were for higher than 1.0 and the pollution degree was very serious. Priority remediation site for these mines was Wondong. As Results of danger indices, it was showed that exchangeable form in Wondong and Fe-Mn oxide form in the rest mines should be removed preferentially.

Investigation on Reaction Products From Oxidative Coupling Reactions of 1-Naphthol By Manganese Oxide (망간산화물에 의한 1-Naphthol의 산화-결합 반응에 따른 반응산물 연구)

  • In this study, abiotic transformation of 1-naphthol(1-NP) via oxidative-coupling reaction and its reaction products were investigated in the presence of Mn oxides. The reaction products were characterized for their relative polarity using solvent extraction experiment and reverse-phase HPLC, and for structure using CCMS and LC/MS, and for absorption characteristics using UV-Vis spectrometry. The reaction products present in aqueous phase were more polar than parent naphthol and comprised of 1,4-naphthoquinon(1,4-NPQ) and oligomers such as dimers and trimers. Hydrophilic component present in water phase after solvent$(CH_2Cl_2)$ extractions was identified as naphthol polymerized products having molecular weight(m/z) ranging from 400 to 2,000, and showed similar UV-Vis. absorption characteristics to that of foil fulvic acid. Transformation of 1,4-NPQ, which is non-reactive to Mn oxide, to the polymerized products via cross-coupling reaction in the presence of 1-NP was also verified. In this experimental conditions(20.5 mg/L, 1-NP, 2.5 g/L $MnO_2$, pH 5), the transformation of 1-NP into the oligomers and polymerized products were about 83% of initial 1-NP concentrations, and more than 30% of the reaction products was estimated to be water insoluble fractions, not extracted by $H_2O$ methanol. Results from this study suggest that Mn oxide-mediated treatment of naphthol contaminated soils can achieve risk reduction through the formation of oligomers md polymer precipitation.

Study On The Robustness Of Face Authentication Methods Under illumination Changes (얼굴인증 방법들의 조명변화에 대한 견인성 비교 연구)

  • This paper focuses on the study of the face authentication system and the robustness of fact authentication methods under illumination changes. Four different face authentication methods are tried. These methods are as fellows; PCA(Principal Component Analysis), GMM(Gaussian Mixture Modeis), 1D HMM(1 Dimensional Hidden Markov Models), Pseudo 2D HMM(Pseudo 2 Dimensional Hidden Markov Models). Experiment results involving an artificial illumination change to fate images are compared with each other. Face feature vector extraction based on the 2D DCT(2 Dimensional Discrete Cosine Transform) if used. Experiments to evaluate the above four different fate authentication methods are carried out on the ORL(Olivetti Research Laboratory) face database. Experiment results show the EER(Equal Error Rate) performance degrade in ail occasions for the varying ${\delta}$. For the non illumination changes, Pseudo 2D HMM is $2.54{\%}$,1D HMM is $3.18{\%}$, PCA is $11.7{\%}$, GMM is $13.38{\%}$. The 1D HMM have the bettor performance than PCA where there is no illumination changes. But the 1D HMM have worse performance than PCA where there is large illumination changes(${\delta}{\geq}40$). For the Pseudo 2D HMM, The best EER performance is observed regardless of the illumination changes.

Antioxidant, Anti-wrinkle and Whitening Effect of Fermented Extracts of Hwangryunhaedoktang (황련해독탕 발효물의 항산화, 항주름 및 미백 효과)

  • Hwangryunhaedoktang (HHT) has been traditionally used as a preventive and therapeutic medicine to treat enervation and diverse chronic diseases. This study was designed to compare the antioxidant, anti-wrinkle and whitening effects of HHT extract and its fermented extract by Leuconostoc mesenteroides (FHHT). FHHT was prepared by inoculation of L. mesenteroides after the extraction procedure with 70% ethanol. HHT and FHHT was investigated via high-performance liquid chromatography (HPLC). Simultaneous qualitative analysis of two bioacitive components, berberine and palmatine. was achieved by comparing their retention times ($t_R$) and UV spectra with those of the standard components. Cell viability test results indicated that both HHT and FHHT were non-toxic. In DPPH radical scavenging ability, $SC_{50}$ values of the FHHT was $68.85{\mu}g/mL$, which is more effective than HHT. Moreover, FHHT showed higher expression in production of procollagen type I than HHT. In nontoxic concentration range, FHHT showed strong melanin production inhibitory effect in ${\alpha}-melanocyte$ stimulating hormone (${\alpha}-MSH$)-stimulated B16F10 cell ($IC_{50}=9.82{\mu}g/mL$). These results suggested that fermented extracts of hwangryunhaedoktang had considerable potential as a cosmetics ingredient with an antioxidant and anti-wrinkle and whitening effects.
