Medicago truncatula is a diploid legume plant related to the forage crop alfalfa. Recently, it has been chosen as a model species for genomic studies due to its small genome, self-fertility, short generation time, and high transformation efficiency. M. truncatula engages in symbiosis with nitrogen-fixing soil bacterium Rhizobium meliloti. M. truncatula mutants that are defective in nodulation and developmental processes have been generated. Some of these mutants exhibited altered phenotypes in symbiotic responses such as root hair deformation, expression of nodulin genes, and calcium spiking. Thus, the genes controlling these traits are likely to encode functions that are required for Nod-factor signal transduction pathways. To facilitate genome analysis and map-based cloning of symbiotic genes, a bacterial artificial chromosome library was constructed. An efficient polymerase chain reaction-based screening of the library was devised to fasten physical mapping of specific genomic regions. As a genomics approach, comparative mapping revealed high levels of macro- and microsynteny between M. truncatula and other legume genomes. Expressed sequence tags and microarray profiles reflecting the genetic and biochemical events associated with the development and environmental interactions of M. truncatula are assembled in the databases. Together, these genomics programs will help enrich our understanding of the legume biology.
This study evaluated the kinetics of acrylamide (AM) biodegradation by mixed culture bacteria and Enterobacter aerogenes (E. aerogenes) in sequencing batch reactor (SBR) systems with AQUASIM and linear regression. The zero-order, first-order, and Monod kinetic models were used to evaluate the kinetic parameters of both autotrophic and heterotrophic nitrifications and both AM and chemical oxygen demand (COD) removals at different AM concentrations of 100, 200, 300, and 400 mg AM/L. The results revealed that both autotrophic and heterotrophic nitrifications and both AM and COD removals followed the Monod kinetics. High AM loadings resulted in the transformation of Monod kinetics to the first-order reaction for AM and COD removals as the results of the compositions of mixed substrates and the inhibition of the free ammonia nitrogen (FAN). The kinetic parameters indicated that E. aerogenes degraded AM and COD at higher rates than mixed culture bacteria. The FAN from the AM biodegradation increased both heterotrophic and autotrophic nitrification rates at the AM concentrations of 100-300 mg AM/L. At higher AM concentrations, the FAN accumulated in the SBR system inhibited the autotrophic nitrification of mixed culture bacteria. The accumulation of intracellular polyphosphate caused the heterotrophic nitrification of E. aerogenes to follow the first-order approximation.
The behaviors of inorganic nitrogen derived from solid animal waste in soil has been received too much concern partly because nitrate which occurred from nitrification can act as a pollutant to soil and groundwater and partly because the loss of nitrogen from surface soil by downward movement of water is disadvantageous in the view of plant nutrient. This present study was conducted to get fundamental imformations on nitrogen behavior and to provide improved basical concepts on the management of animal waste. Fresh or fermented pig manure was mixed with a sandy loam soil in the ratio of 2:1(soil:pig manure), packed into test tube and incubated at $30^+/-1^{\circ}C$ for 8 weeks under aerobic- or anaerobic condition. Sample tubes were taken at the one week interval and analyzed on pH, the amount of $CH_4$ produced under anaerobic condition and inorganic nitrogen. The pH of soil treated with fresh pig manure under anaerobic condition was lowered by 1.87 unit compared to that of under aerobic condition, but at the treatment with fermented pig manure, pH change was very little between aerobic and anaerobic condition. The coefficients of regressional equations which were obtained from pH and incubation time were -0.114 in fresh pig manure and -0.089 in fermented pig manure, and the extent of pH decrease due to incubation was greater in fresh pig manure than that of fermented pig waste. No differences in the amounts of $CH_4$ produced under anaerobic condition between fresh and fermented pig manure was observed until 3 weeks of incubation, however, after that the amount of $CH_4$ produced in fresh pig manure was abruptly increased and cumulative amont of $CH_4$ was reached 8.6 mole/g. K values on $CH_4$ production in fresh and fermented pig manure was 0.211 mole/g/day and 0.046 mole/g/day, respectively, for 5 weeks from the 3rd to the 8th week. $NH_4-N$ concentration at aerobic condition with fresh pig manure treatment was lowered by passing time of incubation, but $NO_3-N$ concentration was elevated from 11.2 ppm at initial state to 67.3 ppm after incubation and this trend on $NH_4-N$, $NO_3-N$ concentration was very similar to the treatment of fermented pig manure. While $NH_4-N$ concentration under anaerobic condition was greatly increased. $NO_3-N$ concentartion was not only very low but also no great changes, that was ranged from 4 to 8 ppm.

In this study, to investigate the effect of chemical pretreatment for livestock wastewater, laboratory scale test for ozonation and linked treatment of sewage were conducted. and the results were obtained as follows. The ozonation of livestock wastewater showed the COD removal rate per hour to be 17%, 78% and 62% at each pH 4, 7 and 10, respectively. With transformation of NBDCOD to biodegradable BDCOD by ozonation, the ratio of SCODcr/TCODcr was increased from 26% to 38%. Accordingly, pretreatment of livestock wastewater affected to the biological post treatment process to elevate removal efficiency by transformation of nonbiodegradable mass to biodegradable mass. As the results of linked treatment of pre-ozonated livestock wastewater and sewage in the MLE process, the treatment efficiencies of TCODcr 93.8%, T-N 74.3%, T-P 89.7%, SS 97.5% were earned at 100% of internal recycle rate. When the internal recycle rate was increased to 150%, the treatment efficiencies of TCODcr 94.5%, T-N 54.5%, T-P 70.8%, SS 98.5% were earned. Also the removal efficiencies of TCODcr 92.6%, T-N 83.1%, T-P 81.9%, SS 98.5% were earned as the internal recycle rate was increased to 200%. Especially, nitrogen removal efficiency in the linked treatment showed 74.3%, 54.5%, 83.1% at 100%, 150% and 200% of internal recycle ratio, respectively, which revealed the tendency of higher removal efficiency than that of sewage treatment.
Hydrogenated amorphous carbon nitride[a-C:H(N)] films were deposited on p-type Si(100) at room temperature with bias voltage of 200 V by DC saddle-field plasma-enhanced chemical vapor deposition. Effects of the ratio of $N_2$ to $CH_4$($N_2/CH_4$), in the range of 0 and 4 on such properties as optical properties, microstucture, relative fraction of nitrogen and carbon, etc. of the films have been investigated. The thickness of the a-C:H(N) film was abruptly decreased with the addition of nitrogen, but at $N_2/CH_4$>0.5, the thickness of the film gradually decreased with the increase of the $N_2/CH_4$. The ratio of N to C(N/C) of the films was saturated at 0.25 with the increase of $N_2CH_4$. N-H, C≡N bonds of the films increased but C-H bond decreased with the increase of $N_2CH_4$.Optical band gap energy of the film decreased from 2.53 eV at the ratio of $N_2CH_4$=4.
The optimum conditions for glucoamylase production, and ethanol productivity of the transformant TSD-14 were investigated as compared with the parental strains. The properties of TSD-14 were comparatively similar to the donor S. diastaticus IFO 1046 as regards the conditions of glucoamylase production and ethanol productivity. The soluble starch was the most effective carbon source for the glucoamylase production. While inorganic nitrogen sources did not prompt cell growth and enzyme production, the organic nitrogen sources generally enhanced both cell growth and glucoamylase production. The metal salts such as FeSO$_4$, MgSO$_4$, MnCl$_2$, and NiSO$_4$were favorable to the enzyme production. And the optium temperature and initial pH for glucoamylase production were 3$0^{\circ}C$ and 5. The transformant TSD-14 produced 8.3%(v/v) ethanol from 15% sucrose medium, 4.8%(v/v) ethanol from 15% soluble starch medium, and 7.5%(v/v) ethanol from 15% liquefied potato starch medium. The corresponding fermentation efficiency were 84% , 45% and 70%, respectively.
For screening thermostable $\alpha$-amylase from thermophiles, various samples from extreme environments such as hot spring and sewage near them, and compoat, wereexamined microbial growth in enrichment culture medium at 55$\circ$C on the assumption that enzymes from thermophiles are inevitable thermostable. One strain showing higher $\alpha$-amylase activity was pure cultured and designated as Bacillus sp. TR-25 from the results of morphological, cultural and physiological characteristics. The most important carbon sourses for the enzyme production were soluble starch, dextrin, potato starch and corn starch. Glucose and fructose had a catabolite repression on the enzyme production. The good nitrogen sources for the enzyme production were yeat extract, nutrient broth, tryptone, corn steep liquor and ammonium sulfate. The enzyme production was accelerated by addition of CaCl$_{2}$. $\cdot $ H$_{2}$O. The optimal medium composition for the enzyme production was soluble starch 2.0%, yeast extract 0.55, CaCl$_{2}$ $\cdot $ 2H$_{2}$O 0.015, Tween 80 0.001%, pH8.0, respectively. In jar fermenter culture, this strain shows a rapid growth and required cheaper carbon and nitrogen source. These properties are very useful to fermentation industry. The $\alpha$-amylase of this strain demonstrated a maximum activity at 80$\circ$C, pH 5.0, respectively. And calcium ion did not improve thermostability of the enzyme. At 10$0^{\circ}C$, this enzyme has 235 of relative activity. Transformation was carried out by thermophilic Bacillus sp. TR-25 genomic DNA. As a result, the transformant has increased thermostable $\alpha$-amylase activity.
The increasing of greenhouse gases may change agricultural environment. The agronomic productivity will depend upon change of temperature, precipitation, solar radiation and fertilization. Particularly, nitrogen fertilization considerably influences rice productivity and agricultural environments. This experiment was conducted to study transformation of nitrogen and to determine the primary yield components responsible for yield differences in paddy soil. $NH_4-N$ concentration of NPK plot in surface water of paddy soil was $2.07mg\;L^{-1}$ at 5 days after transplanting, and then was decreased sharply due to rice absorption and loss to environment. $NO_3-N$ concentration of NPK plot in surface water was $3.97mg\;L^{-1}$ at 10 days after transplanting. $NO_3-N$ concentration range of CRF plot in surface water was $3-5mg\;L^{-1}$ at 30th after transplanting. The accumulation of $NH_3$ volatilization in NPK plot was $22.39kg\;ha^{-1}$, which accounted for 20% of N fertilizer applied but using of CRF fertilizer can reduce $NH_3$ volatilization by 67% in paddy soil. Use efficiency of N fertilizer was not different between CRF70% and CRF100% plot. Rate of N use efficiency were 27.4%, 51.2%, 49.0% in paddy field NPK, CRF70% and CRF100% plots respectively. The yield of CRF70% showed the best effect with 9.3% increase production ratio, compare with NPK plot.

Objective: To assess the potency of calcium propionate (CAP) used as silage additive, an experiment was carried out to evaluate the effect of CAP on the nitrogen transformation, fermentation quality and aerobic stability of alfalfa silages. Methods: Alfalfa was ensiled with four levels of CAP (5, 10, 15, and 20 g/kg of fresh weight [FW]) in laboratory silos for 30 days. After opening, the silages were analyzed for the chemical and microbiological characteristics, and subjected to an aerobic stability test. Results: The increasing proportion of CAP did not affect pH, lactic acid (LA) concentrations and yeast counts, while linearly decreased counts of enterobacteria (p = 0.029), molds (p<0.001) and clostridia (p<0.001), and concentrations of acetic acid (p<0.001), propionic acid (p<0.001), butyric acid (p<0.001), and ethanol (p = 0.007), and quadratically (p = 0.001) increased lactic acid bacteria counts. With increasing the proportion of CAP, the dry matter (DM) loss (p<0.001), free amino acid N (p<0.001), ammonia N (p = 0.004), and non-protein N (p<0.001) contents were linearly reduced, whereas DM (p = 0.048), water soluble carbohydrate (p<0.001) and peptide N (p<0.001) contents were linearly increased. The highest Flieg's point was found in CAP10 (75.9), represented the best fermentation quality. All silages treated with CAP improved aerobic stability as indicated by increased stable hours compared with control. Conclusion: The addition of CAP can suppress the undesirable microorganisms during ensiling and exposure to air, thereby improving the fermentation quality and aerobic stability as well as retarding the proteolysis of alfalfa silage. It is suggested that CAP used as an additive is recommended at a level of 10 g/kg FW.
Transgenic tobacco plants were produced by the transformation of ginseng CAB gene using Agrobacterium tumefaciens LBA4404. The presence of CAB gene in the second generation of transgenic tobacco plant was confirmed by genomic PCR. The photosynthetic ability of transgenic plants was higher than normal tobacco plants and the maximum photosynthetic point of transgenic and normal tobacco plants was 500 $\mu$mol m$^{-2}$ s$^{-1}$ . The photosynthesis of C7, C11, 1, C14 cell lines was higher than normal plants at all the light intensities investigated. The photosynthesis of C2, C11, C14 cell lines in 90% dark condition was higher than normal plants. The chlorophyll contents of transgenic tobacco plants were almost same as normal plants. The % of dry weight, nicotine content, total sugar and nitrogen contents of harvested transgenic tobacco plant leaves were almost same as normal plants.
