RDX 입도에 따른 다기추진제의 연소 및 강내탄도 특성 (Characteristic Property of Combustion and Internal Ballistics of Triple-Based Propellant according to Particle Size of RDX)

  • The important factors in the design of the gun propellant are impetus, flame temperature and pressure. In this paper, we considered a nitrocellulose based propellant composition that replaced sensitive NG(Nitroglycerin) with RDX(Cyclotrimethylenetrinitramine) and DEGDN(Diethylene glycol dinitrate) which high energy and low sensitivity. Particle size and content of RDX are the two main factors that affect the burning stability of RDX-based propellants. Among them, the characteristics of the propellant according to the particle size of RDX were confirmed. The relative combustion rate(R.Q., Relative Quickness) of the propellant changed according to the RDX particle size, and internal ballistics of properties of propellant were also varied. The particle size of RDX can be confirmed as a major factor in the combustion and internal ballistics characteristics of the propellant.

Legionella Pneumophila 검출을 위한 효소면역측정 스트립 센서 (An Enzyme-linked Immunosorbent Assay Strip Sensor for the Detection of Legionella Pneumophila)

  • 본 연구에서는 효소면역 분석법(enzyme-linked immunosorbent assay)과 면역크로마토그래픽 기법을 결합하여 Legionella pneumophila 검출을 위한 면역스트립을 제작하였다. 면역스트립은 4종의 멤브레인을 이용하여 제작하였다. 니트로셀룰로오스 멤브레인은 포획항체를 고정화하여 신호 발생을 일으키기 위해 사용되었고, 두 종류의 유리섬유 멤브레인은 각각 중합체 패드와 시료주입 패드로 사용되었다. 셀룰로오스 멤브레인은 모세관 현상으로 시료흐름을 유도하는 흡수 패드로 이용하였다. 샌드위치 면역반응과 효소반응에 의해 30 min 이내에 생성된 발색신호는 정성 및 정량 분석이 가능하였다. 분석조건 하에서 육안에 의한 정성 검출뿐 아니라, $1.3{\times}10^3-1.3{\times}10^6CFU/mL$ 범위의 L. pneumophila 농도를 디지털카메라와 자체 제작된 소프트웨어를 이용하여 정량적으로 분석할 수 있었다.

Development of Rapid Immune-gold Strip Kit for On-Site Diagnosis of Tomato spotted wilt virus

  • A rapid, user-friendly and simple immune-chromatographic dipstick kit named 'rapid immune-gold strip' (RIGS) kit was developed in a novel single strip format to detect on-site detection of Tomato spotted wilt virus (TSWV). Immunoglobulin G (IgG) from polyclonal antisera raised in rabbits against TSWV was purified through protein-A affinity chromatography and then the purified TSWV-IgG was conjugated to colloidal gold nano-particles which served as a test line on nitrocellulose membrane. Protein A that non-specifically binds to TSWV antibody was used as a control line on the same strip. The diagnosis process with the TSWV-RIGS involves simply grinding the suspect plant sample in a bag that contains the extraction buffer and inserting the strip the bag. Results can be seen in 2-5 minutes. The flow of the complexes of gold particles coated with TSWV-IgG and a crude sap from TSWV-infected pepper, tobacco and tomato plants resulted in intensive color formed on the test lines proportional to the concentrations of TSWV. The RIGS-TSWV kit did not show any cross-reactions against other tomato-infecting viruses unrelated to TSWV. These results indicate that the TSWV-RIGS kit is highly sensitive and is not required for laboratory training and experience prior to testing. The TSWV-RIGS kit is suitable for on-site detection of suspect TSWV-infected plants as well as for laboratory diagnosis.

네일 샵 종사자들의 직무 형태별 취급 유해화학물질 (Task-Specific Hazardous Chemicals Used by Nail Shop Technicians)

  • Objectives: This study was conducted to evaluate the task-specific hazards of chemicals used by nail technicians in Daegu Metropolitan City. Materials: A total of 30 nail shops located in Daegu City were surveyed to investigate the major tasks and practices performed by nail technicians and the ingredients listed in nail care products used in shops. We also collected instructions for use and material safety data sheets(MSDSs) of nail care products and compared CAS Nos. of ingredients with the lists of chemicals regulated by the Industrial Safety and Health Act(ISHA) and Chemical Substances Control Act(CSCA). Results: A total of 125 chemical ingredients were found in 468 nail care products used at the 30 nail shops. The most frequently found ingredients were ethyl acetate(72%), followed by n-butyl acetate(71.8%), isopropanol(56%), benzophenone(51.1%), nitrocellulose(46.4%) and ethanol(45.3%). Comparing six tasks, the task of manicuring used the most products at 222 products containing 91 ingredients. Among the 125 ingredients, there are 31 chemicals with occupational exposure limits(OEL) designated by the Ministry of Employment and Labor(MoEL), eight categorized as carcinogens, one mutagen and two reproductive toxic chemicals. In terms of carcinogens, formaldehyde was identified as the only confirmed human carcinogen(1A). We found that there was one chemical with a permissible limit, one special management substance, 18 workplace monitoring substances and ten special health diagnosis substances regulated by ISHA. For CSCA, nine poisonous substances, six substances requiring preparation for accidents and one restricted substance were identified. Conclusions: Based on these findings, formaldehyde was identified as one of the chemicals that should most strictly be controlled for the protection of the health of nail technicians and customers. At the same time, it is necessary to distribute materials with detailed hazardous information of nail care products for nail shop technicians.

  • 안지오텐신II 수용체(AT1 수용체)는 혈관수축과 체내 전해질이온 조절에 중요한 역할을 한다. AT1 수용체 길항제(ARB)는 고혈압 치료에 이용되며 최근에는 당뇨병을 포함한 대사질환에 효능이 있음이 알려져 있다. 이 연구에서는 ARB 처리 후 세포 내 인산화단백질에 인산화가 일어나는지를 antibody array를 이용하여 실험하였다. 아미노산세린 및 트레오닌에 인산화되는 단백질 6개, 티로신에 인산화되는 단백질 12개에 대한 항체를 선정하여 nitrocellulose membrane에 부착시켰다. AT1 수용체를 발현한 COS-1 세포에 로사틴(losartan)을 처리하였을 때 small GTPase인 RhoA의 세린 잔기에 인산화가 20% 증가함을 관찰하였다. RhoA는 세포골격의 재배열에 중요한 역할을 하며 세린 잔기에 인산화가 되면 활성이 억제된다. 본 연구결과로부터 ARB가 AT1 수용체에 의한 혈관수축을 억제할 뿐만 아니라 새로운 세포 신호룰 생성함을 알 수 있다.

액상 효소반응을 이용한 Membrane Strip 형 Cholesterol 측정시스템의 개발 (Development of Membrane Strip Assay System for Lipoprotein Cholesterol Based on Liquid-Phase Enzyme Reactions)

  • A sensitive membrane strip assay for plasma lipoprotein cholesterol that can be performed without handling reagents has been investigated. We previously developed an assay system with immobilized enzymes (cholesterol esterase and cholesterol oxidase) on the surfaces of nitrocellulose membrane(1). In such a case, the amount of enzymes present on the membrane was limited by its surface area and, thus, the detection capability was relatively poor (> 50 mg/dL cholesterol). To overcome this problem, we devised a new system with non-immobilized enzymes by placing them within interstitial spaces of a celullose membrane pad in a dry state. Upon contact with sample medium, the enzymes were immediately dissolved and participated in the reactions with cholesterol in a liquid phase. We constructed a user-friendly system consisting of four membrane pads fro sample application, cholesterol decomposition, color development as signal, and medium absorption to invoke a continuous flow (sequential location from the bottom). A sample containing lipoproteins was added into the application pad by capillary action and transferred to the next pad for decomposition. The decomposition pad (namely, enzyme pad) contained a detergent (sodium cholate) for the destruction of lipoprotein particles, the two enzymes for cholesterol decomposition, and a chromogen (3,3'-diaminobenzidine). As a consequence of the enzyme reactions, hydrogen peroxide was produced, and then reacted in the presence of the chromogen with horseradish peroxidase immobilized on the signal generation pad. Finally, a colorimetric signal directly proportional to the cholesterol concentration was produced. The detection limit determined from this system under optimal conditions was at least 2 times lower than of the enzyme-immobilized system.

항체가 건조상태로 고정된 면역진단키트의 안정화와 Shelf-Life 결정 (Stabilization of Dry Immuno-Diagnostic Kit with Immobilized Antibody and Determination of Its Shelf-Life)

  • Stabilization of antibody, which is specific to Salmonella typhimurium antigens, present in dry states on membranes was accomplished, and its shelf-life, i.e., duration for maintaining minimum 90% of the initial activity, under optimal conditions was determined. To prepare two major components of an immuno-strip, the antibody was not only immobilized on nitrocellulose membrane surfaces but also placed within the pores of glass fiber membrane after conjugating it with old colloids as signal generator. Among potential stabilizers of the immuno-components, a disaccharide, trehalose, showed a significant protection effect of immunoglobulin structure from thermal energy. Optimal concentrations of trehalose for the respective component were significantly different (8-fold higher for the antibody-gold conjugate than for the immobilized antibody), which probably resulted from distinct densities and configurations of antibody present on the membranes. An additional requirement for the gold conjugate was freeze-drying of this substance such that the conjugate can be readily resolubilized upon contact with aqueous medium. By using the components prepared under optimal conditions, immuno-strips were constructed and exposed to thermal energy. Signals with less than 10% decrease in the intensity were maintained for approximately 21 days at 60$^{\circ}C$. Compared to previous reports, this result represented a 2-year shelf-life at room temperature. it was, however, two times longer if determined from thermal acceleration tests based on the theory of inactivation rate of protein. Such discrepancy between the two estimates could be mainly attributed to errors in accurately controlling temperatures and also to changes in the physical properties of membranes due to a high thermal energy.

분자생물학적 방법인 PCR-REBA를 이용한 대중목욕탕 수질 중 수인성병원성미생물 검출 (Detection of Waterborne Pathogens in Public Bath Houses by PCR-Reverse Blot Hybridization Assay (PCR-REBA))

  • 수인성 병원성 미생물에 의한 공중목욕탕의 오염은 질병발생의 원인이 된다. 본 연구에서는 공중목욕탕내에 존재하는 수인성 병원성미생물들을 확인하고자 하였다. 서울지역내의 30 곳 공중목욕탕에서 욕조수 시료를 채수하여 진행하였다. 수인성 병원성미생물의 검출은 0.45 ${\mu}m$의 여과막을 이용하여 전통적인 배양방법으로 분리 및 동정하였다. 분자생물학적 기법을 사용하기 위해 미생물학적인 배양을 하지 않고 핵산을 추출하여 16S rRNA유전자를 표적으로 polymerase chain reaction-reverse blot hybridization (PCR-REBA)을 실시하였다. 미생물학적 배양방법에서는 지표세균인 Escherichia coli와 Shigella spp.가 검출되었으며, 분자생물학적 기법인 PCR-REBA을 수행한 결과 E. coli, Shigella spp., Salmonella spp., Pseudomonas spp., Mycobacterium spp. 등의 수인성 병원성미생물이 7곳에서 검출되었다. 본 연구결과를 토대로 공중목욕탕의 욕조수내에 수인성병원성미생물에 의한 감염을 줄이기 위해 적절한 위생관리과 E. coli를 포함한 유해미생물을 선정하여 지속적인 모니터링이 필요할 것으로 사료된다.

CVD법을 이용한 적색 인조진주 코팅 및 제조 (Preparation and Coating of Red Colored Artificial Pearl by CVD Method)

  • 현대인의 눈높이에 맞는 친환경적인 고급품질의 인조진주 제품개발이 세계의 악세사리 및 생활용품 시장에서 요구되고 있다. 본 연구는 고품질의 인조진주 제품을 경제적으로 대량생산하기 위한 방법으로 유기안료를 인조진주 제조용 친환경 무기안료로 대체하고 기존 제조 방식에서 사용하는 주재료였던 니트로셀룰로오스를 우레탄 바인더로 대체하여 광택도가 73.4%에서 86.7%로 증가하였으며 CVD 마감처리 후에는 96%의 높은 광택도를 가졌다. 색차분석 결과 CVD 코팅으로 인하여 빛의 간섭효과 때문에 a*와 b*값이 각각 +37.7에서 +31.9로, +24.5에서 +14.2로 감소하면서 다양한 색을 발현하여 영롱한 빛깔의 인조진주가 제조되었다. 마감 증착된 고품질의 인조진주의 내화학성, 광택도, 색차계, 표면 거칠기, 내마모성, 중금속함량검사, 염수분무테스트 등을 분석하여 품질향상 및 인체 무해성을 확인하였다.

국내 단기추진제 탄약의 저장수명 예측에 관한 연구 : 105미리 고폭탄 추진체를 중심으로 (A Study on the Shelf-Life Prediction of the Domestic Single Base Propellants Ammunition : Based on 105mm High Explosive Propellants)

  • Domestic 105mm HE (High Explosive) shell is composed of three parts that are Fuze, Projectile and Propellants. Among three parts, propelling charge of propellants part consists of single base propellants. It has been known that the lifespan of single base propellants is affected by a storage period. These are because Nitrocellulose (NC) which is the main component of propelling gunpowder can be naturally decomposed to unstable substances similar with other nitric acid ester. Even though it cannot be prevented fundamentally from being disassembled, a decomposition product ($NO_2$, $NO_3$, and $HNO_3$) and tranquillizer DPA (Diphenylamine) having high reactivity are added into a propellant to restrain induction of automatic catalysis by a decomposition product. The decay rate of the tranquillizer is also affected by a production rate of the decomposition product of NC. Therefore, an accurate prediction of the Self-Life is required to ensure against risks such as explosion. Hereupon, this paper presents a new methodology to estimate the shelf-life of single base propellants using data of ASRP (Ammunition Stockpile Reliability Program) to domestic 105mm HE (propelling charge of propellants part). We selected four attributes that are inferred to have influence on distribution of the DPA amount in a propellant from the ASRP dataset through data mining processes. Then the selected attributes were used as independent variables in a regression analysis in order to estimate the shelf-life of single base propellants.
