  • Recently, jawbone osteonecrosis has been largely reported as a potential adverse effect of bisphosphonate (BP)administration. Currently available published incidence data for BRONJ are based on retrospective studies and estimates of cumulative incidence range from 0.8 to 12%. The mandible is more commonly affected than the maxilla (2:1 ratio), and 60-70% of cases are preceded by a dental surgical procedure. The signs and symptoms that may occur before the appearance of clinical evident osteonecrosis include changes in the health of periodontal tissues, non-healing mucosal ulcers, loose teeth and unexplained soft-tissue infection. Tooth extraction as a precipitating event is a common observation. The significant benefits that bisphosphonates offer to patients clearly surpass the risk of potential side effects; however, any patient for whom prolonged bisphosphonate therapy is indicated, should be provided with preventive dental care in order to minimize the risk of developing this severe condition.

Design of Nonuniform Coupled Line-Type Transversal Filters Using Improved Woodward-Lawson Sampling Method (개선된 Woodward-Lawson 샘플링법을 사용한 불균일 결합선로형 트랜스버설 필터 설계)

  • The design method of the transversal filter using continuously cascaded directional couplers is presented. The coupler can be treated for a continuously varying nonuniform coupled transmission line. The design method is based on the optimum extraction of desired coupling factor by the control of null positions which are inherent to the coupling spectra pattern. In the optimization process, the improved Woodward-Lawson sampling method is applied to easily synthesize the distributed delay and weighting elements for transversal filter properties. For application, the microstrip transversal filter is fabricated and optimum dielectric overlay is introduced for the mode phase velocity compensation for non-TEM coupler nodes by using SDA(Spectral Domain Approach). Experiment results confirm the validity of the proposed method.

Chromaticity Analysis of Natural Dyes Extracted from Sappan Wood, Gardenia, and Mugwort (소목, 치자, 쑥으로부터 추출된 천연염료의 색도분석)

  • Extracting natural dyes have been widely studied since the needs of eco-friendly and non-toxic natural dyes increased. In this paper, the natural dyes were extracted from sappan wood, gardenia, and mugwort containing brazilein, crocin, and chlorophyll process. After the extraction with variables of pH of solvent and reaction time, the chromaticity of extracted natural dyes was analyzed using quantitative values from brightness and color coordinate (L, a, b) based on the target colors of red, yellow and green. For the case of brazilein and crocin, the cadmium red and cadmium yellow, respectively were extracted. In the case of sappan wood and gardenia, the red pigment under pH 12 (${\Delta}$ = 18.2) and the yellow pigment at pH 9 (${\Delta}$ = 18.4) were extracted respectively. However, the color of extracted chlorophyll from mugwort was different from the target chrome green.

Effect of Aging on the Chemical Forms and Phytotoxicity of Arsenic in Soil (비소 오염기간이 토양 내 비소의 존재형태와 식물독성에 미치는 영향)

  • This study investigates effects of an aging period on arsenic (As) chemical forms in soils and phytotoxicity using artificially As-contaminated soils with a range of As concentrations (0-300 mg/kg) and aging periods (0 and 3 months). A sequential extraction procedure showed that the increasing As concentration in soils increased the ratio of non-specifically and specifically bound As, which are known to be bioavailable. This resulted in increasing As uptake by tomatoes with increasing As concentration (R2=0.87 for exponential fitting); however, the seed germination was not sensitive to the As concentrations of the soil samples. The seed germination was also statistically similar in the soils with 75 and 150 mg-As/kg regardless of the aging period. The time taken until the seed germination (i.e., lag phase), on the other hand, decreased from 10 d to 3 d with aging for 3 months. This can be attributed to the decreased amount of bioavailable As with aging. Overall, this study shows that when the toxic effects of the As-contaminated soils are assessed using tomato plants, it is better to use more sensitive methods than seed germination such as the As accumulation or the lag phase for seed germination.

Simultaneous Quantification of Oleins (triolein, diolein and monoolein) in Mouse Feces using Liquid Chromatography-Electrospray Ionization/Mass Spectrometry

  • Fat malabsorption is an important cause of poor growth in infancy and childhood. Steatorrhea tests have been developed using various methods. Traditional measurements of stool fat, however, require large samples and it often takes as a week to complete the analysis. In this paper, a liquid chromatography-electrospray ionization/mass spectrometry (LC-ESI/MS) method was developed for simultaneous quantitative analysis of triacylglycerols, triolein, diolein and monoolein, in mouse feces. Moreover, the procedure was rapid, simple as well as compatible with LC-ESI/MS. Chloroform-isopropyl alcohol solution was used for fat-soluble sample extraction. After centrifugation and filtration, an analytical solution was prepared. Triolein, diolein and monoolein were separated using non-aqueous reversed-phase column with the mobile phase consisting of A (methanol) and B (acetone-isopropyl alcohol). The precision (% CV) and accuracy (% bias) of the assay were 3.8-14.7% and 85.2-114.9%, respectively. This method has been successfully applied to simultaneous determination of triolein, diolein and monoolein in feces from 30 mice. This method can therefore be applied to measure triacylglycerols in mouse feces accurately and precisely by LC-ESI/MS, thereby helping to predictive biomarker in fat malabsorption and diagnostic research.

Low Expression of the bcl2 Gene in Gastric Adenocarcinomas in Mazandaran Province of Iran

  • Background: Gastric cancer accounts for about 8% of the total cancer cases and 10% of total cancer deaths worldwide. It is the second lethal cancer after esophageal cancer and is considered the fourth most common cancer in north and northwest Iran. The bcl2 family has a key role in the regulation of apoptosis and change in its expression can contribute to cancer. This study initially scheduled to determine the expression of bcl2 gene in tissue samples of adenocarcinoma cancer patients. Materials and Methods: A total of 10 samples of gastric adenocarcinoma and 10 of normal tissues from Sari hospital were selected and after DNA extraction from tissues, bcl2 gene expression assayed by real-time PCR. Results: Our results demonstrated higher expression of the bcl2 gene in control compared with cancer and marginal cancer tissues. Conclusions: On one hand BCL2 plays an important role as an oncogene to inhibit apoptosis; on the other hand, it can initiate cell cycle arrest at G0 stage. Our observed association between its expression and patient survival is quite conflicting and may be tissue-specific. The data suggest expression both tumoural and non-tumoral(marginal) groups have lowered expression than controls (P>0.05). Due to the low number of samples we could not examine the relationship with clinicopathological features. However, bcl-2 expression may be important for prognostic outcome or a useful target for therapeutic intervention.

Apoptosis-Inducing Activity of HPLC Fraction from Voacanga globosa (Blanco) Merr. on the Human Colon Carcinoma Cell Line, HCT116

  • Voacanga globosa (Blanco), a plant endemic to the Philippines, is traditionally used especially by indigenous people of Bataan in the treatment of ulcers, wounds and tumorous growths. This study aimed to provide scientific evidence to therapeutic properties by determining cytotoxic and pro-apoptotic activity of HPLC fractions from leaves on HCT116 human colon carcinoma and A549 human lung carcinoma cell lines. Ethanolic extraction was performed on V globosa leaves followed by hexane and ethyl acetate partitioning. Silica gel column chromatography and high performance liquid chromatography (HPLC) produced MP1, MP2 and MP3 fractions. Cytotoxic activity of the fractions was determined through MTT assay against the cancer cell lines HCT116 and A549 and the non-cancer AA8 Chinese hamster ovarian cell line. Pro-apoptotic activities of the most active fractions were further assessed through DAPI staining, TUNEL assay and JC-1 mitochondrial membrane potential assay with HCT116 cells. While the MPI fraction exerted no significant activity against all cell lines tested, MP2 and MP3 fractions demonstrated high toxicity against HCT116 and A549 cells. The MP3 fraction induced formation of apoptotic bodies, condensed DNA and other morphological changes consistent with apoptosis of HCT116 cells and TUNEL assay showed significant increase in DNA fragmentation over time. In these cells, the MP3 fraction also induced mitochondrial membrane destabilization, which is generally associated with the beginning of apoptosis. Phytochemical analysis demonstrated the presence only of saponins and terpenoids in the MP3 fraction. The results indicate that the MP3 fraction exerts cytotoxic activity on HCT116 cells via induction of apoptosis triggered by loss of mitochondrial membrane potential crucial for cell survival.

Effects of Extraction Methods on Histidine-containing Low-molecular Weight Peptides and Pro-oxidants Contents in Tuna Thunnus Extracts (다랑어(Thunnus) 추출물 중의 Histidine 함유 저분자 펩타이드 및 산화촉진물질 함량에 미치는 추출방법의 영향)

  • We investigated methods for extracting histidine-containing low-molecular-weight (LMW) peptides such as anserine, carnosine and histidine from the edible meat of tuna byproducts. Extracts were treated by several methods including heat treatment ($80^{\circ}C$, 10 min), DOWEX ion exchange (IEC), ultrafiltration (UF), and carboxymethyl (CM)-cellulose column chromatography (IEC+CMC); then the levels of protein, total iron, histidine, carnosine, and anserine were measured. Extracts treated with IEC+CMC using CM-cellulose were analyzed for total iron, protein, histidine, and anserine content, which were $6.27{\pm}0.26mg/mL$, $5.20{\pm}0.21{\mu}g/mL$, 0.80 mg/mL, 0.208 mg/mL, and 4.40 mg/mL, respectively, in yellowfin tuna; and $9.05{\pm}0.82mg/mL$, $4.06{\pm}0.20{\mu}g/mL$, 1.62 mg/mL, 0.012 mg/mL, and 7.28 mg/mL in bigeye tuna. By comparison in IEC-UF treated extracts, protein, total iron, and histidine content decreased by 43%, 73%, and 27% in yellowfin and 0.4%, 54%, and 23% in bigeye tuna, wheres carnosine and anserine content increased by 22% and 17%, respectively. Freeze-dried (FD) extracts exhibited similar trends as non-dried extracts, i.e., dipeptide content increased with purification steps, whereas pro-oxidant (total iron and protein) content decreased. IEC+CMC treated FD extracts had the highest anserine, content, and the greatest reductuion in pro-oxidants.

Anaphoricity Determination of Zero Pronouns for Intra-sentential Zero Anaphora Resolution (문장 내 영 조응어 해석을 위한 영대명사의 조응성 결정)

  • Identifying the referents of omitted elements in a text is an important task to many natural language processing applications such as machine translation, information extraction and so on. These omitted elements are often called zero anaphors or zero pronouns, and are regarded as one of the most common forms of reference. However, since all zero elements do not refer to explicit objects which occur in the same text, recent work on zero anaphora resolution have attempted to identify the anaphoricity of zero pronouns. This paper focuses on intra-sentential anaphoricity determination of subject zero pronouns that frequently occur in Korean. Unlike previous studies on pair-wise comparisons, this study attempts to determine the intra-sentential anaphoricity of zero pronouns by learning directly the structure of clauses in which either non-anaphoric or inter-sentential subject zero pronouns occur. The proposed method outperforms baseline methods, and anaphoricity determination of zero pronouns will play an important role in resolving zero anaphora.

Analysis of Radiolytic Compounds of Lipids for the Detection of Irradiation in Dried Mytilus coruscus (건조 홍합의 방사선 조사여부를 확인하기 위한 지방분해산물 분석)

  • Radiolytic compounds of lipids, as hydrocarbons and 2-alkylcyclobutanones, were determined to detect the irradiated dried Mytilus coruscus. The detection methods were composed of fat extraction by Soxtec apparatus from dried Mytilus coruscus, isolation of hydrocarbons and 2-alkylcyclobutanones with a florisil column chromatography and identification of GC/MS. Concentrations of the radiation-induced hydrocarbons and 2-alkylcyclobutanones increased with the irradiation dose. The major hydrocarbons in irradiated Mytilus coruscus were pentadecane and 1-tetradecene originated from palmitic acid, and heptadecane and 1-hexadecene originated from stearic acid. 2-(5'- Tetradecenyl)cyclobutanone of 2-alkylcyclobutanones was high relatively. The radiation - induced hydrocarbons and 2-alkylcyclobutanones from dried Mytilus coruscus were detected at 0.5 kGy over and not detected at the non-irradiated.
