Evaluation of nutritive value of chestnut hull for ruminant animals using in vitro rumen fermentation (밤 가공 부산물의 반추가축용 사료 가치 평가: in vitro 반추위 배양)

  • During the manufacturing process of chestnut, 50% of biomass is produced as chestnut shell (CS) or chestnut hull (CH), a forestry by-product. Due to its high fiber content and economic benefit, there is a possibility of using chestnut hull as a supplement for a ruminant diet. Few studies, however, have been conducted on evaluating nutritive value of chestnut hull for ruminant animals. The objective of this study were thus to analyze chemical composition of CS, a by-product after the first processing of chestnut, and CH, a by-product after the second processing, and access in vitro rumen fermentation characteristics of them. For the in vitro fermentation using strained rumen fluid obtained from a fistulated Hanwoo steer, commercial total mixed ration (TMR) for dairy goat was used as a basal diet and was replaced with different proportions of chestnut shell and hull. A total number of 13 treatments were carried out in this study: 100% TMR, 100% CS, 100% CH, a mix with 50% CS and 50% of CH (MIX), TMR replaced with 5%, 10%, or 15% of CS, CH, or MIX, respectively. For each treatment, in vitro dry matter digestibility (IVDMD) and pH after 48 hours of rumen fermentation were measured. Gas production at 6, 12, 24, 48 hours of incubation was also analyzed. Compared to CH, CS contains higher level of fiber (NDF, ADF, lignin) and consequently has a lower amount of non-fiber carbohydrate, but no difference was observed in the other nutrients (i.e. crude protein, crude fat, and ash). IVDMD was significantly (p<0.05) the highest in 100% CH (71.97%) and the lowest in 100% CS (42.80%). Addition of CH by replacing TMR did not affect IVDMD, while an increase in the proportion of CS tended to decrease IVDMD. The total gas production after 48 hours of incubation and the rate of gas production were also the highest in 100% CH and the lowest in 100% CS (P<0.05). Likewise, the pH after 48 hours of fermentation was significantly (p<0.05) the lowest in 100% CH (6.33) and the highest in 100% CS (6.50), and no significant difference in gas production was observed when TMR was replaced with CS or CH up to 15% (P>0.05). In conclusion, CH may successfully be used for a supplement in a ruminant diet. The nutritive value of CS is relative low, but can replace, if not 100%, low quality forage. This study provides valuable information about the nutritive value of CS and CH. An in vivo trials, however, is needed for conclusively accessing the nutritive value of CS and CH.

Comparison of Ileal and Fecal Digestibility of Canola Meal and Wheat Bran in Ducks (채종박과 밀기울에 대한 오리의 분과 회장 소화율 비교)

  • This work was carried out to evaluate the feces and ileal digestibility of canola meal and wheat bran in ducks. Ninety Pekin ducks (6-week-old, average 3.4 kg) were divided into three treatments (6 replications/treatment, 5 birds/replication) by feedstuffs (basal diet, canola meal, and wheat bran) and were fed on these diets in individual metabolic cages for one week. After one week, the samples for analysis were collected from feces and ileum. On comparison of treatments and collected sites, the feces digestibility of basal diet was the highest among treatments on chemical compositions except crude protein and crude fat and ileal digestibility of canola meal was the lowest (P<0.01). On comparison of treatments, the digestibility of basal diet was higher and that of wheat bran was lower when compared to other treatments (P<0.05). The feces digestibility was lower than the ileal digestibility in crude protein (P<0.05) and crude fat (P<0.01). The digestibility of amino acid was the highest at feces of control group among treatments (P<0.01). No significant difference was detected in terms of digestibility of isoleucine, leucine and phenylalanine among essential amino acids, and alanine, glycine and tyrosine among non-essential amino acids between control and wheat bran, and wheat bran and canola meal. However amino acid digestibility was higher in control group when compared to canola meal group (P<0.05). Other amino acid digestibilities were not significantly differ by treatments. These results may provide the basic data on amino acid digestibility of ducks to evaluate feedstuffs and to formulate optimum diet for commercial Pekin duck.

An Investigation of the Sources of Nitrate Contamination in the Kyonggi Province Groundwater by Isotope Ratios Analysis of Nitrogen (질소 동위 원소 분석을 이용한 경기도 지역 지하수 중 질산태 질소 오염원 구명)

  • $^{15}N$-Isotope concentrations of groundwater from l4 wells with different land-use types in Kyonggi Province were measured to investigate the nitrate contamination sources. Water samples were collected monthly from January to December 1997 and analyzed for pH. PC, anions (fluoride, chloride, nitrate, sulfate, inorganic phosphate, and bicarbonate), and canons (calcium, magnesium, potassium, and sodium). For the analysis of the $^{15}N/^{14}N$ ratio as ${\delta}^{15}N$, $N_2$ samples were prepared through Kjeldahl-Rittenberg method and were analyzed using an isotope ratio mass spectrometer (VG Optima IRMS). Reproducibility of the method and precision of the IRMS were below 1.0‰ and 0.1‰, respectively. The ionic composition of each groundwater sample was only slightly different according to the land-use type. The nitrate concentrations of groundwater in cropland or livestock farming areas were higher than those in the residential area. The percentages of nitrate to total anions of groundwater samples from the livestock farming area were higher than those of other areas. The ${\delta}^{15}N$ values of ammonium sulfate, urea, groundwater sample in the non-contaminated area, and water from the animal manure septic tank were -2.7, 1.4, 5.5, and 27.2‰, respectively. Based on the ${\delta}^{15}N$ values, the sources of nitrate could be classified as originated from chemical fertilizers with ${\delta}^{15}N$ values below 5% and as from animal manure or municipal waste with ${\delta}^{15}N$ values over 10‰. In most cases, contamination sources investigated from ${\delta}^{15}N$ values of groundwater samples were correlated with the specific sources according to the land-use types. However, some ${\delta}^{15}N$ values did not matched the apparent land-use types, and there were seasonal variations of ${\delta}^{15}N$ values within the same well. These results suggest that the groundwater quality was affected by two or more contamination sources and the contribution of each source to the groundwater quality varied depending on the sampling season.

Establishment of analytical methods for HPHC list of mainstream cigarette smoke (담배 주류연 중 7개 그룹의 유해성분(HPHC) 분석법 확립 및 유효성 평가)

  • Harmful and potentially harmful constituents (HPHCs) are chemical compounds in tobacco smoke that cause harm to smokers and non-smokers. This study established and validated methods for the analysis of HPHCs from mainstream cigarette smoke. The analyzed HPHCs were categorized into seven groups: aromatic amines, volatile organic compounds (VOCs), heavy metals, tobacco specific nitrosamines (TSNAs), benzo[a]pyrene (B[a]P), ammonia, and carbonyl compounds. The methods were validated by specificity, linearity, limit of detection (LOD), accuracy, precision, and recovery. These validated methods were then applied to the reference cigarettes (1R5F, 3R4F). The correlation coefficients (r2) for the calibration curves of the seven groups were over 0.995. The LODs showed values of 0.01-0.04 ng/cig cig for aromatic amines, 0.01-0.16 μg/cig for VOCs, 0.01-1.27 ng/cig for heavy metals, 0.06-0.28 ng/cig for TSNAs, 0.04 ng/cig for benzo[a]pyrene, 0.08 μg/cig for ammonia, and 0.78-1.77 μg/cig for carbonyl compounds. The precisions obtained from the intra and inter-day batches were less than 15%. The accuracy and the recovery range were less than 15% and 79.2-117.5%, respectively. The proposed methods can therefore be applied for determining HPHCs in tobacco mainstream smoke.

Effects of Acanthopanax Senticosus Leaves Supplementation on Performance, Nutrient Digestibility and Blood Characteristics in Korean Black Goats (가시오갈피 지엽의 급여가 흑염소의 생산성, 영양소 소화율 및 혈액성상에 미치는 영향)

  • This study was conducted not only to investigate effects of different supplementation levels of Acanthopanax senticosus leaves on feed intakes, nutrient digestibility, nitrogen retention, and blood metabolites in Korean black goats but also to obtain basic data for nutritive value of Acanthopanax senticosus leaves and production of high quality functional animal products. Twelve Korean black goats were allotted to treatments in four groups of three goats per treatment and then they were housed in individual metabolism cages with completely random arrangements for 21 days. Four treatments were separated into controls and three different supplementation levels of Acanthopanax senticosus such as 10, 20 and 30%, respectively. For the chemical composition of experimental diets, crude protein (CP) contents of controls were 13.39% and those of supplementation treatments were lowered with increasing levels of Acanthopanax senticosus. Control groups tended to have higher acid detergent fiber (ADF) and neutral detergent fiber (NDF) contents as compared to supplementation groups, but non-fibrous carbohydrate (NFC) contents tended to be higher far supplementation groups than those for controls. Feed intakes had no significant difference among treatments, but digestible dry matter amounts were significantly higher for 20 and 30% supplementation groups than those for other treatments. Dry matter intakes per metabolic body weight and their ratio per body weight were highest in 20% group, but those of 40% groups were lowest (p<0.05). Crude protein, ADF and NDF digestibilities were significantly higher in Acanthopanax senticosus supplementation groups in comparison to controls (p<0.05), and in particular, they were increased with increasing supplementation levels of tranthopanax senticosus. Nitrogen retention was significantly higher for 20 and 30% treatments than those fur controls (p<0.05). Plasma urea nitrogen concentration was significantly lower for 30% supplementation group than those fur controls (p<0.05). Furthermore, plasma triglyceride concentration was significantly lower for Acanthopanax senticosus groups than those for controls (p<0.05). The results showed that supplemental level of 20% Acanthopnnax senticosus leaves to Korean black goats improved nutrient digestibility, nitrogen retention, and plasma urea nitrogen and triglyceride concentrations. Consequently, it might contribute to more efficient production of Korean black goats.

Heavy Metal Contamination around the Abandoned Au-Ag and Base Metal Mine Sites in Korea (국내 전형적 금은 및 비(base)금속 폐광산지역의 중금속 오염특성)

  • The objectives of this study we to assess the extent and degree of environmental contamination and to draw general conclusions on the fate of toxic elements derived from mining activities in Korea. 인t abandoned mines with four base-metal mines and four Au-Ag mines were selected and the results of environmental surveys in those areas were discussed. In the base-metal mining areas, the Sambo Pb-Zn-barite, the Shinyemi Pb-Zn-Fe, the Geodo Cu-Fe and the Shiheung Cu-Pb-Zn mine, significant levels of Cd, Cu, Pb and Zn were found in mine dump soils developed over mine waste materials, tailings and slag. Furthermore, agricultural soils, stream sediments and stream water near the mines were severely contaminated by the metals mainly due to the continuing dispersion downstream and downslope from the sites, which was controlled by the feature of geography, prevailing wind directions and the distance from the mine. In e Au-Ag mining areas, the Kubong, the Samkwang, the Keumwang and the Kilkok mines, elevated levels of As, Cd, Cu, Pb and Zn were found in tailings and mine dump soils. These levels may have caused increased concentrations of those elements in stream sediments and waters due to direct dis-charge downstream from tailings and mine dumps. In the Au-Ag mines, As would be the most characteristic contaminant in the nearby environment. Arsenic and heavy metals were found to be mainly associated with sulfide gangue minerals, and mobility of these metals would be enhanced by the effect of oxidation. According to sequential extraction of metals in soils, most heavy metals were identified as non-residual chemical forms, and those are very susceptible to the change of ambient conditions of a nearby environment. As application of pollution index (PI), giving data on multi-element contamination in soils, over 1.0 value of the PI was found in soils sampled at and around the mining areas.

Production and Characterization of Extracellular Polysaccharide Produced by Pseudomonas sp. GP32 (Pseudomonas sp. GP32에 의해 생산된 세포 외 다당류의 생산 및 특성)

  • A strain GP32 which produces a highly viscous extracellular polysaccharide was conducted with soil samples and identified as Pseudomonas species. The culture flask conditions for the production of extracellular polysaccharide by Pseudomonas sp. GP32 were investigated. The most suitable carbon and nitrogen source for extracellular polysaccharide production were galactose and (NH4)2SO4. The optimum carbon/nitrogen ratio for the production of extracellular polysaccharide was around 50. The optimum pH and temperature for extracellular polysaccharide production was 7.5 and 32℃, respectively. In batch fermentation using a jar fermentor, the highest extracellular polysaccharide content (15.7 g/l) was obtained after 70 hr of cultivation. The extracellular polysaccharide produced by Pseudomonas sp. GP32 (designated Biopol32) was purified by ethanol precipitation, cetylpyridinium chloride (CPC) precipitation, and gel permeation chromatography. Biopol32, which has an estimated molecular weight of over 3×107 datons, is a novel polysaccharide derived from sugar components consisting of galactose, glucose, gulcouronic acid and galactouronic acid in an approximate molar ratio of 1.85 : 3.24 : 1.00 : 1.42. The solution of Biopol32 showed non-Newtonian characteristics. The viscosity of Biopol32 exhibited appeared to be higher at all concentration compared to that of zooglan from Zoogloea ramigera. An analysis of the flocculating efficiency of Biopol32 in industry wastewater (food, textile, and paper wastewater) revealed chemical oxygen demand (COD) reduction rates 58.4-67.3% and suspended solid (SS) removal rates 82.6-91.3%. Based on these results, Biopol32 is a possible candidate for industrial applications such as wastewater treatment.

Comparisons of the Forage Productivity and Mineral Contents Between the Newly Reclaimed and Arable Pasture Soils II. Changes in the soil properties, vigour at early growth, yields, and mineral nutrients of forages as affected by lime application (신개간 및 기경작 초지토양간 목초의 생산성과 무기양분 함량 비교 II. 석회 시용에 따른 토양특성, 목초의 초기생육, 수량 및 무기양분 함량 변화)

  • This pot experiment was conducted to find out the forage productivity and contents of mineral nutrients in an orchardgrass sward affected by the compost lime, and phosphorus applications on the newly reclaimed and arable pasture soils. This 2nd part was concerned with the effect of lime application(L$_{0}$=control, L$_1$=279, L$_2$=54g CaCO$_3$/pot). The results obtained are summarized as follows: 1. There were considerable differences between the newly reclaimed and arable pasture soils in the soil chemical properties as follows in general; poor/common in the newly reclaimed soil longrightarrow and common/good in the arable pasture soil in terms of soil fertility standard. 2. It was recognized that the lime application was resulted in somewhat decrease of the concentrations of available P$_2$O$_{5}$ and increase of P$_2$O$_{5}$ absorbtion coefficient in the newly reclaimed soil, whereas these negative effects were positively enhanced by both lime and compost applications. The concentrations of exchangeable Ca and mg in both soils were increased by lime application. 3. The vigour at early growth stage of orchardgrass was better in the arable pasture soil than newly reclaimed soil. It was worsened by the heavy lime application(L$_2$), but this negative effect was positively enhanced by both lime and compost application. 4. Compared with the non-lime application, the forage yields of L$_1$ treatment were increased in both soils by 15~18%. whereas it of L$_2$ treatment was similar or lower than it of L$_1$. 5. The concentrations of Ca, mg, and P in orchardgrass were relatively higher in the arable pasture soil than the newly reclaimed soil, whereas there it was no difference in the K concentration. It was recognized that the concentrations of mg and Ca in orchardgrass were higher in the L$_1$ treatment than the Lo treatment in both soils, whereas there were no difference in it between L$_1$ and L$_2$ treatments.ments.

Effect of Annual and Basal Dressing with Liquid Pig Manure on Growth and Quality of Rice in Double Cropping System of Rice-Malting Barley (벼.맥주보리 작부체계에서 돈분액비 연용이 벼 생육과 미질에 미치는 영향)

  • In order to investigate the effects of continual pre-plant application of liquid pig manure (LPM) on growth and quality of rice in double cropping system of rice and malting barley, the liquid pig manure was applied after harvesting rice and malting barley for 3 years. Field experiment was designed with non-fertilizer, chemical fertilizer (CF) 100% recommended by soil testing, rice (LPM 50%+CF 50%)+malting barley (CF 100%), rice (LPM 50%+CF 50%)+malting barley (LPM 50%+CF 50%), rice (LPM 100%)+malting barley (CF 100%) and rice (LPM 100%)+malting barley (LPM 100%). The yield of rice was increased by 13% with increasing culm length and No. of panicle per hill as 482 kg $10a^{-1}$ in rice (LPM 100%)+malting barley (LPM 100%) plot compare with 427 kg $10a^{-1}$ in rice (CF 100%)+malting barley (CF 100%) plot. Whereas, lodging index was high in rice (LPM 100%)+malting barley (LPM 100%) plot by increasing of internodes length from 3rd to 4th and decreasing of breaking strength. When the quality of milled rice was compare with rice (CF 100%)+malting barley (CF 100%) plot, it was decreased by increasing of protein contents and decreasing of rate of perfect grain in rice (LPM 100%)+malting barley (LPM 100%) plot. The content of amylose was not significant by 19.4~22.0%. Toyo-taste value was not different in LPM 100% and CF 100% plot. Rice could be grown with only liquid pig manure 100%, but considering yield and quality of rice and lodging risk, basal fertilization by LPM 50% and top-dressing by CF 50% application was recommended for rice cultivation.

Evaluation of Soil Redox Capacity using Chromium Oxidation-reduction Reactions in Volcanic Ash Soils in Jeju Island (크롬산화환원반응을 이용한 제주도 화산회토양 내 토양산화환원능 평가)

  • The soil developed from volcanic ash in Jeju Island, Korea, were classified as typical Andisols. The soils had acidic pH, high water contents, high organic matters and clay-silty textures. The crystalline minerals of the samples were mainly composed of ferromagnesian minerals such as olivine and pyroxene, and iron oxides such as magnetite and hematite derived from basaltic materials. A large amount of gibbsite was found at the subsurface horizon as a secondary product from the migration of excessive aluminum. In addition, our study has shown that considerable amounts of poorly ordered minerals like allophane and ferrihydrite were present in Jeju soils. The contents of $SiO_2$ were lower than those of other soil orders, but $A1_2O_3$ and $Fe_2O_3$ contents were higher. These results are some of the important chemical properties of Andisols. The contents of heavy metals were in the range of $84{\sim}198$ for Zn, $56{\sim}414$ for Ni, $38{\sim}150$ for Co, $132{\sim}1164\;mg\;kg^{-1}$ for Cr, which are higher than the worldwide values in most of the soils. Some soil samples contained relatively high levels of Cr exceeding 1000 mg/kg. Mean reduction capacity of the Jeju soils was $6.53\;mg\;L^{-1}$ reduced Cr(VI), 5.1 times higher than that of the non-volcanic ash soils from inland of Korea. The soil reduction capacity of the inland soils had a good correlation with total carbon content (R = 0.90). However, in spite of 20 times higher total carbon contents in the Jeju soils, there was a week negative correlation between the reduction capacity and the carbon content (R = -0.469), suggesting that the reduction capacity of Jeju soils is not mainly controlled by the carbon content and affected by other soil properties. Correlations of the reduction capacity with major elements showed that Al and Fe were closely connected with the reduction capacity in Jeju soil (R = 0.793; R = 0.626 respectively). Moreover, the amounts of Ni, Co and Cr had considerable correlations with the reduction capacity (R = 0.538; R = 0.647; R = 0.468 respectively). In particular, in relation to the behavior of redox-sensitive Cr, the oxidation of the trivalent chromium to mobile and toxic hexavalent chromium can be restricted by the high reduction capacity in Jeju soil. The factors controlling the reduction capacity in Jeju soils may have a close relation with the andic soil properties explained by the presence of considerable allophane and ferrihydrite in the soils.
