The Expression Analysis of Complement Component C3 during Early Developmental Stages in Olive Flounder (Paralichthys olivaceus)

  • Fish larvae are immediately exposed to microbes from hatching to maturation of their lymphoid organs, therefore effective innate mechanisms is very important for survival in such an environment. The key component of innate immune system, C3 is central protein of all activation pathways of the complement system, leading to inflammatory reactions, such as opsonisation, chemotaxis, and cell lysis of pathogens. Although, innate mechanisms is essential for survival in the early stage of development, little is known about defence mechanisms. In this study, the alignment analysis showed that amino acid sequence of C3 from olive flounder liver EST homologous to other known C3 sequences with 73-99% identity. Also, we examined the tissue distribution of olive flounder C3 and analyzed expression pattern from the fertilized egg until 28 days post hatching. As a result, olive flounder C3 mRNA was expressed only in the liver and the mRNA level more increased as developmental proceed during the early stage. These results may suggest that olive flounder C3 plays an important function in the early immune response of olive flounder larvae.

Expression of Perforin Gene for Early Development of Nephrons in Olive Flounder (Paralichthys olivaceus)

  • The innate immune system is the only defense weapon that invertebrates have, and it is the fundamental defense mechanism for fish. The innate immune response is important in newly hatched flounders because it is closely involved in the initial feeding phase, which is why it is essential for survival during the juvenile period. The expression analysis of genes involved in the innate immune response in the olive flounder (Paralichthys olivaceus) in the days after hatching is incomplete. Therefore, we have begun to examine the expression patterns of genes specifically induced during the development of the innate immune system in newly hatched flounders. Microscopic observation showed that pronephron formation corresponded with the expression of perforin-encoding gene. These results suggest that perforin plays a vital role in the innate immunity of the kidney during developmental stages. Perforin expression was strong at the start of the development of the innate immune response, and continued throughout all the development stages. Our findings have important implications with respect to perforin's biological role and the evolution of the first defense mechanisms in olive flounder. Further studies are required to elucidate the perforin-mediated innate immunity response and to decipher the functional role of perforin in developmental stages.

  • 인체의 항상성 유지에 필수적인 물질로 알려진 산화질소는 인체를 침입한 미생물로부터 보호하는 면역반응에서 매우 중요한 역할을 담당하고 있다. 산화질소는 미생물에 직간접적으로 작용하여 다양한 기전으로 항균작용을 나타낸다. 항생제의 심각한 내성이 심각한 사회적 문제로 대두되면서 새로운 계열의 항생제 개발이 절실한 시점에서 항균물질로서의 산화질소에 대한 연구가 활발히 진행되고 있다. 하지만 매우 짧은 반감기와 기체분자인 산화질소를 항생제로 이용하기 위해서는 산화질소의 저장과 방출을 제어할 전략 필요하다. 본 총설에서는 산화질소의 체내에서의 생화학적 특성과 항균작용을 나타내는 다양한 기전에 대해 설명하였다. 또한 산화질소 방출을 조절하여 항균작용을 향상시키는 최근의 연구에 대해 알아보았다.

Comparative analysis of fat and muscle proteins in fenofibratefed type II diabetic OLETF rats: the fenofibrate-dependent expression of PEBP or C11orf59 protein

  • Fenofibrate, an agonist of $PPAR{\alpha}$, plays an important role in activating many proteins catalyzing lipid metabolism, and it also has a considerable effect on improvement of insulin sensitivity in the diabetic condition. To investigate fenofibrate-dependent expression of peripheral tissue proteins in diabetes, we analyzed whole muscle or fat proteins of fenofibrate-fed OLETF rats, an animal model of type II diabetes, using 2-dimensional gel electrophoresis. We found that many proteins were specifically expressed in a fenofibrate-dependent manner in these diabetic rats. In particular, a functionally unknown C11orf59 protein was differentially expressed in the muscle tissues (about 5-fold increase) in fenofibrate-fed OLETF rats as compared to control rats. Additionally, the signal proteins phosphatidylethanolamine binding protein and IkB interacting protein were differentially regulated in the fenofibrate-treated adipose tissues. We suggest here that these proteins might be involved in controlling lipid or carbohydrate metabolism in diabetes via $PPAR{\alpha}$ activation.

Expression Analysis of Visual Arrestin gene during Ocular Development of Olive Flounder (Paralichthys olivaceus)

  • Olive flounder (Paralichthys olivaceus) is one of the commercial important flatfish species in Korea. The ocular signal transduction pathway is important in newly hatched flounders because it is closely involved in the initial feeding phase thus essential for survival during the juvenile period. However, the study of gene expression during ocular development is incomplete in olive flounder. Therefore we examined the expression analysis of specifically induced genes during the development of the visual system in newly hatched flounders. We searched ocular development-involved gene in the database of expressed sequence tags (ESTs) from olive flounder eye and this gene similar to arrestin with a partial sequence homology. Microscopic observation of retinal formation corresponded with the time of expression of the arrestin gene in the developmental stage. These results suggest that arrestin plays a vital role in the visual signal transduction pathway of the retina during ocular development. The expression of arrestin was strong in the ocular system during the entirety of the development stages. Our findings regarding arrestin have important implications with respect to its biological role and evolution of G-protein coupled receptor (GPCR) signaling in olive flounder. Further studies are required on the GPCR-mediated signaling pathway and to decipher the functional role of arrestin.

  • 핸드폰, PMP, MP3 플레이어 등으로 대표되는 개인 모바일 장치는 빠르게 발전하고 있다. 이런 모바일 환경의 발전은 사용자의 일상을 이해하고자 하는 라이프로그 연구를 활성화하고 있다. 즉, 모바일 장치의 다양한 센서들을 활용하여 개인의 일상을 기록한 라이프로그는 복잡한 일상에 대한 기억 보조 도구로 사용가능하기에 현재 활발히 연구되고 있다. 본 논문에서는 GPS에 의한 실외 위치뿐만 아니라 무선랜에 의한 실내 위치를 고려하여 상황을 추론하고 시각화하는 방법을 제시한다. 실내에서 GPS가 제대로 동작하지 않더라도 무선랜을 기반으로 한 실내 위치 추정을 통해 위치 정보를 획득하고 상황 인식 성능을 향상시켰다. 또한, 지도 인터페이스 및 블로그를 기반으로 하는 시각화 방법을 개발하였다. 실험에서는 모바일 장비를 가지고 실제 데이터를 수집한 뒤 제안하는 방법을 통해 상황을 인식하고 시각화 서비스를 제공하는 성능을 평가하였다.

참돔의 lipoprotein lipase 유전자 다형성 (Polymorphisms of the Lipoprotein Lipase Gene of Red Seabream, Pagrus major)

  • Polymorphism of the lipoprotein lipase (LPL) gene which plays an important role in regulation of lipid deposition was analysed in two red seabream (pagrus major) populations (KF4, cultured KORDI line, n=100 : JPN, imported from Japan, n=100). We amplified a DNA fragment (1,091 bp) including the exon 2 region of the LPL gene, and conducted PCR-RFLP analysis using MspI and AluI. The PCR products were also sequenced. Two alleles (A and B) were found in MspI digestion and Sve alleles (A, B, C, D and E) in AluI digestion. The sequenced data revealed four nucleotide substitutions including one transversion at the MspI recognition site (nt 2,235, $C{\rightarrow}10$) and three transitions at the AluI recognition sites (nt 1,721, $A{\rightarrow}G;$ nt 2,319, $C{\rightarrow}T;$ nt 2,319, $T{\rightarrow}C$). Among them, substitutions at the nt 2,235 and 2,319 sites which are located in the exon 2 were proved to be silent point mutations. MspI polymorphism resulted in 3 genotypes, and the allele frequency was significantly different between the two fish populations, KF4 and JPN. In the case of AluI polymorphism, the 5 alleles (A, B, C, D, E) comprised 12 genotypes of the 5 alleles. KF4 population, alleles D and I were specific to the LPL gene Polymorphisms would be useful DNA markers for red seabream population.

Characteristics of Semi-diurnal and Diurnal Currents at a KOGA Station over the East China Sea Shelf

  • The long-term mooring performed at a KOGA station, located at about $30^{\circ}20^{\prime}N$, $126^{\circ}12^{\prime}E$ in the East China Sea shelf, shows some different behaviors between "semi-diurnal" and "diurnal currents" defined as the currents with periods around, respectively, a half day and a day. They appear to be predominantly tidal having significant coherences with sea level changes around the semi-diurnal and diurnal frequencies. The "semi-diurnal current" is strongly barotropic all year round. However, contrastingly, it is largely baroclinic in summer in the area about 70 km nearer to the continental slope, referred to as the "slope-area", as was found in previous current observations. The "diurnal current" of tidal origin is strongly barotropic in winter. In spring and summer, however, it becomes more baroclinic although it still remains largely barotropic, also showing more of its barotropic nature than in the "slope-area". The inertial oscillation contributing to the "diurnal current" appears to be more prominent when the current is baroclinic, indicating the important role played by stratification in generation of inertial oscillations. Downward energy propagation of inertial oscillation is not observed, suggesting that it is not created at the surface by wind. Considering that the study area is both near a critical latitude and proximity to the continental slope, it is suggested that parametric subharmonic instability (PSI) plays a significant role in creating the baroclinic inertial oscillation.

증권회사 홈트레이딩 시스템(HTS)의 서비스 품질과 고객만족에 관한 연구 (The Service Quality of Home Trading System: Its Impact on Customer Satisfaction and Loyalty)

  • 이 연구는 홈 트레이딩 시스템(HTS)의 서비스 품질요인을 파악하고, 파악된 품질요인과 HTS사용자의 만족도, 충성도와의 관계, 더 나아가 이런 요인들이 증권회사에 대한 고객 만족도와 충성도에 어떠한 영향을 미치는지에 대해 고찰하였다. (1) HTS 서비스 품질요인으로 PZB(1985)의 확신성, 신뢰성, 유형성, 반응성, 공감성의 다섯 가지 요인을 채택하였으며, (2) 이 요인들은 HTS 사용자 만족도와 증권회사에 대한 만족충성도에 유의한 영향을 미치는 것으로 나타났다. (3) 또한, 홈 트레이딩 시스템에 대한 사용자 만족도는 증권사에 대한 만족충성도에 유의한 영향을 미치는 것으로 나타났다. 본 연구는 현대 증권거래의 필수적 거래수단이 되고 있는 HTS의 품질요인과 이러한 요인들이 HTS와 증권회사에 대한 서비스 만족도와 충성도에 미치는 영향을 파악함으로써 증권회사의 실적 제고와 고객 서비스 향상에 기여하고자 한다.

삼차원 불연속면 연결구조 해석 및 가시화 소프트웨어 모듈 개발 (Development of the 3-D Fracture Network Analysis and Visualization Software Modules)

  • 본 연구는 삼차원 지질 모델링 소프트웨어 개발의 일환으로 불연속면의 구조기하형태에 대한 삼차원적 해석 및 가시화를 수행 할 수 있는 새로운 소프트웨어 모듈을 개발하는 데에 목적이 있다. 불연속면 연결구조의 삼차원 해석 및 가시화 알고리즘을 바탕으로 개발된 소프트웨어는 C++언어 기반의 MFC 및 OpenGL 라이브러리를 응용하여 Microsoft Visual Studio 상에서 제작되었다. 개발된 소프트웨어는 BOUNDARY, DISK3D, FNTWK3D, CSECT, BDM 등의 모듈을 포함하며 각각의 모듈은 해석영역 구성, 불연속면 네트워크 시스템의 가시화, 등가파이프의 산정, 단면도 작성, 시추공 자료 관리 등 절리성 암반의 삼차원 구조기하형태 해석에 관련된 다양한 기능을 수행한다. 이 연구에서 개발한 불연속면 연결구조의 삼차원 가시화 및 해석 소프트웨어는 불연속체 기반의 암반강도 및 변형성에 관한 연구, 수리지질학적 특성에 관한 연구 및 사면안정 연구를 수행함에 있어서 활용도가 높을 것으로 판단되며 국내 소프트웨어 산업의 경쟁력을 높이는데 기여할 수 있을 것으로 기대된다.
